본 발명의 실시예들에 따른 그래핀 바이오 전자 장치는 내시경 팁 부분에 배치되어 생체 내부를 진단하고 치료하는 기능을 수행할 수 있다. 그래핀 바이오 전자 장치는 투명도가 높아 카메라 위에 배치될 수 있다. 이에 의해, 내시경 팁의 표면적을 증가시키지 않고도 그래핀 바이오 전자 장치가 내시경에 배치될 수 있고, 내시경 시스템이 고집적화 될 수 있다. 그래핀 바이오 전자 장치는 지속적이고 정확한 종양 센싱 및 pH 센싱이 가능하고, 어블레이션 치료 및 피드백 모니터링이 가능하여 치료 및 진단 효과가 우수하다. 상기 그래핀 바이오 전자 장치는 초박막으로 형성될 수 있어 우수한 기계적 변형 능력과 우수한 기계적 안정성을 가질 수 있다. 또, 상기 그래핀 바이오 전자 장치는 우수한 열적 안정성과 전기적 안정성을 가질 수 있다.
본 발명의 실시예들에 따른 치료진단 나노입자는 광역학 치료, 광열 치료, 및 화학 치료의 복합 치료가 가능하여 치료 효과가 우수하다. 상기 치료진단 나노입자에 로딩된 화학 약물의 방출을 제어할 수 있어 표적화된 치료가 가능하고 부작용을 최소화할 수 있다. 또, 암세포 등의 이미징에 의한 추가적인 진단이 가능하고, 능동적 표적화에 의해 치료 효과를 향상시킬 수 있다.
본 발명의 실시예들에 따르면, 투명한 그래핀 바이오 전자 장치를 채용함으로써 치료 및 진단 효과가 우수한 고집적 내시경 시스템이 구현될 수 있다. 상기 내시경 시스템은 적색 레이저와 근적외선 레이저에 의해 상기 치료진단 나노입자의 활성화를 제어함으로써 치료 및 진단 효과를 최적화할 수 있다.

도 4를 참조하면, 베이스 기판(250) 위에 제1 희생층(260)이 형성된다. 예를 들어, 베이스 기판(250)은 실리콘 기판일 수 있고, 제1 희생층(260)은 열 증착 공정을 이용하여 니켈로 형성될 수 있다.
제1 희생층(260) 위에 제1 보호층(211)이 형성된다. 제1 보호층(211)은 에폭시와 같은 투명한 소재로 제1 희생층(260) 위에 층을 형성한 후 패터닝하는 것에 의해 형성될 수 있다.
제1 보호층(211) 위에 그래핀층(221a)이 형성된다. 그래핀층(221a)은 화학기상증착 공정을 이용하여 구리 포일(Cu foil) 위에 형성된 후 제1 보호층(211) 위로 전사되는 것에 의해 형성될 수 있다.
그래핀층(221a) 위에 은 나노와이어층(222a)이 형성된다. 은 나노와이어층(222a)은 은 나노와이어 용액을 그래핀층(221a) 위에 스핀 코팅한 후 어닐링하는 것에 의해 형성될 수 있다.
도 5를 참조하면, 포토리소그래피 공정을 수행하여 그래핀층(221a)과 나노와이어층(222a)이 패터닝되고, 제1 그래핀층(221)과 은 나노와이어층(222)이 형성된다. 이때, 온도 센서의 활성 위치에 있는 은 나노와이어층(222a)과 그래핀층(221a)이 선택적으로 제거될 수 있다.

도 11을 참조하면, 실리콘 기판 위에 열 증착을 이용하여 니켈 희생층(100nm)이 형성된다. 니켈 희생층 위에 에폭시층을 형성한 후 패터닝하여 하부 에폭시층(Bottom epoxy)이 형성된다. 하부 그래핀층(Bottom graphene)은 화학기상증착 과정을 통해 구리 포일(Cu foil) 위에 형성된 후 구리 포일이 습식 식각에 의해 제거되고 PMMA에 의해 상기 실리콘 기판으로 전사된다. 은 나노와이어층(Ag NW)은 나노와이어 용액(0.5wt%)을 2000rpm으로 스핀 코팅하고, 1분 동안 어닐링하는 것에 의해 형성된다. Ag NW/GP층들은 포토리소그래피 공정에 의해 패터닝된다. 온도 센서의 활성 위치에 있는 은 나노와이어는 온도 모니터링 민감도를 향상시키기 위해 선택적으로 제거된다. 또 다른 GP층이 Ag NW/GP층들 위로 전사된다. 상부 그래핀층(Top graphene)은 패터닝된 후 상부 에폭시층(Top epoxy)에 의해 인캡슐레이션된다. 상기 상부 에폭시층은 활성 위치에서 GP/Ag NW/GP 전극을 노출시키도록 패터닝될 수 있다.
