Abstract
본 발명은 리오바이러스(reovirus)를 유효성분으로 함유하는 혈관신생 억제용 조성물에 관한 것으로, 구체적으로 리오바이러스(reovirus)는 암세포주에 의한 혈관 내피세포의 파괴 마이크로 유체칩 모델에서 리오바이러스가 혈관내피세포의 세포조직층을 파괴하는 효과를 나타내며, 자연 발생하도록 유도된 종양에 대하여 유의적인 혈관 신생 억제효과를 나타내므로, 상기 리오바이러스는 혈관신생 억제용 약학적 조성물 및 건강식품용 조성물의 유효성분으로 유용하게 이용될 수 있다.

리오바이러스를 유효성분으로 함유하는 혈관신생 억제용 조성물 【기술분야】
본 발명은 리오바이러스 (reovirus)를 유효성분으로 함유하는 혈관신생 억제용 약학적 조성물, 및 혈관신생 관련질환 예방 및 개선용 건강식품에 관한 것이다. 【배경기술】
야생형 종양파괴 바이러스 (wild type oncolytic virus)는 내재적으로 강력한 종양파괴능력을 가지는 바이러스로 (Roberts et al. , 2006; Liu et al . , 2007; Bluming et al. , 1971), 일부 야생형 종양파괴 바이러스는 세포 내 항ᅳ바이러스 면역이 결핍 또는 약화된 암세포를 특이적으로 감염 및 증식함을 통해 종양 세포의 파괴를 유도하는 것으로 알려져 있다 (Kim et al. , 2010; Parato et al., 2005) .
야생형 종양파괴 바이러스를 이용한 암 치료의 연구는 기존의 치료 유전자를 바이러스 내에 삽입하여 치료유전자의 발현에 의한 종양살상 효과를 목표로 하는 유전자 치료 (gene therapy)와 구별되며, 자연 감염된 여러 종류의 바이러스로 인한 자연적 암의 치료는 오래전부터 보고되고 있으나, 본격적으로 야생형 종양파괴 바이러스에 의한 종양 특이적 살상 기작에 관한 연구가 진행되고 있는 대표적인 예로는 oncolyticbiotech사에서 임상 3상의 적용에까지 이른 야생형 리오바이러스 (Reovirus)를 이용한 암 치료 연구가 있다.
야생형 종양파괴 바이러스를 이용한 암 치료의 연구는 자연계에 이미 존재하는 바이러스를 사용하기 때문에 우수한 안전성을 가져 임상 적용이 빠르게 진행된다는 장점올 가진다. 또한 암세포만을 선택적으로 증식 및 세포사멸을 유도하므로, 최근 다양한 종류의 야생형 종양살상 바이러스를
이용한 전임상 및 임상연구가 활발하게 진행되고 있다 (Roberts et al. , 2006) . 현재 한국을 포함하여 미국, 유럽 등에서 10여 개가 넘는 다양한 항암 바이러스가 임상시험중에 있다. 리오바이러스는 2011년 현재 3개국 (미국, 영국, 벨기에)에서 임상 3상이 진행 중이며, 그 외에도 Wellstat Biologies사에서 가금류 바이러스의 일종인 뉴캐슬병 바이러스 (Newcastle disease virus)를 이용한 임상 1상의 적용을 마쳤고, 콕사키 바이러스 (Coxsackie virus)(Viralytics사) 및 폭스바이러스 (Poxvirus)의 일종인 백시니아 바이러스 (Vaccinia virus) (Generex 및 Genelux사)는 임상 2상의 적용을 마친 상태이다. 특히, 상기 바이러스의 대부분은 임상적용시험에서 심각한 독성이 확인되지 않았으며, 최대허용용량 (maximum tolerated dose; MTD)이 설정되지 않아, 높은 수준의 안정성을 보인다 (Roberts et al., 2006; Liu et al . , 2007)., 따라서, 항암 바이러스를 임상에서 광범위하게 사용하기 위해서는 항암 바이러스의 면역능 증가에 대웅하여 면역학적 방어를 최소화할 수 있는 제형 연구, 기존 항암제 등과의 병용 요법 연구, 안정성 연구 등이 필수적으로 진행되어야 한다. 리오바이러스 (Respiratory Enteric Orphan Virus! Reovirus)는 전체 게놈이 10 개의 이중 -가닥 RNA 단편으로 구성되는 비 -외피 바이러스로ᅳ 일반적인 환경에 편재하며 (ubiquitous), 면역 기능을 가지는 숙주에서 증상을 보이지 않는 무증후성 바이러스이다. 사람에게 감염성이 약하여 감염 부위는 상부 호흡기 및 위장관에 국한되는 것으로 알려져 있다 (Tylerᅳ 2001).
