시험 물질의 세포 재생 효과를 알아보기 위해 HaCaT 10000세포수/웰, 정상 인간 섬유아세포(normal human fibroblast) 6000 세포수/웰로 96-웰 플레이트에 심었다. 물질을 처리하기 전에 배지를 FBS가 첨가되지 않은 배지로 교환하고 1시간 동안 배양하였다. 이후 배지를 제거하고 FBS가 1% 함유된 배지에 상기 제조예 2의 발효 청국장 추출 분획물을 0.1, 1, 5, 10, 50, 100ppm의 농도별로 녹인 후 처리하였다. 이때 10% FBS가 함유된 배지는 양성대조군으로 사용하였다. 상기 제조예 2의 발효 청국장 추출 분획물 처리 후 48시간 동안 배양 후, WST-1을 100㎕/웰로 처리하였다. WST-1을 처리하고 3시간 동안 37℃ 5% CO2배양기에서 반응시켰다. 3시간 동안 배양 후 마이크로플레이트 리더기(microplate reader)를 이용하여 450nm에서 흡광도를 측정하여 세포 재생 효과를 확인하였다. 그 결과는 도 2 및 3에 나타내었다.