니켈 식각액에 의해 니켈 희생층이 제거되고, GP/Ag NW/GP 전극은 PMMA와 PDMS에 의해 픽업된 후 얇은 PDMS층으로 전사된다. GP/AgNW/GP 전극은 20mM AuCl3 용액에 10분 동안 담겨지고 상기 활성 위치의 노출된 부분에 금 도핑층이 형성된다. 전기 도금을 이용하여 금 도핑층 위에 이리듐 산화물층이 형성된다. 금 도핑층에 의해 이리듐 산화물층 증착의 균일성이 향상될 수 있다.
전기 도금을 위한 이리듐 산화물 용액은 150mg의 이리듐 4염화물을 100mL의 초고순도 증류수에 20분간 저어 용해시킴으로써 준비될 수 있다. 30% H2O2 수용액 1mL의 부분 표본(aliquot)을 첨가하고, 10분 동안 저어준다. 이어서, 500mg의 옥살산 2수화물을 첨가하고, 또 10분 동안 저어준다. 마지막으로, 무수 탄산칼륨을 이용하여 상기 용액의 pH가 10.5로 조절된다. 상기 용액은 이리듐 이온을 안정화하기 위해 상온에서 1주 동안 저장되고, 밝은 보라색을 갖는 용액이 형성된다. 전기 증착(electrodeposition)은 전기화학 분석 장치를 이용하여 3전극 방법에 의해 수행된다. 대시간 전위차법(Chronopotentiometry)은 이리듐 산화물 용액 내 Ag/AgCl 기준 전극, 그래핀 하이브리드 작업 전극 및 백금선 상대 전극을 가로질러 0.7V에서 5분 동안 수행된다.
그래핀 위에 전기 증착된 이리듐 산화물층은 낮은 계면 임피던스, 최소 부피 팽창, 및 높은 pH 민감도를 가진다. 그러므로 이리듐 산화물층은 작은 전기 신호의 감지, 어블레이션 에너지 전달, pH 변화의 측정, 및 조직으로의 전하 주입에 적합하다.
도 12는 본 발명의 일 실시예에 따른 그래핀 바이오 전자 장치의 제조 공정에서 금 도핑의 효과를 나타낸다. 도 12에서 왼쪽에 정렬된 도면은 금 도핑을 하지 않은 경우를 나타내고, 오른쪽에 정렬된 도면은 금 도핑을 한 경우를 나타낸다.
도 12를 참조하면, GP/Ag Nw/GP 구조의 금 도핑은 물 접촉각을 감소시키고 이리듐 산화물 전구체를 포함하는 수용액에서의 표면 적심(wetting)을 향상시킨다. 이에 의해 도면에서 나타나는 바와 같이 이리듐 산화물의 균일한 코팅이 이루어진다. 또, 이리듐 산화물 증착에 의해 형성되는 낮은 임피던스 성질이 신호 대 잡음 비와 그래핀 바이오 전자 장치의 전하 주입 효율을 향상시킨다.
[치료진단 나노입자]
도 13은 본 발명의 실시예들에 따른 따른 치료진단 나노입자를 개략적으로 나타낸다.
도 13을 참조하면, 치료진단 나노입자(300)는 금속 나노막대(310), 다공성 쉘(320), PDT(photodynamic, 광역학) 염료(321), 형광 염료(322), 화학 약물(323), 고분자 캡슐층(330), 및 항체(331)를 포함할 수 있다.
금속 나노막대(310)은 근적외선 레이저의 조사에 의해 활성화될 수 있는 소재, 예를 들어, 금으로 형성될 수 있고, 근적외선 레이저의 조사에 의해 PTT(photothermal, 광열) 치료를 유도할 수 있다.
다공성 쉘(320)은 금속 나노막대(310)와 함께 코어-쉘 구조체를 형성하고, 금속 나노막대(310)를 둘러쌀 수 있다. 다공성 쉘(320)은 금속 나노막대(310)와 반응하여 그 표면에서 성장할 수 있는 소재, 예를 들어, 다공성 실리카로 형성될 수 있다. PDT(Photodynamic) 염료(321), 형광 염료(322), 및 화학 약물(323)은 다공성 쉘(320)에 로딩될 수 있다. 고분자 캡슐층(330)은 금속 나노막대(310)와 다공성 쉘(320)의 코어-쉘 구조체를 둘러쌀 수 있다. 고분자 캡슐층(330)은 다공성 쉘(320)의 표면에서 중합될 수 있는 감열성 고분자, 예를 들어, 폴리(N-이소프로필아크릴아미드)(poly(N-isopropylacrylamide), PNIPAAm)로 형성될 수 있다. 고분자 캡슐층(330)은 부피 변화에 대한 감열 특성을 갖는다. 예를 들어, 고분자 캡슐층(330)은 열을 받지 않으면 그 부피를 유지하여 치료진단 나노입자(300)의 내부에 로딩된 화학 약물(323)을 밖으로 방출시키지 않지만, 열을 받으면 축소되어 부피가 감소하여 화학 약물(323)을 밖으로 방출시킨다. 따라서, 치료진단 나노입자(300)가 생체 내 암세포 등 타겟 위치에 도달한 후 열을 받으면 고분자 캡슐층(330)의 부피가 감소하여 내부에 로딩되어 있는 화학 약물(323)을 타겟 위치로 방출할 수 있다. 이에 의해 암세포 등을 효과적으로 제거할 수 있다. 화학 약물(323)이 타겟 위치가 아닌 다른 장기로 우연히 방출되는 것을 방지하기 위하여 고분자 캡슐층(330)의 부피 변화에 대한 감열성의 임계 온도는 체온(36.5℃)보다 높은 것이 바람직하다.
