Description
即ち、本発明は以下の態様を含むものである:
{1}被検動物におけるミトコンドリアtRNA(mt-tRNA)中に含まれる修飾ヌクレオシドの量を決定する方法であって、以下の工程:
(1)該被検動物由来の体液試料及び該被検動物由来の細胞の培養上清からなる群より選択される試料中の修飾ヌクレオシドの量を、タンデム質量分析法を用いて決定する工程、および
(2)工程(1)により決定された試料中の修飾ヌクレオシド量が、被検動物におけるmt-tRNAにおける該修飾ヌクレオシドの量に関連付けられる工程、
を含む方法。
{2}前記タンデム質量分析法が、前処理として液体クロマトグラフィー(LC)を含みイオン化源がエレクトロスプレーイオン化源(ESI)であるLC-ESI-MS/MSであり、かつ、使用される質量分析モードが選択反応モニタリングである、上記{1}に記載の方法。
{3}前記試料が、被検動物由来の尿を含む上記{1}又は{2}に記載の方法。

本明細書において「及び/又は」は、いずれか一方、あるいは、両方を包含する意味で使用される。
例えば、本発明の測定対照である修飾ヌクレオシドとしては、これに限定されないが、上記5-taurinomethyl-2-thiouridine(τm5s2U)、5-taurinomethyluridine(τm5U))、2-methylthio-N6-isopentenyl adenosine(ms2i6A)に加え、2-methylthio-N6-threonylcarbamoyl adenosine(ms2t6A)、N6-methyladenosine (m6A)などをあげることができる。
本明細書中、「mt-tRNA中に含まれるタウリン修飾ウリジン量」とは、mt-tRNAに含まれるタウリン修飾されたウラシル基の量を意味し、「mt-tRNAがタウリン修飾ウリジンを含む」とは、mt-tRNAがタウリン修飾されたウラシル基を有することを意味する。
本発明の方法1は、液体クロマトグラフィー(LC)に供してタウリン修飾ウリジンが濃縮された画分を得る工程を含みうる。
タウリン修飾ウリジンが濃縮された画分を得られる限り限定されるものではないが、本発明に好ましく用いられるLCとしては、RPLCが挙げられる。
本明細書中、逆相液体クロマトグラフィーとは、極性移動相及び非極性固定相を使用するクロマトグラフィーを意味する。
好ましい実施態様では、分析カラムは、直径が約2μmの粒子を含有する。
工程(a)において、逆相HPLCを用いる場合、分析カラムに用いる充填剤としては、例えば、シリカゲルやポリマーゲル基材に炭化水素鎖を結合した充填剤が挙げられる。工程(a)において、逆相HPLCを用いる場合、分析カラムに用いる好ましい充填剤としては、シリカゲル基材にオクタデシル基を結合した充填剤(ODS又はC18充填剤とも称する)が挙げられる。

が挙げられるが、これに限定されない。上記の条件下で四重極質量分析器LCMS(LCMS-8050;島津製作所)を用いてタウリン修飾ウリジンの検出を行った場合、τm5Uは2分±10%でピークが検出され、τm5s2Uは3分±10%でピークが検出され得る。
タウリン修飾ウリジンが濃縮された画分のイオン化の方法としては、エレクトロスプレーイオン化法(ESI)、大気圧化学イオン化法(APCI)、大気圧光イオン化法(APPI)、電子イオン化法(EI)、高速電子衝撃(FAB)/液体二次イオン化法(LSIMS)、マトリクス支援レーザー脱離イオン化法(MALDI)、フィールドイオン化法、フィールド脱離法、熱スプレー/プラズマスプレーイオン化法及びパーティクルビーム・イオン化法などが挙げられる。当業者であれば、測定する分析物、試料の種類、検出器の種類、正イオンモード又は負イオンモードの選択等に基づいて、イオン化法を選択することができる。タウリン修飾ウリジンが検出され得る限り特に限定されるものではないが、本発明の方法1においては、ESIを用いて行うことが好ましい。
