Abstract
La presente invención se refiere a un material nanoestructurado compuesto de nanopartfculas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos y un agente bioactivo unido por un enlace covalente hidrolizable y, opcíonalmente, moléculas de un PEG, un agente fluorescente, un radionúcleo y/o una sustancia directora a receptores, células o tejidos específicos, unidas por enlace covalente, y su uso en el tratamiento o profilaxis de una determinada patología.
Description
NANOPARTÍCULAS DE SILICE PARA DIFUSIÓN INTRACELULAR DE AGENTES BIOACTIVOS POCO SOLUBLES
CAMPO TÉCNICO DE LA INVENCIÓN
La presente invención se enmarca dentro de las aplicaciones de las nanopartículas de sílice para nanotecnología y biotecnología. La reducida solubilidad de numerosos agentes bioactivos complica su difusión en fluidos biológicos, lo que limita su actividad terapéutica. La unión de una molécula bioactiva mediante un enlace covalente hidrolizable a una nanopartícula de sílice cuya superficie se encuentra modificada con grupos aniónicos, mejora la solubilidad y reduce su toxicidad in vitro e in vivo, a la vez que favorece la acumulación de la molécula bioactiva en tejidos patógenos por el efecto de permeabilidad de mejora y conservación (EPR).
ESTADO DE LA TÉCNICA ANTERIOR A LA INVENCIÓN
La camptotecina (20(S)-camptotecina; CPT) es un derivado de la quinolina con amplio espectro de actividad antitumoral en modelos animales (Ann. Oncol. 1997, 8, 837-855; Rothenberg, M.L.). La forma lactona es esencial para la actividad terapéutica. Por el contrario, la apertura del anillo de lactona origina el correspondiente carboxilato, el cual resulta inactivo. Desafortunadamente, la solubilidad de la lactona en medio acuoso es muy reducida, mientras que el carboxilato está favorecido a pH fisiológico.
La sustitución en posición 20-OH de la CPT estabiliza el anillo de lactona. Así, se ha unido CPT a través de este punto (20-OH) a polímeros solubles en agua como el poli(etilenglicol), poli-N-(2-hidroxipropil)meta- crilamida y ácido poli-L-glutámico. Sin embargo, sólo en determinados casos estos conjugados han permitido solubilizar la forma bioactiva del agente terapéutico, así como la preparación de formulaciones que liberen de forma
controlada el fármaco, evitando complicaciones asociadas con una elevada concentración sistémica del mismo.
En la solicitud WO2004/022099 2004; Cheng, J.; Davis, M.E.; Khin, K.T., se presenta un nuevo tipo de composición farmacéutica basada en polímeros de ciclodextrinas, capaces de transportar pequeñas moléculas con actividad terapéutica, como la CPT. Estos polímeros conjugados son solubles en agua y biocompatibles, y pueden incorporar la CPT. No obstante, en un estudio paralelo (Bioconjugate Chem. 2003, 14, 1007-1017; Cheng, J.; Khin, K.T.; Jensen, G.S.; Liu A.; Davis, M.D.) los mismos autores indican que el tiempo de vida media del polímero con CPT es inferior a 2 h en plasma humano, esto es, en menos de 2 h libera el 50% de la carga de fármaco antitumoral transportada, lo que puede limitar considerablemente su utilización en organismos vivos, ya que se reduce la posibilidad de acumulación de la droga en el tumor por efecto EPR.
En la solicitud WO2009/078924 2009, Liong, M.; Lu, J.; Tamanoi, F.; Zink, J.l.) describe un material basado en nanopartículas de sílice mesoporosa cuya superficie se encuentra cubierta de grupos aniónicos que confieren carácter hidrofílico a las estructuras submicrónicas, a la vez que provocan la aparición de fuerzas electrostáticas repulsivas entre partículas que permiten la preparación de coloides estables en medio acuoso. En el interior de los poros del material se pueden incorporar moléculas con actividad biológica, como la CPT y el paclitaxel. Estos se liberarían por simple difusión molecular a través de los poros. Esta difusión ocurrirá en su mayor parte en el plasma, produciéndose la liberación prematura del agente terapéutico. La principal limitación de este sistema es el escaso control sobre la liberación de los agentes farmacológicos, así como la ausencia de estímulos específicos que provoquen dicha liberación en los objetivos terapéuticos.
Todo lo anterior conduce a la necesidad de desarrollar nuevos sistemas para el transporte y liberación de sustancias bioactivas de baja solubilidad en fluidos biológicos, como CPT, paclitaxel y doxorrubicina, que reúnan las
siguientes propiedades: i) Biocompatibilidad, es decir, no provocar respuesta inmune inespecífica o específica; ii) Estricto control de la liberación del fármaco en función de un determinado estímulo fisiológico, p.e., la actividad de una enzima intracelular, un cambio de pH o la acción de sustancias reductoras intracelulares; iii) Estabilidad en plasma humano, evitando la liberación prematura del agente terapéutico antes de alcanzar los tejidos diana; iv) Elevada capacidad de carga y transporte de las moléculas terapéuticas, lo que permite alcanzar valores de DE5o con cantidades bajas del preparado farmacéutico.
En nuestro caso, hemos encontrado que, sorprendentemente, nanopartículas de sílice modificadas a las que se une el agente bioactivo mediante unión covalente y que disponen de un grupo químico capaz de hidrolizarse y liberar el fármaco bajo la acción de enzimas intracelulares, resultan extraordinariamente estables en plasma, liberándose muy preferentemente en el citosol de las células objetivo.
DESCRIPCIÓN DE LA INVENCIÓN
La presente invención se refiere a un material nanoestructurado basado en sílice que contiene agentes bioactivos y su utilización en aplicaciones biomédicas, por ejemplo, imagen, diagnóstico, tratamiento o profilaxis de una patología.