리오바이러스는 특정 타입의 형질 변환된 세포에 감염되었을 때, 강력한 세포파괴 (cytocidal) 활성을 나타내는 것으로 오래전부터 알려져 있어 (Duncan et al, 1978; Hashiro et al , 1977), 이를 활용하기 위한 다양한 연구가 진행되고 있다. 야생형 리오바이러스에 감염된 건강한 숙주에서는 증상이 나타나지 않으나, 숙주의 면역체계가 손상된 경우 위험도가 증가하게 되므로, 암 환자와 같은 환자에서 리오바이러스의 임상적 가능성이 제한될 수 있다. 그러나, 리오바이러스의 종양파괴 활성에 대한 연구가 계속 되면서 종양을 유발하는
Ras-신호전달 경로를 포함하는 세포가 실험실 내 (in vitro) 및 생체 내 (in vivo)에서 리오바이러스 (3형 Dearing 주) 감염에 민감한 것이 보고되면서 (Coffey et al, 1998; Strong et al, 1998; Norman et al , 2004) , 면역 체계 손상의 제한을 가지지 않는 암 환자를 대상으로 하는 리오바이러스 요법이 현재 임상에서 사용되고 있다 (Norman etal, 2005) . 신생혈관의 유도현상은 기존의 혈관으로부터 새로운 혈관이 형성되는 과정으로, 정상적인 발생, 즉 태아의 발생 (embryonic development), 생식주기 (female reproductive cycle)(D. M. Noden , Am. Rev. Respir . Dis. 140, 1097-1103(1989)), 성장 그리고 상처 치유 시 필수적일 뿐 아니라, 각종 암, 관절염, 당뇨병성 망막증, 죽상경화증 등의 질병과도 깊은 관련을 맺고 있다 (O'Brien, E. R. et al. , AM. J. Pathol. 145, 883-894(1994)). 특히 악성종양으로 발전하는 과정 중의 하나인 암의 전이 시 생혈관 유도현상은 필수적인 과정으로 여겨진다.
따라서, 신생 혈관의 형성을 억제함을 통해 암의 성장 기전을 밝히고, 암의 조기진단, 예방 및 치료에 사용할 수 있을 뿐만 아니라, 관련 질병인 류마티스성 관절염, 만성염증, 당뇨병성 망막증, 혈관종 등의 질병 치료에도 광범위하게 사용될 수 있으므로, 지속적인 연구 및 개발이 계속되고 있으며 (W.Auerbach et al ., Pharnac . Ther . , 61, 265-311(1994)), 몇 가지의 억제제는 활성검증을 위해 임상 시험이 진행 중에 있다 (R. Bicknel et al., TiPS, 16, 57(1995)).
FVB/NJ 쥐는 다른 마우스종에 비해 화학물질에 의해 유도돠는 피부암의 발생율이 현저하게 높은 것으로 알려져 있다. FVB/NJ 쥐에 7,12-디메틸벤조 [ α]안트라센 (7,12-dimethylbenz[ a]anthracene; DMBA) 100 및 포르볼 -12-미리스테이트 -13-아세테이트 (Phorbol -12-myr i st at e_13— acetate; TPA) 5 //g을 피부에 처리하였을 때 마우스당 11.8개의 유두종 (papillomas)이 생성되었고, 상기 유두종이 악성종양인 편평상피암 (Squamous Cell Carcinomas;
sec)로 악성화되는 비율이 다른 마우스 종에 비해 현저히 높은 것으로 보고되어 있다 1" 10 61 3 14:11, 2353-2358) . 이에, 본 발명자들은 새로운 혈관신생 억제 활성을 갖는 바이러스를 찾기 위해 노력한 결과, 암세포주에 의한 혈관 내피세포의 파괴 마이크로 유체칩 모델에서 리오바이러스가 혈관내피세포의 세포조직층을 파괴하는 효과를 나타내며, 화학물질을 이용하여 마우스의 피부에 자연 발생하도록 유도된 종양에 대하여, 리오바이러스의 현저한 혈관 신생 억제효과를 확인함으로써, 상기 리오바이러스를 혈관신생 억제용 조성물의 유효성분으로 사용될 수 있음을 확인함으로써, 본 발명을 완성하였다.
【발명의 상세한 설명】
【기술적 과제]
본 발명의 목적은 리오바이러스 (reovirus)를 유효성분으로 함유하는 혈관신생 억제용 약학적 조성물, 및 혈관신생 관련질환 예방 및 개선용 건강식품을 제공하는 것이다.