本発明において好ましい解裂手段としては、衝突励起が挙げられる。本発明の好ましい一態様としては、前駆イオンの衝突誘起解離(CID:Collision Induced Dissociation)によって断片化された生成物イオン量を決定する工程を含む。本明細書中、衝突誘起解離とは、選択した前駆イオンと中性分子との衝突を起こすことにより、前駆イオンの一部の結合を破壊することを指す。
本発明の方法1において、タンデムMSに用いるアナライザーの例としては、トリプル四重極アナライザー、イオントラップアナライザー及び飛行時間型アナライザーなどが挙げられる。本発明の方法1においてタンデムMSに用いる好ましいアナライザーとしては、トリプル四重極アナライザー又は四重極飛行時間型(QTOF)アナライザーが挙げられる。本発明の方法1においては、SRMアッセイのために有利な市販の機器プラットフォームは、多くの場合、トリプル四重極アナライザーを用いているという観点から、トリプル四重極アナライザーを用いてタンデムMSを行うことがより好ましい。ここでいう「トリプル四重極」は、当業者が通常理解するように、四重極のみならず、四重極の代わりに多重極や積層電極を用いる場合も含む意味である。
本発明において、質量分析は、負イオンモードで行ってもよい。あるいは、質量分析は、正イオンモードで行ってもよい。本明細書で使用される場合、用語「正イオンモード」とは、正イオンが生成及び検出される質量分析法を指し、用語「負イオンモード」とは、負イオンが生成および検出される質量分析法を指す。
本発明の方法1の好ましい一実施態様において、タウリン修飾ウリジンは、負イオンモードで分析される。本発明の方法1のより好ましい一実施態様において、タウリン修飾ウリジンは、ESIによりイオン化され、負イオンモードで分析される。また、本発明の好ましい一態様において、ms2i6修飾アデノシンの検出は、正イオンモードで分析される。
上記工程(c)で決定されたイオンの量が、試料中のタウリン修飾ウリジンの量に関連づけられる。一実施態様において、試料中のタウリン修飾ウリジンの量の決定は、相対的定量であり得る。一実施態様において、試料中のタウリン修飾ウリジンの量の決定は、絶対的定量であり得る。
(c1)工程(b)によりイオン化されたイオンのうち、タウリン修飾ウリジン前駆イオンと同一のm/zを有するイオン(好ましくは、τm5s2Uの検出には396±0.5 m/zを有する負イオン、及び/又はτm5Uの検出には380±0.5 m/zを有する負イオン)を選択する工程と、
(c2)工程(c1)において選択されたイオンを、該タウリン修飾ウリジン前駆イオンがタウリン修飾ウリジン生成イオン(好ましくは124±0.5のm/zを有するタウリン修飾ウリジン生成負イオン)を発生する条件下で、衝突反応に付す工程と、

を含み、工程(c3)で決定されたイオンの量が、試料中のタウリン修飾ウリジンの量に関連づけられる。
一実施態様において、タウリン修飾ウリジンイオンの量は、内部標準物質との比較により、試料中のタウリン修飾ウリジンの量と関連付けることができる。
SRMを用いる別の一実施態様において、タウリン修飾ウリジンの相対的な定量は、例えば、タウリン修飾ウリジンのSRM特徴ピーク面積を、同一試料中の異なる物質(例えば、未修飾ウリジン)からのSRM特徴ピーク面積と比較することによって、行うことができる。
一実施態様において、内部標準は、1以上の重同位体で標識された、合成タウリン修飾ウリジンである。適切な同位体標識内部標準は、質量分析で分析された時に、それが天然のタウリン修飾ウリジン特徴ピークとは異なりかつ区別できるため対照ピークとして使用できる、予測可能で一貫性のあるSRM特徴ピークを生成するように合成され得る。従って、試料に既知量の内部標準を添加し、質量分析で分析した場合に、同一試料中の天然タウリン修飾ウリジンのSRM特徴ピーク面積を、内部標準のSRM特徴ピークと比較することができる。