항체(331)가 고분자 캡슐층(330) 표면에 결합되어 배치된다. 항체(331)는 암세포 등의 항원과 결합 특이성을 가질 수 있으며, 예를 들어 세툭시맙일 수 있다. 상기 세툭시맙은 세포 표면의 상피 세포 증식 인자 수용체에 결합하여 세포 분열을 증진하는 경로를 차단하여 암세포의 증식을 방지할 수 있다. 예를 들어, 상기 세툭시맙은 대장암(HT-29) 세포에서 과발현되는 표피 생장인자 수용체의 능동적 표적화를 가능하게 한다. 즉, 치료진단 나노입자(300)는 항체(331)에 의해 능동적 표적화가 가능하게 된다.
도 14는 본 발명의 일 실시예에 따른 치료진단 나노입자의 구조 및 제조 공정을 나타낸다.
도 14를 참조하면, 치료진단 나노입자는 금 나노막대(Au nanorod, Au NR), 다공성 실리카 쉘(Mesoporous silica shell, MSS), 폴리(N-이소프로필아크릴아미드)(Poly(N-isopropylacrylamide), PNIPAAm) 캡슐층, 클로린 e6(Chlorin e6), 로다민 B(Rhodamin B), 독소루비신(Doxorubicin), 및 세툭시맙(Cetuximab)을 포함할 수 있다.
상기 치료진단 나노입자는 여러 단계 반응과 분리 과정에 의해 형성될 수 있다. 형성 과정은 1) 금 나노막대의 합성, 2) 다공성 실리카 쉘의 합성, 3) PDT 염료와 형광(FL) 염료의 로딩, 4) PNIPAAm 캡슐층의 형성, 및 5) 항체(세툭시맙) 결합과 독소루비신의 로딩으로 구성된다.
1) 금 나노막대의 합성 : 금 씨드 용액(Au seed solution)이 NaBH4용액(600μL, 10mM)을 HAuCl4·H2O(250μL, 10mM)과 세틸트리메틸 암모늄 브로마이드(CTAB)(7.5μL, 100mM)을 포함하는 씨드 수용액에 주입함으로써 제조된다. 성장(growth) 용액은 HAuCl4·3H2O(1.7mL, 10mM)와 AgNO3(250μL, 10mM)을 CTAB 용액(40mL, 100mM)에 첨가함으로써 제조되고, L-아스코르브산(270μL, 100mM)이 주입된다. 이 금 씨드는 씨드 용액(420μL)을 성장 용액에 추가적으로 주입함으로써 금 나노막대로 변환되고, 3시간 동안 반응시킨다. 최종 제품 용액은 2회 원심분리된다.
2) 다공성 실리카 쉘의 합성 : 실리카 쉘은 금 나노막대의 표면에서 성장한다. 테트라에틸 오소실리케이트(tetraethyl orthosilicate)(TEOS)(30μL)가 알칼리 조건(pH 10-11)에서 금 나노막대 용액(50mL)에 주입되고, 금 나노막대와 4시간 동안 반응한다. 실리카 표면의 기능화는 (3-아미노프로필) 트리에톡시실란((3-aminopropyl) triethoxysilane)(10μL)과 3-(메타크릴록시) 프로필 트리에톡시실란(3-(methacryloxy) propyl triethoxysilane)(10μL)을 주입함으로써 획득되고, 상기 용액은 4시간 동안 저어진다. 실리카 코팅 금 나노막대(Au NR@MSS)는 2회 원심분리되고 에탄올에서 확산된다. 실리카 쉘에서 기공을 형성하기 위해서, pH를 1-2로 조절하기 위해 HCl이 나노입자-에탄올 서스펜션에 추가되고, CTAB 템플레이트를 제거하기 위해 환류된다. 결과물인 실리카 코팅 금 나노막대(Au NR@MSS)는 2회에 걸쳐 원심분리되고 물에 확산된다.
3) PDT 염료와 형광(FL) 염료의 로딩 : 클로린 e6(Ce6)는 N-(3-디메틸아미노프로필)-N'-에틸카보디이미드 하이드로클로라이드(EDC)와 N-하이드록시숙신이미드(NHS)와 같은 몰 양으로 반응한다. 그리고 기능화된 클로린 e6는 실리카 코팅 금 나노막대(Au NR@MSS)와 12시간 동안 반응한다. 형광(FL) 염료를 로딩하기 위해서, 로다민 B 이소티오시아나이트(isothiocyanate)가 혼합되고 12시간 동안 반응한다. 결합 후에, 실리카 코팅 금 나노막대(Au NR@MSS)는 원심 분리되고 물에 확산된다.
