【배경기술】
<2> 일반적으로 만성 피부 염증은 표피의 탄력을 저해하므로 주름살 생성의 주요 원인이 된다. 또한 피부에 염증이 생긴 후에는 멜라닌 색소의 침착이 증가하여 염 증 부위가 질은 갈색으로 변하기도 한다. 이와 같이 염증 반응은 피부 노화를 촉진 한다. 한편, 노화된 피부는 면역력, 특히 피부 세포의 면역력이 저하되어 있어 각 종 피부 감염에 취약하며, 혈청 내 자가 항체의 발생 빈도가 높아 자가 면역 피부 질환의 빈도도 높아진다. 또한 랑게르한스 세포의 수는 젊은 사람의 피부 보다 내 인성 노화가 일어난 피부에 더 적으며, 광노화가 일어난 피부에는 더욱 더 적다. 따라서 노화된 피부는 약한 자극에도 염증이 발생할 가능성이 높으며, 발생한 염증 은 또 다시 노화를 야기하므로 악순환이 계속된다.

<18> 본 명세서에서 "피부 "라 함은, 동물의 체표를 덮는 조직을 의미하는 것으로 서, 얼굴 또는 바디 등의 체표를 덮는 조직뿐만 아니라, 두피와 모발을 포함하는 최광의의 개념이다.
<19> 본 명세서에서, "염증성 피부 노화"는 염증에 의해 야기되거나, 염증을 동반 하는 피부 노화를 포함한다.

<22> 염증 반웅에 의해 생성되는 TNF— α와 IL— 1β 등과 같은 사이토카인
(cytokine)은 혈관 내피 (vascular endothelium)에 존재하는 수용체에 결합하여 여 러 염증 관련 신호 경로 (signaling pathway)를 활성화시킨다. 또한 이 때 세포 부 착 분자 (cell adhesion moleculre)의 발현이 증가되어 염증성 세포들의 분출 (extravasation)이 일어나게 된다. 이러한 염증성 세포의 침투 (inf i ltrat ion) 후 여러 사이토카인이나 케모카인 등의 케모ᅳ어트랙티브 활성 (chemo-attractive activity)이 있게 되고 이들이 염증이 일어난 부위로 이동하면서 면역 반웅을 일으 키게 된다.
<23> 이러한 염증성 사이토카인들은 매트릭스 메탈로프로테이나아제 (Matrix
Metalloproteinase)의 발현을 촉진하여 콜라겐 분해를 유도하므로 피부 노화를 일 으키는데 결정적인 역할을 하며 생성된 사이토카인은 다시 수용체에 결합하여 연쇄 반웅을 일으키게 된다. 따라서 임의의 물질이 염증성 사이토카인의 생성을 억제하 고, 나아가 염증성 사이토카인에 의한 MMP-1의 생성을 억제할 수 있다면 염증성 피 부 노화 억제 물질로 작용할 수 있다고 판단할 수 있을 것이다.

따라서 염증 자극원을 처리한 면역 세포, 보다 구체적으로는 염증 자극원을 처리한 면역 세포의 배지는 다양한 염증성 사이토카인을 포함하게 된다. 이러한 면역 세포 를 피부 세포에 처리하면 염증성 사이토카인에 의해 피부 세포에서 피부 콜라겐의 분해를 촉진하는 MMP-1의 발현아촉진된다. 상기 살펴본 바와 같이, 염증 및 MMP-1 의 생성은 괴부 노화의 주요 원인이며, 일단 생성되면 계속 연쇄 반웅이 일어난다 는 점에서 더욱 좋지 않다. 따라서 후보 물질을 피부 세포에 전처리하고 이후 염증 성 사이토카인을 포함하는 면역 세포를 처리한 다음 피부 세포의 MMP-1 농도를 측 정함으로써 후보 물질이 염증성 피부 노화를 억제할 수 있는 물질인지 여부를 판단 할 수 있다. 이는 일반적인 MMP-1의 생성을 억제하는 물질을 스크리닝하는 방법과 달리 염증성 사이토카인에 의한 MMP-1 생성을 억제하는 물질을 스크리닝하므로 후 보 물질이 염증성 노화 억제 물질인지 여부를 보다 정확하게 스크리닝할 수 있다.

