본 발명은 금 나노선을 이용하여 생물학적 물질을 수정란의 핵내로 전달하는 방법에 관한 것이다.
재조합 외래 유전자를 동물의 염색체상에 인공적으로 삽입하여 그 동물의 일부 형질을 변화시킨 동물, 즉 형질전환동물(transgenic animals)은 삽입된 외래 유전자의 기능 및 특성을 연구하고, 의학적, 생물공학적 가치가 높은 단백질의 생산에 있어서 유용한 도구이다.
또한, 초기 배아에 외래 유전자를 도입하는 기술은 발생학의 연구 및 형질전환동물의 생산에 있어서도 매우 강력한 도구가 될 수 있다. 현재, 형질전환동물을 생산하는데 있어서 가장 널리 사용되는 접근방법으로는 클로닝한 유전자를 수정란 전핵(pronucleus)안으로 마이크로 주입하는 방법(microinjection), 재조합 배아줄기세포를 배아 안으로 주입하는 방법, 핵 이식법, 및 레트로바이러스를 이용하는 방법 등이 있다.
그러나, 이러한 접근방법들은 여러가지 이유로 어려움을 겪고 있는데, 예를 들면, 마이크로주입법(microinjection)의 경우 형질전환 발생율이 매우 낮고 형질전환동물의 산자에서 모자이크현상이 높은 빈도로 발생한다고 알려져 있다. 또한, 핵이식법의 경우 형질전환 공여 세포의 핵을 수용자 세포질체 안으로 주입하는 단계를 포함하는데 매우 비효율적이며 고비용의 방법이고, 배아가 형질전환 줄기세포 또는 생식세포와 혼합되어 있는 키메릭 배아를 통해 생산된 형질전환동물은 키메릭 동물이 만들어진다는 문제가 있다. 또한, 이들 접근방법의 대안으로서 레트로바이러스 매개 형질전환동물 제조방법이 알려져 있으나(한국특허공개 제10-2014-0091119호), 레트로바이러스는 메틸화 등의 후생유전학적 변형이 일어날 뿐만 아니라, 배아발생동안 혹은 출생직후에 정상 유전자의 발현을 비활성화시킬 수 있다는 단점이 있다.
한편, 외래 유전자와 같은 생물학적 물질을 특정한 세포에 정확하게 전달 및 제어하는 것은, 다양한 세포생물학적 현상을 이해하는데 중요한 tool로서, 형질전환동물 제조에 있어서도 핵심 기술에 해당한다. 그러나, 종래 많이 이용하고 있는 마이크로주입법(microinjection)의 경우, 전술한 바와 같이 외래 유전자가 genomic DNA에 융합되지 못하여 형질전환 발생율이 매우 낮고 형질전환동물의 산자에서 모자이크현상이 발생한다는 문제가 있었다.
또한, 종래 일반적인 세포에 외래 유전자를 도입하기 위하여 통상적으로 사용하는 트랜스펙션 시약(예: Lipofectamine)을 수정란에 그대로 적용하는 것은 곤란한데, 트랜스펙션 시약의 독성으로 인해 수정란이 정상적으로 발달이 되지 않기 때문이다.
그러나, 이러한 문제에도 불구하고, 종래에 금나노선을 이용하여 단일세포(신경아세포종 세포주 SK-N-CH)에 DNA를 전달하는 기술이 보고된 바는 있으나, 현재까지 수정란의 핵에 유전자와 같은 생물학적 물질을 정확하게 전달함으로써 모자이크 현상 없이 모든 할구에서 외래 유전자의 발현이 가능하도록 한 예는 보고된 바 없다.
