 しかしながら、脱塩処理は煩雑である。また、特許文献1のように、電極とゲルとの間に水で湿らせたろ紙を挟んで等電点電気泳動を行った場合、当該ろ紙が等電点電気泳動に次の様な悪影響を及ぼす場合がある。
 図7の(a)は、従来技術に係る等電点電気泳動器具9の側方断面図である。等電点電気泳動器具9は、pH勾配が形成されたゲル2、電極3、電極4、ろ紙7およびろ紙8を備えており、ゲル2と電極3との間にろ紙7が、ゲル2と電極4との間にろ紙8が挟まれている。そのため、電流は、電極3-ろ紙7-ゲル2-ろ紙8-電極4という経路で流れる。ゆえに、電気泳動によって、サンプルが、ゲル2からろ紙7またはろ紙8に流出する場合がある。ろ紙に接するゲル部は等電位になるため、ろ紙7またはろ紙8に流出したサンプルは、ろ紙7またはろ紙8中に拡散し、一部は、再度ゲル2に入る。これによって、ゲルにおけるろ紙7またはろ紙8が接触している部位の等電点が、図7の(b)に示すようにフラットになり、分離範囲が狭くなってしまう。また、ろ紙7、8を設置および除去する工程も煩雑であり、ろ紙7、8をゲル2から除去する際にゲル2を破損するおそれもある。
 これに対し、図7の(c)に示す等電点電気泳動器具10のように、ゲル2に直接電極3および電極4が接触している構成では、電流が、電極3-ゲル2-電極4という経路で流れるため、電気泳動によって、サンプルがゲル2からろ紙等に流出することはない。また、ろ紙の設置および除去も不要である。しかしながら、等電点電気泳動器具10の構成では、サンプルに含まれたイオン性夾雑物が電極に堆積して、クリアな泳動結果が得られない場合がある。
 本発明は、上記課題に鑑みてなされたものであり、等電点電気泳動において、ゲルに接触している電極にイオン性夾雑物が堆積することを防止して、クリアな泳動結果を得るための技術を提供することを主たる目的とする。
 上記の課題を解決するために、本発明の一態様に係る等電点電気泳動器具は、特定の方向に沿ってpH勾配が形成されているゲルと、該ゲルに対し、該特定の方向に沿って電圧を印加する第一および第二の電極と、を備え、第一の電極は、該ゲルに接触しており、溶媒を保持するとともに、該溶媒を第一の電極および該ゲルに接触させる溶媒保持部をさらに備えている。
 本発明の一態様によれば、等電点電気泳動において、ゲルに接触している電極にイオン性夾雑物が堆積することを防止して、クリアな泳動結果を得ることができる。

 本発明の態様1に係る等電点電気泳動器具は、特定の方向に沿ってpH勾配が形成されているゲルと、該ゲルに対し、該特定の方向に沿って電圧を印加する第一および第二の電極と、を備え、第一の電極は、該ゲルに接触しており、溶媒を保持するとともに、該溶媒を第一の電極および該ゲルに接触させる溶媒保持部をさらに備えている。
 上記の構成によれば、第一の電極はゲルに接触しており、溶媒保持部が溶媒を第一の電極およびゲルに接触させている。これにより、ゲルに接触している第一の電極近傍に移動したイオン性夾雑物を当該溶媒に拡散させて、第一の電極にイオン性夾雑物が堆積することを防止することができる。これにより、イオン性夾雑物が等電点電気泳動に及ぼす影響を抑制して、クリアな泳動結果を得ることができる。
 〔実施形態1〕
 (等電点電気泳動器具)
 図1は、本発明の一実施形態(実施形態1)に係る等電点電気泳動器具1の概略構成を模式的に示す側方断面図である。等電点電気泳動器具1は、ゲル2、第一の電極4、第二の電極3、溶媒保持部6および溶媒保持部5を備えている。
 ゲル2としては、特定の方向に沿ってpH勾配が形成されているものであれば特に限定されず、等電点電気泳動において一般的に用いられている公知のゲルを用いることができ、例えば、アクリルアミド誘導体によって形成されるIPG(Immobilized pH gradient、固定化pH勾配)ゲルや、アンフォライトによってpH勾配が形成されたチューブゲル等を用いることができる。
 ゲル2の形状は特に限定されないが、本実施形態では、長手方向が、pH勾配の方向である板状のゲルである。このようなゲル2としては、例えば、市販のIPGゲルを用いることができる。