<28> 본 발명의 일측면에서, 염증 자극원에 의해 생성되는 사이토카인은 다양한 염증성 사이토카인을 포함한다. 본 발명의 다른 일측면에서, 사이토카인은 GROa (CXCL1, Chemokine(C-X-C motif) ligand 1), I-309(CCL-1, chemokine(C-C motif) 1 igand 1), sICAM-KSoluble cell adhesion molecules-1) , IL(Interleukin)— 1β , IL-lra, IL-6, IL-8, IL-23, IP— 10, MCP-K monocyte chemoat tract ant protein-1), MIF(Macrophage migrat ion inhibitory factor) , MIP(Macrophage inflammatory protein)-l α , ΜΙΡ_1β, Sepr in El, RANTES( regulated on activation, normal T— eel 1 expressed, and secreted) 및 TNF( tumor necrosis factor)- α 중 하나 이상을 포함하나, 이에 제한되는 것은 아니다.
<29> 본 발명의 일측면에서, 면역 세포는 염증 자극원 처리에 의해 염증성 사이토 카인을 생성하는 세포를 포함한다. ^ 발명의 다른 일측면에서, 면역 세포는 THP-1 세포 (Human acute monocytic leukemia cell line)를 포함한다. 단핵식균세포들은 면역계를 구성하는 주요한 세포 집단 중 하나로, 식균 작용 (phagocytosis)을 하는 것으로 알려져 있다. 식균 세포 증 혈액에 있는 세포를 단구 (monocytes)라고 하며, 조직에 존재하는 세포를 대식 세포 (macrophage)라고 부른다. THP-1 세포는 인간 유 래 세포로, 조직에서 대식 세포로 분화될 수 있는 단핵구이며 배양 가능 기간이 길 어 세포 실험에 유리하다는 장점을 가지므로, 본 발명에 보다 적합할 수 있다.
<30> 본 발명의 일측면에서, 피부 세포는 정상 인간 피부 섬유 아세포 (normal human fibroblast, NHF)를 포함한다. 진피층에 분포하는 섬유 아세포는 창상 치유 에 중요한 역할을 하며, 조직 손상에 의한 염증 반응에 의해 수일 내에 빠르게 증 식한다. 섬유 아세포는 세포외기질 (extracellular matrix; ECM) 단백질의 주된 근 원이며, 육아조직 (granulation)을 형성하는 교원질 (col lagen)과 섬유결합소 (fibronectin)를 생산한다. 또한모세관 및 기타 혈관과 네트워크를 형성하여 염증 성 사이토카인에 빠르게 반웅할 수 있다. 따라서, 본 발명에 보다 적합할 수 있다.
<31> 본 발명의 일측면에서, 염증 자극원은 염증성 사이토카인을 생성할 수 있는 물질을 포함한다. 본 발명의 다른 일측면에서, 염증 자극원은 LPS(Lipopolysaccharide) 및 INF-γ (Interferon- γ )를 포함한다.
<32> LPS는 그람 음성 세균 표층의 펩티드글리칸을 둘러싸는 외막의 중요 구성 성 분이다. LPS는 NF-κΒ를 핵으로 이동시키며, NF-κΒ는 주로 염증 및 면역 반웅에 관여하는 여러 유전자의 프로모터 부위에 결합하여 이들 유전자를 발현시킨다. 따 라서 LPS는 사람에게서 면역 조절 물질, 염증 유발 물질 등을 자극하는 것으로 알 려져 있으며, 그 유래 균주에 따라 자극을 유도하는 정도가 다르다. 본 발명의 일 측면에서, LPS는 균 {Escherichia coli) 및 살모넬라 (5a½o/2e//a) 속균 중 하나 이상에서 유래한 LPS를 포함한다. 본 발명의 다른 일측면에서, 대장균에서 유래한 LPS는 E.coli 0111:B4, E.coli 055:B5 및 E.coli EH100 중 하나 이상에서 유래한 LPS를 포함한다. 본 발명의 또 다른 일측면에서, 살모넬라 속균에서 유래한 LPS는 살모넬라 티포사 (5. typhosa) 및 살모넬라 티피뮤리움 (5. typhi murium) 중 하나 이 상에서 유래한 LPS를 포함한다.
<33> 본 발명의 일측면에서, 세포 독성을 나타내지 않으면서도 염증성 사이토카인 의 충분한 생성이 이루어지도록 하기 위해, LPS는 0.001 μΜ 내지 20 μΜ, 구체적 으로 0.01 μΜ 내지 15 μΜ, 더 구체적으로 0.1 μΜ 내지 10 μΜ일 수 있다.
<34> 본 발명의 일측면에서, INF-Y (Interferon-Y)는 직접 염증 반웅을 매개하여 염증성 사이토카인의 생성을 촉진하기 보다는 LPS를 보조하여 LPS에 의한 염증 자 극 반웅을 강화하는 역할을 한다. 따라서, LPS와 INF-Y를 함께 처리하는 경우 염 증성 사이토카인이 보다 잘 생성될 수 있으며, 나아가 염증성 피부 노화 억제 물질 을 보다 민감하게 스크리닝할 수 있다.
<35> 본 발명의 일측면에서, 세포 독성을 나타내지 않으면서도 본 발명이 의도하 는 효과를 잘 나타내기 위해, INF-y는 1 내지 2000 UNITs, 구체적으로 5 내지 1500 UNITs , 더 구체적으로 10 내지 1000 UNITs일 수 있다.