Un objeto de la presente invención es un material nanoestructurado que comprende al menos dos componentes:
- un soporte inorgánico, que comprende nanopartículas de sílice cuya superficie se encuentra cubierta de grupos aniónicos terminales, y
- un agente bioactivo unido por enlace covalente a un grupo aniónico terminal de la superficie del soporte inorgánico.
Según la presente invención, la sílice puede ser porosa o no porosa.
Según realizaciones particulares adicionales la sílice es no estructurada y no porosa. Según realizaciones particulares adicionales la sílice es no estructurada y porosa, con diámetro de poro comprendido entre 1 y 100 nm.
Una realización particular adicional de la presente invención es un material nanoestructurado que comprende al menos dos componentes:
- un soporte inorgánico, que comprende nanopartículas de sílice no estructurada y no porosa, cuya superficie se encuentra cubierta de grupos aniónicos terminales, y
- un agente bioactivo unido por enlace covalente a un grupo aniónico terminal de la superficie del soporte inorgánico.
Según otra realización particular adicional el material es un material nanoestructurado que comprende al menos dos componentes:
- un soporte inorgánico, que comprende nanopartículas de sílice no estructurada y porosa, con diámetro de poro comprendido entre 1 y 100 nm, cuya superficie se encuentra cubierta de grupos aniónicos terminales, y
- un agente bioactivo unido por enlace covalente a un grupo aniónico terminal de la superficie del soporte inorgánico. Otra realización particular adicional de la presente invención es un material nanoestructurado que comprende al menos dos componentes:
- Un soporte inorgánico, que comprende nanopartículas de sílice estructurada y porosa, con diámetro de poro comprendido entre 1 y 30 nm. Preferentemente dicho soporte es mesoporoso y la superficie se encuentra cubierta de grupos aniónicos terminales, y
- Un agente bioactivo unido por enlace covalente a un grupo aniónico terminal de la superficie del soporte inorgánico.
Otra realización particular adicional de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice estructurada y porosa y presenta orden a larga distancia. Preferentemente, según esta realización, el diámetro de poro está comprendido entre 1 y 30 nm. De modo más preferente aún, el material puede presentar una estructura tipo MCM-41 , MCM-48, MCM-50, FSM-16, HMS, KIT-1 , MSU-1 , MSU-2, MSU-3, MSU-V, SBA-1 , SBA-2, SBA-3, SBA-8, SBA-1 1 , SBA-12, SBA-14, SBA-15, SBA-16, STAC-1 , Ma y PCH. Cuando la matriz de sílice es estructurada y porosa, la distancia centro de poro a centro de poro en la matriz puede estar comprendida entre 3,4 y 4,2 nm, y el espesor de la pared oscila entre 1 y 2 nm.
En el material de la presente invención las nanopartículas de sílice pueden presentar forma esférica, hexagonal o alargada, preferentemente esférica.
Según la presente invención las nanopartículas de sílice pueden presentar un diámetro medio comprendido entre 5 y 500 nm, preferentemente, entre 5 y 200 nm.
Según una realización particular de la presente invención, el agente bioactivo del material es una molécula terapéutica unida por enlace covalente a un grupo aniónico terminal de la superficie del soporte inorgánico. En el material de la presente invención, los grupos aniónicos terminales proporcionan un potencial zeta negativo a la suspensión de nanopartículas de sílice, lo que confiere carácter hidrofílico a las estructuras submicrónicas, a la vez que provoca la aparición de fuerzas electrostáticas repulsivas entre partículas que permiten la preparación de coloides estables en medio acuoso, facilitando la difusión intracelular por endocitosis.
Una realización particular adicional preferente de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, en el que los grupos aniónicos terminales son grupos carboxilato y una molécula de un agente bioactivo unida por enlace covalente a dicho grupo carboxilato.
Una realización particular adicional más preferente de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, y un agente bioactivo unido por enlace covalente con un grupo aniónico terminal a través de un enlace bio-hidrolizable seleccionado entre un éster, amido, carbamato o carbonato.
Otra realización particular adicional de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, y un agente bioactivo unido por enlace covalente a un grupo aniónico terminal en el que el agente bioactivo, preferentemente la molécula terapéutica, puede incorporar un ligando orgánico para facilitar la unión covalente a un grupo aniónico terminal. Mediante la selección de diferentes ligandos orgánicos es posible establecer un control de la liberación, con vistas a obtener una máxima eficacia terapéutica. De modo preferente el ligando orgánico puede estar seleccionado entre un aminoácido o un oligopéptido. Una realización particular adicional de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, y un agente bioactivo unido por enlace covalente a un grupo aniónico terminal en el que el agente bioactivo es una molécula terapéutica que puede estar seleccionada entre al menos una molécula pequeña, una macromolécula, un anticuerpo, un péptido, una proteína, una enzima, un ácido nucleico o un polímero orgánico o un derivado de estos que incorpore un ligando orgánico que facilite su unión
covalente a un grupo aniónico terminal, y combinaciones de los mismos. El material de la presente invención puede contener un tipo de molécula terapéutica o puede contener dos o más tipos. En el campo de la presente invención, se entiende por molécula pequeña cualquier molécula de origen biológico o sintético cuyo peso molecular sea inferior a 3.000 Da.
Según una realización particular adicional preferente dicha molécula terapéutica es una molécula pequeña e hidrofóbica. De modo preferente, pero sin limitar el campo de aplicación de la presente invención, dicha molécula puede ser un antitumoral (como la CPT, el paclitaxel o la doxorrubicina), un antibiótico, un antimicótico o un antiviral o un derivado de éstos que incorpore un ligando orgánico que facilite su unión covalente a un grupo aniónico terminal.