本明細書において、「体液」とは、個体の身体から単離できる任意の液体を意味する。体液の例としては、血液、血漿、血清、胆汁、唾液、尿、涙、汗、脳脊髄液(CSF)などが挙げられる。好ましくは、体液は、尿又は血清であり、最も好ましくは尿である。
本明細書において、「細胞培養上清」とは、任意の細胞(好ましくは動物細胞、より好ましくは哺乳動物細胞)を、培地中で一定期間培養することにより得られる培養物の上清を意味する。培養上清の調整に用いることのできる細胞の例としては、培養細胞、動物個体の身体から単離された細胞、又は動物個体の身体から単離された細胞由来の多能性幹細胞もしくは該多能性幹細胞から分化した細胞などの、動物個体の身体から単離された細胞由来の細胞などが挙げられる。細胞の培養条件は、タウリン修飾ウリジンを検出し得る限り特に限定されるものではないが、例えば、105~107の細胞を、細胞培養用培地2 mLを有する培養皿に播種し、1日以上培養した培養物の培養上清などが例示される。

本発明は、さらに、被検動物におけるmt-tRNA中に含まれるタウリン修飾ウリジン量を決定する方法であって、該被検動物由来の細胞の培養上清及び該被検動物由来の体液試料からなる群より選択される試料中のタウリン修飾ウリジン量を決定することを含み、決定された試料中のタウリン修飾ウリジンの量が、被検動物におけるmt-tRNAのタウリン修飾ウリジンの量に関連付けられる、方法(本明細書中、本発明の方法2とも称する)を提供する。
本発明の方法2の一実施態様において、試料中のタウリン修飾ウリジンの量の決定は、
(b’)質量分析により検出し得るタウリン修飾ウリジンイオンを発生させるのに適した条件下で、タウリン修飾ウリジンが濃縮された画分をイオン化源に供する工程と、
(c’)質量分析によりタウリン修飾ウリジンイオンの量を決定する工程とを含み、工程(c’)で決定されたタウリン修飾ウリジンイオンの量が、前記試料中のタウリン修飾ウリジンの量に関連づけられ、すなわち被検動物におけるmt-tRNAのタウリン修飾ウリジンの量に関連付けられる。
タウリン修飾ウリジンの量が決定できる限り特に限定されるものではないが、本発明の方法2において質量分析に用いるアナライザーとしてはトリプル四重極アナライザー又はQTOFアナライザーが好ましく、SRMアッセイを用いる場合、SRMアッセイのために有利な市販の機器プラットフォームは、多くの場合、トリプル四重極アナライザーを用いているという観点から、アナライザーとしてはトリプル四重極アナライザーを用いて行うことが好ましい。
被検動物におけるmt-tRNA中に含まれるタウリン修飾ウリジン量の決定は、例えば試料中に提供される1以上の内部標準を用いて行うことができる。
一実施態様において、タウリン修飾ウリジンイオンの量は、内部標準物質との比較により、被検動物におけるmt-tRNA中に含まれるタウリン修飾ウリジン量と関連付けることができる。
あるいは、被検動物におけるmt-tRNA中に含まれるタウリン修飾ウリジン量の決定は、例えば試料中に提供される1以上の外部参照標準を用いて行うことができる。
内部標準又は外部標準を用いた定量は、あらかじめ作成した検量線を用いて行ってもよい。

が挙げられる。
本発明は、さらに本発明の方法2により決定された被検動物におけるmt-tRNA中に含まれるタウリン修飾ウリジン量と、ミトコンドリア病の発症可能性との間の負の相関に基づき、ミトコンドリア病の発症可能性を検査する方法(本明細書中、本発明の方法3とも称する)を提供する。
また、例えば、検出された値における診断効率(全症例数に対する、ミトコンドリア病のヒトを「ミトコンドリア病」と正しく判定した症例と、ミトコンドリア病を発症していないヒトを「ミトコンドリア病を発症していない」と正しく判定した症例との合計数の割合)を求め、最も高い診断効率が算出される値をカットオフ値とすることができる。
各用語の定義は、特に言及しない限り、上記1又は2に記載したものと同一である。
