Abstract

La présente invention concerne une méthode pour diminuer le risque de contamination du lait et de la viande lors de la traite et de l'abattage de ruminants porteurs de STEC. L'invention concerne en particulier une méthode pour diminuer le portage des STEC chez les ruminants, caractérisée par l'administration à un ruminant d'au moins une souche de Hafnia alvei. L'invention concerne également une composition comprenant au moins une souche d'Hafnia alvei en combinaison avec une souche de Saccharomyces boulardii.

UTILISATION D'HAFNIA AL VEI POUR REDUIRE LE PORTAGE DES ESCHERICHIA COLI PRODUISANT DES SHIGA TOXINES (STEC) CHEZ LES RUMINANTS
DOMAINE DE L'INVENTION
La présente invention concerne l'utilisation d'une souche de Hafnia alvei comme additif alimentaire en nutrition animale, pour réduire le portage des Escherichia coli producteurs de Shiga-toxines chez les ruminants.
ETAT DE LA TECHNIQUE
Les Escherichia coli producteurs de Shiga-toxines (STEC), aussi nommées Escherichia coli entérohémorragiques (FEHEC), et plus particulièrement le sérogroupe 0157 :H7, sont reconnus actuellement comme des pathogènes émergents importants en santé publique. Ces bactéries ont été détectées chez un grand nombre d'espèces animales sans que ces derniers ne présentent des signes cliniques de maladie. Les principaux « réservoirs » ou « porteurs » sont les ruminants et plus particulièrement les bovins. La prévalence des STEC dans les fèces des bovins varie de 4 à 36%. La persistance des souches de STEC dans les cheptels est due en partie au 'portage intestinal' par ces animaux. Le portage varie selon les saisons, la prévalence étant plus importante dans la période printemps-été.
Le risque de contamination des denrées d'origine animale, destinées à l'homme, est lié aux procédures d'hygiène dans les abattoirs et les ateliers de transformation, mais également à l'importance du portage animal. Ingérées par l'homme, ces bactéries provoquent le plus fréquemment des colites hémorragiques. Un syndrome hémolytique et urémique peut également apparaître chez l'enfant et le sujet âgé, ainsi qu'un purpura thrombotique thrombocytopénique chez l'adulte.
Pour diminuer le portage des STEC par les ruminants, il a été proposé de leur administer des bactéries comme additif alimentaire, telles que des souches de Lactobacillus acidophilus qui diminuent la prévalence de E. coli 0157 dans les fèces (Stephens et al., 2007 ; Tabe et al, 2008). Il a également été montré in vitro qu'une souche de Lactobacillus acidophilus et une souche de Saccharomyces cerevisiae diminuent la période de survie des STEC dans le fluide de la panse (ou rumen), le premier des compartiments du système digestif complexe des ruminants (Chaucheyras-Durand et al., 2010). Hafnia alvei est une entérobactérie qui se trouve dans la flore digestive humaine et animale. Cette bactérie est utilisée dans l'industrie fromagère, pour reproduire le goût des fromages au lait cru dans des fromages préparés à base de lait pasteurisé (Delbès-Paus et al., 2012) ou encore pour permettre de réduire la croissance d'E.coli 026 :H1 1 dans le fromage (Delbès-Paus et al., 2013). Hafnia alvei est en particulier utilisée dans une composition pour l'aromatisation des fromages, comprenant également des Lactobacillus et au moins une levure (demande de brevet WO 99/43214). Hafnia alvei peut également être utilisée pour conserver des produits alimentaires, par inoculation, afin d'inhiber de manière compétitive le développement de bactéries pathogènes (US 5,869,1 13).