Según otra realización particular adicional más preferente la molécula terapéutica pequeña e hidrofóbica es CPT o un derivado de ésta que incorpora un ligando orgánico que facilite su unión covalente a un grupo aniónico terminal. De modo aún más preferente, el ligando orgánico es glicina, que se une por un extremo a la CPT en posición 20-OH mediante enlace éster (Gly- CPT) y por el extremo opuesto a un grupo carboxilato terminal mediante enlace amido. En el material de la invención, la molécula terapéutica aumenta su solubilidad y estabilidad en medio acuoso, a la vez que se reducen las interacciones con otras moléculas terapéuticas y con componentes del plasma sanguíneo. Igualmente, se reduce o elimina la respuesta inmune, se le protege del metabolismo, se modula la cinética de liberación, se aumenta el tiempo de residencia plasmática y la vida media, se reduce la toxicidad y se aumenta la eficacia, se normaliza su metabolismo en sujetos de diferentes especies y
etnias y/o razas, y se promueve la difusión dirigida a tejidos y/o células específicos.
En el material de la invención, la concentración de la molécula terapéutica puede oscilar entre el 0,1 y el 40% en peso.
Una realización particular adicional de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, una molécula de un agente bioactivo unida por enlace covalente a un grupo aniónico terminal, y una molécula de un poli(etilenglicol) (PEG) unida por enlace covalente. La molécula de PEG aumenta la solubilidad y estabilidad del material en medio acuoso, permitiendo la preparación de coloides estables a la vez que reduce las interacciones de las nanopartículas con componentes del plasma sanguíneo, aumenta el tiempo de residencia plasmática, reduce o elimina la respuesta inmune, reduce la toxicidad y aumenta la eficacia, y promueve la difusión dirigida a tejidos y/o células específicos.
En el material de la presente invención, el peso molecular de la molécula de PEG puede oscilar entre 200 y 20.000 Da y su concentración puede oscilar entre el 1 y el 30% en peso.
Según una realización particular adicional de la presente invención el material nanoestructurado comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, una molécula de un agente bioactivo unida por enlace covalente a un grupo aniónico terminal, y una molécula de un agente fluorescente unido por enlace covalente. La molécula del agente fluorescente permite el estudio de la biodistribución del preparado farmacéutico. De manera preferente, el agente fluorescente está seleccionado entre 5-amino fluoresceina, fluoresceina isotiocianato, NHS-fluoresceina, rodamina B isotiocianato, tetrametilrodamina B isotiocianato, NHS-Cy5 y/o
NHS-Cy5.5 o un derivado de éstos que incorpore un ligando orgánico que facilite la unión covalente con el soporte inorgánico.
En el material de la presente invención, la concentración del agente fluorescente puede oscilar entre el 0,01 y el 10% en peso.
Una realización particular adicional de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, una molécula de un agente bioactivo unida por enlace covalente a un grupo aniónico terminal, y un radionúcleo de 18F unido por enlace covalente a la matriz inorgánica. El isótopo radioactivo permite el estudio de la biodistribución y la farmacocinética del preparado farmacéutico. En el material de la presente invención, la concentración del radionúcleo de 18F puede oscilar entre el 0,0001 y el 1 % en peso.
Una realización particular adicional de la presente invención es un material nanoestructurado que comprende nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales, una molécula de un agente bioactivo unida por enlace covalente a un grupo aniónico terminal, y una molécula directora a receptores específicos, células o tejidos, unida por enlace covalente. La molécula directora conduce selectivamente las nanopartículas a su objetivo terapéutico, favoreciendo, asimismo, el acceso por endocitosis a las células. La molécula directora puede ser sintética, semi- sintética o natural. Las sustancias que pueden ejercer de moléculas directoras están seleccionada entre proteínas (incluyendo anticuerpos, fragmentos de anticuerpos, aptámeros, péptidos, polipéptidos y aminoácidos), carbohidratos, lípidos, ácidos nucleicos (incluyendo nucleósidos, nucleótidos y polinucleótidos) y pequeñas moléculas como hormonas y análogos hormonales, vitaminas, co- factores enzimáticos y metabolitos de la síntesis de proteínas, o un derivado de
éstos que incorpore un ligando orgánico que facilite la unión covalente el soporte inorgánico.
Un objeto adicional de la presente invención es el uso del material nanoestructurado definido anteriormente en una aplicación biotecnológica, comprendiendo dicho uso poner en contacto una o más células con las nanopartículas del material.
Dicha aplicación biotecnológica es preferentemente un tratamiento o profilaxis de una patología.
Se ha comprobado que las nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales difunden al interior celular sin efecto citotóxico alguno. La unión de una molécula terapéutica o un derivado de ésta que incorpora un ligando orgánico que facilite su unión covalente a un grupo aniónico terminal mejora la solubilidad del fármaco y favorece su difusión intracelular, dando lugar a una acción citotóxica que conduce a la muerte celular, lo que permite llevar a cabo un tratamiento de quimioterapia.
Cuando la unión covalente entre la molécula terapéutica y el soporte inorgánico es un enlace éster la liberación de la molécula terapéutica en el entorno intracelular y su actividad citotóxica depende de la acción de esterasas citoplasmáticas.
En una realización preferida de la presente invención la concentración equivalente de molécula terapéutica en el medio celular necesaria para provocar una acción citotóxica oscila entre 0,001 μg ml_"1 y 10 μg ml_"1. Un objeto particular adicional de la presente invención es el uso del material nanoestructurado definido anteriormente en un método de tratamiento o profilaxis de una patología in vivo que comprende poner en contacto los
tejidos afectados o supuestamente afectados por la patología con nanopartículas del material definido.
La patología mencionada consiste preferentemente en una infección viral, infección fúngica, infección bacteriana o tumor.
La unión de una molécula terapéutica a una nanopartícula de sílice, cuya superficie se encuentra cubierta de grupos aniónicos terminales mejora la solubilidad del fármaco y reduce su citotoxicidad in vivo, a la vez que favorece la acumulación del agente terapéutico en el tumor mediante el efecto de permeabilidad de mejora y conservación (EPR).
La concentración de nanopartículas en el tejido patógeno varía entre 1 y 1000 μg cm"3. , y preferentemente entre 5 y 100 μg cm"3.