【기술적 해결방법】
상기 목적올 달성하기 위하여, 본 발명은 리오바이러스 (reovirus)를 유효성분으로 함유하는 혈관신생 억제용 약학적 조성물을 제공한다.
또한, 본 발명은 혈관신생 억제용 외래 유전자를 추가적으로 리오바이러스를 유효성분으로 함유하는 혈관신생 억제용 약학적 조성물을 제공한다.
또한, 본 발명은 리오바이러스를 유효성분으로 함유하는 혈관신생 관련질환 예방 및 개선용 건강식품을 제공한다.
또한, 본 발명은 리오바이러스과에 속하는 리오바이러스를 유효성분으로 함유하는 약학적 조성물을 필요로 하는 개체에 투여하는 단계를 포함하는 혈관신생 억제 방법을 제공한다.
또한, 본 발명은 혈관신생 억제용 약학적 조성물로 사용하기 위한 리오바이러스과에 속하는 리오바이러스의 용도를 제공한다 .

[유리한 효과]
본 발명은 리오바이러스 (reovirus)는 자연 발생하도록 유도된 종양에 대하여, 유의적인 혈관 신생 억제효과를 나타내므로, 혈관신생 억제용 약학적 조성물 및 건강식품용 조성물의 유효성분으로 유용하게 이용될 수 있다.

도 1 및 도 2는 유방암 세포주인 MCF7가 자라면서 혈관내피세포층 (Vascular endothelial cell layer)을 파괴 (Invasion)하는 마이크로 유체칩 모델에서, 리오바이러스에 의한 암세포주 생장 억제 및 혈관내피세포 조직층 파괴 효과를 나타내는 도이다.
도 3은 FVB/NJ 마우스에 포르볼 -12-미리스테이트 -13-아세테이트 (Phor bo 1-12— myri stat e-13-acet ate; TPA)를 도포하여 6 주 후 유도된 유두종 (Papilloma) 조직을 나타내는 도이다. 도 4는 FVB/NJ 마우스에 TPA를 도포하여 10 주 후 성장한 유두종 조직을 나타내는 도이다.

【발명의 실시를 위한 최선의 형태】
이하, 본 발명을 상세히 설명한다. 본 발명은 리오바이러스 (reovirus)를 유효성분으로 함유하는 혈관신생 억제용 약학적 조성물을 제공한다.
또한, 본 발명은 리오바이러스과에 속하는 리오바이러스를 유효성분으로 함유하는 약학적 조성물을 필요로 하는 개체에 투여하는 단채를 포함하는 혈관신생 억제 방법을 제공한다.
또한, 본 발명은 혈관신생 억제용 약학적 조성물로 사용하기 위한 리오바이러스과에 속하는 리오바이러스의 용도를 제공한다.
상기 리오바이러스는 야생형 (wild type) 바이러스인 것이 바람직하나 이에 한정되지 않는다.
상기 리오바이러스는 임의의 리오바이러스로부터 유도될 수 있으며 , 다양한 소스로부터 수득될 수 있는 리오바이러스과 (Reoviridea family)의 구성원인 것이 바람직하고, 혈청 타입 l(Lang), 타입 2( Jones) 또는 타입 3(Dealing 및 Aneny) 리오바이러스인 것이 보다 바람직하나 이에 한정되지 않는다. 또한, 상기 리오바이러스는 인간을 제외한 영장류 (챔팬지, 고릴라, 짧은 꼬리 원숭이, 원숭이 등), 설치류 (생쥐, 쥐, 게리빌스쥐, 햄스터, 토끼, 기나아피그 등), 개, 고양이, 일반 가축 (소, 말, 돼지, 염소)을 포함하는 기타 포유류 종의 세포에 대해 향성 (tropism)을 나타내는 하나 또는 그 이상의 리오바이러스로 부터 유도될 수 있으나, 이에 한정되지 않는다.