選択反応モニタリングを応用した質量分析によるτm5Uとτm5s2Uの検出法を検証するために、野生型の胚性幹(ES)細胞(WT cells)、及びタウリン修飾酵素であるMto1を欠損したES細胞(Mto1-KO cells)からtotal RNAを抽出し、島津製作所製四重極質量分析器LCMS(LCMS-8050)に注入し、選択反応モニタリング法でタウリン修飾を検討した。尚、Mto1-KO細胞は、相同組換え法によってMto1遺伝子の第3-4エクソンを欠失させることによって作製した。
106個の細胞を1 mLのTrizol液(Invitrogen社)で懸濁し、0.2 mLのクロロフホルムを加えて12,000 x gで15分遠心し、total RNAを水相に分離した。その後、0.5 mLのイソプロパノールを加えてtotal RNAを析出させ、12,000 x gで15分遠心し、total RNAを沈殿させた。沈殿を70%エタノールで一度洗い、total RNAを完全に乾燥させた後、適当量の純水でtotal RNAを再溶解した。
分離カラムは逆相カラムであるInertsil ODS-3(150 mm×2.1mm I.D., 2μm)を使用した。移動相A:30 mM Ammonium Acetate (pH 5.8)、移動相B:60% Acetonitrileを用い、グラジェント分析(移動相B: 0min 1%→10min 35%→15min 100%→20min 100%→25min 1%)を行った。流速0.4mL/min,カラムオーブン温度50℃に設定した。試料注入量2μLとした。測定は負イオンモードにて行った。
質量分析は以下の条件で行った。
イオン化法(ESI)

τm5Uに分析にあたっては、m/z値 380である前駆イオンを選択し、これを不活性ガスとの衝突により開裂反応を起こしてさらに断片化して、生成物イオンを示すm/z 124のピークを観察した。また、τm5s2Uに分析にあたっては、m/z値 396である前駆イオンを選択し、これを不活性ガスとの衝突により開裂反応を起こしてさらに断片化して、生成物イオンを示すm/z 124のピークを観察した。
その結果、正常の細胞由来のRNAにおいて、分析開始後3分と2分後にτm5s2Uとτm5Uに対応するピークを検出した。これらのピークがタウリン修飾酵素であるMto1を欠損した細胞に由来するRNAでは消失していたことから、これらのピークがτm5s2Uとτm5Uに対応するものであることが実証された(図4)。
次に、細胞培養上清を用いてタウリン修飾の検出を行った。ヒト由来の培養細胞であるHeLa細胞(1.5 x 105 個)を直径3.5 cmの培養皿に播種し、2mLのDMEM(Dulbecco’s Modified Eagle Medium)培地で一晩培養した。細胞培養上清100μLにメタノール500μLを加え、タウリン修飾されたヌクレオシドの抽出を行った。遠心エバポレーターで試料を15000 rpmで10分遠心し乾燥させた後、100μLの蒸留水を加え、再溶解した。2μLの試料を島津製作所製四重極質量分析器LCMS(LCMS-8050)に注入し、実施例1と同様の条件にて選択反応モニタリング法によりタウリン修飾を検討した。
実施例2の結果を踏まえ、生体試料を用いたタウリン修飾の検出を試みた。ヒト尿検体100μLを採取し、500μLのメタノールを加えて、除タンパクを行った。次に、15000 rpmで10分遠心し、上清を遠心濃縮装置で乾燥させた。最後に沈殿を100μLの蒸留水で溶解し、2μLを質量分析装置(島津製作所LCMS-8050)で分析した。その結果、多量のτm5Uと
ミトコンドリア病の一つである赤色ぼろ線維・ミオクローヌスてんかん症候群(MERRF)の患者2名から尿を採取し、尿中のタウリン修飾(τm5s2U)を質量分析法で分析した。対象として健常人2名から尿を採取し、同様にタウリン修飾(τm5s2U)を質量分析法で分析した。結果を図7に示す。
健常人と比べてMERRF患者ではτm5s2Uが顕著に低下した。一方、未修飾ヌクレオシドであるグアノシン(G)はMERRF患者と健常人では差が見られなかった。
