Abstract

본 발명은 미량의 타겟물질도 빠른 시간 내에 검출할 수 있는 바이오센서 및 이를 이용한 타겟물질 검출방법을 위하여, 수용체가 고정될 수 있으며 강자성체물질을 포함하는 수용체고정부와, 상기 수용체고정부에 방향이 가변하는 자기장을 인가할 수 있는 자기장인가부를 구비하는, 바이오센서 및 이를 이용한 타겟물질 검출방법을 제공한다.

Description

바이오센서 및 이를 이용한 타겟물질 검출방법
본 발명은 바이오센서 및 이를 이용한 타겟물질 검출방법에 관한 것으로서, 더 상세하게는 미량의 타겟물질도 빠른 시간 내에 검출할 수 있는 바이오센서 및 이를 이용한 타겟물질 검출방법에 관한 것이다.
일반적으로 바이오센서는 관심대상인 타겟물질의 물리적 양 등에 대한 정보를 효소와 같은 생체유래물질 및/또는 미생물 등을 이용하여 검출하고 전기신호로 변환하여 출력할 수 있는 것으로, 타겟물질에 대한 선택성이 우수하고 감도가 예민하기에 분석장치로서 주목받고 있다.
그러나 이러한 종래의 바이오센서에는 관심대상인 타겟물질의 양이 적을 경우 이를 검출하는데 장시간이 소요된다는 문제점이 있었다. 즉, 종래의 바이오센서의 경우 관심대상인 타겟물질이 확산 등의 과정을 거쳐 바이오센서의 검지영역에 도달되면 이를 감지하는 방식인바, 따라서 관심대상인 타겟물질의 양이 매우 적을 경우 관심대상인 타겟물질의 확산 등의 속도가 매우 느려 감지에 소요되는 시간이 매우 길다는 문제점이 있었다.
본 발명은 상기와 같은 문제점을 포함하여 여러 문제점들을 해결하기 위한 것으로서, 미량의 타겟물질도 빠른 시간 내에 검출할 수 있는 바이오센서 및 이를 이용한 타겟물질 검출방법을 제공하는 것을 목적으로 한다. 그러나 이러한 과제는 예시적인 것으로, 이에 의해 본 발명의 범위가 한정되는 것은 아니다.
본 발명의 일 관점에 따르면, 수용체가 고정될 수 있으며 강자성체물질을 포함하는 수용체고정부와, 상기 수용체고정부에 방향이 가변하는 자기장을 인가할 수 있는 자기장인가부를 구비하는, 바이오센서가 제공된다.
상기 자기장인가부는, 상기 수용체고정부의 수용체가 고정될 면을 가로지르는 방향으로 자기장을 인가할 수 있다. 구체적으로, 상기 자기장인가부는, 상기 수용체고정부의 수용체가 고정될 면을 가로지르되 시간이 흐름에 따라 일 방향과 그 반대 방향으로 가변하는 자기장을 인가할 수 있다.
한편, 상기 강자성체물질은 철, 코발트 및 니켈 중 적어도 어느 하나를 포함할 수 있다.
상기 수용체고정부에 고정된 수용체를 더 포함할 수 있다. 이 경우 상기 수용체는, DNA, RNA, DNA탐침, 펩타이드, 단백질, 압타머 및 항체로 구성된 군에서 선택된 것일 수 있다. 나아가 상기 수용체는, DNA, RNA, 펩타이드, 단백질, 항원, 항체, 합텐(hapten), 유기화합물 및 무기화합물로 구성된 군에서 선택된 타겟물질과 결합하도록 할 수 있다. 이때, 상기 타겟물질은 자성나노입자에 고정된 것일 수 있다.
한편, 제1전극과, 상기 제1전극과 이격되어 배치되는 제2전극과, 상기 제1전극과 상기 수용체고정부 사이를 연결하는 제1반도체층과, 상기 제2전극과 상기 수용체고정부 사이를 연결하는 제2반도체층을 더 구비할 수 있다.
이때, 상기 제1반도체층과 상기 수용체고정부 사이의 제1영역과 상기 제2반도체층과 상기 수용체고정부 사이의 제2영역을 포함하는 검지영역 중 적어도 일부를 감싸는 웰(well)이 형성되도록 하는 벽(壁)을 더 포함할 수 있다.
본 발명의 다른 일 관점에 따르면, 강자성체물질을 포함하는 수용체고정부를 갖는 바이오센서의 수용체고정부에 수용체를 고정하는 단계와, 자성나노입자를 타겟물질이 존재하는 용액에 공급하여 타겟물질의 적어도 일부를 자성나노입자 표면에 흡착시키는 단계와, 자성나노입자가 공급된 타겟물질이 존재하는 용액을 수용체고정부에 공급하는 단계와, 수용체고정부에 방향이 가변하는 자기장을 인가하는 단계를 포함하는, 타겟물질 검출방법이 제공된다.