En una realización preferida de la presente invención la concentración equivalente de molécula terapéutica administrada al huésped necesaria para provocar una acción terapéutica oscila entre 0,0001 mg Kg"1 h"1 y 10 mg Kg"1 h" 1, durante 1 a 2000 h.
Las nanopartículas del material nanoestructurado definido anteriormente son administradas al huésped preferentemente por vía oral, parenteral, respiratoria o transdérmica en concentración suficiente para alcanzar de forma no selectiva los tejidos patógenos. El aumento de la permeabilidad capilar asociado a muchas lesiones permite la extravasación de las nanopartículas a nivel de los mismos (efecto EPR), acumulándose en la matriz extracelular.
Alternativamente las nanopartículas del material nanoestructurado definido anteriormente son administradas al huésped mediante inyección directa en la zona lesionada, acumulándose en el tejido intersticial.
Un objeto particular adicional de la presente invención es el uso del material nanoestructurado definido anteriormente en un método de tratamiento
o profilaxis de un paciente por administración de nanopartículas del material definido por vía oral, parenteral, respiratoria o transdérmica, en concentración suficiente para alcanzar de forma no selectiva los tejidos patógenos. El aumento de la permeabilidad capilar asociado a muchas lesiones permite la extravasación de las nanopartículas a nivel de los mismos (efecto EPR), acumulándose en la matriz extracelular.
Alternativamente las nanopartículas del material nanoestructurado definido anteriormente son administradas al paciente mediante inyección directa en la zona lesionada, acumulándose en el tejido intersticial.
De manera preferida, la patología a tratar por quimioterapia es una infección viral, infección fúngica, infección bacteriana o tumor.
La concentración de nanopartículas en el tejido patógeno varía entre 1 y 1000 μg cm"3, y preferentemente entre 5 y 100 μg cm"3.
En una realización preferida de la presente invención la concentración equivalente de la molécula terapéutica administrada al paciente necesaria para provocar una acción terapéutica oscila entre 0,0001 mg Kg"1 h"1 y 10 mg Kg"1 h" 1, durante 1 a 2000 h.
Por tanto, la presente invención se refiere a un material nanoestructurado compuesto de nanopartículas de sílice, cuya superficie se encuentra cubierta de grupos aniónicos y un agente bioactivo unido por un enlace covalente hidrolizable y, opcionalmente, moléculas de un PEG, un agente fluorescente, un radionúcleo y/o una sustancia directora a receptores, células o tejidos específicos, unidas por enlace covalente, y su uso en el tratamiento o profilaxis de una determinada patología.
A lo largo de la descripción y las reivindicaciones la palabra "comprende" y sus variantes no pretenden excluir otras características técnicas, aditivos,
componentes o pasos. Para los expertos en la materia, otros objetos, ventajas y características de la invención se desprenderán en parte de la descripción y en parte de la práctica de la invención. Los ejemplos que se proporcionan con a modo de ilustración, y no se pretende que sean limitativos de la presente invención.

MODOS DE REALIZACIÓN DE LA INVENCIÓN
Los ejemplos siguientes pretenden ilustrar características relacionadas con invención.
Materiales
EJEMPLO 1 : Preparación de Gly-CPT (modificado de Bioorg. Med. Chem. 1998, 6, 551 -562; Greenwald, R.B.; Pendri, A.; Conover, C.D.; Lee, C; Choe, Y.H.; Gilbert, C; Martínez, A.; Xia, J.; Wu, D.; Hsue, M.-M.).
En un matraz de 50 mL con un imán agitador se introducen 1 15 mg de t- Boc-glicina (0,66 mmol). A continuación, se inyecta 10 mL de diclorometano anhidro (DCM). Se añaden 80 μΐ de Ν,Ν'-diisopropilcarbodiimida (DIPC, 0,52 mmol) y 50 mg de 4-(dimetilamino)piridina (DMAP, 0,41 mmol) y se mantiene la disolución en agitación durante 30 min a 0 °C. Transcurrido ese tiempo se añaden 160 mg de CPT (0,46 mmol, 1 ), se deja calentar la mezcla a temperatura ambiente y se mantiene en agitación durante 16 h. Transcurrido este tiempo se añaden otros 40 mL de DCM y se neutraliza con HCI 0.1 M. A continuación se lava la mezcla con salmuera. La fase orgánica se separa, se seca con sulfato de magnesio anhidro y se evapora el disolvente a vacío (50 torr), obteniéndose un sólido blanco. El sólido blanco se disuelve en la mínima cantidad posible de metanol caliente y se mantiene a 4 °C durante 72 h. El sólido recristal izado se filtra y se añaden 0,5 mL de DCM y 0,5 mL de ácido trifluoroacético, y se agita a temperatura ambiente durante 30 min. Transcurrido este tiempo se evapora el sobrenadante a vacío (50 torr), obteniendo un sólido blanco.
El sólido blanco se disuelve en la mínima cantidad posible de metanol caliente, se añaden dos volúmenes de dietiléter y se mantiene en agitación
durante 4 h a 4 °C. Transcurrido este tiempo se filtra y se obtienen 145 mg de un sólido blanco (2). 1H NMR (DMSO-c/6) 8,7 (s); 8,4 (ws); 8,2 (m); 7,9 (m); 7,7 (m); 7,4 (s); 5,6 (s); 5,4 (s); 4,4 (m); 2,2 (m); 1 ,8 (m); 1 ,0 (t); 13C NMR (DMSO- c/6)172,4; 166,9; 157,8; 152,5; 147,9; 146,0; 144,6; 131 ,5; 130,3; 129,7; 128,7; 128,4; 127,9; 127,7; 1 19,0; 95,1 ; 77,4; 66,3; 50,2; 30,6; 7,7.