본 발명은, 형질전환동물의 제조를 위해 생물학적 물질을 수정란의 핵내로 전달하는 방법을 제공하는 것을 목적으로 하며, 특히 수정란 세포를 손상시키지 않으면서 전달 물질의 양이나 전달시간 등 정밀한 제어에 의해 전달효율을 극대화시키면서, 모자이크 현상 없이 모든 할구에서 외래 유전자의 발현이 가능하도록 하는 방법을 제공하는데 그 목적이 있다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기와 같은 과제를 해결하기 위하여, 본 발명은, (a) 생물학적 물질을 금 나노선을 포함하는 나노 인젝터(nanoinjector)에 접촉시켜 금 나노선에 생물학적 물질이 부착되도록 하는 단계; (b) 상기 생물학적 물질이 부착된 금 나노선 부분을 in vitro에서 수정란 핵내에 주입하는 단계; 및 (c) 상기 수정란 핵내에 주입된 금 나노선으로부터 상기 생물학적 물질을 분리시켜 전달하는 단계를 포함하는, 수정란 핵내로의 생물학적 물질 전달방법을 제공한다.
본 발명의 일 구현예로, 상기 금 나노선은 직경이 150 내지 850 nm이고, 그 길이가 1 내지 100 μm인 것을 특징으로 한다.
본 발명의 다른 구현예로, 상기 금 나노선은 원기둥 형태인 것을 특징으로 한다.
본 발명의 또 다른 구현예로, 상기 (c) 단계의 분리는, 기준 전극으로 포화 카로멜 전극 사용시 -0.5 내지 -1.5 V의 범위로 30초 내지 5분의 전기펄스를 적용함으로써 행하는 것을 특징으로 한다.
본 발명의 또 다른 구현예로, 상기 전기펄스는 기준 전극으로 포화 카로멜 전극 사용시 -0.8V로 2분 적용하는 것을 특징으로 한다.
본 발명의 또 다른 구현예로, 상기 (c) 단계의 분리는, 물리적 확산에 의한 것임을 특징으로 한다.

본 발명의 또 다른 구현예로, 상기 (a) 단계의 물질 부착은 금나노선과 생물학적 물질 사이에 링커(linker) 역할을 할 수 있는 화학분자, 페이스트(paste), 또는 담체(carrier) 물질을 도입하여 행하는 것임을 특징으로 한다.
본 발명의 또 다른 구현예로, 상기 (a) 단계의 물질 부착은 금나노선과 생물학적 물질 사이에 직접적인 결합에 의한 것임을 특징으로 한다.
본 발명의 또 다른 구현예로, 상기 수정란은 투명대가 존재하거나 존재하지 않는 것을 특징으로 한다.
본 발명에 의하면, 수정란 세포를 손상시키지 않으면서 생물학적 물질의 양이나 전달시간 등 전달조건의 정밀한 제어에 의해 전달효율을 극대화시킬 뿐만 아니라, 모자이크 현상 없이 모든 할구에서 외래 유전자의 발현이 가능하기 때문에, 형질전환동물의 제작에 유용하게 이용될 수 있다.

본 발명은, 생물학적 물질을 금 나노선을 포함하는 나노 인젝터(nanoinjector)에 접촉시켜 금나노선에 생물학적 물질이 부착되도록 하는 단계; 상기 생물학적 물질이 부착된 금 나노선 부분을 in vitro에서 수정란 핵내에 주입하는 단계; 및 상기 수정란 핵내에 주입된 금 나노선으로부터 상기 생물학적 물질을 분리시켜 전달하는 단계를 포함하는, 수정란 핵내로의 생물학적 물질 전달방법을 제공한다.
본 발명의 금나노선 인젝터 장치는, 금 나노선을 포함하는 나노 인젝터를 포함하는 작동 전극, 상기 작동전극에 대응하는 상대(counter)전극, 전기펄스를 제공하기 위한 기준(reference) 전극, 상기 작동전극내의 금 나노선과 생물학적 물질이 주입되는 수정란을 관찰하는 광학 현미경, 수정란을 고정시키기 위한 장치 및 상기 나노인젝터내 금 나노선에 전기신호를 제공하는 전기신호 공급부를 포함하여 이루어질 수 있다.