 第一の電極4および第二の電極3は、何れか一方が陽極であり他方が陰極である。第一および第二の電極の素材は特に限定されないが、例えば、白金、金、銀、銅等の金属や、カーボン等であり得る。第一の電極4および第二の電極3は、ゲル2(例えば、ゲル2の板面)に接触しており、ゲル2に対して、ゲル2のpH勾配の方向に沿って電圧を印加するための電極となる。
 溶媒保持部6は、溶媒を、第一の電極4およびゲル2に接触させるように保持するものである。また、溶媒保持部5は、溶媒を、第二の電極3およびゲル2に接触させるように保持するものである。
 「溶媒」は、イオン性夾雑物(例えば、塩)を溶解することができる溶媒であれば特に限定されず、水系の溶媒または有機溶媒であり得る。また、溶媒には、粘度を高めるための添加剤(例えば、グリセロール)が含まれていてもよい。
 ゲル2にサンプルを導入し、第一の電極4および第二の電極3を用いてゲル2に電圧を印加することにより、等電点電気泳動器具1を用いて等電点電気泳動を行うことができる。ゲル2にはpH勾配が形成されているため、第一の電極4および第二の電極3の間に電圧が印加されると、サンプル中の各成分(例えば、タンパク質)は、その等電点に対応するゲル2中の位置に泳動される。
 このとき、サンプル中には、イオン性夾雑物が含まれていることがある。例えば、生体内から抽出したサンプルには、イオン性夾雑物(塩)が多く含まれる。また、難溶性の生体分子を溶解させるために塩を含んだ溶媒に溶解させてサンプルとする場合もある。このイオン性夾雑物が、電気泳動によって、第一の電極4または第二の電極3の近傍まで移動し、第一の電極4または第二の電極3に堆積すると、泳動が乱れ、クリアな泳動結果が得られない。
 ここで、本実施形態によれば、溶媒保持部6が、溶媒を第一の電極4に接触させるため、第一の電極4近傍に移動したイオン性夾雑物は当該溶媒に拡散する。これにより、第一の電極4にイオン性夾雑物が堆積することを防止することができる。これにより、イオン性夾雑物が等電点電気泳動に及ぼす影響を抑制して、クリアな泳動結果を得ることができる。また、溶媒保持部5も同様の効果を奏する。
 なお、等電点電気泳動器具1では、第一の電極4および第二の電極3は、ゲル2に直接接触している。そのため、電流は、溶媒保持部5および6を介さずに、第二の電極3-ゲル2-第一の電極4という経路で流れる。ゆえに、電気泳動によって、ゲル2中のサンプルが、ゲル2から溶媒保持部5または6に流出することは殆ど生じない。ゆえに、特許文献1に記載の技術のように分離範囲が狭まることがなく、電極近傍まで等電点スポットをクリアに検出することができる。
 また、本実施形態において、溶媒保持部6は、ゲル2のpH勾配の方向に垂直かつ第一の電極4における第二の電極3側の外縁を含む平面Aに対して、第二の電極3とは反対側の領域内に配置されており、平面Aに対して、第二の電極3とは反対側の領域内において溶媒を保持している。
 これを、別の観点からみれば、第一の電極4、および、溶媒保持部6における第二の電極側3の外縁がともに平面A上にあり、イオン性夾雑物を溶解する溶媒は、第一および第二の電極の電極間から完全に排除されている。
 これにより、イオン性夾雑物を溶解する溶媒は、第一の電極4および第二の電極3の電極間外において保持される。これにより、溶媒中に溶解したイオン性夾雑物が、再度、第一の電極4方向に移動することを抑制し、イオン性夾雑物が等電点電気泳動に及ぼす影響をさらに抑制することができる。また、溶媒保持部5も同様の効果を奏する。
 また、本実施形態では、ゲル2と溶媒保持部6とで第一の電極4を挟んでいる。これにより、第一の電極4近傍に移動したイオン性夾雑物を、溶媒保持部6が保持する溶媒に首尾よく拡散させることができる。また、溶媒保持部5も同様の効果を奏する。
 (溶媒保持部)
 図2は、本実施形態における溶媒保持部6のバリエーションを模式的に示す平面図である。
 図2の(a)は、溶媒保持部6における第一の電極4およびゲル2に対向する面に溝6aが設けられている構成を示す。このように、溶媒保持部6における第一の電極4およびゲル2に対向する面に、溶媒を保持するための溝6aが形成されていることによって、溶媒を首尾よく保持することができる。