4) PNIPAAm 캡슐층의 형성 : PNIPAAm 캡슐층을 형성하기 위해서, 실리카 코팅 금 나노막대(Au NR@MSS) 용액(5mL)은 N-이소프로필아크릴아미드(NIPAAm)(12mL, 100mM), 아크릴산(1.4mL, 100mM), N,N'-메틸렌비스(아크릴아미드)(1.2mL, 100mM), 아크릴레이트-PEG-NHS(20mg), 소듐 도데실 설페이트(Sodium dodecyl sulfate)(200μL)와 반응한다. 상기 용액은 산소를 제거하기 위해 70℃까지 가열되고 아르곤으로 버블된다(bubbled). 30분 후에, 과황산 칼륨(potassium persulfate)(1mL, 20mM)이 중합을 시작하기 위해 주입된다. Au NR@MSS@PNIPAAm 용액은 반응하지 않은 화학성분들을 제거하기 위해 2회 원심분리된다.
5) 항체 결합과 독소루비신(Dox)의 로딩 : 세툭시맙(항체)(2mL, 5mg/mL)이 결합을 위해 AuNR@MSS@PNIPAAm 용액에 첨가된다. NHS 말단기는 PEG 말단기와 반응한다. 이러한 항체가 결합된 AuNR@MSS@PNIPAAm은 원심분리되고 PBS에서 확산된다. 이어서 독소루비신(Dox)(1mL, 0.6mg/mL) 용액을 나노입자 용액(5mL)에 첨가하고 하루 동안 저어준다. 과잉 독소루비신(Dox)은 나노입자를 원심분리 함으로써 제거될 수 있다.
도 15는 본 발명의 일 실시예에 따른 치료진단 나노입자의 생체 내 독성 시험 결과를 나타낸다.
도 15를 참조하면, 치료진단 나노입자가 주입된 쥐와 정상 쥐의 여러 장기에서 조직 이미지를 분석한 결과 상기 치료진단 나노입자는 각각의 장기에서 아무런 염증을 유발하지 않았고, 이는 상기 치료진단 나노입자는 독성을 갖고 있지 않다는 것을 의미한다.
내시경 시스템의 개략적인 구조 및 이미지
도 16은 본 발명의 일 실시예에 따른 내시경 시스템의 개략적인 구조와 대응 이미지를 나타내고, 도 17은 내시경 위에 그래핀 바이오 전자 장치의 집적 과정을 나타내며, 도 18은 내시경 위에 설치되는 그래핀 바이오 전자 장치의 외부 연결을 나타낸다.
도 16 내지 도 18을 참조하면, 암세포가 감지되고 확인되면, 암세포 조직의 큰 부위는 겸자(forceps)를 통해 절제되고, 이어서 그래핀 바이오 전자 장치를 이용한 고주파 어블레이션이 수행된다. 이 어블레이션 치료의 피드백 조절(feedback modulations)은 온도, 접촉, 및 세포/조직 생존력의 지속적인 모니터링에 기반을 둔다. 다공성 실리카 셀(MSS)에 로딩된 PDT 염료(chlorin e6; Ce6), 금 나노막대(Au NR), 및 화학 약물(독소루비신, Dox)에 의해 유도된 광역학(PDT) 치료, 광열(PTT) 치료, 및 화학 치료는 조사된 적색 또는 근적외선 레이저를 통해 활성화 되어 치료 부위 주변의 잔류 암세포들을 효과적으로 제거할 수 있다. 감열성 폴리(N-이소프로필아크릴아미드)(PNIPAAm) 캡슐층은 독소루비신(Dox)이 근적외선 레이저의 조사 없이 방출되는 것을 방지한다. 치료 및 진단 과정을 진행하기에 앞서, 상기 그래핀 바이오 전자 장치는 세척되고, 살균되어 내시경에 부착된다(도 17의 a,b). 상기 그래핀 바이오 전자 장치는 그래핀의 플렉시블 성질, 초박막 구조, 및 중성 역학층 설계(neutral mechanical plane designs)에 의해 획득되는 고도의 기계적 변형성으로 인해 설치, 제거, 및 배선 동안 균열을 발생시키지 않고 기계적으로 구부러지거나 휘어질 수 있다.
그래핀 바이오 전자 장치의 특성

도 20을 참조하면, 그래핀 바이오 전자 장치는 멸균 처리기(120℃, 200kPa, 15min)에서 뜨거운 포화 증기를 이용하여 살균 처리를 거친 후에도 살균 처리 전과 비교하여 임피던스가 비슷하게 나타난다. 따라서 상기 그래핀 바이오 전자 장치는 우수한 열적 안정성을 가짐을 알 수 있다.