상기 자기장을 인가하는 단계는, 수용체고정부의 수용체가 고정될 면을 가로지르는 방향으로 자기장을 인가하는 단계일 수 있다. 구체적으로, 상기 자기장을 인가하는 단계는, 수용체고정부의 수용체가 고정될 면을 가로지르되 시간이 흐름에 따라 일 방향과 그 반대 방향으로 가변하는 자기장을 인가하는 단계일 수 있다.
한편, 수용체고정부가 포함하는 강자성체물질은 철, 코발트 및 니켈 중 적어도 어느 하나를 포함할 수 있다.
상기 바이오센서의 수용체고정부에 수용체를 고정하는 단계는, DNA, RNA, DNA탐침, 펩타이드, 단백질, 압타머 및 항체로 구성된 군에서 선택된 것을 바이오센서의 수용체고정부에 고정하는 단계일 수 있다. 이 경우, 상기 타겟물질의 적어도 일부를 자성나노입자 표면에 흡착시키는 단계는, DNA, RNA, 펩타이드, 단백질, 항원, 항체, 합텐(hapten), 유기화합물 및 무기화합물로 구성된 군에서 선택된 것의 적어도 일부를 자성나노입자 표면에 흡착시키는 단계일 수 있다.
한편, 수용체고정부와 제1전극을 연결하는 제1반도체층과 수용체고정부 사이의 제1영역과, 수용체고정부와 제2전극을 연결하는 제2반도체층과 수용체고정부 사이의 제2영역을 포함하는 검지영역 중 적어도 일부를 감싸는 벽(壁)에 의해 한정된 웰(well)에 컨트롤용액을 공급하는 단계를 더 포함하고, 상기 자성나노입자가 공급된 타겟물질이 존재하는 용액을 수용체고정부에 공급하는 단계는, 자성나노입자가 공급된 타겟물질이 존재하는 용액을 웰(well) 내에 공급하는 단계일 수 있다.
이 경우, 상기 컨트롤용액을 공급하는 단계는, 타겟물질이 존재하는 용액의 용매와 동일한 성분의 물질을 공급하는 단계일 수 있다.
상기한 바와 같이 이루어진 본 발명의 일 실시예에 따르면, 미량의 타겟물질도 빠른 시간 내에 검출할 수 있는 바이오센서 및 이를 이용한 타겟물질 검출방법을 구현할 수 있다. 물론 이러한 효과에 의해 본 발명의 범위가 한정되는 것은 아니다.

이하, 첨부된 도면들을 참조하여 본 발명의 실시예를 상세히 설명하면 다음과 같다. 그러나 본 발명은 이하에서 개시되는 실시예에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 수 있는 것으로, 이하의 실시예는 본 발명의 개시가 완전하도록 하며, 통상의 지식을 가진 자에게 발명의 범주를 완전하게 알려주기 위해 제공되는 것이다. 또한 설명의 편의를 위하여 도면에서는 구성 요소들이 그 크기가 과장 또는 축소될 수 있다. 예컨대, 도면에서 나타난 각 구성의 크기 및 두께는 설명의 편의를 위해 임의로 나타내었으므로, 본 발명이 반드시 도시된 바에 한정되지 않는다.
이하의 실시예에서, x축, y축 및 z축은 직교 좌표계 상의 세 축으로 한정되지 않고, 이를 포함하는 넓은 의미로 해석될 수 있다. 예를 들어, x축, y축 및 z축은 서로 직교할 수도 있지만, 서로 직교하지 않는 서로 다른 방향을 지칭할 수도 있다.
한편, 층, 막, 영역, 판 등의 각종 구성요소가 다른 구성요소 "상에" 있다고 할 때, 이는 다른 구성요소 "바로 상에" 있는 경우뿐 아니라 그 사이에 다른 구성요소가 개재된 경우도 포함한다.
도 1은 본 발명의 일 실시예에 따른 바이오센서를 개략적으로 도시하는 단면도이다. 본 실시예에 따른 바이오센서는 수용체고정부(110)와 자기장인가부(500)를 구비한다. 수용체고정부(110)는 실리콘웨이퍼, 글래스재, 플라스틱재 등의 물질을 포함할 수 있는 기판(100) 상에 형성될 수 있는데, 기판(100)은 경우에 따라서는 금속을 포함하고 절연체로 코팅된 구조를 가질 수도 있다. 나아가 도시된 것과 같이 제1전극(210)과, 이 제1전극(210)과 이격되어 배치되는 제2전극(220)과, 제1전극(210)과 수용체고정부(110) 사이를 연결하는 제1반도체층(310)과, 제2전극(220)과 수용체고정부(110) 사이를 연결하는 제2반도체층(320)이 기판(100) 상에 배치될 수 있다.