II convient cependant de distinguer les propriétés de Hafnia alvei sur un produit animal qui est un aliment de son effet désiré chez l'animal sur pied. En particulier, un effet antagoniste mis en évidence en surface de viande ou en matrice fromagère ne permet pas de prédire un effet inhibiteur de la croissance-survie de STEC dans les conditions du tractus digestif animal ou d'un système modèle in vitro représentatif. En effet, les conditions physicochimiques d'une matrice fromagère et d'un contenu digestif animal sont très différentes (potentiel d'oxydo-réduction beaucoup plus bas dans le tube digestif qui est un milieu essentiellement anaérobie ; pH plus acide dans les fromages ; activité de l'eau (aw) plus basse en fromage, dû à l'ajout de sel ; substrats disponibles, à savoir lactose et autres sucres, protéines, matières grasses dans le lait alors que ce sont essentiellement des polysaccharides végétaux dans le tube digestif des ruminants). Les microorganismes qui y sont présents sont aussi très différents en termes d'espèces microbiennes présentes, de diversité et de niveaux de population ; les bactéries lactiques étant majoritaires dans le lait cru et dans les fromages (Masoude et al, 2012) alors que dans les écosystèmes digestifs des ruminants les principaux genres bactériens sont Prevotella, Ruminococcus, Butyrivibrio, Clostridium etc. (Jami & Mizrahi, 2012 ; de Olivera et al, 2013).
Ainsi, les inventeurs ont montré que les microorganismes suivants par exemple présentent un effet inhibiteur de la croissance des STEC (au moins égal à un log : différence de croissance d'une souche EHEC avec ou sans la souche inhibitrice) en matrice fromagère (PPNC ou pâte pressée non cuite), et en particulier des souches 0157 :H7 ou 026 :H1 1 , alors qu'ils n'ont pas d'effet inhibiteur sur ces souches incubées dans du contenu digestif animal in vitro : Leuconostoc pseudomesenteroides, souche MSE7 ; Leuconostoc citreum, souche MSE2 ; Leuconostoc citreum, souche MSE30.5 ; Marinilactibacilli psychotolerans, souche CRBM29 ; Carnobacterium mobile, souche CRBM18 ; Enterococcus faecalis, souche N688 ; Macrococcus caseolyticus, souche RP8 ; Lactococcus lactis lactis, souche 1 147 ; Lactococcus lactis lactis, souche 278m ; Lactococcus lactis cremoris, souche N718 ; Lactococcus lactis, souche D5.3 ; Lactobacillus plantarum, souche FH3. EXPOSE DE L'INVENTION
A ce jour, dans les élevages industriels, il n'existe pas de méthode pour diminuer le portage des STEC chez les ruminants. Le seul moyen de limiter la contamination du lait ou de la viande est de s'assurer d'une excellente hygiène lors de la traite ou de l'abattage des animaux. Une diminution du portage permettrait de diminuer les risques de contamination directe de l'homme par contact avec l'animal, ses produits dérivés ou via la consommation de végétaux souillés.
La présente demande est relative à une méthode pour diminuer les risques de contamination du lait ou de la viande issus de ruminants, lors de la traite ou de l'abattage des animaux porteurs de STEC. Les risques de contamination sont diminués en particulier en réduisant le portage de STEC par ces ruminants.
La présente invention est en particulier relative à une méthode pour diminuer le portage des STEC chez les ruminants, caractérisée par l'administration à un ruminant d'une ou plusieurs souches de l'espèce Hafnia alvei. Cette administration se fait aisément par voie orale, sous la forme d'un additif alimentaire.
DESCRIPTION DETAILLEE DE L'INVENTION
L'invention concerne une méthode pour diminuer le portage des STEC chez les ruminants, en particulier des sérotypes 0157:1-17 et 026:1-11 1 , caractérisée par l'administration à un ruminant d'au moins une souche de Hafnia alvei.