도 21을 참조하면, 다양한 곡률 반경에서 구부리고 비트는 변형 후에 그래핀 바이오 전자 장치의 저항 변화가 거의 나타나지 않는다. 상기 그래핀 바이오 전자 장치의 초박막 구성은 더욱 우수한 기계적 변형 능력을 가질 수 있게 한다. 따라서, 상기 그래핀 바이오 전자 장치는 우수한 기계적 안정성을 가짐을 알 수 있다.
도 22는 본 발명의 일 실시예에 따른 그래핀 바이오 전자 장치의 재료 특성을 나타낸다.
도 22를 참조하면, 그래핀 바이오 전자 장치의 그래핀 구성은 라만 분광법에 의해 특징지어진다. 상기 그래핀 구성은 2D와 G밴드의 상대적인 피크 강도(I2D/G=1.01)에 기초하여 2-3 그래핀층들로 구성된다. 결함 관련 D 밴드 피크가 강하게 억제되어, 고품질의 그래핀 합성을 나타낸다. 은 나노와이어가 X선 회절(XRD)에 의해 분석된다. 산화은(Ag2O)의 피크의 감지(θ=32°)는 열 어닐링을 하는 동안 표면에서 형성된 적은 양의 산화은을 나타낸다. 추가적인 산화는 상부 그래핀과의 패시베이션(passivation)에 의해 최소화된다. 전기 증착된 이리듐 산화물 필름은 X선 광전자 분광법(XPS)에 의해 이리듐과 산소 원자 사이의 결합을 나타내는, 62.1과 65.0eV의 이리듐 4f7

/2 피크 에너지로 특징지어진다. 금속 이리듐(61.1 과 64.1 eV)과 이리듐 산화물 표준(62.7과 65.7 eV)의 특징적인 XPS 피크를 비교하면, 전기 증착된 이리듐 산화물은 고도로 산화된 형태로 존재한다.
도 23은 본 발명의 일 실시예에 따른 그래핀 바이오 전자 장치의 전기적 안정성을 나타낸다.
도 23을 참조하면, 내시경 사용 과정에서 센서와 작동기로서 적합하기 위해, 전자 장치의 재료는 고주파 어블레이션 동안의 온도 변화 및 바이오 유체 환경에서 다중 전기화학 사이클에의 노출에 견뎌야 한다. 그래핀 바이오 전자 장치는 PBS 내 20-50℃의 온도 범위에서 안정한 임피던스 값을 유지한다. 인터커넥션의 저항 변화는 작다. 그래핀 바이오 전자 장치는 PBS에서 다중 순환 전압전류법 테스트 후에도 전기화학적 안정성을 유지한다. 소 태아혈청(Life technologies, 16000)에서 임피던스의 안정성은 6시간에 걸쳐 확인되고, 바이오 유체 환경에서의 안정적인 전기화학 작용을 나타낸다. 산화 및 환원 과정에서 새로운 물질이 형성되지 않는다.
종양 센싱
도 24는 본 발명의 일 실시예에 따른 그래핀 바이오 전자 장치의 종양 센싱 특성을 나타내고, 도 25는 본 발명의 일 실시예에 따른 그래핀 바이오 전자 장치의 pH 센싱 특성을 나타낸다.
도 24 및 도 25를 참조하면, 종양 센서는 임피던스 차이에 따라 정상 조직에 대하여 암 조직을 구별할 수 있다. 암 조직은 건강한 조직과 비교하여 상당히 낮은 임피던스를 나타낸다. 이러한 임피던스의 차이는 종양 조직 및 종양 성장을 감지하는데 사용될 수 있다. 바이오 임피던스 리코딩(Bio-impedence recordings)은 크기, 간격, 및 밀도에 매우 의존적이고, 이는 조직에서 전기 신호와 전하 저장을 변경할 수 있다. 특정 주파수 범위도 바이오 임피던스 분석에 근거한 조직 구별에 중요하다. 임피던스 기반 종양 감지는 유방암, 식도암, 전립선암, 및 뇌종양의 진단에도 사용될 수 있다. 본 발명에서 제공된 내시경 센싱은 쥐(BALB/c-nude mouse)의 피하 대장암(HT-29) 모델의 체내 분석을 나타낸다. 바이오센서는 암 조직과 정상 피부 조직 사이의 임피던스 차이를 측정하며, 체외 실험과 체내 실험 모두, 대장암(HT-29) 조직은 정상 조직보다 낮은 임피던스를 나타낸다.