수용체고정부(110)는 강자성체물질을 포함하는데, 예컨대 철, 코발트 및 니켈 중 적어도 어느 하나를 포함할 수 있다. 이러한 강자성체물질을 포함하는 수용체고정부(110)는 후술하는 것과 같이 강자성체물질의 특성을 이용해 타겟물질이 특정 위치에 집중되도록 하여 검출 효율성일 획기적으로 높일 수 있다. 수용체고정부(110)는 (도 1의 x축 방향을 따라) 대략 3um 정도의 길이로 형성할 수 있다.
수용체고정부(110)에는 타겟물질인 특정물질과 결합할 수 있는 수용체가 고정될 수 있다. 수용체의 수용체고정부(110) 상에의 고정은 다양한 방법을 통해 이루어질 수 있다. 예컨대 수용체고정부(110)를 이작용성 라디칼(bifunctional radical, B)로 표면처리한 후, 수용체를 공급할 수 있다. 이 경우 이작용성 라디칼(B)의 일측은 수용체고정부(110)에 고정되고, 타측은 수용체와 결합할 수 있다.
수용체는 DNA, RNA, DNA탐침, 펩타이드, 단백질, 압타머 및 항체로 구성된 군에서 선택된 것일 수 있다. 이러한 수용체는 DNA, RNA, 펩타이드, 단백질, 항원, 항체, 합텐(hapten), 유기화합물 및 무기화합물로 구성된 군에서 선택된 타겟물질과 결합할 수 있다.
자기장인가부(500)는 수용체고정부(110)에 방향이 가변하는 자기장을 인가할 수 있다. 이러한 자기장인가부(500)는 예컨대 솔레노이드를 포함할 수 있다. 솔레노이드의 경우 솔레노이드의 코일에 흐르는 전류의 방향을 바꿈으로서 솔레노이드에서 생성되는 자기장의 방향을 반대로 바꿀 수 있다. 물론 본 발명이 이에 한정되는 것은 아니며, 통상의 영구자석과 이 영구자석을 회전시킬 수 있는 플립부를 구비하여, 플립부가 영구자석을 180도 회전시킴으로써 수용체고정부(110)에 인가되는 자기장의 방향이 변하도록 할 수도 있는 등 다양한 변형이 가능하다.
제1전극(210)과 이 제1전극(210)과 이격되어 배치되는 제2전극(220)은 전도성 물질로 형성될 수 있는데, 예컨대 팔라듐이나 금 등을 포함할 수 있다.
제1반도체층(310)은 제1전극(210)과 수용체고정부(110) 사이를 연결하고 제2반도체층(320)은 제2전극(220)과 수용체고정부(110) 사이를 연결한다. 이러한 제1반도체층(310)과 제2반도체층(320)은 예컨대 탄소나노튜브를 포함할 수 있다. 제1반도체층(310)과 제2반도체층(320) 각각은 (도 1의 x축 방향을 따라) 대략 10um 정도의 길이로 형성할 수 있다.
이러한 제1반도체층(310)은 도시된 것과 같이 제1전극(210)에서 수용체고정부(110) 방향으로 연장된 형상을 가질 수 있다. 나아가 제1반도체층(310)은 제1전극(210)에서 수용체고정부(110) 방향으로 연장된 형상의 복수개의 제1연장부들을 구비하되, 제1연장부들 각각이 대략 상호 평행하며 y축 방향으로 상호 이격되어 배치되도록 할 수도 있다. 제2반도체층(320) 역시 도시된 것과 같이 제2전극(220)에서 수용체고정부(110) 방향으로 연장된 형상을 가질 수 있다. 나아가 제2반도체층(320)도 제2전극(220)에서 수용체고정부(110) 방향으로 연장된 형상의 복수개의 제2연장부들을 구비하되, 제2연장부들 각각이 대략 상호 평행하며 y축 방향으로 상호 이격되어 배치되도록 할 수도 있다. 이를 통해 수용체고정부(110)의 (xy평면에서의) 면적을 넓힐 수 있으며, 이는 후술하는 것과 같이 바이오센서의 검지영역을 넓히는 역할을 할 수 있다.
수용체고정부(110)는 전술한 바와 같이 강자성체물질, 즉 금속을 포함하기에, 이러한 제1반도체층(310)과 수용체고정부(110), 그리고 제2반도체층(320)과 수용체고정부(110) 사이에는 쇼트키배리어가 형성된다. 이러한 상황에서 타겟물질이 수용체고정부(110)에 고정된 수용체에 결합하게 되면, 쇼트키배리어의 크기가 변하게 되면서 제1전극(210)과 제2전극(220) 사이에서 흐르는 전류의 크기가 달라지는바, 이를 통해 타겟물질의 존재여부를 검출할 수 있다. 이에 대해서는 후술한다.