Le terme « portage » ou « portage animal » désigne le fait que les bactéries STEC résident de façon naturelle dans l'organisme des animaux qui les « portent », sans leur causer de préjudice, notamment sans qu'aucun signe clinique de maladie ne soit décelable, bien que les fèces des animaux comprennent ces STEC. Bien que les animaux portent les bactéries STEC, la présente invention n'est pas relative à une méthode de traitement des animaux, mais à une méthode pour diminuer la quantité de STEC présentes dans les poches intestinales de ces animaux, et ainsi réduire le risque de contamination du lait lors de la traite, de la viande lors de l'abattage desdits animaux ou de leurs fèces.
Le terme « STEC » désigne des bactéries Escherichia coli producteurs de Shiga-toxines ; ce groupe (tel que défini dans le rapport de l'AFSSA de 2003) comprend en particulier les sérogroupes 0157:1-17 et 026:1-11 1. Le terme « ruminants » désigne des animaux herbivores capables de régurgiter de la nourriture afin de la remastiquer, autrement dit de pratiquer la rumination. Par ruminants, on comprend essentiellement les animaux du sous-ordre des Ruminantia, et notamment les bovins, ovins et caprins. La présente invention concerne en particulier les bovins de rente.
Le terme « administration » désigne à la fois une administration par voie orale, par voie intraveineuse ou intramusculaire. La présente invention est néanmoins essentiellement relative à une administration par voie orale.
Le terme « souche(s) de Hafnia alvei » désigne toute souche, ou association de souches, appartenant à cette sous-famille des Hafnia, connue depuis les années 1960. Des souches Hafnia ont en particulier été isolées à partir de lait cru et de fromage. Deux grandes familles existent : les Hafnia alvei et les Hafnia paralvei. Certaines souches peuvent être virulentes (Abbott et al., 201 1 ). Naturellement, la présente invention se rapporte à l'utilisation de souche(s) d'Hafnia alvei non virulente(s), non-pathogénique(s).
Selon un aspect préféré de l'invention, la ou les souche(s) est (sont) comprise(s) dans un additif alimentaire destiné aux ruminants. Un « additif alimentaire » se distingue d'un aliment par le fait qu'il est utilisé à faible dose dans l'alimentation des animaux. Les additifs alimentaires destinés à l'élevage animal font l'objet d'une réglementation en Europe (règlement CEn°1831/2003 du Parlement et du Conseil). Les industriels peuvent dans certaines conditions ajouter à la nourriture des animaux :
- agents dits «additifs technologiques» ; conservateurs, antioxygènes, émulsifiants, stabilisants, épaississants, gélifiants, liants, substances pour le contrôle de contamination de radionucléides, anti-agglomérants, correcteurs d'acidité, additifs pour l'ensilage, dénaturants ;
additifs dits « sensoriels » ; colorants (dopant ou modifiant la couleur d'aliments pour animaux ; des substances qui, incluses dans l'alimentation des animaux donneront une couleur aux denrées alimentaires produites à partir de ces animaux. Certains pigments ayant un effet positif sur la couleur des poissons ou oiseaux d'ornement sont aussi autorisés) ;
- substances aromatiques ;
- additifs nutritionnels, dont vitamines, provitamines et « substances à effet analogue chimiquement bien définies », composés d'oligo-éléments, acides aminés (et leurs sels et « produits analogues », ainsi que de l'urée et ses dérivés ; additifs dits «zootechniques», autorisés comme améliorateurs de digestibilité, stabilisateurs de la flore intestinale, substances ayant un «effet positif sur l'environnement», et dVcautres additifs zootechniques».
Préférentiellement, l'additif alimentaire comprenant la (les) souche(s) d'Hafnia alvei sera mélangé à un aliment destiné aux ruminants. De préférence, la (les) souche(s) de Hafnia alvei est sous forme lyophilisée, et notamment sous forme de poudre, pour être additionnée aisément à cet aliment.