본 발명에서 상기 나노 인젝터는 일단부에 상기 금 나노선을 포함하는 금속팁이 부착되며, 내부에는 상기 금속팁으로 외부의 전기신호가 전달되도록 배선이 연결되고, 외부는 절연되어 있는 본체를 추가로 포함할 수 있다. 이때, 상기 본체의 길이는 0.5 내지 20 cm의 범위일 수 있고, 상기 금속팁의 길이는 1 mm 내지 20 mm의 범위일 수 있다.
또한, 상기 나노 인젝터는 기판을 포함하며, 상기 금나노선을 포함하는 금속팁이 기판상에 고정되고, 상기 기판상 또는 기판내에 형성된 배선이 상기 금속팁과 연결되고, 상기 금속팁과 연결되는 반대쪽의 배선을 외부와 연결함으로써 상기 나노 인젝터가 구성될 수 있다.
또한, 상기 나노선 인젝터는 전기적 자극을 이용하지 않고 물리적 확산을 이용하여 생물학적 물질을 전달하는 경우, 위와 같은 전기 신호 공급부 및 배선에 연결되지 않고 독립적으로 혹은 지지대 역할을 하는 물체에 부착되어 조작될 수 있다.
본 발명에서 금나노선의 길이와 직경은 제조공정에 따라 달라질 수 있으나, 바람직하게는 직경이 150 내지 850 nm이고, 그 길이는 1 내지 100 μm일 수 있다. 또한, 금나노선은 원기둥(실린더) 형태인 것이 바람직하다.
본 발명에서의 상기 금나노선은 단결정 형태로 대단히 강하면서도 동시에 유연하기 때문에, 수정란 관통이 쉬우며 손상을 줄일 수 있는 장점이 있다. 즉, 본 발명의 나노 인젝터내 금나노선은 종래의 microinjector와는 달리 세포에 눈에 띄는 손상을 일으키지 않고 삽입될 수 있으며, 금 나노선의 높은 전기 전도성에 의해 전기적 신호에 따라 신속하고 정량적인 물질의 전달이 가능할 뿐만 아니라, 확산(diffusion)과 같은 물리적 방법에 의해서도 생물학적 물질을 전달할 수 있다.
또한, 본 발명에서 나노 인젝터에 부착되거나 분리시킬 수 있는 생물학적 물질로는, 생체내 또는 생체밖에서 생리활성을 가지는 물질이면 그 종류에 무관하게 사용가능하나, 바람직하게는 DNA, RNA, 단백질일 수 있다. 이때, DNA 및 RNA의 경우, 이는 DNA 백신 및 유전자 치료를 위한 연구에 응용될 수 있고, 또한 유전자 기능을 조사하고 기초적인 생물학적 메커니즘을 이해하기 위한 연구에 있어 유용하게 응용될 수 있다.
본 발명의 방법에 있어서, 금 나노선에 생물학적 물질이 부착되도록 하는 단계는 상기 생물학적 물질내 포함된 황원자가 금 나노선의 금 표면내 금원자와 결합함으로써 단결정 금나노선에 생물학적 물질이 부착되거나, 생물학적 물질이 전하를 띄는 경우 금 나노선 표면에 생물학적 물질과 반대의 전하를 띄는 물질을 붙인 뒤, 정전기적 인력으로 생물학적 물질이 금 나노선에 부착될 수 있다.
또한, 상기 생물학적 물질로서 선형의 DNA와 원형의 플라스미드를 모두 단결정 금나노선에 부착하여 수정란으로 전달할 수 있다. 상기 선형의 DNA와 원형의 플라스미드는 생물학 및 의학 연구에서 전형적으로 이러한 두 형태로 DNA가 세포 안에 전달되기 때문에 중요한 의미를 갖는다.