후술하는 바와 같이 타겟물질의 검출은 용액 내에 타겟물질이 존재하는지 여부 등을 확인하는 경우일 수 있다. 따라서 제1반도체층(310), 제2반도체층(320), 제1전극(210) 및 제2전극(220)을 용액으로부터 보호하기 위해, 도시된 것과 같이 제1반도체층(310), 제2반도체층(320), 제1전극(210) 및 제2전극(220)의 적어도 일부를 덮는 패시베이션층(410)이 형성되도록 할 수 있다. 이러한 패시베이션층(410)은 포토리지스트물질로 형성할 수도 있고, 실리콘나이트라이드나 실리콘옥사이드 등과 같은 절연성 물질로 형성할 수도 있다.
아울러 타겟물질의 검출은 대부분 제1반도체층(310)과 수용체고정부(110)가 만나는 부분 및 그 주위, 그리고 제2반도체층(320)과 수용체고정부(110)가 만나는 부분 및 그 주위에서 이루어진다. 따라서 용액 내에 타겟물질이 존재하는지 여부를 확인할 시 그 용액이 제1반도체층(310)과 수용체고정부(110)가 만나는 부분 및 그 주위, 그리고 제2반도체층(320)과 수용체고정부(110)가 만나는 부분 및 그 주위에 존재하도록 하기 위해, 도시된 것과 같이 벽(壁, 420)이 형성되도록 할 수 있다. 이러한 벽(420)은 검지영역의 적어도 일부, 즉 제1반도체층(310)과 수용체고정부(110) 사이의 제1영역과 제2반도체층(320)과 수용체고정부(110) 사이의 제2영역을 포함하는 검지영역 중 적어도 일부를 감싸는 웰(well, 420a)을 정의할 수 있다.
벽(420)은 도시된 것과 같이 패시베이션층(410) 상에 위치하되 패시베이션층(410)의 수용체고정부(110) 방향의 끝단에 위치할 수도 있고, 이와 달리 벽(420)이 패시베이션층(410) 상에 위치하되 패시베이션층(410)이 벽(420)에 의해 정의되는 웰(420a)의 저면의 일부분을 더 덮을 수도 있다. 또는 벽(420)이 패시베이션층(410)의 수용체고정부(110) 방향의 끝단과 이격되어 제1반도체층(310)이나 제2반도체층(320)과 직접 컨택하도록 배치될 수도 있다.
한편, 도면에서는 벽(420)이 패시베이션층(410)과 별도인 것으로 도시하고 있으나, 필요에 따라서는 일체(一體)로 형성될 수도 있다. 아울러 벽(420)은 바이오센서의 다른 구성요소들에 대해 착탈가능한 것일 수도 있다. 예컨대 벽(420)은 링(ring) 또는 원통형쉘(cylindrical shell) 형상을 가진 것으로, 관심대상인 타겟물질의 검출에 앞서 패시베이션층(410) 상에 배치되는 것일 수 있다. 이러한 경우 벽(420)은 아크릴 등과 같은 플라스틱물질을 포함할 수 있다.
이와 같은 바이오센서에 있어서, 자기장인가부(500)는 도 2에 도시된 것과 같이 수용체고정부(110)의 수용체가 고정될 면을 가로지르는 방향(대략 +z 방향)으로 자기장을 인가할 수 있다. 나아가 자기장인가부(500)는 도 2에 도시된 것과 같이 수용체고정부(110)의 수용체가 고정될 면을 가로지르는 방향(대략 +z 방향)으로 자기장을 인가하다가, 도 3에 도시된 것과 같이 그 반대 방향(대략 -z 방향)으로 자기장을 인가할 수 있다. 즉, 자기장인가부(500)는, 수용체고정부(110)의 수용체가 고정될 면을 가로지르되 시간이 흐름에 따라 일 방향과 그 반대 방향으로 가변하는 자기장을 인가할 수 있다. 자기장인가부(500)에서 인가하는 이와 같은 자기장과 수용체고정부(110)가 포함하는 강자성체물질에 의해, 검출하고자 하는 타겟물질을 자성나노입자에 고정하면, 검출하고자 하는 타겟물질의 양이 매우 미량이라 하더라도 매우 신속하게 타겟물질을 검출할 수 있다. 이하에서는 이에 대해 상세히 설명한다.
도 4는 도 1의 바이오센서의 자기장인가부(500)에 의해 수용체고정부(110)에 인가되는 자기장의 크기와 방향이 바뀔 시 나타나는 수용체고정부(110)에서의 자화도를 개략적으로 도시하는 그래프이다. 그래프에서 가로축은 자기장인가부(500)에서 인가하는 외부자기장의 세기를 의미하고, 세로축은 외부자기장에 의해 발생하는 수용체고정부(110)의 자화도의 크기를 의미한다. 수용체고정부(110)는 강자성체물질을 포함하기에, 도시된 것과 같이 강자성체물질이 갖는 자기이력현상을 수용체고정부(110) 역시 갖게 된다.
