Estado de la técnica anterior
Es bien sabido que la oxidación de las membranas celulares lipídicas es uno de los eventos farmacológicos más importantes que tienen lugar durante el ictus, que conduce y se traduce en la muerte del tejido cerebral, y neuronas (Brouns, R.; De Deyn, P. P. The complexity of neurobiological processes in acute ischemic stroke. Clin. Neurol. Neurosurg. 2009, 111, 483-495). Consecuentemente, una de las áreas de investigación más activas para el tratamiento de la isquemia cerebral, o el ictus, se centra en la búsqueda de nuevos agentes permeables de potente capacidad antioxidante y fuerte acción neuroprotectora, capaces de bloquear el diverso tipo de radicales libres oxigenados (ROS), responsables del estrés oxidativo que produce el ictus, una grave patología en rápido aumento en nuestras sociedades avanzadas, para la que no existe un tratamiento eficiente, y que es la cuarta causa de mortalidad después del cáncer, enfermedades coronarias, y Alzheimer (Chan, P. Es en este contexto donde los compuestos orgánicos del tipo nitrona, por su estructura y propiedades, han jugado un papel clave en los últimos treinta años, aunque desgraciadamente su presunta actividad beneficiosa en los numerosos ensayos clínicos a los que se han sometido una amplia gama de las mismas, dista mucho de estar contrastada (Floyd, R. Ejemplos típicos de esta familia de compuestos son:
(Z)-iV-[(2-Cloro-6-metilquinolin-3-il)metileno]-2-metilpropan-2-amina óxido (1), y
(Z)-A7-[(2-Cloro-6-metilquínolin-3-il)metileno]- 1 -fenilmetanamína óxido (2), preparados a partir del aldehido comercial, 2-cloro-6-metilquínolina-3-carbaldehído, por reacción con er -butil hydroxilamina, y ALbencilhidroxilamma comerciales, de acuerdo con el método A o B (ver esquema), para rendir la nitronas deseadas con un rendimiento del 28% y 64%, respectivamente.

Los cálculos teóricos han demostrado que las nitronas 1 y 2 son permeables, y presentan alta capacidad de neuroprotección (tabla 1).

La invención se ilustra con los siguientes ejemplos, que no limitan el alcance de la misma.
Métodos Experimentales
Los puntos de fusión fueron determinados en un equipo Koffier y no están corregidos. Los espectros de 1H NMR y 1 C NMR fueron obtenidos a temperatura ambiente, a 300, 400 ó 500 MHz y a 75, 100 o 125 MHz, respectivamente, utilizando CDC13 ó DMSO-dó como disolventes y los picos de estos disolventes deuterados como referencias internas (CDC13: 7.27 (£>), 77.2 (C) ppm; D20: 4.60 ppm y DMSO-d6: 2.49 (D). 40 (Q). La asignación de los desplazamientos químicos de los compuestos están determinados de acuerdo con los datos obtenidos en experimentos de RMN estándar ((¾, 13C-DEPT, 1H-COSY, gHSQC, gHMBC). Los análisis de espectrometría de masas se llevaron a cabo en un equipo de GC/MS con una fuente de ionización del tipo API-ES. Los microanálisis se realizaron en el CQO (CSIC, Madrid). La cromatografía de capa fina se llevó a cabo en placas de silicagel F254 y para su visualización se utilizó luz ultravioleta o los reveladores ninhidrina, anísaldehído y ácido fosfomolíbdico-H2S04. Todas las reacciones se realizaron empleando disolventes secos. Las columnas de cromatografía se llevaron a cabo
utilizando silicagel de 0,06 mm (230 mesh). Determinación de la capacidad antioxidante: Inhibición de la peroxidación lipídica. La producción del dieno hidroperóxido conjugado por oxidación del ácido linoleico en una dispersión acuosa se monitorizó a 234 nm. Se usó AAPH como radical libre iniciador. 10 μΐ, de una solución acuosa de la sal sódica de ácido linoleico (16 mM) se añadieron a una cubeta de UV conteniendo 0.93 mL de solución de un tampón fosfato (0.05 M) a pH 7.4, a 37 °C. La oxidación se inició a 37 °C, al aire, por la adición de 50 de una solución de AAPH 40 mM. La velocidad de la oxidación a 37 °C se determinó midiendo el incremento en la absorción a 234 nm producida por el dieno hidroperóxido conjugado. Determinación de la capacidad antioxidante por el método del radical DPPH. El ensayo se realizó en cubetas de 96 pocilios, con 200 GL en cada uno. Una solución en metanol de DPPH (1.35xl0~4 M, concentración final) (2.7 mL) se mezcló con las nitronas a diferentes concentraciones (0, 100, 200, 300, 400 and 500 ΠΜ, concentración final) (0.3 mL). La mezcla resultante se incubó por 2 h a temperatura ambiente. En ese momento, 200 G L se inyectaron en los 96 pocilios de la microplaca, y se midió la absorbancia del radical DPPH a 517 nm. La actividad antioxidante se determinó como RSA%. Competición de las nitronas con DMSO por el radical hidroxilo. Los radicales hidroxilo se han generado en el sistema Fe3+/ácido ascórbico por medida del formaldehído producido por la oxidación del DMSO. La mezcla de reacción contenía EDTA (0.1 mM), Fe3+ (167 μΜ), DMSO (33 mM) en un tampón fosfato (50 mM, pH 7.4), las nitronas (O. lmM) y ácido ascórbico (10 mM). Después de 30 min de incubación, a 37 °C, la reacción se paró por adición de CCI3COOH (17 % w/v). Atrapamiento del anión radical superóxido. El anion-radical superóxido se ha generado por el sistema xantina-xantína oxidasa, y se midió con el método del
NBT. A la mezcla de reacción se añadió un tampón fosfato a pH 7.4 (0.1M) conteniendo xantina, NBT y las nitronas (0.1 mM, concentración final), xantina oxidasa 40 μΐ (1.4 mL/ 60 mg, 0.07U/mL). Después de incubar por 10 min a temperatura ambiente, la absorbancia se registró a 560 nm. Inhibición de la lipoxigenasa de soja in vitro. Las nitronas se disolvieron en DMSO y se incubaron con linoleato de sodio (0.1 mL) y 0.2 mL de una solución del enzima (O. lmg/mL). La conversión del linoleato de sodio en ácido 13-hídroperoxilinoleico a 234 nm se midió y comparó con NDGA. Las nitronas se añadieron al sustrato a diferentes concentraciones. Las reacciones empezaron por adición de la misma cantidad del enzima a cada solución del sustrato conteniendo las nitronas.
Ejemplo 1. Procedimiento general para la síntesis de las nitronas. Método A. A una disolución del aldehido en EtOH (0.14M) se añadió metil-2-nitropropano (2 equiv) y zinc (3 equiv). La mezcla de reacción se enfrió a 0 °C, y AcOH glacial (6 equiv) se añadió gota a gota, se agitó a temperatura ambiente durante 14 h, y a 0 °C durante 2 h. El precipitado se filtró, lavó con EtOH, y purificó por cromatografía en columna. Método B. En un tubo de vidrio (20 mL), equipado con un septum, el aldehido, Na2S04 (3 equiv) y trietilamina (2 equiv) se suspendieron en THF seco. A continuación, se añadió el hidrocloruro de hidroxilamina (2 equiv), la mezcla se agitó 30 sec, y se irradió en un aparato de microondas (250 W) a 90 °C por 1 h. Siguiendo el Método B, la reacción de 2-cloro-6-metilquinolina-3- carbaldehído (200 mg, 0.97 mmol), Na2S04 (410 mg, 2.92 mmol), Et3N (0.30 mL, 1.95 mmol) e hidrocloruro de N-benycilhidroxilamina (310 mg, 1.95 mmol) en THF (4 mL), después de column cromatografía en columna (hexano/EtOAc, 7:3), dió la nitrona 2 (192 mg, 64%): pf 154-6°C; IR (KBr) vmax 3436, 2918, 1569, 1555, 1429,