Selon un aspect particulier de l'invention, ledit aliment ou composition alimentaire ou additif alimentaire comprend au moins environ 108 UFC de Hafnia alvei par jour et par animal, avantageusement d'environ 108 UFC à environ 1012 UFC de Hafnia alvei par jour et par animal. Chez les bovins, la dose préférée est d'environ 1010 UFC par jour et par animal et chez les ovins et caprins, la dose préférée est d'environ 109 UFC par jour et par animal. Le terme « UFC » signifie « Unité Formant Colonie » et est l'unité de mesure généralement reconnue par l'homme du métier pour quantifier les bactéries capables de fonder une colonie.
En particulier, la composition alimentaire pourra comprendre d'autres additifs alimentaires choisis parmi les additifs cités précédemment, et/ou parmi les principes actifs suivants : des hormones et/ou agents anti-inflammatoires et/ou des agents bactéricides et/ou des agents antifongiques.
Selon un aspect de l'invention, la souche de Hafnia alvei administrée à un ruminant est choisie parmi les souches déposées auprès de la DSMZ sous les numéros d'accès 30098, 30099 ou 30163.
Selon un aspect préféré de l'invention, la souche de Hafnia alvei administrée à un ruminant est la souche B16 déposée à la CNCM sous le numéro CNCM 1-4710 en date du 20/12/2012. Cette souche, précédemment dénommée 'CC16', a été isolée en 2009 à partir d'un fromage livarot (Mounier et a/., 2009).
Selon un aspect spécifique de l'invention, la souche de Hafnia alvei est administrée en combinaison avec au moins une autre souche de microorganisme, et notamment une autre souche de Hafnia alvei et/ou une levure. Dans le cas d'une combaison avec une levure, la levure est en particulier la souche de Saccharomyces boulardii déposée sous le numéro d'accès 1-1079 à la CNCM.
Selon un autre aspect spécifique de l'invention, plusieurs souches de Hafnia alvei sont associées pour être administrées à un ruminant dans le cadre de la méthode selon l'invention. L'invention est également relative à une composition comprenant une souche d'Hafnia alvei en combinaison avec au moins une autre souche de microorganisme. En particulier, cette autre souche de microorganisme est une autre souche de Hafnia alvei et/ou une levure. De manière préférée, cette levure est de l'espèce particulière Saccharomyces boulardii. En particulier la souche de Saccharomyces boulardii présente dans la composition est la souche déposée sous le numéro d'accès 1-1079 à la CNCM.
En particulier, la souche de Hafnia alvei présente dans ladite composition est la souche B16 déposée sous le numéro d'accès 1-4710 à la CNCM, en association avec la souche de Saccharomyces boulardii déposée sous le numéro d'accès 1-1079 à la CNCM.
Selon un autre mode de réalisation de l'invention, la composition comprend une souche de Hafnia alvei, notamment l'une des souches déposées sous le numéro d'accès 30098, 30099 ou 30163 auprès de la DSMZ, et la souche de Saccharomyces boulardii déposée sous le numéro d'accès 1-1079 à la CNCM.
Selon un autre mode de réalisation de l'invention, la composition comprend de plus au moins une autre souche de microorganisme. Cet autre microorganisme peut être en particulier une souche de Hafnia alvei, notamment l'une des souches déposées sous le numéro d'accès 30098, 30099 ou 30163 auprès de la DSMZ, ou tout autre microorganisme approprié.
Selon un autre mode de réalisation, la composition comprend la souche B16 déposée sous le numéro d'accès 1-4710 à la CNCM, au moins une souche déposée sous le numéro d'accès 30098, 30099 ou 30163 auprès de la DSMZ, et la souche de Saccharomyces boulardii déposée sous le numéro d'accès 1-1079 à la CNCM.
L'invention est également relative à une utilisation de l'une des compositions telles que définies ci-dessus, comme additif alimentaire pour des ruminants. Cette composition sera préférentiellement ajoutée à un aliment pour les animaux. Cette composition sera utilisée pour diminuer le portage des STEC chez les animaux ruminants, nourris avec cette composition.
Enfin, l'invention concerne l'utilisation d'une souche de Hafnia alvei, seule ou en combinaison avec d'autres souches de microorganismes, pour diminuer le portage des STEC chez les ruminants.