종양 센서는 암 조직 주변에서의 pH 수준의 변화를 측정하여 암 조직을 감지할 수 있다. 암의 빠fms 신진 대사에 기인한 종양 주변에서의 pH 수준의 변화는 종양을 감지하는 중요한 마커(marker)로서 기능한다. 암 조직에서 세포외 pH 수준은 증가된 젖산 생성과 감소된 세포간 유체 완충 작용에 기인하여 정상 조직에서의 pH보다 낮다. 그래핀 복합층의 표면 제타 전위가 pH 의존도를 가지기 때문에 개방회로 전위를 측정함으로써 pH를 감지할 수 있다. 측정된 개방회로 전위 값은 캘리브레이션 커브에 근거하여 pH 값으로 변환된다. 본 발명의 실시예들에 따른 pH 센서는 종양 조직과 정상 조직을 구별하기 위해 종양 근처의 pH수준을 측정한다. 종래의 pH 센서는 용액상 pH 측정을 위한 3-전극 방법을 사용했다. 그러나 본 발명의 실시예들에 따른 pH 센서는 2-전극 기반이다. 즉, 상기 pH 센서는 이리듐 산화물이 전기 증착된 그래핀 복합층 부분의 작업 전극과 금 도핑된 그래핀 복합층 부분의 상대 전극으로 구성된다. 이는 전기 증착된 이리듐 산화물이 pH에 민감한 성질을 갖기 때문이다. 상기 pH 센서는 완충 용액의 pH 변화 동안에도 재현 가능하고, 암 조직과 정상 조직의 pH 값은 생체 내에서 측정될 수 있다.
고주파 어블레이션 및 피드백 모니터링

도 29는 본 발명의 일 실시예에 따른 치료진단 나노입자의 표적화, 이미징, 및 치료 특성을 나타낸다.
도 29를 참조하면, 그래핀 바이오 전자 장치와 결합되어 사용되는 치료진단 나노입자는 추가적인 암 진단과 표적화된 치료 방법을 제공한다. 도 29b의 세포 TEM 이미지는 암세포에 의한 나노입자의 표적화된 흡수를 나타내고, 이는 도 29c에 있는 형광 이미지와 도 29d에 있는 유동 세포분석 데이터에 의해 입증된다. 상기 치료진단 나노입자들은 활성 산소종(ROS) 발생, 광유도 발열, 및 제어된 약물 방출을 통해 암세포와 싸운다. 상기 치료진단 나노입자의 광 활성화는 레이저 조사 부위에 국한되고, 레이저의 강도를 조절하는 것에 의해 제어된다. 내시경을 통해 전달되는 레이저 빛의 직접적인 제어는 빛의 투과 깊이와 관련하여 많은 문제들을 해결한다. 유동 세포분석 결과에서 나타난 바와 같이, 대조군과 비교했을 때, 상기 치료진단 나노입자 상의 PDT 염료는 더욱 효과적으로 암세포에 전달되고 흡수된다. 적색 레이저(파장 670nm)에 의해 조사되면, PDT 염료는 활성 산소종을 발생시키고, 세포 생존력은 감소한다. 온도는 입자 농도, 근적외선 레이저 강도(808nm), 및 조사 시간를 변화시키는 것에 의해 광열적으로 조절되고, 암세포 생존력을 감소시키기 위해 최적화된다. 온도를 증가시키는 것은 PNIPAAm 캡슐층의 수축에 의해 상기 치료진단 나노입자의 유체역학적 직경을 약 290nm에서 약 110 nm로 변화시키고, 이것은 상기 치료진단 나노입자에 로딩된 독소루비신(Dox)의 방출을 유도한다. 방출 온도는 체온보다 높게 설계되는 것이 바람직하다. PNIPAAm 블록 공중합체는 레이저 조사가 없으면 약물 방출을 억제하고, 독소루비신(Dox)의 부작용을 최소화한다. PDT 치료, PTT 치료, 및 화학 치료의 복합 치료(GaAs 펄스 레이저를 사용함; 파장 690nm, 전력 30mW의 적색 레이저, 파장 808nm, 전력 30mW의 근적외선 레이저)을 수행한 후의 암세포의 생존력 시험은 복합 치료의 효과가 우수하다는 것을 나타낸다.
내시경 시스템을 이용한 대장암 치료
도 30은 본 발명의 일 실시예에 따른 내시경 시스템을 이용한 종양 치료 과정을 개략적으로 나타낸다.
도 30을 참조하면, 내시경 시스템의 대표적인 의학 적용 사례는 대장암 치료를 포함한다. 상기 치료는 치료진단 나노입자를 정맥에 투입함으로써 시작되고, 상기 치료진단 나노입자의 표면 위에 결합된 특정 항체(세툭시맙)는 대장암 세포(HT-29)를 표적으로 한다. 상기 치료진단 나노입자에 로딩된 형광 염료의 이미징은 암세포의 공간 분포에 관한 시각적인 정보를 제공한다. 상기 내시경은 상기 치료진단 나노입자에 노출된 의심 위치에 레이저 빛이 접근할 수 있도록 해준다. 이러한 부위들은 가시범위에서 80%의 총 투과율을 가지는 그래핀 바이오 전자 장치에서도 쉽게 관찰될 수 있다. 그래핀 바이오 전자 장치와 이와 관련된 센서들은 종양 분포의 추가적인 전기화학적 분석을 제공한다.