 図2の(b)は、溶媒保持部6における第一の電極4およびゲル2に対向する面に、溶媒を保持する多孔質体6bが設けられている構成を示す。このように、溶媒保持部6における第一の電極4およびゲル2に対向する面に、溶媒を保持する多孔質体6bが設けられていることによって、溶媒を首尾よく保持することができる。また、毛管現象によって溶媒保持部6からゲル2へ溶媒が移動してゲル2が膨らみ、等電点電気泳動に悪影響を与えることを抑制することができる。
 なお、溶媒保持部6全体を多孔質体6bで構成してもよい。また、多孔質体6bとしては、溶媒を保持し得るものであれば特に限定されないが、例えば、多孔質樹脂または繊維であり得る。繊維としては、例えば、ろ紙であってもよい。多孔質体6bをろ紙とすることで、簡易かつ安価に溶媒保持部6を構成することができる。
 図2の(c)は、溶媒保持部6における第一の電極4およびゲル2に対向する面に複数の微細な溝6cが設けられている構成を示す。複数の微細な溝6cは、一実施形態において、キャピラリ電気泳動において使用されるような微細な溝であり得る。このように、溶媒が微細な溝6c中に保持されていることによって、毛管現象によって溶媒保持部6からゲル2へ溶媒が移動してゲル2が膨らみ、等電点電気泳動に悪影響を与えることを抑制することができる。
 なお、図2の(c)に示す構成では、溶媒保持部6における第一の電極4およびゲル2に対向する面に設けられる複数の微細な溝の形状は、ストライプ状であるが、本実施形態はこれに限定されず、図2の(d)に示すような同心円状の微細な溝6dであってもよいし、図2の(e)に示すような格子状の微細な溝6eであってもよい。なお、図2の(e)に示すような格子状の微細な溝6eを設けることにより、溶媒の保持量を増加させることができる。
 また、溶媒保持部6は、等電点電気泳動器具1から取り外し可能であってもよい。これにより、等電点電気泳動を行っているとき、イオン性夾雑物が十分に溶媒に溶解した後に、溶媒保持部を取り外して、等電点電気泳動器具1内から、イオン性夾雑物を除去することができる。これにより、イオン性夾雑物の等電点電気泳動に対する影響をさらに抑制して、よりクリアな泳動結果を得ることができる。なお、本実施形態における溶媒保持部6は、特許文献1に記載のろ紙とは異なり、電極とゲルとの間に挟まれているものではないため、電極とゲルとの間の位置関係をずらすことなく、取り外すことができる。
 溶媒保持部5のバリエーションについては、溶媒保持部6のバリエーションと同様であるため、説明を省略する。また、溶媒保持部5も、等電点電気泳動器具1から取り外し可能であってもよい。
 (等電点電気泳動方法)
 本実施形態に係る等電点電気泳動方法は、等電点電気泳動器具1のゲル2にサンプルを導入する導入工程と、第一の電極4および第二の電極3の間に電圧を印加して等電点電気泳動を行う泳動工程と、を包含する。
 サンプルの導入方法は、特に限定されず、公知の方法を用いることができる。なお、ゲル2としてIPGゲルを用いる場合には、サンプルの導入の前に、ゲル2を膨潤させておくことが好ましい。
 また、電圧印加の条件も特に限定されず、例えば、等電点電気泳動方法において一般的に使用される条件を用いればよい。
 なお、一実施形態において、導入工程の後、泳動工程の前に、第一の電極4および第二の電極3の間に電圧を印加した後に、溶媒保持部6を等電点電気泳動器具1から取り外す予備泳動工程をさらに包含してもよい。これにより、予備泳動工程において、イオン性夾雑物が溶媒に溶解した後に、溶媒保持部6を取り外して、等電点電気泳動器具1内から、イオン性夾雑物を除去することができる。これにより、イオン性夾雑物の等電点電気泳動に対する影響をさらに抑制して、よりクリアな泳動結果を得ることができる。