일반적으로 물체에 자기장을 가하면 물체는 자화되고 자화도는 물체에 가해 준 자기장의 크기와 비례한다. 물체가 자기포화상태가 되면 자기장을 가해도 더 이상 자화되지 않으며 이때 자기장을 줄이면 자화도도 줄어들기 시작하여 자기장이 사라지면 자화도도 0이 된다. 자기장인가부(500)가 수용체고정부(110)에 자기장을 인가하면 수용체고정부(110) 역시 자화가 일어나고 특정한 자기장 이상에서는 자기포화상태가 되어 최대의 자화도를 갖는다(도 4의 "1" 부분의 원점에서 시작하는 실선 참조).
하지만 수용체고정부(110)가 강자성체물질을 포함하기에, 그와 같은 상태에서 자기장인가부(500)가 인가하는 자기장의 세기를 줄이더라도 수용체고정부(110)의 자화도는 상대적으로 느리게 줄어들게 되고, 자기장인가부(500)가 자기장을 인가하지 않더라도 자화도는 남아 있는 상태가 된다(도 4의 "1" 부분 점선 참조). 따라서 그 상황에서 자기장인가부(500)가 최초 자기장을 인가할 시의 방향과 반대인 방향으로 자기장을 인가하면(도 4의 "2" 부분 점선 참조), 수용체고정부(110)의 자화도에 의한 내부자기장과 자기장인가부(500)가 인가한 외부자기장의 방향이 반대가 되어, 수용체고정부(110) 인근에서는 토탈자기장의 세기가 약하게 된다.
물론 자기장인가부(500)가 더 센 자기장을 인가하면(도 4의 "3" 부분 위쪽 점선 참조) 수용체고정부(110)의 자화도에 의한 내부자기장 역시 자기장인가부(500)가 인가하는 방향으로 되어 수용체고정부(110) 인근에서의 토탈자기장의 세기가 강하게 된다. 다시 자기장인가부(500)가 인가하는 자기장의 세기가 줄어들더라도, 수용체고정부(110)의 자화도는 상대적으로 느리게 줄어들게 된다(도 4의 "3 "부분 아래쪽 점선 참조). 그 상황에서 자기장인가부(500)가 최초 자기장을 인가할 시의 방향으로 자기장을 인가하면(도 4의 "4" 부분 점선 참조), 수용체고정부(110)의 자화도에 의한 내부자기장과 자기장인가부(500)가 인가한 외부자기장의 방향이 반대가 되어, 수용체고정부(110) 인근에서는 토탈자기장의 세기가 약하게 된다.
도 5a 내지 도 5d는 도 1의 바이오센서에서 타겟물질이 검출되는 과정을 개략적으로 도시하는 개념도들이다.
자기장인가부(500)가 수용체고정부(110)에 (+z 방향으로) 자기장을 인가하면 수용체고정부(110) 역시 자화가 일어나고 특정한 자기장 이상에서는 자기포화상태가 되어 최대의 자화도를 갖는다(도 4의 "1" 부분의 원점에서 시작하는 실선 참조). 이때 도 5a에 도시된 것과 같이 자기장인가부(500)에 의한 외부자기장(BE)의 방향과 수용체고정부(110)가 포함하는 강자성체물질의 자화에 의한 내부자기장(BM)의 방향이 일치하기에, 총자기장(BT)은 그 크기가 매우 크며 이에 따라 타겟물질(TM)이 고정요소(B)에 의해 고정된 자성나노입자(MP)가 큰 자기력에 의해 수용체고정부(110)로 접근하게 된다. 이에 따라 수용체고정부(110) 상의 수용체(A)와 타겟물질(TM)이 가까워지면 상호 결합하게 된다.
이후 자기장인가부(500)가 최초 자기장을 인가할 시의 방향(+z 방향)과 반대인 방향(-z 방향)으로 자기장을 인가하면(도 4의 "2" 부분 점선 참조), 도 5b에 도시된 것과 같이, 수용체고정부(110)의 자화도에 의한 내부자기장(BM)은 여전히 최초 방향(+z 방향)을 유지하는 가운데 자기장인가부(500)가 인가한 외부자기장(BE)의 방향(-z 방향)이 수용체고정부(110)의 자화도에 의한 내부자기장(BM)의 방향과 반대가 되기에, 수용체고정부(110) 인근에서는 토탈자기장(BT)의 세기가 약하게 된다. 이에 따라 타겟물질(TM)이 고정요소(B)에 의해 고정된 자성나노입자(MP)가 약한 자기력에 의해 수용체고정부(110)로부터 멀어지게 된다. 이때, 수용체고정부(110) 상의 수용체(A)와 이미 결합된 타겟물질(TM)의 경우 그 결합력에 의해 수용체고정부(110)로부터 멀어지지 않고 그대로 결합된 채 수용체고정부(110) 근방에 머물게 된다.