본 발명에서는 서로 다른 방법을 이용하여 이 두 종류의 DNA를 금 나노선에 부착할 수 있다. 선형 DNA의 경우, 선형 DNA의 5' 끝을 thiol로 수정한 이후에, Au-S 결합을 이용하여, 생물학적 물질(DNA)내 포함된 황원자가 금 나노선의 금 표면내 금원자와 직접적으로 결합함으로써 금 나노선에 부착할 수 있다. 원형 플라스미드의 경우, thiolated 플라스미드는 만들기가 어렵고 비효율적이므로 대신에 정전기적 부착을 이용할 수 있는데, 금과도 결합할 수 있고 말단은 양전하를 띄는 분자(예; cysteamine)를 이용하여 금나노선 표면을 개질(modification)하고 이를 정전기적 인력에 의해 음전하를 띄는 플라스미드가 이 금 나노선에 부착할 수 있다.
또한, 본 발명에서 금나노선에 생물학적 물질을 부착하는 방법으로는, 상기 직접적인 결합 이외에도, 링커(linker), 페이스트(paste), 또는 담체(carrier)를 통한 간접적인 부착도 가능하다.
본 발명에서 수정란의 핵내에 주입된 금 나노선으로부터 생물학적 물질을 분리하는 방법은 특별한 제한은 없으나, 바람직하게는 전기펄스를 이용한 전기적 방법이나, 확산을 이용한 물리적인 방법을 적용할 수 있다.
본 발명에서 상기 전기펄스를 이용하는 경우에 전압범위는 기준 전극으로 포화 카로멜 전극 사용시 -0.5 내지 -1.5 V의 범위일 수 있고, 펄스를 가하는 시간은 30초 내지 5분 적용할 수 있다. 다만, 상기 전기 펄스의 최적조건은 사용되는 생물학적 물질의 종류와 개별적 분리조건에 따라 달라질 수 있으나, DNA의 경우에는 금 나노선 전극을 통해 2분 동안 기준 전극으로 포화 카로멜 전극 사용시 -0.8 V의 전기 펄스를 가하는 것이 바람직하다.
본 발명에서 이용되는 수정란에는 제한이 없으며, 예를 들면 투명대가 존재하거나 존재하지 않는 것일 수 있으며, 또한 투명대를 얇아지게 처리한 수정란일 수 있다.
생물종 마다 수정란의 크기, 수정란 핵의 크기, 수정란 투명대의 두께 등이 다르기 때문에, 수정란에 최고의 침투 효율을 가지면서 동시에 수정란 손상을 최소화할 수 있는 금나노선 인젝터의 디멘젼(길이 및 직경)의 최적화가 필요하다.
종래, 수정란 내에 생물학적 물질 전달을 위한 물리적 접근(physical access) 방법 중 하나인 마이크로주입법에서는, tapered 형태의 유리 마이크로 피펫을 이용하는데, 이 경우 직경이 sub-μm 또는 수-μm의 가느다란 tip 부분 뿐만 아니라 수정란 내에 삽입될수록 직경이 굵어짐에 따라 수정란의 투명대 및 세포막에 홀(hole)이 커지게 되어 막통합성(membrane integrity)을 잃게 될 수 있다. 그 결과, 수정란에 큰 손상이 가해지게 되고 심한 경우 수정란의 용해(lysis)가 초래될 수도 있다.