実施例2と同様に試料を調整し、正イオンモードで質量分析を行った。τm5s2Uの検出に際しては、398 m/zを有する前駆正イオンを選択し、126 m/zのプロダクト正イオンを検出した。τm5s2Uの検出に際しては、380 m/zを有する前駆正イオンを選択し、126 m/zのプロダクト正イオンを検出した。
正イオンモードにおいてもタウリン修飾は検出されたが、正イオンモードでの検出を行った場合、負イオンモードで行った場合と比較して、その検出感度は低かった。
他のミトコンドリア病である慢性進行性外眼筋麻痺症候群(CEPO)の患者を対象として、修飾ヌクレオシドである、ms2i6A、ならにびτm5s2U及びτm5Uの量を測定した。
CEPOは外眼筋麻痺や眼臉下垂、四肢筋力低下を主な症状とするミトコンドリア病の一種である。CEPOの70%にミトコンドリアDNAの欠失もしくは重複を認める。欠失するミトコンドリアDNAの領域は患者によって様々である。CEPOの遺伝子診断には、mtDNAの欠失や重複を検出するサザンブロット法とLong PCR法を併用する。一方、CEPOの場合は、患部である眼筋でしかミトコンドリアDNAの欠失が見つからないことがあるため、血液のみならず、眼筋の生検も必要であり、患者の負担が大きいという問題がある。
100μLのヒト尿検体に500μLのメタノールを加えて、その後、15,000 rpmで10分遠心し、上清を回収することにより除タンパクを行った。回収した上清を遠心エバポレーターで乾燥させ、最後に沈殿を100μLの蒸留水で溶解し、2μLを質量分析装置(島津製作所LCMS-8050)で分析した。 ms2i6Aは、選択反応モニタリング法によって検出した。検出パラメーターは次の通りである。ms2i6A:前駆正イオン m/z 382、生成物イオン m/z 182; G:前駆正イオン m/z 284、生成物イオン m/z 152。τm5s2U及びτm5Uの検出パラメーターは、実施例1と同様である。結果を図8に示す。
健常人と比べてCIPOF患者ではms2i6Aが顕著に低下した。一方、同じミトコンドリアtRNA由来のτm5s2U及びτm5U、また未修飾ヌクレオシドであるグアノシン(G)はCIPO患者と健常人では差が見られなかった。
Claims (23)
- 被検動物におけるミトコンドリアtRNA(mt-tRNA)中に含まれる修飾ヌクレオシドの量を決定する方法であって、以下の工程:
(1)該被検動物由来の体液試料及び該被検動物由来の細胞の培養上清からなる群より選択される試料中の修飾ヌクレオシドの量を、タンデム質量分析法を用いて決定する工程、および
(2)工程(1)により決定された試料中の修飾ヌクレオシド量が、被検動物におけるmt-tRNAにおける該修飾ヌクレオシドの量に関連付けられる工程、
を含む方法。 - 前記タンデム質量分析法が、前処理として液体クロマトグラフィー(LC)を含みイオン化源がエレクトロスプレーイオン化源(ESI)であるLC-ESI-MS/MSであり、かつ、使用される質量分析モードが選択反応モニタリングである、請求項1に記載の方法。
- 前記試料が、被検動物由来の尿を含む請求項1又は2に記載の方法。

工程(c)におけるタンデム質量分析が、382±0.5の質量対電荷比(m/z)を有する前駆正イオンを、ms2i6A前駆正イオンが182±0.5のm/zを有するms2i6A生成正イオンを発生する条件下で、衝突反応に付し、該衝突反応により生じた182±0.5のm/zを有する生成イオン量を決定することを含み、
工程(c)で決定されたイオンの量が、前記試料中のms2i6Aの量に関連づけられる、方法。 - 前記イオン化源がエレクトロスプレーイオン化源である、請求項17~19のいずれか一つに記載の方法。
- 試料が体液試料又は細胞培養上清を含む、請求項17~20のいずれか一項に記載の方法。
- 試料が尿を含む、請求項17~20のいずれか一項に記載の方法。
- 試料が、除タンパク処理された体液試料又は細胞培養上清である、請求項17~22のいずれか一項に記載の方法。