 特に、第一の電極4および第二の電極3の間に電圧を印加した後において第一の電極4および第二の電極3の間に流れる電流値が減少した後に、溶媒保持部6を等電点電気泳動器具1から取り外すことが好ましい。第一の電極4および第二の電極3の間に流れる電流値は、ゲル2中のイオン性夾雑物の量の指標となる。そのため、第一の電極4および第二の電極3の間に流れる電流値が減少し、ゲル2中に存在したイオン性夾雑物の多くが溶媒中に溶解した後に、溶媒保持部6を取り外すようにすることで、等電点電気泳動器具1内から、イオン性夾雑物を十分に除去することができる。これにより、イオン性夾雑物の等電点電気泳動に対する影響をさらに抑制して、よりクリアな泳動結果を得ることができる。
 なお、予備泳動工程において、溶媒保持部6に加えて、または、溶媒保持部6の代わりに溶媒保持部5を取り外すようにしてもよい。
 (変形例)
 なお、本実施形態では、第一の電極4および第二の電極3のそれぞれに溶媒を接触させる溶媒保持部6および溶媒保持部5が設けられており、第一の電極4および第二の電極3の両方がゲル2に接触しているが、本発明はこれに限定されず、溶媒保持部6および溶媒保持部5のうちの一方は、省略してもよい。
 〔実施形態2〕
 本発明の他の実施形態について、図3に基づいて説明すれば、以下のとおりである。なお、説明の便宜上、前記実施形態にて説明した部材と同じ機能を有する部材については、同じ符号を付記し、その説明を省略する。
 図3に示すように、溶媒保持部6の一部が、ゲル2のpH勾配の方向に垂直かつ第一の電極4における第二の電極3側の外縁を含む平面Aに対して、第二の電極3側の領域内にも配置されていてもよく、平面Aに対して、第二の電極3側の領域内においても一部の溶媒を保持しているものであってもよい。すなわち、別の観点からみれば、第一の電極4、および、溶媒保持部6における第二の電極側3の外縁は同一平面上になくともよい。
 このような構成であっても、ゲル2に接触している第一の電極4近傍に移動したイオン性夾雑物を溶媒に拡散させて、第一の電極4にイオン性夾雑物が堆積することを防止することができる。これにより、イオン性夾雑物が等電点電気泳動に及ぼす影響を抑制して、クリアな泳動結果を得ることができる。
 また、同様に、溶媒保持部5の一部が、ゲル2のpH勾配の方向に垂直かつ第二の電極3における第一の電極4側の外縁を含む平面に対して、第一の電極4側の領域内にも配置されていてもよい。
 〔実施形態3〕
 本発明のさらに他の実施形態について、図4に基づいて説明すれば、以下のとおりである。なお、説明の便宜上、前記実施形態にて説明した部材と同じ機能を有する部材については、同じ符号を付記し、その説明を省略する。
 図4に示すように、第一の電極4は、溶媒保持部6に埋め込まれていてもよい。すなわち、第一の電極4は、ゲル2および溶媒保持部6の両方に接触していればよく、ゲル2と溶媒保持部6とに挟まれている必要は無い。
 このような構成であっても、ゲル2に接触している第一の電極4近傍に移動したイオン性夾雑物を溶媒に拡散させて、第一の電極4にイオン性夾雑物が堆積することを防止することができる。これにより、イオン性夾雑物が等電点電気泳動に及ぼす影響を抑制して、クリアな泳動結果を得ることができる。
 なお、第二の電極3も、同様に、溶媒保持部5に埋め込まれていてもよい。すなわち、第二の電極3は、ゲル2および溶媒保持部5の両方に接触していればよく、ゲル2と溶媒保持部5とに挟まれている必要は無い。
 (まとめ)
 以上の様に、本発明の態様1に係る等電点電気泳動器具は、特定の方向に沿ってpH勾配が形成されているゲルと、該ゲルに対し、該特定の方向に沿って電圧を印加する第一および第二の電極と、を備え、第一の電極は、該ゲルに接触しており、溶媒を保持するとともに、該溶媒を第一の電極および該ゲルに接触させる溶媒保持部をさらに備えている。
 上記の構成によれば、第一の電極はゲルに接触しており、溶媒保持部が溶媒を第一の電極およびゲルに接触させている。これにより、ゲルに接触している第一の電極近傍に移動したイオン性夾雑物を当該溶媒に拡散させて、第一の電極にイオン性夾雑物が堆積することを防止することができる。これにより、イオン性夾雑物が等電点電気泳動に及ぼす影響を抑制して、クリアな泳動結果を得ることができる。