<37> 본 발명의 일측면에 따른 염증성 노화 억제 물질 스크리닝 방법은 피부 세포 의 匪 P— 1 농도를 측정하는 단계 이후, 후보 물질을 처리한 피부 세포의 MMP-1의 농 도가 후보 물질을 처리하지 않은 피부 세포의 MMP-1의 농도보다 낮은 경우, 후보 물질을 염증성 노화 억제 물질이라고 판단하는 단계를 더 포함할 수 있다. 본 발명 의 다른 일측면에서, 후보 물질을 처리한 피부 세포의 MMP-1의 농도가후보 물질을 처리하지 않은 피부 세포의 ffiP-1의 농도보다 높은 경우, 후보 물질을 염증성 노화 억제 물질이 아니라고 판단하는 단계를 더 포함할 수 있다. 본 발명의 또 다른 일 측면에서, 후보 물질을 처리한 피부 세포의 MMP-1 농도가 후보 물질을 처리하지 않 은 피부 세포의 匪 P-1의 농도보다 낮을수록 염증성 노화 억제 효과가 높은 물질이 라고 판단하는 단계를 더 포함할 수 있다. <38> 본 발명의 일측면에서, 후보 물질은 염증성 노화 억제 물질인지 여부를 판단 하는 대상 물질을 의미한다. 본 발명의 다른 일측면에서, 후보 물질은 식물을 예로 들 수 있는 천연물 및 그 추출물 또는 화학 합성물을 모두 포함할 수 있다.

<42> [실험예 1] 자극원으로 적절한 LPS 균주의 확인
<43> LPS는 그 유래 균주에 따라 자극을 유도하는 정도가 다르므로, 염증성 피부 노화 억제 물질 스크리닝에 적절한 LPS를 선택하고자 아래와 같은 실험을 실시하였 다.
<44> THP-1 세포 (한국 세포주 은행에서 입수)를 6-웰에 5xl05개씩 처리하고 37°C,
5% C02 조건 하 10% (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 /ml를 포함하는 RPM 1-1640 배지 (L0NZA에서 입수)에서 24시간 배양시켰다. 이에 0, 0.1 및 1 /g/ml의 대장균에서 유래한 LPS 3종 (각각 E.coli 0111:B4, E.coli 055 :B5 및 E.coli EH100에서 유래) 및 살모넬라 속균에서 유래한 LPS 2종 (각각 살모넬라 티포 4(5. y sa)와 살모넬라 티피뮤리움 . typhimurium예 ^ 유래) (시그마 알드리 치에서 입수)과 0, 250,- 500 및 1000 UNITs의 INF-γ (시그마 알드리치에서 입수)를 처리하고 24시간 배양하였다. 이후 배지를 lOOOrpm으로 3분 동안 원심 분리하고 상 등액을 취한 뒤 TNF- α EL ISA 키트를 이용하여 세포 배양액에 포함되어 있는 TNF- a의 농도를 구했으며, 이를 도 1에 나타내었다.
<45> 도 1에서 볼 수 있듯이, LPS를 처리한 경우, E.coli 0111:B4, E.coli
055 :B5, E.coli EH100, 살모넬라 티포사, 살모넬라 티피뮤리움 순으로 TNF- a의 생 성량이 많았으며, 처리한 LPS 농도가 다른 경우에도 상기 순서는 유지되었다. 한 편, 0.1 //g/ml의 LPS를 처리한 경우보다 1 /ml의 LPS를 처리한 경우에 TNF- a의 절대적인 생성량이 많았다. 이를 통해, 대장균, 구체적으로 E.coli 0111:B4 유래 LPS가 염증성 사이토카인 생성에 있어 인간 면역 세포에 가장 민감함을 알 수 있 다.
<46> 또한, IFN-Y의 경우 농도에 상관없이 TNF- a의 생성에 영향을 미치지 않았 다. 하지만 .5종의 LPS 1 g/ml와 IFN-Y 500 UNITs를 함깨 처리한 경우에 LPS를 단 독으로 처리한 경우보다 TNF- a 생성량이 현저하게 증가하였으며, 그 생성량은 E.coli 0111:B4, E.coli 055:B5, E.coli EH100, 살모넬라 티피뮤리움, 살모넬라 티 포사 순으로 높았다. 이를 통해 IFN-Y가 LPS에 의한 자극을 강화함을 알 수 있다.