이에 반하여, 본 발명의 금 나노선 인젝터는 수정란에 삽입되는 부분이 처음부터 끝까지 완벽한 원기둥 형태를 이루고 있어 기존 마이크로인젝터와는 다른 형태일 뿐만 아니라, 금 나노선 인젝터는 마이크로 유리피펫에 비해 매우 작은 직경을 갖기 때문에 수정란의 투명대, 세포막, 핵막 등에 훨씬 작은 홀(hole)을 만들면서 삽입된다. 금 나노선 인젝터가 제거된 이후 이 작은 홀은 쉽고 빠르게 회복되어 세포에 큰 손상을 주지 않게 된다. 또한, 이와 같이 가느다란 직경은 수정란 접근에 있어 또 다른 우수성을 갖는데, 그것은 전핵 내의 다른 소기관(예: 인)과 접촉하지 않는다는 점이다. 즉, 수정란 내의 소기관(예: 인)과 접촉하게 되는 경우 수정란에 손상(chromosomal damage 등)이 가해지게 되어 수정란의 생존률이 현저히 낮아지게 되고, 인젝션 후 수정란이 생존하더라도 이후의 발생과정이 정상적으로 이뤄지지 않게 된다. 따라서 가느다란 직경으로 얻게 되는 높은 공간 분해능으로 전핵 내의 소기관을 피하면서 정확하게 핵내에 물질이 삽입될 수 있다. 이로써 핵 내로 유전 물질의 정확하고 안전한 전달을 통해, 수정란의 생존률이 높아지며 이후 정상 발생을 통해 형질 전환된 개체를 얻을 수 있는 확률 또한 증가되게 된다.
세포 내로의 외부 물질 전달을 위해 최대 효율을 내면서 세포 손상을 최소로 하는 인젝터의 디멘젼은 세포(종류)마다 다르다. 게다가 수정란의 경우, 세포막에 더하여 투명대라는 외층(outer layer)이 존재하므로 인젝터가 이를 침투할 수 있는 것이 중요하다.
이에, 본 발명자들은, 수정란의 손상을 최소로 하면서 투명대를 통과할 수 있는 나노선 인젝터의 디멘젼(길이와 직경)을 구현하였다. 수정란의 전핵 내로 직접 물질 전달을 위해, 1-100μm 길이일 때, 바람직하게는 20-50μm 길이일 때 나노선 인젝터가 전핵 내에 접근 가능함을 확인하였는데, 이는 평균 100μm 직경을 갖는 수정란 내에 랜덤하게 위치한 전핵에 효과적으로 접근할 수 있기 때문이다. 또한, 상기 길이 범위에서 150-850 nm 직경을 가질 때, 투명대 및 세포막을 100% 효율로 통과할 수 있음을 확인하였다. 만약, 20-50 μm 길이 범위 안에 있으면서 금 나노선 인젝터의 직경이 너무 굵은 경우 (>850 nm) 투명대는 침투할 수 있지만, 세포막을 통과하지 못하고 세포막에서 L-shape으로 휘는 것을 확인하였다. 또한, 수정란 인젝션에 적절한 길이 범위를 가지면서 직경이 너무 가는 경우(<150 nm), 투명대조.차 침투하지 못하고 L-shape으로 금 나노선 인젝터가 휘어짐을 확인하였다.
종래, 나노디멘젼을 갖는 인젝터가 수정란 내에 삽입된 것을 보고한 연구에서는 핵이 아닌 세포질에 삽입되었으며, tapered 형태를 갖고 있었기 때문에, 본 발명은, 수정란의 핵에 원기둥 형태를 갖는 나노 수준의 인젝터가 도입된 최초의 사례이다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1: 수정란 준비
4주 이상의 암컷 마우스에 PMSG(Sigma G-4877)와 hCG(sigma CG-5) 호르몬 주사를 48시간 간격으로 복강주사하여 과배란(super ovulation)을 유도한 후, 수컷 마우스와 교배시키고 다음날 아침 질전(vaginal plug)을 확인하였다.
그 다음, 암컷 마우스를 희생하여 난관(oviduct)을 추출하고 팽대부를 터트려 zygote를 회수한 후 난구세포(cumulus cell) 제거를 위해 히알루로니다아제(hyaluronidase)(sigma H3884)를 처리한 후 M2 배지(sigma M7167)로 세척하였다. 최종 얻어진 수정란은, M16 배지(sigma M7292)에서 5% CO2, 37℃ 조건의 incubator에서 배양하였다(도 1 참조).