 本発明の態様2に係る等電点電気泳動器具では、上記態様1において、上記溶媒保持部は、上記特定の方向に垂直かつ第一の電極における第二の電極側の外縁を含む平面に対して第二の電極とは反対側の領域内において上記溶媒を保持していてもよい。
 上記の構成によれば、イオン性夾雑物を溶解する溶媒は、第一および第二の電極の電極間外において保持される。これにより、溶媒中に溶解したイオン性夾雑物が、再度、電極方向に移動することを抑制し、イオン性夾雑物が等電点電気泳動に及ぼす影響をさらに抑制することができる。また、第一および第二の電極の間に、ろ紙が接して等電位になるゲル部がないため、分離範囲が狭くなることがない。
 本発明の態様3に係る等電点電気泳動器具では、上記態様1または2において、第一の電極および上記溶媒保持部における第二の電極側の外縁が同一平面上にあってもよい。
 上記の構成によれば、イオン性夾雑物を溶解する溶媒は、第一および第二の電極の電極間から完全に排除される。これにより、溶媒中に溶解したイオン性夾雑物が、再度、電極方向に移動することを抑制し、イオン性夾雑物が等電点電気泳動に及ぼす影響をさらに抑制することができる。
 本発明の態様4に係る等電点電気泳動器具では、上記態様1~3において、上記ゲルと上記溶媒保持部とで第一の電極を挟んでいてもよい。
 上記の構成によれば、第一の電極近傍に移動したイオン性夾雑物を、溶媒保持部が保持する溶媒に首尾よく拡散させることができる。
 本発明の態様5に係る等電点電気泳動器具では、上記態様1~4において、上記溶媒保持部における第一の電極および上記ゲルに対向する面には、上記溶媒を保持するための溝が形成されていてもよい。
 上記の構成によれば、溶媒保持部は、溶媒を首尾よく保持することができる。
 本発明の態様6に係る等電点電気泳動器具では、上記態様5において、上記溝が、複数の微細な溝から構成されていてもよい。
 上記の構成によれば、溶媒が微細な溝中に保持されていることによって、毛管現象によって溶媒保持部からゲルへ溶媒が移動してゲルが膨らむことを抑制することができる。
 本発明の態様7に係る等電点電気泳動器具では、上記態様1~4において、上記溶媒保持部における第一の電極および上記ゲルに対向する面には、上記溶液を保持する多孔質体が設けられていてもよい。
 上記の構成によれば、溶媒が多孔質体中に保持されていることによって、毛管現象によって溶媒保持部からゲルへ溶媒が移動してゲルが膨らむことを抑制することができる。
 本発明の態様8に係る等電点電気泳動器具では、上記態様7において、上記多孔質体が、ろ紙であってもよい。
 上記の構成によれば、簡易かつ安価に溶媒保持部を構成することができる。
 本発明の態様9に係る等電点電気泳動器具では、上記態様1~8において、上記溶媒保持部は、当該等電点電気泳動器具から取り外し可能であってもよい。
 上記の構成によれば、溶媒保持部を、等電点電気泳動器具から取り外すことができるため、イオン性夾雑物が十分に溶媒に溶解した後に、溶媒保持部を取り外して、等電点電気泳動器具内から、イオン性夾雑物を除去することができる。これにより、イオン性夾雑物の等電点電気泳動に対する影響をさらに抑制して、よりクリアな泳動結果を得ることができる。
 本発明の態様10に係る等電点電気泳動器具では、上記態様1~9において、上記ゲルは、長手方向が上記特定の方向である板状のゲルであり、第一の電極は、上記ゲルの板面に接触していてもよい。
 上記の構成によれば、ゲルとして、市販のIPGゲルを首尾よく用いることができる。
 本発明の態様11に係る等電点電気泳動方法では、態様1~10の等電点電気泳動器具における上記ゲルにサンプルを導入する導入工程と、第一および第二の電極間に電圧を印加して等電点電気泳動を行う泳動工程と、を包含する。
 上記の構成によれば、態様1~10の等電点電気泳動器具と同等の効果を奏する。
 本発明の態様12に係る等電点電気泳動器具では、上記態様11において、上記導入工程の後、上記泳動工程の前に、第一および第二の電極間に電圧を印加した後に上記溶媒保持部を上記等電点電気泳動器具から取り外す予備泳動工程をさらに包含してもよい。