도 31은 본 발명의 일 실시예에 따른 내시경 시스템을 이용한 생체 내 대장암 치료를 설명하기 위한 도면이다.
도 31을 참조하면, 내시경에 탑재된 투명한 그래핀 바이오 전자 장치와 유도 레이저에 의해 활성화되는 치료진단 나노입자는 생체 내 모델로 적용될 수 있다. 쥐의 피하 표면에서 자란 대장암(HT-29)의 내시경 치료는 상기 치료진단 나노입자를 꼬리 정맥을 통해 정맥 주사로 주입하면서 시작된다. 대장암 모델은 큰 동물에서는 이용하기 어렵고 작은 동물들의 위장관을 보기엔 내시경이 크기 때문에, 쥐 피하의 대장암 모델을 이용하여 체내 연구를 진행한다. 상기 치료진단 나노입자에 로딩된 독소루비신의 방출이 PNIPAAm 캡슐화에 의해 억제되기 때문에, 독소루비신(Dox)으로 인한 부작용은 최소화된다. 절제된 장기의 형광 이미지와 생물학적 분배 분석 데이터는 성공적인 표적화를 보여준다. 많은 수의 치료진단 나노입자는 주입 후 6시간 안에 종양에 축적된다. 대부분의 치료진단 나노입자는 짧은 순환 시간 때문에 하루 안에 피에서 제거된다.
나노입자-표적화된 종양의 형광 광학 이미지는 치료진단 나노입자 주입 후 6시간 후에 체내에서 획득될 수 있고, 종양 의심부분과 종양이 없는 부분이 확인될 수 있다. 자세한 사항은 내시경에 설치된 카메라를 통해 시각적으로 관찰될 수 있다. 내시경 카메라에 의해 캡쳐된 종양이 자란 표면의 이미지는, 투명 그래핀 바이오 전자 장치가 시각적 관찰을 방해하지 않는 반면, 금속 기반 전자 장치는 심각한 방해를 야기하는 것을 나타낸다. 암에 걸린 세포를 확인하면, 고주파 어블레이션 치료를 이용하여 종양을 제거한다. 이 경우, 접촉 센서는 그래핀 바이오 전자 장치와 조직 간 콘포말 접촉(conformal contact)을 감지하기 위해 사용된다. 고주파 어블레이션 동안의 접촉모드에서, 종양의 시각적 관찰은 불가능하다. 그러므로 임피던스 기반 종양 센서는 종양 세포의 낮은 임피던스와 pH 수준에 따라 암 조직의 위치를 찾아내는데 사용된다. 온도 변화의 모니터링은 고주파 어블레이션 동안 추가적인 안내를 제공한다. 최종적으로, 어블레이션 치료를 확인하기 위해 조직 생존력이 측정된다.
그래핀 바이오 전자 장치를 통한 물리적 치료와 함께 암세포는 치료진단 나노입자로 치료된다. 이 치료진단 나노입자는 적색 및 근적외선 레이저를 이용하여 국부적으로 활성화되어 PDT 치료, PTT 치료, 및 화학 치료를 유도한다. 이러한 다중 조정(multiple interventions)의 효과는 시각적 관찰을 기반으로 종양 부피(HT-29) 변화를 추적함으로써 생체 내에서 확인된다. 대조군(치료 없음)에서 종양 부피는 2주 후에 증가하나, 치료를 받은 그룹은 종양 부피가 감소한다. 쥐에서 자라는 종양은 나노입자 주입 없이 레이저를 조사한 경우 또는 나노입자 없이 화학 약물만으로 치료된 경우에 종양의 부피가 감소하지 않지만, 복합 치료(PDT, PTT, 및 화학 치료)는 종양 부피의 급격한 감소를 보였다. 치료 후 종양의 H&E(헤마톡실린과 에오신) 염색 이미지는 암세포의 세포 소멸 때문에 불규칙적인 구조를 나타낸다. TUNEL(Terminal deoxynucleotidyl transferase dUTP nick end labeling) 분석은 복합 치료 후의 암세포의 세포 자멸을 보여준다.
이제까지 본 발명에 대한 구체적인 실시예들을 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
본 발명의 실시예들에 따른 그래핀 바이오 전자 장치는 내시경 팁 부분에 배치되어 생체 내부를 진단하고 치료하는 기능을 수행할 수 있다. 그래핀 바이오 전자 장치는 투명도가 높아 카메라 위에 배치될 수 있다. 이에 의해, 내시경 팁의 표면적을 증가시키지 않고도 그래핀 바이오 전자 장치가 내시경에 배치될 수 있고, 내시경 시스템이 고집적화 될 수 있다. 그래핀 바이오 전자 장치는 지속적이고 정확한 종양 센싱 및 pH 센싱이 가능하고, 어블레이션 치료 및 피드백 모니터링이 가능하여 치료 및 진단 효과가 우수하다. 상기 그래핀 바이오 전자 장치는 초박막으로 형성될 수 있어 우수한 기계적 변형 능력과 우수한 기계적 안정성을 가질 수 있다. 또, 상기 그래핀 바이오 전자 장치는 우수한 열적 안정성과 전기적 안정성을 가질 수 있다.