실시예 2: 수정란 핵내로의 유전자 주입
본 발명의 금나노선 나노인젝터와 종래의 마이크로인젝터에 의한 DNA 전달 효과를 비교확인하기 위하여, 녹색 형광단백질(GFP)을 인코딩하는 유전자를 마우스 수정란의 전핵에 주입하여 발생을 유도한 후 공초점 현미경으로 녹색형광신호를 관찰하였다. 이때, 발생 유도는, 유전자가 도입된 수정란을 M16 배지(sigma M7292)와 미네랄 오일을 도입하여 배양기 안에서 배반포기(blastocyst)까지 유도하였다(도 2 참조).
구체적인 실험방법은 각각 다음과 같다.
2-1. 마이크로인젝터
마이크로주입법은 종래의 알려진 방법으로 microinjector를 이용하여 마우스 수정란에 용액 상태의 GFP 유전자를 주입하였다(도 3 참조).
그 결과, 도 4에 나타낸 바와 같이, DNA를 전달한 수정란 10개 중 1개의 수정란에서만 녹색형광이 관찰되었으며, 더욱이 이 1개의 수정란도 단지 하나의 할구에서만 형광이 나타났다. 즉, microinjector를 이용하여 DNA를 전달할 경우에는, 전달효율이 낮을 뿐만 아니라, 수정란의 모든 할구에 동일하게 DNA가 전달되지 않고 한 할구에 치우쳐 전달됨을 확인할 수 있었다.
2-2. 금나노선 인젝터
본 발명의 금나노선 인젝터를 이용하여, 마우스 수정란에 GFP 유전자가 암호화된 벡터(phMGFP, progma)를 주입하는 과정을 도 5에 나타내었다.
구체적으로, Au-S 결합을 이루는 linker 분자를 금나노선(150-850 nm 직경)에 부착 후, 정전기적 인력으로 금나노선 표면에 DNA(10-100 nM)를 부착시켰다(도 5a). 그 다음, M2 배지내에서 금나노선 인젝터를 수정란의 핵내로 침투하기에 적절하게 위치시킨 뒤 수정란의 투명대, 세포막, 핵막을 통과시키고 전기적 펄스를 가하여 수정란 핵내로 직접 DNA 분자만을 분리 방출시켰다. 이때, 금나노선 인젝터의 높은 공간분해능으로 인해 핵내 소기관(예; 인)과 금 나노선 인젝터가 접촉하지 않으면서 핵내로 삽입될 수 있었다(도 5b). M2 배지 안에는, 상대전극, 기준전극 및 수정란이 담겨져 있고, 금 나노선 인젝터을 포함하는 금속 지지대가 배지 안에 담겨져 있다(도 5c).
GFP 유전자 주입후 배발생을 유도한 결과, 도 6에 나타낸 바와 같이, 금 나노선 인젝터로 도입한 수정란의 경우, 실험에서 시도했던 모든 조건의 수정란에서 녹색형광이 관찰되었다. 더 나아가, 실시예 2-1의 microinjector와 달리, 금나노선 인젝터 인젝션 후 배반포기 수정란의 모든 할구에서 녹색 형광을 관찰할 수 있었다. 이는 종래의 마이크로인젝터에 의한 DNA 전달보다 전달효율이나 안정성이 훨씬 우수함을 의미하는 것이다(도 7 및 표 1 참조).

실시예 3: 금나노선의 최적의 디멘젼 분석
본 발명의 금나노선 인젝터에 대하여, 수정란의 투명대, 세포막, 핵막 등을 통과하기에 적합한 형태와 dimension(길이 및 직경) 조건을 조사하였다.
도 8은 본 발명의 금나노선이 원기둥(실린더) 형태를 가짐을 보여주는 전자현미경 사진(a, b) 및 광학 사진(c)이다.
도 9a는 본 발명의 금나노선이 직경 150-850 nm이고 길이 1-100 ㎛일 때 수정란의 투명대, 세포막, 핵막 등을 통과하기에 적합함을 보여주는 수정란 핵내 삽입 이미지이며, 도 9b는 본 발명의 금나노선이 150-850 nm의 가는 직경범위로 인하여 높은 공간 분해능을 가지면서 핵내 소기관(예; 인)과의 접촉을 효율적으로 피함을 보여주는 수정란 핵내 삽입 이미지이다.