 上記の構成によれば、予備泳動工程において、イオン性夾雑物が溶媒に溶解した後に、溶媒保持部を取り外して、等電点電気泳動器具内から、イオン性夾雑物を除去することができる。これにより、イオン性夾雑物の等電点電気泳動に対する影響をさらに抑制して、よりクリアな泳動結果を得ることができる。
 本発明の態様13に係る等電点電気泳動器具では、上記態様12において、上記予備泳動工程では、第一および第二の電極間に所定の電圧を印加した状態において第一および第二の電極間に流れる電流値が減少した後に、上記溶媒保持部を上記等電点電気泳動器具から取り外してもよい。
 上記の構成によれば、第一および第二の電極間に流れる電流値は、ゲル中のイオン性夾雑物の量の指標となる。そのため、第一および第二の電極間に流れる電流値が減少し、ゲル中に存在したイオン性夾雑物の多くが溶媒中に溶解した後に、溶媒保持部を取り外すようにすることで、等電点電気泳動器具内から、イオン性夾雑物を十分に除去することができる。これにより、イオン性夾雑物の等電点電気泳動に対する影響をさらに抑制して、よりクリアな泳動結果を得ることができる。
 本発明は上述した各実施形態に限定されるものではなく、請求項に示した範囲で種々の変更が可能であり、異なる実施形態にそれぞれ開示された技術的手段を適宜組み合わせて得られる実施形態についても本発明の技術的範囲に含まれる。さらに、各実施形態にそれぞれ開示された技術的手段を組み合わせることにより、新しい技術的特徴を形成することができる。
 (実施例1)
 図1に示すような等電点電気泳動器具1を用いて等電点電気泳動を行った。ゲル2としては、pH4~7の範囲でpH勾配が形成されている膨潤されたIPGゲルを用いた。サンプルとしては、IC-5によって標識されたMLL(マウス肝臓サンプル)2μgを尿素、チオ尿素、3-(3-cholamidepropyl)dimethylammonio-1-propanesulphonate(CHAPS)、ジチオトレイトール(DTT)、両性電解質(Ampholyte)を含む溶液に、塩として80mM NaClを加えた溶解させたものを用いた。溶媒保持部5、6としては、蒸留水によって湿らせたろ紙を用いた。
 等電点電気泳動では、図5の折れ線5に示す電圧を第一の電極4と第二の電極3との間に印加した。すなわち、ステップ1として、5分間、200Vの定電圧を印加した。続いて、ステップ2として、5分間、1000Vまで非線形に変化させながら電圧を印加した。続いて、ステップ3として、5分間、1000Vの定電圧を印加した。続いて、ステップ4として、10分間、7000Vまで線形に変化させながら電圧を印加した。続いて、ステップ5として、10分間、7000Vの定電圧を印加した。
 次いで、等電点電気泳動が行われたゲル2をSDS平衡化し、10%ポリアクリルアミドゲルを用いて二次元目の電気泳動を行った。得られたゲルを蛍光イメージスキャナー(Tyhoon:GE ヘスルケア社)で撮像し、スポットを可視化した。
 (比較例1)
 図7の(c)に示すような等電点電気泳動器具10を用いたこと以外は、実施例1と同様に等電点電気泳動を行った。次いで、実施例1と同様に二次元目の電気泳動および撮像を行った。
 (実施例2)
 実施例1において、ステップ1の前に予備泳動を行ったこと以外は、実施例1と同様に等電点電気泳動を行った。予備泳動では、ステップAとして、5分間、200Vの定電圧を印加した。続いて、ステップBとして、5分間、1000Vまで非線形に変化させながら電圧を印加した。続いて、ステップCとして、5分間、1000Vの定電圧を印加した。その後、溶媒保持部5、6を等電点電気泳動器具1から取り外し、ステップ1~5を行った。
 次いで、実施例1と同様に二次元目の電気泳動および撮像を行った。
 (結果)
 図5は、実施例1、2および比較例1に係る等電点電気泳動における電流値の変化を比較するグラフである。折れ線1は、実施例1における電流値の変化を示す。折れ線2は、比較例1における電流値の変化を示す。折れ線3は、実施例2のステップ1~5における電流値の変化を示す。折れ線4は、実施例2のステップA~Cにおける電流値の変化を示す。折れ線5は、実施例1、2および比較例1のステップ1~5における電圧の変化を示す。