<48> [실험예 2] 자극원의 적정 농도 확인
<4 > 염증성 피부 노화 억제 물질 스크리닝에 적절한 자극원의 농도를 결정하고자 아래와 같은 실험을 실시하였다ᅳ
<50> THP-1 세포 (한국 세포주 은행에서 입수)를 96-웰에 3xl04개씩 처리하여 37
°C, 5% C02 조건 하 10% (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 m
/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)에서 24시간 배양시켰다. 이에 0, 0.01, 0.1, 1 및 10 //g/ml의 대장균 (E.coli 0111:B4) 유래 LPS (시그마 알드리치에 서 입수) , 0, 50, 100, 500, 및 1000 UNITs의 IFN-γ (시그마 알드리치에서 입수)를 흔합 처리하여 24시간 배양시켰다. 세포 계수 키트 시약을 웰 당 배지 부피의 1/10 씩 처리하고 2시간 배양한 후 스펙트라 맥스 (spectraMAX)로 O.D. 값을 측정하여 세 포 생육력 (viability)을 계산하였다. 이를 각각 도 2 내지 도 4에 나타내었다.
<5i> 도 2에서 볼 수 있듯이, 0, 0.01, 0.1, 1 및 10 /g/ml의 LPS를 처리한 경우 모두 세포 독성이 나타나지 않으며, 농도 의존적으로 세포 증식을 촉진한다. 도 3 에서 볼 수 있듯이 , 0, 50, 100, 500 및 1000 UNITs의 IFN-γ를 처리한 경우 모두 세포 독성이 나타나지 않는다. 또한 도 4에서 볼 수 있듯이 , LPS와 IFN-Y를 함께 처리한 경우 모든 농도에서 세포 독성이 나타나지 않으며, 세포 증식 정도는 LPS의 농도와 양의 상관 관계를 나타낸다.

<53> [실험예 3] 자극원 처리시 염증성 사이토카인 생성 확인
<54> 자극원 처리시 염증 반웅에 의한 염증성 사이토카인 생성을 확인하고자 아래 와 같은 실험을 실시하였다.
<55> THP-1 세포 (한국 세포주 은행에서 입수)를 6-웰에 5xl05개씩 처리하여 37°C,
5% C02 조건 하 10% (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 ig/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)에서 24시간 배양시켰다. 이에 1 //g/ml의 LPSCE.coli 0111:B4) (시그마 알드리치에서 입수)와 500 UNITs의 IFN-γ (시그마 알 드리치에서 입수)를 처리한 후 24시간 배양시켰다. 배지를 1000 rpni으로 3분 동안 원심 분리하여 상등액을 취한 뒤 사이토카인 분석 키트 (Cytokine array kit)를 이 용하여 세포 배양액 중 대조군에 비해 증가한 염증성 사이토카인의 종류를 확인하 였다. 도 5는 염증성 사이토카인의 발현 정도를 나타내며, 아래 표는 각 좌표에 해 당하는 염증성 사이토카인의 목록이다.