바뀐 방향으로 자기장인가부(500)가 더 센 자기장을 인가하면(도 4의 "3" 부분 위쪽 점선 참조) 도 5c에 도시된 것과 같이 수용체고정부(110)의 자화도에 의한 내부자기장(BM) 역시 자기장인가부(500)가 인가하는 방향(-z 방향)으로 되어 수용체고정부(110) 인근에서의 토탈자기장(BT)의 세기가 다시 강하게 된다. 이에 따라 타겟물질(TM)이 고정요소(B)에 의해 고정된 자성나노입자(MP)가 큰 자기력에 의해 수용체고정부(110)로 다시 접근하게 된다. 그 결과 수용체고정부(110) 상의 수용체(A)와 타겟물질(TM)이 가까워지면 상호 결합하게 된다.
이후 자기장인가부(500)가 인가하는 자기장의 방향을 다시 바꿔 최초 자기장을 인가할 시의 방향(+z 방향)으로 자기장을 인가하면(도 4의 "4" 부분 점선 참조), 도 5d에 도시된 것과 같이 수용체고정부(110)의 자화도에 의한 내부자기장(BM)과 자기장인가부(500)가 인가한 외부자기장(BE)의 방향이 반대가 되어, 수용체고정부(110) 인근에서는 토탈자기장(BT)의 세기가 약하게 된다. 이에 따라 타겟물질(TM)이 고정요소(B)에 의해 고정된 자성나노입자(MP)가 약한 자기력에 의해 수용체고정부(110)로부터 멀어지게 된다. 이때, 수용체고정부(110) 상의 수용체(A)와 이미 결합된 타겟물질(TM)의 경우 그 결합력에 의해 수용체고정부(110)로부터 멀어지지 않고 그대로 결합된 채 수용체고정부(110) 근방에 머물게 된다.
이와 같이 자기장인가부(500)가 인가하는 자기장의 방향이 바뀜에 따라, 도 5a 내지 도 5d에 도시된 것과 같은 자성나노입자의 캡처링, 릴리징, 캡처링 및 릴리징이 일어난다. 따라서 캡처링과 릴리징이 반복되도록 함으로써, 용액 내에 타겟물질(TM)이 존재하는지 여부 등을 확인할 시 용액 내에 타겟물질(TM)이 미량 존재하더라도 수용체고정부(110)로 타겟물질(TM)이 집중되도록 할 수 있으며, 이에 따라 신속한 검출이 가능하도록 할 수 있다.
여기서 릴리징이 필요한 이유는, 타겟물질(TM)이 고정된 자성나노입자(MP)도 존재하지만 타겟물질(TM)이 고정되지 않은 자성나노입자(MP)도 존재할 수 있는바, 그러한 타겟물질(TM)이 고정되지 않은 자성나노입자(MP)가 수용체고정부(110) 인근에 몰려있으면 수용체고정부(110) 상의 수용체(A)와 타겟물질(TM)의 결합이 용이하지 않을 수 있기 때문이다. 따라서 릴리징 후 다시 캡처링이 이루어지도록 함으로써, 타겟물질(TM)이 고정된 자성나노입자(MP)가 수용체고정부(110) 인근에 효과적으로 접근하여 고농도로 모이도록 할 수 있다.
타겟물질(TM)이 수용체고정부(110) 상의 수용체(A)와 결합하게 되면, 도 1에 도시된 것과 같은 바이오센서의 경우 제1전극(210)과 제2전극(220) 사이의 전류세기가 바뀔 수 있다. 전술한 바와 같이 제1반도체층(310)과 수용체고정부(110), 그리고 제2반도체층(320)과 수용체고정부(110) 사이에는 쇼트키배리어가 형성된다. 이러한 상황에서 타겟물질(TM)이 수용체고정부(110)에 고정된 수용체(A)에 결합하게 되면, 쇼트키배리어의 크기가 변하게 되면서 제1전극(210)과 제2전극(220) 사이에서 흐르는 전류의 크기가 달라진다. 이는 타겟물질(TM)에 포함된 전하에 의해 제1반도체층(310)과 수용체고정부(110), 그리고 제2반도체층(320)과 수용체고정부(110) 사이에 형성된 쇼트키배리어의 크기가 변하기 때문이다. 예컨대 타겟물질(TM)이 단백질을 포함한다면 이 단백질은 아미노산을 포함하게 되며, 이러한 아미노산은 전하를 갖기에, 수용체(A)와 타겟물질(TM)이 결합하게 되면 쇼트키배리어의 크기가 변하게 된다. 따라서 그러한 전류 크기 변화를 확인함으로써 검출하고자 하는 타겟물질(TM)의 존재여부나 그 양 등을 확인할 수 있다.