 図6の(a)は、比較例1における二次元電気泳動の結果の写真を示す。図6の(b)は、実施例1における二次元電気泳動の結果の写真を示す。図6の(c)は、実施例2における二次元電気泳動の結果の写真を示す。
 図6の(a)と(b)とを比較すれば分かるように、実施例1では、比較例1に比べて、スポットの分解能(集束)が格段に向上しており、よりクリアな結果が得られた。
 図6の(b)と(c)とを比較すれば分かるように、実施例2では、実施例1に比べて、スポットの分解能(集束)がさらに向上しており、よりクリアな結果が得られた。また、折れ線1と折れ線3とを比較すれば分かるように、実施例2では、実施例1とは異なり、泳動初期~中期(ステップ1~3)において殆ど電流が流れていなかった。これは、実施例2の予備泳動(ステップA~C 折れ線4)が、電流値が0に近いところまで減少してから終了していることから、予備泳動において、ゲル2中のイオン性夾雑物の大部分が、溶媒内に溶解して除去されたことを示している。
 本発明は、生体分子等の分析機器の製造分野および生体分子等の分析分野において利用可能である。

Claims (13)

  1.  特定の方向に沿ってpH勾配が形成されているゲルと、
     該ゲルに対し、該特定の方向に沿って電圧を印加する第一および第二の電極と、を備え、
     第一の電極は、該ゲルに接触しており、
     溶媒を保持するとともに、該溶媒を第一の電極および該ゲルに接触させる溶媒保持部をさらに備えていることを特徴とする等電点電気泳動器具。
  2.  上記溶媒保持部は、上記特定の方向に垂直かつ第一の電極における第二の電極側の外縁を含む平面に対して第二の電極とは反対側の領域内において上記溶媒を保持していることを特徴とする請求項1に記載の等電点電気泳動器具。
  3.  第一の電極および上記溶媒保持部における第二の電極側の外縁が同一平面上にあることを特徴とする請求項1または2に記載の等電点電気泳動器具。
  4.  上記ゲルと上記溶媒保持部とで第一の電極を挟んでいることを特徴とする請求項1~3の何れか一項に記載の等電点電気泳動器具。
  5.  上記溶媒保持部における第一の電極および上記ゲルに対向する面には、上記溶媒を保持するための溝が形成されていることを特徴とする請求項1~4の何れか一項に記載の等電点電気泳動器具。
  6.  上記溝が、複数の微細な溝から構成されることを特徴とする請求項5に記載の等電点電気泳動器具。
  7.  上記溶媒保持部における第一の電極および上記ゲルに対向する面には、上記溶媒を保持する多孔質体が設けられていることを特徴とする請求項1~4の何れか一項に記載の等電点電気泳動器具。
  8.  上記多孔質体が、ろ紙であることを特徴とする請求項7に記載の等電点電気泳動器具。
  9.  上記溶媒保持部は、当該等電点電気泳動器具から取り外し可能であることを特徴とする請求項1~8の何れか一項に記載の等電点電気泳動器具。
  10.  上記ゲルは、長手方向が上記特定の方向である板状のゲルであり、
     第一の電極は、上記ゲルの板面に接触していることを特徴とする請求項1~9の何れか一項に記載の等電点電気泳動器具。
  11.  請求項1~10の何れか一項に記載の等電点電気泳動器具における上記ゲルにサンプルを導入する導入工程と、
     第一および第二の電極間に電圧を印加して等電点電気泳動を行う泳動工程と、を包含することを特徴とする等電点電気泳動方法。
  12.  上記導入工程の後、上記泳動工程の前に、第一および第二の電極間に電圧を印加した後に上記溶媒保持部を上記等電点電気泳動器具から取り外す予備泳動工程をさらに包含することを特徴とする請求項11に記載の等電点電気泳動方法。
  13.  上記予備泳動工程では、第一および第二の電極間に所定の電圧を印加した状態において第一および第二の電極間に流れる電流値が減少した後に、上記溶媒保持部を上記等電点電気泳動器具から取り外すことを特徴とする請求項12に記載の等電点電気泳動方法。