상기 결과에서 볼 수 있듯이, LPS와 IFN-Y 처리 후 THP-1 세포에서 총 16종 의 사이토카인, 즉 GROa, 1-309, sICAM-1, IL-Ιβ, IL-lra, IL-6, IL-8, IL-23, IP-10, MCP-1, MIF, MIP-1 a , MIP-1 β , Sepr in El, RANTES 및 TNF- a이 증가되었 다. 즉, LPS와 IFN-Y를 처리하는 경우, THP-1 세포에서 다양한 염증성 사이토카인 의 생성됨을 확인할 수 있다.
[실험예 4] THP-1 배양 배지에 의한 MMP-1 생성 확인
자극원을 처리한 THP-1 배양 배지에 의해 醒 P-1이 생성되는지 여부를 확인하 고자 아래와 같은 실험을 실시하였다.
THP-1 세포 (한국 세포주 은행에서 입수)는 10% (v/v) FBS, 페니실린 100 U/ml 및 스트랩토마이신 100 /g/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)를 포함하는 6-웰에 5xl05개씩, NHF(Normal human fibroblast) 세포 (L0NZA에서 입수)는
10% (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 /g/ml를 포함하는 DMEM 배지 (L0NZA에서 입수)를 포함하는 24-웰에 3xl04개씩 처리하여 37°C, 5% C02 조건에 서 24시간 배양시켰다. THP-1 세포에는 LPS(E.coli 0111:B4) (시그마 알드리치에서 입수) 1 /g/m 및 IFN-Y (시그마 알드리치에서 입수) 500 UNITs을 처리하거나 (자극 배지), 아무 것도 처리하지 않고 (무자극 배지), NHF 세포는 무혈청 배지로 교환하 여 24시간 배양시켰다. THP-1의 배지를 1000 rpm으로 3분 동안 원심 분리하여 상등 액을 취했다. NHF 세포에 무자극 THP-1 배지 및 자극 THP-1 배지를 처리한 것을 각 각 무자극 배지 처리군 및 자극 배지 처리군으로 하고, 세럼 -없는 (serum free) DMEM을 처리한 것을 무처리 군으로 하였다. MMP-1 분석 키트로 NHF 세포의 丽 P-l 농도를 측정하고, 무처리군 대비 무자극 배지 처리군과 자극 배지 처리군의 匪 P-1 의 농도비를 구하여 도 6에 나타내었다.
<63> 도 6에서 볼 수 있듯이, 무자극 배지 처리군의 경우 무처리군과 비슷한 농도 의 MMP-1 생성이 확인된다. 반면, 자극 배지 처리군의 경우 무처리군에 비해 3배 가량 높은 醒 Pᅳ 1 생성이 확인된다. 즉, 자극원을 처리한 THP-1 배양 배지에는 여러 염증성 사이토카인이 존재하므로 이에 의해 NHF 세포에서 MMP-1이 생성됨을 확인할 수 있다.

°C, 5% C02 조건 하 10% (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 g
/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)에서 24시간 배양시켰다. 스크리닝 후보 물질로 염증성 피부 노화 억제 물질로 알려진 0.001, 0.01, 0.1, 1 및 10 μΜ 의 글리시테인 (Glycitein)을 처리하였으며, 선택적으로 자극원인 LPS(E.coli 0111:B4) (시그마. 알드리치에서 입수) 1 /g/ml 및 IFN-γ (시그마 알드리치에서 입 수) 500 UNITs을 처리한 후 24시간 배양시켰다. 세포 계수 키트 시약을 웰 당 배지 부피의 1/10씩 처리하고 2시간 배양한 후 스펙트라맥스로 O.D. 값을 측정하고, 대 조군 대비 생육률을 도 7과 도 8에 나타내었다.
<68> 도 7에서 볼 수 있듯이 , THP-1 세포에서 글리시테인은 10 μΜ 이하의 농도에 서 세포 독성을 나타내지 않는다. 도 8에서 볼 수 있듯이, 글리시테인과 자극원인 LPS 및 IFN-Y를 함께 처리한 경우에도 세포 독성을 나타내지 않는다.