상기 약학적 조성물은 혈관신생에 의해 유발되는 질환을 예방 또는 치료할 수 있는 것이 바람직하고,상기 질환은 암, 혈관종, 혈관섬유종, 관절염, 당뇨병성 망막증 (diabetic ret ionpathy), 조숙아의 망막증, 신생혈관성 녹내장, 신생혈관에 의한 각막질환, 퇴화반 /황반변성 (macular degeneration), 반점의 변성, 익상편, 망막변성, 후수정체 섬유증식증, 과립성 결막염, 건선ᅵ 모세관 확장증, 화농성 육아종, 건선, 지루성 피부염 및 여드름으로 이루어지는 군으로부터 선택되는 어느 하나 이상의 질환인 것이 바람직하며, 상기 암은
간암, 위암, 결장암, 유방암, 폐암, 비소세포성폐암, 골암, 췌장암, 피부암, 두부 또는 경부암, 피부 또는 안구 내 혹색종, 자궁경부암, 난소암, 대장암, 소장암, 직장암 ,항문부근암,나팔관암종,자궁내막암종,자궁경부암종,질암종,음문암종, 호지킨병 (Hodgkin's disease), 식도암, 소장암, 임파선암, 방광암, 담낭암, 내분비선암, 갑상선암, 부갑상선암, 부신암, 연조직 육종, 요도암, 음경암, 전립선암, 만성 또는 급성 백혈병, 림프구 림프종, 방광암,신장 또는 수뇨관 암, 신장세포 암종, 신장골반 암종, 중추신경계 (CNS; central nervous system)종양, 1차 CNS 림프종, 척수 종양, 뇌간 신경교종 및 뇌하수체 선종으로 이루어지는 군으로부터 선택되는 어느 하나 이상인 것이 바람직하나 이에 한정되지 않는다.
상기 리오바이러스는 추가적으로 외래 유전자 (heterologous gene)를 리오바이러스 유전자 내 복제필요 유전자부위 (essential region) 또는 복제불필요 유전자부위 (non-essential region)에 포함할 수 있다. 상기 외래 유전자 (heterologous gene)라는 용어는 바이러스 게놈 (viral genome)에서 발견되지 않는 어떤 유전자를 받아들인다는 의미이다. 외래유전자는 야생형 유전자의 대립형질 변이체 (allelic variant)이거나 돌연변이 유전자일 수 있다. 외래유전자는 생체 내 조건의 세포에서 전기 외래유전자를 발현하게 하는 조절서열 (control sequence)에 작동 가능하도톡 연결되는 것이 바람직하다. 따라서, 본 발명의 바이러스는 외래유전자가 발현될 수 있는 생체 내 조건의 세포에 외래유전자 /유전자들을 전달하도록 사용될 수 있다.
상기 외래 유전자는 항-혈관신생 유전자, 종양 억제인자 유전자, 항원성 유전자, 세포독성 유전자, 세포증식억제 (cytostatic) 유전자, 자살 유전자, 항-혈관및 면역 -조절 유전자인 것이 바람직하다.

상기 "항원성 유전자"는 표적 세포에서 그의 발현이 면역 시스템에 의해 인지될 수 있는 세포 표면 항원 단백질의 생성올 야기하는 뉴클레오티드 서열이다ᅳ 상기 항원성 유전자의 예로는, 암태아성 항원 (CEA), CD3, CD133, CD44 및 p53(국제 특허 공개 TO94/02167호)이 포함된다. 면역 시스템에 의한 용이한 인지를 위해, 항원성 유전자는腿 C I형 항원과 결합될 수 있다.

바이러스 티미딘 키나제, 수두 티미딘 키나제, 사이토신 디아미나제, 퓨린 뉴클레오시드 포스포릴라제, 베타 -락타나제, 카복시펩티다제 G2, 사이토크롬 P450-2B1, 니트 로리덕타제, 베타—글루쿠로니다제, TRAIL TNF 관련 세포자멸 -유도 리간드) 등이 포함되나 이에 한정되지 않는다.
상기 "면역 -조절 유전자 "는 세포에서 발현될 때 체액 및 세포성 면역 반응을 조절하는 뉴클레오티드 서열이다. 면역 -조절 유전자는 CD16, CTLA-4, IL24, GM-CSF 등을 암호화하는 유전자가 포함되나 이에 한정되지 않는다.
상기 리오바이러스는 바이러스 운반용 세포 (Virus carrier cells: Molecular Therapy. 2009 Vol 17 :1667-1676 에 탑재될 수 있으며, 이를 통해 생체 내에서 치료효과를 더욱 상승시킬 수 있으나 이에 한정되지 않는다. 본 발명의 구체적인 실시예에서, 시험관 내 (In vitro)에서 리오바이러스의 혈관 생성 저해 효과를 확인하기 위해, 유방암 세포주인 MCF7가 자라면서 혈관내피세포층 (Vascular endothelial cell layer)을 파괴 (Invasion)하는 마이크로 유체칩 모델을 제조하고, 리오바이러스를 감염시켜 배양한 결과, 리오바이러스가 혈관내피세포층이 거의 사라지면서 MCF7 세포주의 생장 역시 억제하는 것을 확인하였다 (도 1 및 도 2 참조).