본 발명의 실시예들에 따른 치료진단 나노입자는 광역학 치료, 광열 치료, 및 화학 치료의 복합 치료가 가능하여 치료 효과가 우수하다. 상기 치료진단 나노입자에 로딩된 화학 약물의 방출을 제어할 수 있어 표적화된 치료가 가능하고 부작용을 최소화할 수 있다. 또, 암세포 등의 이미징에 의한 추가적인 진단이 가능하고, 능동적 표적화에 의해 치료 효과를 향상시킬 수 있다.
본 발명의 실시예들에 따르면, 투명한 그래핀 바이오 전자 장치를 채용함으로써 치료 및 진단 효과가 우수한 고집적 내시경 시스템이 구현될 수 있다. 상기 내시경 시스템은 적색 레이저와 근적외선 레이저에 의해 상기 치료진단 나노입자의 활성화를 제어함으로써 치료 및 진단 효과를 최적화할 수 있다.
Claims (36)
- 둘 이상의 그래핀층을 포함하는 그래핀 복합층을 포함하는 그래핀 바이오 전자 장치.
- 제 1 항에 있어서,상기 그래핀 복합층은,제1 그래핀층,상기 제1 그래핀층 위에 배치되는 은 나노와이어층, 및상기 은 나노와이어층 위에 배치되는 제2 그래핀 층을 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 2 항에 있어서,상기 제2 그래핀층 위에 이격되어 배치되는 복수개의 금속 산화물층을 더 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 3 항에 있어서,상기 금속 산화물층은 이리듐 산화물층을 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 3 항에 있어서,상기 제2 그래핀층과 상기 금속 산화물층 사이에 배치되는 금속 도핑층을 더 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 5 항에 있어서,상기 금속 도핑층은 금 도핑층을 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 3 항에 있어서,상기 그래핀 복합층과 상기 금속 산화물층에 의해 형성되는 센서 및 어블레이션 전극을 더 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 7 항에 있어서,상기 센서는, 종양 센서, 접촉 센서, 및 생존력 센서 중에서 적어도 하나 이상을 더 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 8 항에 있어서,상기 종양 센서는, 작업 전극과 상대 전극을 포함하고,상기 작업 전극은 상기 그래핀 복합층과 상기 금속 산화물층을 포함하고,상기 상대 전극은 상기 그래핀 복합층은 포함하고, 상기 금속 산화물층은 포함하지 않는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 8 항에 있어서,상기 제2 그래핀층의 패터닝된 일부만으로 구성되는 온도 센서를 더 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 1 항에 있어서,상기 그래핀 바이오 전자 장치는 내시경의 카메라 위에 부착되어 사용되는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제1 보호층;상기 제1 보호층 위에 배치되는 제1 그래핀층;상기 제1 그래핀층 위에 배치되는 은 나노와이어층;상기 은 나노와이어층 위에 배치되는 제2 그래핀층; 및상기 제2 그래핀층 위에 배치되는 제2 보호층을 포함하는 그래핀 바이오 전자 장치.
- 제 12 항에 있어서,상기 제1 보호층 및 상기 제2 보호층은 투명 에폭시로 형성되는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 12 항에 있어서,상기 제2 그래핀층 위에 배치되는 금속 산화물층을 더 포함하고,상기 제2 보호층은 상기 제2 그래핀층의 적어도 일부를 노출시키고,상기 금속 산화물층은 상기 제2 그래핀층의 노출된 일부 위에 배치되는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 제 14 항에 있어서,상기 제2 그래핀층과 상기 금속 산화물층 사이에 배치되는 금속 도핑층을 더 포함하는 것을 특징으로 하는 그래핀 바이오 전자 장치.
- 금속 나노막대;상기 금속 나노막대를 둘러싸는 다공성 쉘; 및상기 다공성 쉘을 둘러싸고, 감열성을 갖는 고분자 캡슐층을 포함하는 치료진단 나노입자.
- 제 16 항에 있어서,상기 금속 나노막대는 근적외선 레이저의 조사에 의해 활성화되어 광열 치료를 유도하는 것을 특징으로 하는 치료진단 나노입자.
- 제 16 항에 있어서,상기 다공성 쉘은 광역학(PDT) 염료, 형광 염료, 및 화학 약물 중에서 하나 이상을 포함하는 것을 특징으로 하는 치료진단 나노입자.
- 제 18 항에 있어서,상기 광역학 염료는 적색 레이저의 조사에 의해 활성 산소종을 발생시키는 것을 특징으로 하는 치료진단 나노입자.