<7ΐ> 염증성 피부 노화 억제 스크리닝 후보 물질 및 자극원 처리시 TNF-α 생성을 확인하고자, 아래와 같은 실험을 실시하였다. <72> THP-1 세포 (한국 세포주 은행에서 입수)를 6-웰에 5x10개씩 처리함과 동시 에 0, 0.1, 1 및 10 μΜ의 글리시테인을 처리하였다. 1 (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 /g/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)에 서 37°C, 5% C02 조건 하 24시간 배양시킨 뒤 LPS(E.coli 0111:B4) (시그마 알드리치 에서 입수) 1 ig/ml 및 IFN-Y (시그마 알드리치에서 입수) 500 UNITs를 자극원으 로 처리하였다. 다시 24시간 배양시킨 뒤 배지를 1000 rptn으로 3분 동안 원심 분리 하고 상둥액을 취하였다. TNF-α EL ISA 키트를 이용하여 세포 배양액에 포함되어 있는 TNF-a의 농도를 구하고 이를 도 9에 나타내었다.
<73> 도 9에서 볼 수 있듯이, 글리시테인의 처리 농도가 높을수록 자극원에 의한
TNF-a의 농도가 낮아짐을 알 수 있다. 즉, TNF-a의 농도 측정을 통해 스크리닝 후보 물질의 염증 억제 효과 정도를 판단할 수 있다.

<75> [실험예 7] 스크리닝 후보 물질 및 자극원 처리시 염증성 사이토카인 생성 확인
<76> 염증성 피부 노화 억제 스크리닝 후보 물질 및 자극원 처리시 염증성 사이토 카인 생성을 확인하고자, 아래와 같은 실험을 실시하였다.
<77> THP-1 세포 (한국 세포주 은행에서 입수)를 6-웰에 5xl05개씩 처리함과 동시 에 글리시테인 10 μΜ을 처리 하였다. 10% (v/v) FBS, 페니실린 100 U/ml 및 스트 렙토마이신 100 g/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)에서 37°C, 5% C02 조건 하 24시간 배양시킨 뒤 LPS(E.coli 0111:B4) (시그마 알드리치에서 입수) 1 ig/ml 및 IFN-Y (시그마 알드리치에서 입수) 500 UNITs를 자극원으로 처리하고 다 시 24시간 배양시켰다. 한편, 아무 것도 처리하지 않거나 자극원만을 처리한 세포 를 대조군으로 하였다. 배지를 lOOOrpm으로 3분 동안 원심 분리하고 상등액을 취하 였다. 사이토카인 어레이 키트 (R&D 센터)를 이용하여 세포 배양액에 포함되어 있는 염증성 사이토카인을 확인하였다. 양성 대조군 대비 각 처리군의 사이토카인 양을 도 10에 나타내었다.
<78> 도 10에서 볼 수 있듯이, 자극원 처리에 의해 생성된 염증성 사이토카인들은 글리시테인에 의해 그 정도가 감소된다. 즉, 염증성 사이토카인의 농도 측정을 통 해 스크리닝 후보 물질의 염증 억제 효과 정도를 판단할 수 있다.

<80> [실험예 8] 스크리닝 후보 물질 및 자극원의 복합 처리시 적정 농도 확인 <81> 염증성 피부 노화 억제 스크리닝 후보 물질 및 자극원의 복합 처리시 그 적 정 농도를 확인하고자, 아래와 같은 실험을 실시하였다.
<82> NHF 세포 (L0NZA에서 입수)를 96-웰에 lxl()4개씩 처리하여 10% (v/v) FBS, 페 니실린 100 U/ml 및 스트랩토마이신 100 /zg/ml를 포함하는 DMEM 배지 (L0NZA에서 입 수)에서 37°C, 5% C02 조건 하 24시간 배양시켰다. 이에 자극원인 LPS(E.coli
0111:B4) (시그마 알드리치에서 입수) 1 zg/ml와 IFN-y (시그마 알드리치에서 입수) 500 UNITs, 및 글리시테인 0, 0.1, 0.5, 1, 5 및 10 μΜ을 농도별로 처리한 후 24 시간 배양시켰다. 세포 계수 키트 시약을 웰 당 배지 부피의 1/10씩 처리하고 2시 간 배양한 후 스펙트라맥스 (spectraMAX)로 O.D. 값을 측정하여 음성 대조군 대비 생육률 (viability)(¾)를 계산하여 도 11에 나타내었다.
<83> 도 11에서 볼 수 있듯이, 염증성 피부 노화 억제 물질인 글리시테인과 자극 원을 복합 처리하는 경우 세포 독성이 나타나지 않는다.