물론 본 발명의 바이오센서가 도 1에 도시된 것과 같은 구조에 한정되는 것은 아니다. 즉, 도 4 및 도 5a 내지 도 5d를 참조하여 전술한 것과 같이, 강자성체물질을 포함하는 수용체고정부(110)와 자기장인가부(500)를 구비하고, 자기장인가부(500)가 수용체고정부(110)에 인가하는 자기장의 방향을 바꿈에 따라 타겟물질(TM)이 고정된 자성나노입자(MP)가 수용체고정부(110)에 접근하거나 멀어지도록 함으로써, 캡처링과 릴리징의 반복을 통해 효과적으로 신속하게 타겟물질(TM)이 수용체고정부(110)에 접근하여 수용체고정부(110)의 수용체(A)와 결합하도록 하는 바이오센서라면, 본 발명의 범위에 속한다고 할 것이다.
아울러 전술한 것과 같이 벽(420)은 바이오센서의 다른 구성요소들에 대해 착탈가능한 것일 수도 있어, 관심대상인 타겟물질을 포함하는 용액을 수용체고정부(110), 제1반도체층(310) 및/또는 제2반도체층(320)에 공급하기 직전에 벽(420)이 패시베이션층(410) 등의 상에 배치되고, 관심대상인 타겟물질의 검출 등이 완료된 후에 벽(420)이 제거되도록 할 수 있다.
이와 같은 본 실시예에 따른 바이오센서는 예컨대 특정 항원을 타겟물질(TM)로 하고, 그 특정 항원에 대한 항체를 수용체(A)로 하여, 항체를 수용체고정부(110)에 고정시키고 상기 특정 항원을 자성나노입자(MP)에 결합한 후 그러한 특정 항원에 대한 항체의 작동여부 등을 확인하는데 사용될 수 있다. 또는 항체를 이용하여 그 항체가 작용하는 특정 항원의 존재여부를 확인하는데 사용될 수도 있다. 물론 본 실시예에 따른 바이오센서의 사용이 이에 한정되는 것은 아니며, 이는 단순한 예시일 뿐이다.
한편, 벽(420)에 의해 한정되는 웰(420a) 내에 타겟물질(TM)을 포함하는 용액을 공급하고 제1전극(210)과 제2전극(220) 사이의 전류 변화를 살필 시, 그 전류 변화가 용액 내에 존재하는 타겟물질(TM)에 의한 것일 수도 있지만 용액의 용매에 의한 것일 수도 있다. 따라서 그러한 용매에 의한 변화가 존재한다면 그러한 변화를 사전에 확인하는 것이 필요하다.
이를 위해, 벽(420)에 의해 한정되는 웰(420a) 내에 타겟물질(TM)을 포함하는 용액을 공급하기에 앞서, 웰(420a) 내에 컨트롤용액을 공급하고 이 컨트롤용액에 의한 제1전극(210)과 제2전극(220) 사이의 전류 변화를 확인하는 것이 바람직하다. 이 컨트롤용액은 타겟물질(TM)을 포함하는 용액의 용매와 동일한 성문의 물질일 수 있다. 이와 같이 웰(420a)에 컨트롤용액을 공급하고 그 후에 타겟물질(TM)을 포함하는 용액을 공급함으로써, 타겟물질(TM)에 의한 제1전극(210)과 제2전극(220) 사이의 전류 변화를 정확하게 확인할 수 있다. 컨트롤용액은 예컨대 물이나 PBS(Phosphate Buffered Saline), 또는 물과 PBS 등의 혼합물일 수 있다.
한편, 타겟물질(TM)은 자성나노입자(MP)에 고정된다고 하였는바, 이는 다양한 방법을 통해 이루어질 수 있다. 예컨대 산화철 등을 포함하는 자성나노입자(MP)를 이작용성 라디칼(bifunctional radical, B)로 표면처리한 후, 타겟물질(TM)이 존재하는 용액에 공급한다. 이작용성 라디칼(B)의 일측은 자성나노입자(MP)에 고정되고, 타측은 용액 내의 타겟물질(TM)과 결합할 수 있다. 따라서 용액 내에서 타겟물질(TM)의 적어도 일부는 자성나노입자(MP)에 결합되게 된다. 물론 자성나노입자(MP) 중에서도 타겟물질(TM)과 결합되지 않은 것이 존재할 수 있다.
지금까지 바이오센서에 대해 설명하였으나 본 발명이 이에 한정되는 것은 아니다. 즉, 상술한 것과 같은 바이오센서의 원리를 이용한 타겟물질 검출방법 역시 본 발명의 범위에 속한다.
도 6은 본 발명의 다른 일 실시예에 따른 타겟물질 검출방법을 개략적으로 도시하는 플로우챠트이다.
도시된 것과 같이 강자성체물질을 포함하는 수용체고정부를 갖는 바이오센서의 수용체고정부에 수용체를 고정하는 단계(S10)를 거치고, 아울러 자성나노입자를 타겟물질이 존재하는 용액에 공급하여 타겟물질의 적어도 일부를 자성나노입자 표면에 흡착시키는 단계(S30)를 거친다. 물론 S10 단계와 S30 단계는 그 순서가 바뀔 수도 있고 병행하여 이루어질 수도 있다. 또한 S10 단계가 완료되어 준비된 상태에서 S30 단계부터 시작할 수도 있다.
