- the "Single Cell Gel Electrophoresis” test is a fast agarose microgel electrophoresis technique that allows the visualization of single and double-strand DNA breaks on individual cells.
- the cells After treatment, the cells are trapped in an agarose gel and lysed in a high salinity buffer containing detergents.
- the DNA is then denatured by alkaline bath followed by a short electrophoresis, and is then evidenced by propidium iodide.
- the DNA of an altered cell stretches toward the anode in proportion to the number of breaks, taking more or less the shape of a comet.
- the highly degraded DNA is found in the "tail" of the comet. A intact cell remains round, the remaining DNA compacted at the level of the "head" of the comet.
- DNA damage is assessed using software that determines the percentage of DNA degradation.
- the results show that nutrient-starved cells are stressed and their DNA degraded.
- the results demonstrate that DNA protection of cells treated with cotton fiber extract is increased by 45% over control cells (i.e. cells not treated with extract but having suffered stress). The cotton fiber extract therefore plays an important role in the protection of DNA.
- Example 6 Hydration test on volunteers (Clinical test)
- the cotton fiber extract according to Example 1 was administered topically at 1.5% in a standard cosmetic formulation to obtain a cosmetic composition. Only one application was performed by an expert.
- the skin was observed by a non-invasive technique, using a Vivascope TM.
- the quantitatively evaluated criteria were the thickness of the stratum corneum and the thickness of the stratum granulosum. Result: A significant decrease in the thickness of the stratum corneum was observed after 3 hours of application of the cosmetic composition comprising the cotton fiber extract.
- the cotton fiber extract according to Example 1 was administered topically at 1.5% in a standard cosmetic formulation to obtain a cosmetic composition. Only one application was performed by an expert.
- the evaluation method used is the CM825 Corneasometer.