<87> THP-1 세포 (한국 세포주 은행에서 입수)는 10% (v/v) FBS, 페니실린 100
U/ml 및 스트렘토마이신 100 /g/ml를 포함하는 RPMI-1640 배지 (L0NZA에서 입수)를 포함하는 6-웰에 5χ·105개씩, NHF 세포 (L0NZA에서 입수)는 10% (v/v) FBS, 페니실린 100 U/ml 및 스트렙토마이신 100 /g/ml를 포함하는 DMEM 배지 (LONZA에서 입수)를 포함하는 24-웰에 3xl04개씩 처리하여 37°C, 5% C02 조건 하에서 24시간 배양시켰 다. THP-1 세포에 LPS(E.coli 0111:B4) (시그마 알드리치에서 입수) 1 /g/ml와 IFN- γ(시그마 알드리치에서 입수) 500 UNITs를 처리하여 자극 처리 배지로 하고, 자극 원을 처리하지 않은 배지를 무처리 배지로 하였다. NHF 세포에는 글리시테 을 무 혈청 배지로 10 μΜ씩 처리하여 24시간 배양시켰다. 상기 처리한 THP-1의 배지를 1000 rpm으로 3분 동안 원심 분리하여 상둥액을 취한 후 NHF 세포에 처리하고, 무 처리군에는 세럼 없는 (serum free) DMEM(L0NZA에서 입수)을 처리하였다. 24시간 배 양 후 MMP-1 분석 키트로 NHF 세포의 MMP-1 농도를 측정하고, 아무 것도 처리하지 않은 군 대비 MMP-1의 농도비를 도 12에 나타내었다.
<88> 도 12에서 볼 수 있듯이, 자극 처리 배지에서는 무처리 배지에 비해 3배 가 량 높은 匪 P-1이 생성된다. 또한 글리시테인을 처리하는 경우 자극 배지 처리에 의 한 MMP-1 생성이 감소된다. 어떠한 물질이 자극원에 의한 MMP-1 생성을 억제할 수 있다는 것은, 염증에 의한 콜라겐 분해를 억제할 수 있으며 나아가 염증성 피부 노화를 억제할 수 있음 을 의미한다. 즉', 염증성 피부 노화 억제 후보 물질을 처리하여 피부 세포의 匪 P-1 생성이 억제되는지 여부를 확인함으로쎄 그 후보 물질이 염증성 피부 노화를 억제 할 수 있는지 여부를 확인할 수 있다. 특히 이는 특정 염증성 사이토카인이 아닌 여러 사이토카인에 의한 MMP— 1 생성 억제 여부를 판단할 수 있다는 점에서 여러 사 이토카인에 기인한 염증성 피부 노화를 억제하는 물질을 한 번에 간편하고 효율적 으로 판단할 수 있다.

면역 세포에 염증 자극원을 처리하는 단계;
피부 세포에 후보 물질을 처리하는 단계;
후보 물질을 처리한 피부 세포에 염증 자극원을 처리한 면역 세포를 처리하 는 단계; 및
면역 세포를 처리한 피부 세포의 MMP-l(Matrix Metal loproteinase-1) 농도를 측정하는 단계를 포함하는 염증성 피부 노화 억제 물질 스크리닝 방법.

제 1 항에 있어서,
면역 세포는 THP-1 세포 (Human acute monocytic leukemia cell)를 포함하는 것을 특징으로 하는 염증성 피부 노화 억제 물질 스크리닝 방법.

피부 세포는 정상 인간 피부 섬유 아세포 (normal human fibroblast, NHF)를 포함하는 것을 특징으로 하는 염증성 피부 노화 억제 물질 스크리닝 방법.

염증 자극원은 LPS(Lipopolysaccharide) 및 INF- γ (Interferon- γ )를 포함하 는 것을 특징으로 하는 염증성 피부 노화 억제 물질 스크리닝 방법.