또한, 본 발명자들은 생체 내 (In vivo)에서 리오바이러스의 혈관 생성 저해 효과를 확인하기 위해, FVB/NJ 마우스에 7,12-디메틸벤조[ (1 ]안트라센(7,12—(1^ 1 11 112[ 01 ]3^^^61½; DMBA) 및 포르볼 -12-미리스테이트 -13-아세테이트 (Phorbol-12-myr i state_13— acetate; TPA)로 유두종 (papilloma)을 유도하고 (도 3 및 도 4 참조), 리오바이러스를 동물실험용으로 정제한 다음, 유두종 성장 억제능을 확인하기 위하여 유두종이 유도된 마우스에서 리오바이러스를 투여하여 유두종 조직의 성장을 확인한 결과, 리오바이러스를 투여한 경우, 대조군에 비교하였을 때 유두종의 성장에는 큰 차이가 없는 것을 확인하였다 (도 5 참조).
또한, 리오바이러스의 신생혈관 형성 억제능을 확인하기 위하여, 유두종 조직의 뒷면을 관찰하여 신생혈관 형성을 확인한 결과, 리오바이러스를 투여한
경우, 대조군에 비해 혈관의 형성이 유의적으로 감소 또는 둔화되는 것을 확인하였다 (도 6 참조).

본 발명의 조성물에서 사용될 수 있는 붕해제는 정제가 수성 환경에 노출되었을때 붕해되도록 하기 위하여 사용된다. 너무 많은 붕해제를 포함한 정제는 보관 중에 붕해될 수 있는 반면, 너무 적게 포함한 정제는 바람직한 조건 하에서 바람직한 속도로 붕해되지 않는다. 따라서 활성 성분의 방출을 조절하는데 나쁘지 않도록 너무 많지도 너무 적지도 않은 충분한 붕해제의

상기 유효량은 바이러스의 약동학적 특성, 투여 방법, 연령, 환자의 건강상태 및 체중, 질병 상태의 특성 및 범위, 치료 횟수 및 최근의 치료 형태와 같은 수많은 인자에 의해 달라질 수 있으며 , 가령 바이러스의 병독성 및 역가에 따라서도 달라질 수 있다. 당업자라면 상기 인자에 기초하여 적정량을 조절할 수 있다. 바이러스는 처음에 환자의 임상적 반응에 의존해 필요에 따라 적정량으로 투여될 수 있다. 바이러스의 유효량은 경험적으로 결정할 수 있으며 안전하게 투여될 수 있는 바이러스의 최대량 및 바람직한 결과를 유발하는 바이러스의 최소량에 따라 결정할 수 있다.
바이러스를 전신에 투여할 때 , 질병 부위에 바이러스를 주입함으로써 실현되는 것과 유사한 임상적 효과를 유발하기 위해, 상당히 높은 용량의 바이러스 투여가 요구될 수 있다. 그러나, 적당한 용량 레벨은 바람직한 결과를 실현할 수 있는 최소량이 되어야 한다.

상기 리오바이러스는 암세포주에 의한 혈관 내피세포의 파괴 마이크로 유체칩 모델에서 리오바이러스가 혈관내피세포의 세포조직층을 파괴하는. 효과를 나타내며, 자연 발생하도록 유도된 종양에 대하여 유의적인 혈관 신생
억제효과를 나타내므로, 혈관신생 관련질환 예방 및 개선용 건강식품에 유효성분으로 유용하게 사용될 수 있다. 이하, 본 발명을 실시예 및 제조예에 의해 상세히 설명한다.
단, 하가 실시예 및 제조예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 제조예에 한정되는 것은 아니다.
<실시예 1>야생형 및 약독화된 리오바이러스 (reovirus)의 증식 및 수득
본 발명에서 사용할 리오바이러스를 준비하기 위하여, 야생형 리오바이러스를 제조 및 약독화 (attenuation)한 후, 수득하였다.
구체적으로, 야생형 리오바이러스 인간 타입 3(Dealing)를 미국세포주은행 (american type culture collection; ATCC, VR-824)에서 입수하였다. 상기 리오바이러스의 1955 년경에 설사를 일으키는 어린이의 분변에서 최초 분리되어 Albert Sab in 박사에 의해 기증되었다. 야생형 리오바이러스를 증식하기 위해 BHK-21 세포주, HEK 293 또는 L929 세포에 감염다중도 (Multiplicity of infection; MOD 약 1 내지 10로 바이러스를 접종하여 48내지 72시간 동안 37°C에서 배양하였다. 그런 다음, 상기 세포를 동결융해 (Freezing and Thawing)하여 파쇄한 후, 원심분리하여 세포 파괴물 (cell debris)을 제거하고, 유리된 리오바이러스의 역가를 L929 단분자막 (mono layer)를 이용해 용균반검사 (Plaque assay)법을 사용하여 확인하여 영하 80°C에 보관하면서 사용하였다. 수득한 야생형 리오바이러스를 약독화하기 위해 HT108 세포에 접종하였으며, 야생형 리오바이러스의 증식 및 수득과 동일한 방법으로 약독화된 리오바이러스를 증식 및 수득하였다 (Kim Met al, Oncogene 26 :4124-34, 2007; Kim M et al . , British Journal of Cancer . 104, 290-299, 2011).