Le portage des STEC chez les ruminants est évalué par des méthodes classiques, bien connues de l'homme du métier. En particulier, l'importance du portage in vivo est évaluée en mesurant la prévalence des STEC présentes au sein d'un troupeau, et le taux et/ou la durée d'excrétion des STEC dans les fécès des animaux, tel que décrits dans (Tabe et al., 2008) par exemple. La méthode selon l'invention permet d'obtenir une diminution de l'excrétion fécale d'au moins 10% par rapport à des ruminants n'ayant pas absorbé au moins une souche d'Hafnia alvei ; préférentiellement la diminution est d'au moins 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, 100%, 120%, 150%, 200%, et même de 300%. De manière préférée, la prévalence des STEC et en particulier des sérotypes 0157:1-17 et 026:1-11 1 chez les animaux auxquels au moins une souche d'Hafnia alvei a été administrée est diminuée d'environ 30% par rapport à la prévalence des STEC chez des animaux n'ayant pas été traités selon la méthode de l'invention. L'effet d'inhibition de croissance des STEC par une souche d'Hafnia alvei peut également être démontré in vitro, par croissance des souches dans un jus iléal bovin, tel que cela est présenté dans les exemples ci-dessous.

EXEMPLES
Exemple 1. Effet de Hafnia alvei B16 et d'autres souches d'Hafnia alvei sur la croissance d'E. coli 0157:H7 FCH6 en jus iléal bovin
Les incubations in vitro ont été réalisées en jus iléal obtenu chez deux vaches canulées recevant une ration (80/20 respectivement) de foin de prairie naturelle et de concentré [concentré INRA 863-50183 GV, comprenant pulpe de betteraves, blé, orge, tourteau de colza, tourteau de soja, mélasse de canne à sucre, phosphate bicalcique, sel, magnésie, vitamines, provitamines et oligo-éléments]. Les tubes contenant le jus iléal ont été incubés pendant 5 heures à 39°C, sans agitation, en présence des souches d'intérêt.
L'effet inhibiteur potentiel de chaque souche candidate a été tout d'abord testé sur une souche 0157:1-17 (souche FCH6), rendue résistante à la rifampicine. Le dénombrement de la souche FCH6 avant et après 5 heures d'incubation a été réalisé sur milieu LB Agar supplémenté en rifampicine.
Dans notre étude, un ajout de 500 μΙ d'une culture de 24h d'Hafnia alvei B16 en milieu BHI (non agité) dans 5 ml de jus iléal inoculé avec 104 ufc/ml de FCH6 Rif a permis de réduire de façon significative (P<0.001 ) et reproductible la croissance de la souche STEC en 5 heures d'incubation (-0.55 Log de population en moyenne sur 11 incubations indépendantes, voir figure 1 ). Les autres souches d'Hafnia alvei testées (souches DSMZ 30098, 30099 et 30163) présentent également un effet inhibiteur.
Les résultats présentés en figure 1 indiquent que la souche Hafnia alvei B16, présente une bonne capacité antagoniste de la croissance de la souche E. coli 0157:1-17.
Exemple 2. Effet de Hafnia alvei B16 et d'autres souches d'Hafnia alvei sur la croissance d'E. coli 026 :H11 21765 en jus iléal bovin
Des résultats comparables ont été obtenus avec une autre souche STEC de sérotype 026 :H11 (souche 21765 RifR). Une baisse de croissance significative (P<0.05) de -0.66 Log de population (moyenne sur 9 incubations indépendantes, figure 2) a été en effet mesurée en 5 heures d'incubation en jus iléal bovin avec la souche B16. Les autres souches de Hafnia alvei testées (souches DSMZ 30098, 30099 et 30163) présentent également un effet inhibiteur sur la souche 21765.