EJEMPLO 2: Preparación de nanopartículas de sílice amorfa no porosa cuya superficie se encuentra cubierta de grupos carboxilato terminales
En un matraz de 100 mL con un imán agitador se dispone 1 ,00 g de nanopartículas preformadas de sílice amorfa no porosa con un diámetro total comprendido entre 5 y 20 nm (Aldrich), y se deshidratan a 350 °C en vacío (8 torr) durante 3 h. A continuación se introducen 10 mL de tolueno anhidro, se calienta a temperatura de reflujo en agitación, se introducen 78 μί de 3- cianopropiltriclorosilano (0,50 mmol) y se mantiene en estas condiciones durante 3 h. Se obtiene un sólido que se filtra, se lava con 50 mL de tolueno y 100 mL de metanol y se seca a vacío (8 torr) durante 16 h.
El sólido obtenido en la etapa anterior se dispersa en 150 mL de ácido sulfúrico 60% y se calienta a 150 °C durante 3 h. A continuación, se añaden 200 mL de agua desionizada, se filtra, se lava con 50 mL de tolueno y 100 mL de metanol y se seca a 60 °C en vacío (8 torr).
EJEMPLO 3: Preparación de nanopartículas de sílice amorfa no porosa cuya superficie se encuentra cubierta de grupos carboxilato terminales sobre los que se une Gly-CPT por enlace amido (SNP-Gly-CPT)
500 mg de nanopartículas obtenidas en el Ejemplo 2 se deshidratan a 60
°C en vacío (8 torr) durante 6 h. A continuación se introducen 10 mL de DCM, se añaden 122 mg de N-(3-dimetilaminopropil)-N'-etilcarbodiimide clorhidrato (EDCi, 0,64 mmol) y 130 mg de 1 -hidroxibenzotriazol (HOBT, 0,96 mmol), y se mantiene a 0 °C en agitación. Transcurridos 30 min se añaden 65 mg de Gly- CPT (0.125 mmol) preparados según el Ejemplo 1 , se introducen 87 μΐ de Ν,Ν'-diisopropiletilamina (DIPEA, 0,50 mmol), se deja evolucionar a temperatura ambiente y se mantiene la agitación durante 16 h. Transcurrido
este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava con metanol y se centrifuga sucesivas veces hasta ausencia total de Gly- CPT en el sobrenadante (UV-Vis, Α36δ), y se liofiliza a -55 °C durante 16 h.
Se obtienen nanopartículas de sílice amorfa no porosa con un diámetro total comprendido entre 5 y 20 nm cuya superficie está cubierta de grupos carboxilato terminales a los que se une Gly-CPT por enlace amido (Figura 1 ).
En la Figura 2 se muestra el espectro UV-Vis de este material.
EJEMPLO 4: Preparación de nanopartículas de sílice amorfa no porosa cuya superficie se encuentra cubierta de grupos carboxilato terminales sobre los que se une Gly-CPT por enlace amido (SNP-Gly-CPT) con una concentración menor de CPT
500 mg de nanopartículas obtenidas en el Ejemplo 2 se deshidratan a 60 °C en vacío (8 torr) durante 6 h. A continuación se introducen 10 mL de DCM, se añaden 122 mg de EDCi (0,64 mmol) y 130 mg de HOBT (0,96 mmol), y se mantiene a 0 °C en agitación. Transcurridos 30 min se añaden 6,5 mg de Gly- CPT (0.0125 mmol) preparados según el Ejemplo 1 , se introducen 87 μΙ_ de DIPEA (0,50 mmol), se deja evolucionar a temperatura ambiente y se mantiene la agitación durante 16 h. Transcurrido este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava con metanol y se centrifuga varias veces hasta ausencia total de Gly-CPT en el sobrenadante (UV-Vis, Α36δ), y se liofiliza a -55 °C durante 16 h.
EJEMPLO 5: Preparación de nanopartículas de sílice mesoporosa estructurada con diámetro de poro entre 2 y 4 nm cuya superficie se encuentra cubierta de grupos carboxilato terminales sobre los que se une Gly-CPT por enlace amido (MSN-Gly-CPT)
En un matraz de 100 mL con un imán agitador se dispone 1 ,00 g de nanopartículas de sílice mesoporosa estructurada con un diámetro total comprendido entre 100 y 200 nm y un diámetro de poro comprendido entre 2 y 4 nm preparadas según una receta adaptada (Chem. Mater. 2001 , 13, 258-263;
Caí, Q.; Luo, Z.-S.; Pang, W.-Q.; Fan, Y.W.; Chen , X.H.; Cui, F.-Z.), y se deshidratan a 350 °C en vacío (8 torr) durante 3 h. A continuación se introducen 10 mL de tolueno anhidro, se calienta a temperatura de reflujo en agitación, se introducen 78 μΙ_ de 3-cianopropiltriclorosilano (0,50 mmol) y se mantiene en estas condiciones durante 3 h. Se obtiene un sólido que se filtra, se lava con 50 mL de tolueno y 100 mL de metanol y se seca a vacío (8 torr) durante 16 h.
El sólido obtenido en la etapa anterior se dispersa en 150 mL de ácido sulfúrico 60% y se calienta a 150 °C durante 3 h. A continuación, se añaden 200 mL de agua desionizada, se filtra, se lava con 50 mL de tolueno y 100 mL de metanol y se seca a 60 °C en vacío (8 torr).
500 mg del sólido anterior se deshidratan a 60 °C en vacío (8 torr) durante 6 h. A continuación se introducen 10 mL de DCM, se añaden 122 mg de EDCi (0,64 mmol) y 130 mg de HOBT (0,96 mmol), y se mantiene a 0 °C en agitación. Transcurridos 30 min se añaden 65 mg de Gly-CPT (0.125 mmol) preparados según el Ejemplo 1 , se introducen 87 μΐ de DIPEA (0,50 mmol), se deja evolucionar a temperatura ambiente y se mantiene la agitación durante 16 h. Transcurrido este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava con metanol y se centrifuga varias veces hasta ausencia total de Gly-CPT en el sobrenadante (UV-Vis, Α36δ), y se liofiliza a -55 °C durante 16 h. Se obtienen nanopartículas de sílice mesoporosa estructurada con un diámetro total comprendido entre 100 y 200 nm y un diámetro de poro comprendido entre 2 y 4 nm cuya superficie está cubierta de grupos carboxilato terminales a los que se une Gly-CPT por enlace amido.