그 결과, 100 내지 1000 pfu/세포의 역가를 가지는 야생형 및 약독화된 리오바이러스 시료를 수득하였다.
<실시예 2>시험관 내 (In vitro)에서 리오바이러스의 혈관신생 억제효과 확인 <2-1>마이크로 유체칩 모델와 제조
리오바이러스가 나타내는 혈관신생 억제효과를 시험관 내 수준에서 확인하기 위하여, 유방암 종양세포가 자라면서 내피세포층을 침범 (Invasion)하는 마이크로 유체칩 모델을 제조하였다.
구체적으로, 폴리디메틸실록산 (poly-dimethyl siloxane, PDMS; 제품명: Silgard 184; Dow Cheminal사, 미국)소프트 리소그래피 과정 (soft lithography process)를 이용하여 미세유동 칩 (microf luidic chip)을 제조하였다. 제조한 미세유동 칩을 멸균한 후, 유리 커버슬라이드를 플라즈마 처리 (plasma treatment; Femto Science, 한국)하여 접착함으로써 덮힌 (closed) 미세유동 채널을 형성하였다. 접착 후, 제 1형 콜라겐 (Type I collagen; BD Biosciences , 미국)을 형성된 미세유동 채널로 주입하여 채우고, 유방암 세포주인 MCF-7 구형상체 (spheroid)를 미세유동 채널에 접종한 다음, 암 구형상체를 가두기 (trap)위해서 웰의 측면 (side)를 바닥으로 향하도록 수직적으로 칩을 세워 4°C에서 30 분 동안 배양하였다. 암 구형상체를 가둔 후, 칩 내에 주입했던 콜라겐을 제거하여 (aspirated) 혈관 구조 (vascular structure)을 형성하고, 세포외 기질 (Extracellular matrix, ECM)으로 콜라겐을 가교화 (crosslink)하기 위해서 미세유동 칩을 세포 배양기에 두었다. 그런 다음, 내피세포 (endothelial cell)인 HMVEC Lonza사, 미국)을 미세유동 채널에 2X106 세포 의 세포밀도로 접종하여 1 시간 동안 배양하고, 혈관 구조를 구성하기 위해 상기 미세유동 채널을 세포 배양기에 1 시간 동안 두어, 마이크로 유체칩 모델의 제조를 완성하였다. <2-2>마이크로 유체칩 모델에서 리오바이러스의 혈관신생 억제 효과 확인
리오바이러스가 나타내는 혈관신생 억제효과를 시험관 내 수준에서 확인하기 위하여, 제조된 마이크로 유체칩 모델에서 리오바이러스에 의한 암세포 생장 억제효과를 확인하였다.
구체적으로, 상기 실시예 <2-1>에서 제조한 마이크로 유체칩 모델에 상기 <실시예 1〉에서 수득한 리오바이러스를 2X107 PFU 씩 3 일 동안 연속투여하여 배양하고, 4일 째에 리오바이러스에 의한 혈관내피세포층의 파괴정도를 현미경 관찰을 통해 확인하였다. 음성 대조군으로는, 리오바이러스를 투여하지 않은 마이크로 유체칩에 PBS를 3 일 동안 연속투여하였다. PBS 또는 뫼오바이러스의 투여시에는 PBS 또는 리오바이러스를 칩에 투여한 후, 유속을 막아 1 시간 동안 효율적으로 감염이 일어나도록 유도한 후, 유속을 재개하여 배양하였다.
그 결과, 도 1 및 도 2에서 나타난 바와 같이 음성 대조군에서는 MCF7 세포주가 자가 혈관 내피세포층을 침범하여 이를 파괴하는 것을 확인한 반면, 리오바이러스 투여 후 3 일째 (후감염 2 일째) 또는 4 일째 (후감염 3 일째)의 경우, 리오바이러스가 혈관내피세포층이 거의 사라지면서 MCF7 세포주의 생장 역시 억제함으로써, 본 발명의 리오바이러스가 종양의 억제 뿐만 아니라 혈관 생성을 억제하는 효과를 나타내는 것을 확인하였다 (도 1 및 도 2).