수용체고정부가 포함하는 강자성체물질은 철, 코발트 및 니켈 중 적어도 어느 하나를 포함할 수 있다. 그리고 바이오센서의 수용체고정부에 수용체를 고정하는 단계(S10)는, DNA, RNA, DNA탐침, 펩타이드, 단백질, 압타머 및 항체로 구성된 군에서 선택된 것을 바이오센서의 수용체고정부에 고정하는 단계일 수 있다. 아울러 타겟물질의 적어도 일부를 자성나노입자 표면에 흡착시키는 단계(S30)는, DNA, RNA, 펩타이드, 단백질, 항원, 항체, 합텐(hapten), 유기화합물 및 무기화합물로 구성된 군에서 선택된 것의 적어도 일부를 자성나노입자 표면에 흡착시키는 단계일 수 있다.
그와 같은 상황에서, 검지영역 내의 수용체고정부에 컨트롤용액을 공급하는 단계(S20)를 거친다. 컨트롤용액은 타겟물질이 존재하는 용액의 용매와 동일한 성분의 물질일 수 있다.
검지영역은 전술한 것과 같이 수용체고정부와 제1전극을 연결하는 제1반도체층과 수용체고정부 사이의 제1영역과, 수용체고정부와 제2전극을 연결하는 제2반도체층과 수용체고정부 사이의 제2영역을 포함할 수 있다. 이러한 검지영역의 적어도 일부는 벽(壁)에 의해 감싸일 수 있으며, 이와 같이 벽에 의해 한정된 웰(well) 내에 컨트롤용액이 공급될 수 있다.
이후, 자성나노입자가 공급된 타겟물질이 존재하는 용액을 수용체고정부에 공급하는 단계(S40)를 거친 후, 수용체고정부에 방향이 가변하는 자기장을 인가하는 단계(S50)를 거쳐, 타겟물질의 존재여부, 존재한다면 얼마나 존재하는지 등을 확인할 수 있다.
여기서 자기장을 인가하는 단계(S50)는, 수용체고정부의 수용체가 고정될 면을 가로지르는 방향으로 자기장을 인가하는 단계일 수 있으며, 구체적으로 수용체고정부의 수용체가 고정될 면을 가로지르되 시간이 흐름에 따라 일 방향과 그 반대 방향으로 가변하는 자기장을 인가하는 단계일 수 있다. 이를 통해 수용체고정부 인근에서 자성나노입자의 캡처링과 릴리징이 반복되도록 하여, 타겟물질의 양이 적은 경우에도 신속하고 효과적으로 타겟물질의 존재여부 등을 검출할 수 있다.
물론 본 발명의 타겟물질 검출방법이 도 6에 도시된 것과 같은 모든 단계들을 포함하는 경우에 한정되는 것은 아니다. 강자성체물질을 포함하는 수용체고정부에 타겟물질이 흡착된 자성나노입자가 포함된 용액을 공급하고, 수용체고정부에 방향이 가변하는 자기장을 인가함으로써, 캡처링과 릴리징의 반복을 통해 효과적으로 신속하게 타겟물질이 수용체고정부에 접근하여 수용체고정부의 수용체와 결합하도록 하는 검출방법이라면, 본 발명의 범위에 속한다고 할 것이다. 즉, 방향이 가변하는 자기장을 인가하여 미량의 타겟물질을 검출하는 자기 사이클에 의한 선택적 능동 포획기술이라면, 본 발명의 범위에 속한다고 할 것이다.
본 발명은 도면에 도시된 실시예를 참고로 설명되었으나 이는 예시적인 것에 불과하며, 당해 기술분야에서 통상의 지식을 가진 자라면 이로부터 다양한 변형 및 균등한 다른 실시예가 가능하다는 점을 이해할 것이다. 따라서 본 발명의 진정한 기술적 보호 범위는 첨부된 특허청구범위의 기술적 사상에 의하여 정해져야 할 것이다.
본 발명에 따른 바이오센서는 예컨대 특정 항원을 타겟물질(TM)로 하고, 그 특정 항원에 대한 항체를 수용체(A)로 하여, 항체를 수용체고정부(110)에 고정시키고 상기 특정 항원을 자성나노입자(MP)에 결합한 후 그러한 특정 항원에 대한 항체의 작동여부 등을 확인하는데 사용될 수 있다. 또는 항체를 이용하여 그 항체가 작용하는 특정 항원의 존재여부를 확인하는데 사용될 수도 있다. 물론 본 실시예에 따른 바이오센서의 사용이 이에 한정되는 것은 아니며, 이는 단순한 예시일 뿐이다.