EJEMPLO 6: Preparación de nanopartículas de sílice mesoporosa estructurada con diámetro de poro entre 4 y 25 nm cuya superficie se
encuentra cubierta de grupos carboxilato terminales sobre los que se une Gly-CPT por enlace amido (NS-Gly-CPT)
En un matraz de 100 ml_ con un imán agitador se dispone 1 ,00 g de nanopartículas de sílice mesoporosa estructurada con un diámetro total comprendido entre 80 y 300 nm y un diámetro de poro comprendido entre 4 y 25 nm preparadas según una receta adaptada (J. Phys. Chem. B 2009, 1 13, 1796-1804; Gao, F.; Botella, P.; Corma, A.; Blesa, J.; Dong, L), y se deshidratan a 100 °C en vacío (8 torr) durante 3 h. A continuación se introducen 10 mL de tolueno anhidro, se calienta a temperatura de reflujo en agitación, se introducen 78 μΙ_ de 3-cianopropiltriclorosilano (0,50 mmol) y se mantiene en estas condiciones durante 3 h. Se obtiene un sólido que se filtra, se lava con 50 mL de tolueno y 100 mL de metanol y se seca a vacío (8 torr) durante 16 h. El sólido obtenido en la etapa anterior se dispersa en 150 mL de ácido sulfúrico 60% y se calienta a 150 °C durante 3 h. A continuación, se añaden 200 mL de agua desionizada, se filtra, se lava con 50 mL de tolueno y 100 mL de metanol y se seca a 60 °C en vacío (8 torr). 500 mg del sólido anterior se deshidratan a 60 °C en vacío (8 torr) durante 6 h. A continuación se introducen 10 mL de DCM, se añaden 122 mg de EDCi (0,64 mmol) y 130 mg de HOBT (0,96 mmol), y se mantiene a 0 °C en agitación. Transcurridos 30 min se añaden 65 mg de Gly-CPT (0.125 mmol) preparados según el Ejemplo 1 , se introducen 87 μΐ de DIPEA (0,50 mmol), se deja evolucionar a temperatura ambiente y se mantiene la agitación durante 16 h. Transcurrido este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava con metanol y se centrifuga varias veces hasta ausencia total de Gly-CPT en el sobrenadante (UV-Vis, Α36δ), y se liofiliza a -55 °C durante 16 h.
Se obtienen nanopartículas de sílice mesoporosa estructurada con un diámetro total comprendido entre 80 y 300 nm y un diámetro de poro
comprendido entre 4 y 25 nm cuya superficie está cubierta de grupos carboxilato terminales a los que se une Gly-CPT por enlace amido.
EJEMPLO 7: Preparación de nanopartículas de sílice amorfa no porosa cuya superficie se encuentra cubierta de grupos carboxilato terminales sobre los que se une Gly-CPT y una molécula de PEG3000 por enlace amido (SNP-Gly-CPT-PEG)
500 mg de nanopartículas obtenidas según el Ejemplo 2 se deshidratan a 60 °C en vacío (8 torr) durante 6 h. A continuación se introducen 10 mL de DCM, 122 mg de EDCi (0,64 mmol) y 130 mg de HOBT (0,96 mmol) y se mantiene a 0 °C durante 30 min en agitación. Transcurrido este tiempo se añaden 74 mg de alfa-amino-omega-amino poli(etilenglicol) clorhidrato, (H2N- PEG3ooo-NH2 *HCI, Iris Biotech) y 18 μί de DIPEA (0,10 mmol), se deja evolucionar a temperatura ambiente y se mantiene en agitación durante 16 h. Transcurrido este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava con metanol y se centrifuga varias veces, y se liofiliza a -55 °C durante 16 h.
Se obtienen nanopartículas de sílice amorfa no porosa con un diámetro total comprendido entre 5 y 20 nm cuya superficie está cubierta de grupos carboxilato terminales a los que se unen moléculas de Gly-CPT y de H2N- PEG3000-NH2 por enlace amido.
EJEMPLO 8: Preparación de nanopartículas de sílice amorfa no porosa cuya superficie se encuentra cubierta de grupos carboxilato terminales sobre los que se une Gly-CPT y una molécula de Cy5.5 por enlace amido (SNP-Gly-CPT-Cy5.5)
500 mg de nanopartículas obtenidas según el Ejemplo 2 se deshidratan a 60 °C en vacío (8 torr) durante 6 h. A continuación se introducen 10 mL de DCM, 109 mg de EDCi (0,57 mmol) y 1 17 mg de HOBT (0,87 mmol) y se mantiene a 0 °C durante 30 min en agitación. Transcurrido este tiempo se
añaden 4.7 mg de fenilendiamina (0,04 mmol) y 3 μΙ_ de DIPEA (0,02 mmol), se deja evolucionar a temperatura ambiente y se mantiene en agitación durante 16 h. Transcurrido este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava con metanol y se centrifuga varias veces, y se liofiliza a -55 °C durante 16 h.
Sobre 100 mg del sólido obtenido se añaden 10 mL de DCM y 5 mg de EDCi (0,03 mmol) y agita durante 30 min a 0 °C. Transcurrido este tiempo se añaden 400 μΙ_ de una disolución 0,85 mM de NHS-Cy5.5 en dimetilsulfóxido (DMSO) y 87 mL de DIPEA (0,50 mmol), se deja evolucionar a temperatura ambiente y se mantiene en agitación durante 16 h. Transcurrido este tiempo se evapora el disolvente a vacío (50 torr). El sólido obtenido se lava primero con metanol y se centrifuga varias veces. A continuación, el sólido obtenido se lava con agua y se centrifuga varias veces. Finalmente, el sólido lavado obtenido se liofiliza a -55 °C durante 16 h.