Exemple 3. Effet de Hafnia alvei B16 en combinaison avec une souche de levure sur la croissance d'E. coli 0157 :H7 FCH6 et E. coli 026 :H11 21765 en jus iléal bovin.
Les conditions expérimentales sont identiques à celles décrites dans l'exemple 1 , à l'exception de ce qui suit : les souches inhibitrices sont rajoutées à la quantité de 250 μΙ chacune, et Saccharomyces boulardii est préalablement cultivé en bouillon au malt pendant 24 h à 30°C, sous agitation douce. Les résultats sont présentés en figure 3 : pour la souche FCH6, la diminution de la croissance est de presque 1 log (diminution d'un facteur 10) ; pour la souche 21765, la diminution de la croissance est de -0.5 log.
Exemple 4. Effet d'une combinaison de deux souches de Hafnia alvei (B16 et 30098) sur la croissance d'E. coli 0157:1-17 FCH6 en jus iléal bovin.
Les conditions expérimentales sont identiques à celles décrites dans l'exemple 1 , les deux souches sont ajoutées à raison de 250 μΙ chacune.
Les résultats sont présentés en figure 4 : pour la souche FCH6, la diminution de la croissance est de 1 log (diminution d'un facteur 10) en présence de la combinaison des deux souches d'Hafnia alvei.
Exemple 5. Vérification de la survie de la souche H. alvei B16 dans le contenu iléal in vitro
La survie de la souche Hafnia alvei testée dans les exemples 1 à 4 ci-dessus (B16) dans le contenu iléal in vitro comprenant la flore intestinale endogène a été vérifiée, notamment avec 108 CFU/ml d'Hafnia alvei. Les conditions expérimentales sont identiques à celles décrites dans l'exemple 1 , à l'exception de l'inoculation avec les souches E.coli.
Les résultats obtenus montrent que la quantité de Hafnia alvei B16 se maintient au même niveau dans le jus intestinal in vitro pendant plus de 5 heures (résultats non représentés).

Claims

REVENDICATIONS
1. Méthode pour diminuer le portage des STEC chez les ruminants, caractérisée par l'administration à un ruminant d'au moins une souche de Hafnia alvei.
2. Méthode selon la revendication 1 , caractérisée en ce que ladite souche est comprise dans un additif alimentaire destiné aux ruminants.
3. Méthode selon la revendication 1 ou 2, caractérisée en ce que ladite souche de Hafnia alvei est administrée à une quantité d'au moins environ 108 UFC par jour et par animal.
4. Méthode selon la revendication 3, caractérisée en ce que ladite souche de Hafnia alvei est administrée chez les bovins à une quantité d'environ 1010 UFC par jour et par animal.
5. Méthode selon l'une des revendications 1 à 4, caractérisée en ce que la souche de Hafnia alvei est la souche B16 déposée à la CNCM sous le numéro 1-4710.
6. Méthode selon l'une des revendications 1 à 5, caractérisée en ce que ladite souche est administrée en combinaison avec au moins une autre souche de microorganisme.
7. Méthode selon la revendication 6, caractérisée en ce que l'autre souche est choisie parmi une autre souche de Hafnia alvei et/ou une levure.
8. Méthode selon la revendication 7, caractérisée en ce que la levure est la souche de Saccharomyces boulardii déposée sous le numéro d'accès 1-1079 à la CNCM.
9. Composition comprenant une souche d'Hafnia alvei en combinaison avec une souche de Saccharomyces boulardii.
10. Composition selon la revendication 9, caractérisée en ce que la souche d'Hafnia alvei est la souche B16 déposée à la CNCM sous le numéro 1-4710, et la souche de Saccharomyces boulardii est la souche déposée sous le numéro d'accès 1-1079 à la CNCM.
1 1. Composition selon l'une des revendications 9 ou 10, comprenant de plus au moins une autre souche de microorganisme.
12. Utilisation de la composition selon l'une des revendications 9 à 1 1 comme additif alimentaire pour des ruminants.