<실시예 3> 생체 내 (In vivo)에서 리오바이러스의 혈관신생 억제효과 확인 <3-1>화학물질을 이용한종양의 유도
화학물질을 이용하여 마우스에서 종양을 유도하기 위하여, FVB/NJ 마우스에 7,12-디메틸벤조 [ α]안트라센 (7,12-dimethylbenz[ aknthracene; DMBA) 및 포르블—12-미리스테이트 -13-아세테이트 (Phorbol -12-myr i state— 13-acetate; TPA)로 피부암을 유도하였다.
구체적으로, 7주령의 수컷 FVB/NJ마우스 20마리를 구입하여 등의 털을 제거한 후, 제모로 인한 염증반응이 유도되는지 확인하기 위하여 2 일 동안 관찰하였다. 그런 다음, 일 주일 이내에 25 /g의 DMBA를 FVB/NJ마우스 등에 2 회 도포하고, 일 주일에 2 회씩 최대 10 주 동안 TPA를 도포하여 유두종 (Papilloma)을 유발하였다.
그 결과, 도 3 및 도 4에서 나타난 바와 같이 TPA 도포 6 주 후부터
유두종이 유도되었으며 (도 3), TPA 도포 10 주 후에는 모든 마우스에서 골고루 유두종이 생성된 것을 확인하였다 (도 4).
<실시예 3> 리오바이러스의 유두종 성장 억제능 확인
리오바이러스의 유두종 성장 억제능을 확인하기 위하여 유두종이 유도된 마우스에서 리오바이러스를 투여하여 유두종 조직의 성장을 확인하였다.
구체적으로, 상기 <실시예 2>에서 유두종을 유도한 FVB/NJ 마우스 20 마리를 10 마리씩 실험군 및 대조군으로 나누고, 유두종 성장기간을 위해 10 주간 사육하였고, 상기 <실시예 1〉에서 증식 및 수득한 리오바이러스를 실험군의 유두종 조직에 3일 연속으로 종양 당 ixi09pfu씩 투여한 후, 약 한 달 동안 유두종 조직의 성장을 관찰한 다음, 희생하여 유두종 조직을 분리하였다. 리오바이러스의 유두종 성장 억제능을 비교하기 위하여,
, 대조군에는 리오바이러스를 투여하지 않고 실험군과 동일한 환경에서 유두종 조직의 성장을 관찰한 후, 회생하여 유두종 조직을 분리하여 실험군과 비교하였다.
- 그 결과, 도 5에서 나타난 바와 같이 리오바이러스를 투여한 경우, 대조군에 비교하였을 때 유두종의 성장에는 큰 차이가 없는 것을 확인하였다 (도 5). <실시예 4> 리오바이러스의 신생혈관 형성 억제능 확인
리오바이러스의 신생혈관 형성 억제능을 확인하기 위하여, 유두종 조직의 뒷면을 관찰하여 신생혈관 형성을 확인하였다.
구체적으로, 상기 <실시예 3>에서 희생하여 유두종 조직을 분리한 실험군 및 대조군 마우스의 유두종 조직의 후면을 관찰하여 암 조직과 연계된 신생혈관의 생성 정도를 촬영하여 사진으로 비교하였다.
그 결과, 도 6에서 나타난 바와 같이 리오바이러스를 투여한 경우, 대조군에 비해 혈관의 형성이 유의적으로 감소 또는 둔화되는 것을 확인하였으며 (도 6), 이를 통해 리오바이러스가 종양의 생장에는 영향이
없으나 신생혈관 형성은 현저하게 감소시켜, 악성 종양의 파괴보다는 신생혈관 억제에,대한 유의적인 효과를 나타내는 것을 확인하였다.
<제조예 1>산제의 제조
리오바이러스 (reovirus): 102 내지 1013 pfu o
ΤΓ당: g
상기워 성분을 흔합하고 기밀포에 층진하여 산제를 제조하였다.
<제조예 2>정제의 제조
리오바이러스 (reovirus): 102 내지 1013 pfu 옥수수전분: 100 mg
유당: 100 mg
스테아린산 마그네슘: 2 mg
상기의 성분을 흔합한 후, 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조하였다.
<제조예 3>캡술제의 제조
리오바이러스 (reovirus): 102 내지 1013 pfu 옥수수전분: 100 mg
유당: 100 mg
스테아린산 마그네슘: 2 mg
상기의 성분을 흔합한 후, 통상의 캡슐제의 제조방법에 따라서 젤라틴 캡술에 층전하여 캡슐제를 제조하였다.