Se obtienen nanopartículas de sílice amorfa no porosa con un diámetro total comprendido entre 5 y 20 nm cuya superficie está cubierta de grupos carboxilato terminales y grupos amino terminales a los que se unen, respectivamente, moléculas de Gly-CPT y de Cy5.5 por enlaces amido.
En la Figura 3 se muestra el espectro UV-Vis de este material. Liberación de CPT del material SNP-Gly-CPT EJEMPLO 9: Liberación de CPT en suero fisiológico (PBS)
Se prepara una suspensión en PBS de 10 mg/mL de nanopartículas obtenidas según el Ejemplo 4. Una alícuota de 1 mL de la suspensión se transfiere a un tubo eppendorí de 2 mL y se incuba a 37 °C durante 3 h. La muestra incubada se centrifuga y se liofiliza el sobrenadante a -55 °C durante 16 h. El residuo obtenido se reconstituye con 1 mL de una disolución metanol/HCI 20/1 (v/v). 20 μί de dicha disolución se inyectan en un equipo de
HPLC. Se integra el área del pico de la CPT lactona y se compara con una curva de calibrado.
Se comprueba que el material presenta una gran estabilidad en PBS, con una liberación de CPT inferior al 4% en 3 h. Cuando la liberación de CPT del material SNP-Gly-CPT se llevó a cabo en una solución en PBS de una colesterol esterasa (ChE) o acetilcolinesterasa (AChE), la velocidad de liberación de CPT en el medio se mantiene en el mismo orden de magnitud que en PBS.
EJEMPLO 10: Liberación de CPT en suero humano
Se prepara una suspensión en suero humano estéril (Aldrich) de 10 mg/mL de nanopartículas obtenidas según el Ejemplo 4. Una alícuota de 1 mL de la suspensión se transfiere a un tubo eppendorf estéril de 2 mL y se incuba a 37 °C. Las muestras incubadas a diferentes tiempos se centrifugan y se separa el sobrenadante. 500 μί de suero sobrenadante se transfieren a un vial estéril, se añaden 10 mL de medio CelLytic (Aldrich), se aplican ultrasonidos durante 3 min y se centrifuga a 10 °C. Sobre 500 μΐ de sobrenadante se añaden 500 μΐ de una disolución acuosa de ácido trifluoroacético (TFA) al 5 % a 0 °C, se agita durante 60 segundos y se centrifuga a 4 °C. El sobrenadante se liofiliza a -55 °C durante 16 h. El residuo obtenido se reconstituye con 1 mL de una disolución metanol/HCI 20/1 (v/v). 20 μί de dicha disolución se inyectan en un equipo de HPLC. Se integra el área del pico de la CPT lactona y se compara con una curva de calibrado.
Se comprueba que el material presenta una gran estabilidad en suero humano, con una liberación de CPT inferior al 3% en 3h, e inferior al 9% en 24 h. EJEMPLO 11 : Liberación de CPT a diferentes valores de pH
Se prepara una suspensión acuosa de 10 mg/mL de nanopartículas obtenidas según el Ejemplo 4, ajustando el pH en el rango 2-12. Una alícuota
de 1 mL de la suspensión se transfiere a un tubo eppendorf de 2 mL y se incuba a 37 °C durante 3 h. Las muestras incubadas a diferentes valores de pH se centrifugan y se liofiliza el sobrenadante a -55 °C durante 16 h. El residuo obtenido se reconstituye con 1 mL de una disolución metanol/HCI 20/1 (v/v). 20 μί de dicha disolución se inyectan en un equipo de HPLC. Se integra el área del pico de la CPT lactona y se compara con una curva de calibrado.
En la Tabla 1 se muestra el porcentaje de CPT liberado en el medio acuoso a diferentes valores de pH. Se comprueba que el pH del medio tiene un efecto significativo en la liberación de CPT a partir del material SNP-Gly-CPT. El enlace éster del conjugado glicina-CPT unido al soporte inorgánico resiste bien a valores de pH inferiores a 8, pero se hidroliza rápidamente en medio muy alcalino (p.e., pH=12). Tabla 1 . EJEMPLO 12: Liberación de CPT en función de la concentración
Se prepara una suspensión acuosa de 10 mg mL"1 de nanopartículas obtenidas según el Ejemplo 3. Una alícuota de 1 mL de la suspensión se transfiere a un tubo eppendorf estéril de 2 mL y se incuba a 37 °C. Las muestras incubadas a diferentes tiempos se centrifugan y se liofiliza el sobrenadante a -55 °C durante 16 h. Alternativamente, se prepara una suspensión acuosa de 10 mg mL"1 de nanopartículas obtenidas según el Ejemplo 4. Una alícuota de 1 mL de la suspensión se transfiere a un tubo eppendorf estéril de 2 mL y se incuba a 37 °C. Las muestras incubadas a diferentes tiempos se centrifugan y se liofiliza el sobrenadante a -55 °C durante 16 h. El residuo obtenido se reconstituye con 1 mL de una disolución metanol/HCI 20/1 (v/v). 20 μί de dicha disolución se inyectan en un equipo de
HPLC. Se integra el área del pico de la CPT lactona y se compara con una curva de calibrado.
Se comprueba que la vida media (f½, h) del preparado SNP-Gly-CPT varía en función de la concentración del fármaco en el material, siendo de 51 h en el caso de nanopartículas obtenidas según el Ejemplo 3 y de 8 h para nanopartículas obtenidas según el Ejemplo 4.
EJEMPLO 13: Determinación de la /C50 mediante ensayo MTT
La actividad de la CPT in vitro se determinó mediante el test del bromuro de 3-[4,5-dimetiltiazol-2-il]-2,5-difeniltetrazolio (MTT). En estos ensayos la proliferación de las células vivas se determina vía la actividad de deshidrogenasa mitocondrial: las mitocondrias viables reducen el MTT generando cristales de MTT-formazán cuantificables por espectrofotometría visible.