Claims
Reivindicaciones 1. Derivados de quinolilnitronas de fórmula I

donde, X representa halógeno, grupos amino substituidos o no, alcoxi como metoxi, etoxi, benciloxi, o tiofenilo.
R1, R2 y R3 representan independientemente un radical alquilo C 1-C6 sustituido o no, grupo fenilo, o anillo aromático con uno o varios heteroátomos, y sustituido por grupos hydroxilo, metoxi, trifluorometilo, nitro, ciano, carbaldehído, carboxílico, ester carboxílico, halógeno, o radical alquilo C 1-C6, sustituido o no, y que se encuentran en las posiciones C2', 3' ó 4' del anillo bencénico, o heterocíclico de cinco o seis eslabones, tal como ñirano, tiofeno, pirrol, piridina, píracina, píridacina, pírimidina, indol.
2. Derivados de quinolilnitronas de fórmula I, según la reivindicación 1, que son: (Z)-A-[(2-Cloro-6-metilquinolin-3 1)metileno]-2-metilpropan-2-amina óxido (1),
(Z)-N-[(2-Cloro-6-metilquinolin-3-il)metileno]- 1 -fenilmetanamina óxido (2)
3. Procedimiento para la preparación de quinolilnitronas de fórmula I, según la reivindicación 1, que consiste en hacer reaccionar N-alquil hidroxilaminas II

H II donde R1 representa un radical alquilo C1-C6 sustituido o no, grupo fenilo, o anillo aromático con uno o varios heteroátomos, y sustituido por grupos hydroxilo, metoxi, trifluorometilo, nitro, ciano, carbaldehído, carboxílico, ester carboxílico, halógeno, o radical alquilo C1-C6 sustituido o no, y que se encuentran en las posiciones C2', 3' ó 4' del anillo bencénico, o heterocíclico de cinco o seis eslabones, tal como furano, tiofeno, pirrol, piridina, piracina, piridacina, pirimidina, indol.

4. Composiciones farmacéuticas que comprenden uno o más de los productos de formula I, junto con excipientes farmacéuticamente apropiados, así como sus isómeros, hidratos, sales, polimorfos, y pro-fármacos farmacéuticamente aceptables.
5. Uso de quinolilnitronas de fórmula I, según la reivindicación 1, para la preparación de un medicamento o formulación farmacéutica destinada al tratamiento del ictus, isquemia cerebral, enfermedades neurodegenerativas, tales como la enfermedad de Alzheimer y de Parkinson, o la esclerosis lateral amiotrófica.