이에 반하여, 도 9c는 금나노선이 핵내 침투에는 적절한 길이를 가졌지만, 직경이 850 nm 이상으로 두꺼울 경우, 투명대는 통과가능하나 세포막은 통과할 수 없게 되어 금나노선이 L-shape으로 휘어지고 세포질의 변형이 일어남을 알 수 있다. 또한, 도 9d는 금나노선이 핵내 침투에는 적절한 길이를 가졌지만, 직경이 150 nm 이하로 가는 경우, 투명대조차 통과하지 못하여 L-shape으로 휘어짐을 알 수 있다.
도 10은, 본 발명의 금나노선 인젝터를 수정란 핵내로 삽입하였을 때, 수정란의 생활성에 미치는 영향을 확인한 사진이다. 즉, 직경 440nm, 길이 32μm의 동일한 금나노선 인젝터를 여러 개의 수정란에 30초 동안 삽입 후, 3-4일 뒤 발생 여부를 확인해 본 결과, 수정란의 생활성에 손상이 없음을 확인하였다.
도 11은, 본 발명의 금나노선을 다양한 디멘젼 조건(직경 및 길이)과, 다양한 인젝션 조건(전기적 펄스 유/무 및 수정란내 삽입시간)으로 수정란 핵내 삽입한 뒤, 3일간의 발생을 거친 morula 단계의 수정란 이미지다(배율 × 200). 직경 150-850 nm이고 길이 1-100 ㎛ 범위내의 디멘젼 조건에서는, 수정란에 치명적인 손상을 주지 않음을 확인하였다.
실시예 4: 최적 전달조건 확인
본 발명의 금나노선 인젝터의 최적의 전달조건을 더욱 조사하기 위하여, 전기적 자극을 각각 -0.8 V에서 1분간, -0.8 V에서 2분간 가한 경우, 그리고 전기적 자극 없이 확산방법으로 3분간 주입한 경우를 비교하였다.
그 결과, 도 12에 나타낸 바와 같이, -0.8 V에서 2분간 전기적 자극을 가하여 DNA를 전달한 수정란의 경우 모든 할구에서 녹색형광이 관찰되었다. 즉, Z-stack(focal plane 마다 형광 이미지를 얻은 후 각각의 이미지를 통합하여 3D로 형광을 확인) 기능을 이용하여 3차원적으로 수정란의 GFP 발현을 확인해본 결과, 모든 할구에 DNA가 전달되었음을 확연히 확인할 수 있었다.
결론적으로, 본 발명의 금나노선 인젝터를 이용한 DNA 전달 효율은 마이크로인젝션에 비하여 굉장히 높을 뿐만 아니라, 일부 할구에 치우지지 않고 모든 할구에 동일하게 전달됨을 확인할 수 있었다.
이러한 우수한 효과는, 금나노선 인젝터를 이용할 경우, 마이크로인젝터에 비하여 수정란에 거의 손상을 일으키지 않고 삽입될 수 있으며, 금 나노선의 높은 전기 전도성에 의해 전기적 신호에 따라 신속하고 정량적인 물질의 전달이 가능하기 때문이다.
또한, 기존 마이크로인젝터와 달리 금 나노선 인젝터는 용액 상태로 물질이 전달되지 않고, 물질 자체만을 전달할 수 있기 때문에 host 게놈상에 빠르게 DNA integration이 일어난 것으로 볼 수 있다. 즉, 기존 마이크로인젝터는 세포에 유전물질을 선택적으로 전달하지만 buffer와 함께 전달되므로 host 게놈상에 융합되는 확률이 낮아 일시적 발현 후 사라지나, 본 발명의 금나노선 인젝터에 의하면 유전물질 전달 후 효과적으로 host 게놈상에 융합되어 영속적으로 발현이 유지되므로 분할 및 발생 과정동안에도 잘 보존되는 것을 확인 할 수 있다.