〈제제예 4>주사제의 제조
리오바이러스 (reovirus): 102 내지 1013 pfu 바이러스운반용 세포 (성체줄기세포, 혈액세포등 Molecular Therapy. 2009 Vol 17:1667-1676)
Na2HP0412H20: 26 nig 증류수: 2974 nig 상기의 성분을 흔합한 후, 통상의 제조방법에 따라서 주사제를 제조하였다.
Claims
【청구항 1】
리오바이러스과 (reoviridae)에 속하는 리오바이러스 (reovirus)를 유효성분으로 함유하는 혈관신생 억제용 꺅학적 조성물.
[청구항 2】
제 1항에 있어서, 상기 리오바이러스는 야생형 (wild type) 바이러스인 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.
【청구항 3]
제 1항에 있어서, 상기 라오바이러스는 혈청 타입 (Serotype) 1, 2, 또는 3을 포함하는 타입 l(Lang), 타입 2(Jones) 또는 타입 3(Dearing 및 Abney) 야생형 리오바이러스 및 약독화 리오바이러스인 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.
【청구항 4】
제 1항에 있어서, 상기 약학적 조성물은 혈관신생에 의해 유발되는 질환을 예방 또는 치료할 수 있는 특징으로 하는 혈관신생 억제용 약학적 조성물.
[청구항 5】
제 4항에 있어서, 상기 질환은 암, 혈관종, 혈관섬유종, 관절염, 당뇨병성 망막증 (diabetic retionpathy) , 조숙아의 망막증, 신생혈관성 녹내장, 신생혈관에 의한 각막질환, 퇴화반 /황반변성 (macular degeneration), 반점의 변성, 익상편, 망막변성, 후수정체 섬유증식증, 과립성 결막염, 건선, 모세관 확장증, 화농성 육아종, 건선, 지루성 피부염 및 여드름으로 이루어지는 군으로부터 선택되는 어느 하나의 질환인 것을 특징으로 하는 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.
【청구항 6】
제 5항에 있어서, 상기 암은 간암, 위암, 결장암, 유방암, 폐암, 비소세포성폐암, '골암, 췌장암, 피부암, 두부 또는 경부암, 피부 또는 안구 내 흑색종, 자궁경부암, 난소암, 대장암, 소장암, 직장암, 항문부근암, 나팔관암종ᅳ 자궁내막암종, 자궁경부암종, 질암종, 음문암종, 호지킨병 (Hodgkin's disease), 식도암, 소장암, 임파선암, 방광암, 담낭암, 내분비선암, 갑상선암, 부갑상선암, 부신암, 연조직 육종,요도암,음경암,전립선암, 만성 또는 급성 백혈병, 림프구 림프종, 방광암, 신장 또는 수뇨관 암, 신장세포 암종, 신장골반 암종, 중추신경계 (CNS; central nervous system)종양, 1차 CNS림프종,척수 종양, 뇌간 신경교종 및 뇌하수체 선종으로 이루어지는 군으로부터 선택되는 어느 하나인 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.
【청구항 7】
제 1항에 있어서, 상기 리오바이러스는 혈관신생 억제용 외래 유전자를 추가적으로 포함하는 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.
【청구항 8】
제 7항에 있어서, 상기 신생혈관 억제용 외래 유전자는 리오바이러스 유전자 내 복제필요 유전자부위 (essential region) 또는 복제불필요 유전자부위 (non-essential region)에 삽입되어 혈관신생 억제 활성을 증가시키는 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.
【청구항 9]
제 7항에 있어서, 상기 혈관신생 억제용 외래 유전자는 혈관 내피 성장 인자 (VEGF), 가용성 VEGF 수용체, 안지오스타틴, 엔도스타틴 및 아포리포 단백질 (a) 크링글 영역 (LK8)을 암호화하는 유전자로 이루어진 군에서 선택되는 어느 하나인 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.

제 1항에 있어서, 상기 리오바이러스는 바이러스 운반용 세포 (Virus. carrier cells)에 탑재되어 생체내에서 치료효과가 상승되는 것을 특징으로 하는 혈관신생 억제용 약학적 조성물.

리오바이러스를 유효성분으로 함유하는 혈관신생 관련질환 예방 및 개선용 건강식품.

리오바이러스과에 속하는 리오바이러스를 유효성분으로 함유하는 약학적 조성물을 필요로 하는 개체에 투여하는 단계를 포함하는 혈관신생 억제 방법. 【청구항 13】