또한, 내포작용(endocytosis)을 통해 핵으로 전달될 때에는 DNA 전달이 수동적으로 일어나며, 세포내 효소에 의해 DNA 분해가 일어날 수 있고, 핵막 통과의 실패가 빈번하게 일어난다. 이로 인해 주입된 DNA가 매우 낮은 비율(~ 0.3%)로 핵에 도달하나, 본 발명에 의한 금나노선 인젝터를 이용하여 정량적으로 조절된 DNA를 핵내에 직접 전달하게 되면 세포질에 남게 되는 외래 DNA 의 손실을 최소화 할 수 있다. 
상기에서는 본 발명의 바람직한 실시예를 참조하여 설명하였지만, 해당 기술 분야에서 통상의 지식을 가진 자라면 하기의 특허 청구의 범위에 기재된 본 발명의 사상 및 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.
본 발명에 따른 수정란의 핵내로 생물학적 물질을 전달하는 방법은 모자이크 현상의 발생 빈도를 현저히 줄여 모든 할구에서 외래 유전자의 발현이 가능하며, 정밀한 제어에 의해 생물학적 물질의 전달효율을 극대화 시킨 새로운 방법이므로, 상기와 같은 방법에 의해 제조된 형질전환동물은 유전자의 기능 및 특성 연구, 의학적, 생물공학적 가치가 높은 단백질 생산 등에 유용하게 이용될 것으로 기대된다.

Claims (10)

  1. 하기의 단계를 포함하는, in vitro에서 수정란 핵내로의 생물학적 물질 전달방법:
    (a) 생물학적 물질을 금 나노선을 포함하는 나노 인젝터(nanoinjector)에 접촉시켜 금 나노선에 생물학적 물질이 부착되도록 하는 단계;
    (b) 상기 생물학적 물질이 부착된 금 나노선 부분을 in vitro에서 수정란 핵내에 주입하는 단계; 및
    (c) 상기 수정란 핵내에 주입된 금 나노선으로부터 상기 생물학적 물질을 분리시켜 전달하는 단계.
  2. 제 1 항에 있어서, 상기 금 나노선은 직경이 150 내지 850 nm이고, 그 길이가 1 내지 100 μm인 것을 특징으로 하는, 전달방법.
  3. 제 1 항에 있어서, 상기 금 나노선은 원기둥 형태인 것을 특징으로 하는, 전달방법.
  4. 제 1 항에 있어서, 상기 (c) 단계의 분리는, 기준 전극으로 포화 카로멜 전극 사용시 -0.5 내지 -1.5 V의 범위로 30초 내지 5분의 전기펄스를 적용함으로써 행하는 것을 특징으로 하는, 전달방법.
  5. 제 4 항에 있어서, 상기 전기펄스는 기준 전극으로 포화 카로멜 전극 사용시 -0.8V로 2분 적용하는 것을 특징으로 하는, 전달방법.
  6. 제 1 항에 있어서, 상기 (c) 단계의 분리는, 물리적 확산에 의한 것임을 특징으로 하는, 전달방법.
  7. 제 1 항에 있어서, 상기 생물학적 물질은 DNA, RNA 또는 단백질인 것을 특징으로 하는, 전달방법.
  8. 제 1 항에 있어서, 상기 (a) 단계의 물질 부착은 금나노선과 생물학적 물질 사이에 링커(linker), 페이스트(paste), 또는 담체(carrier)를 통하여 행하는 것임을 특징으로 하는, 전달방법.
  9. 제 1 항에 있어서, 상기 (a) 단계의 물질 부착은 금나노선과 생물학적 물질 사이에 직접적인 결합에 의한 것임을 특징으로 하는, 전달방법.
  10. 제 1 항에 있어서, 상기 수정란은 투명대가 존재하거나 존재하지 않는 것을 특징으로 하는, 전달방법.