Para los ensayos se ha empleado células HeLa de carcinoma de cérvix, U87MG de glioblastoma, HCT-1 16 de cáncer de colon y MDA-MB-231 de cáncer de pecho. Las células se cultivan en medio RPMI (HeLa, HCT-1 16 y MDA-MB-231 ) o DMEM (U87MG) enriquecido con 10% de suero bovino fetal a 37 °C, en atmósfera húmeda de aire (95%) y CO2 (5%). Las células se siembran en placas de 96 pocilios a razón de 2.000 células/pocilio (HeLa), 6.000 células/pocilio (U87MG), 10.000 células/pocilio (HCT-1 16) y 10.000 células/pocilio (MDA-MB-231 ), 18 h antes de incorporar las nanopartículas.
Se preparan alícuotas de 10 mg mL"1 de CPT en DMSO y se mantienen a -20 °C. Cuando necesario se descongela una alícuota, se calienta a 95 °C hasta disolución completa de la CPT y se preparan alícuotas de trabajo (100 μg mL"1) por dilución en medio RPMI no enriquecido. Alternativamente, se preparan alícuotas de 100 μg mL"1 de CPT en medio RPMI no enriquecido y se mantienen a -20 °C hasta necesario.
Se prepara una suspensión de nanopartículas obtenidas según el Ejemplo 3 en medio RPMI no enriquecido con una concentración equivalente de 100 μg mL"1 de CPT y se trata con ultrasonidos (Soniprep 150 sonicator, 23 KHz, 10% amplitud, 3x10 min, 0 °C), para mejorar la dispersión de las nanopartículas. La suspensión se reparte en alícuotas que se mantienen a -20 °C hasta necesario.
Alternativamente, se prepara una suspensión de nanopartículas obtenidas según el Ejemplo 2 en medio RPMI no enriquecido con una concentración equivalente en nanopartículas al caso anterior y se trata con ultrasonidos en idénticas condiciones.
Las células se tratan con CPT o con nanopartículas obtenidas según el Ejemplo 3, en un rango de dosis de CPT entre 2,5 x 10"5 y 2,5 μg mL"1 durante 72 h. Alternativamente, las células se tratan con nanopartículas obtenidas según el Ejemplo 2 durante 72 h. Al final del periodo de incubación se añaden a cada pocilio 5 mg mL"1 de la disolución de MTT y 4 h más tarde se disuelven los cristales de formazán con HCI 0,1 N en isopropanol anhidro o DMSO, y se mide la absorbancia a 590 nm. Se determina el porcentaje de supervivencia celular relativo a las células no tratadas y se calcula los valores de IC5o a partir de las curvas dosis CPT-(%) células vivas (Figuras 4 y 6). Los valores de IC5o se listan en la Tabla 2.

Claims
1 . Un material nanoestructurado caracterizado porque comprende al menos: - Un soporte inorgánico, que comprende nanopartículas de sílice cuya superficie se encuentra cubierta de grupos aniónicos terminales, y
Un agente bioactivo unido por enlace covalente a un grupo aniónico terminal de la superficie del soporte inorgánico.
2. Un material nanoestructurado según la reivindicación 1 , caracterizado porque la sílice es no estructurada y no porosa.
3. Un material nanoestructurado según la reivindicación 1 , caracterizado porque la sílice es no estructurada y porosa, con diámetro de poro comprendido entre 1 y 100 nm.
4. Un material nanoestructurado según la reivindicación 1 , caracterizado porque la sílice es estructurada y porosa, con diámetro de poro comprendido entre 1 y 30 nm.
5. Un material nanoestructurado según la reivindicación 4, caracterizado porque presente orden a larga distancia y cuya estructura está seleccionada entre MCM-41 , MCM-48, MCM-50, FSM-16, HMS, KIT-1 , MSU-1 , MSU-2, MSU-3, MSU-V, SBA-1 , SBA-2, SBA-3, SBA-8, SBA-1 1 , SBA-12, SBA-14, SBA-15, SBA-16, STAC-1 , Ma y PCH.
6. Un material nanoestructurado según la reivindicación 5, caracterizado porque la distancia centro de poro a centro de poro en la matriz está comprendida entre 3,4 y 4,2 nm, y el espesor de la pared oscila entre 1 y 2 nm.
7. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque las nanopartículas de sílice presentan una forma seleccionada entre esférica, hexagonal o alargada.
8. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque las nanopartículas de sílice presentan un diámetro medio comprendido entre 5 y 500 nm.
9. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque el agente bioactivo es al menos una molécula terapéutica.
10. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque los grupos aniónicos terminales son grupos carboxilato.
1 1 . Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque el enlace covalente es un enlace bio- hidrolizable seleccionado entre un éster, amido, carbamato o carbonato.
12. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque el agente bioactivo se une a un grupo aniónico terminal a través de un ligando orgánico.
13. Un material nanoestructurado según la reivindicación 12, caracterizado porque el ligando orgánico está seleccionado entre un aminoácido o un oligopéptido.
14. Un material nanoestructurado según la reivindicación 9, caracterizado porque la molécula terapéutica está seleccionada entre al menos una molécula pequeña, una macromolécula, un anticuerpo, un péptido, una proteína, una enzima, un ácido nucleico, un polímero orgánico, un derivado de éstos que comprende un ligando orgánico y combinaciones de los mismos.
15. Un material nanoestructurado según la reivindicación 14, caracterizado porque la molécula terapéutica es pequeña e hidrofóbica.
16. Un material nanoestructurado según la reivindicación 15, caracterizado porque la molécula terapéutica está seleccionada entre un antitumoral, un antibiótico, un antimicótico o un antiviral o un derivado de estos que incorpore un ligando orgánico.
17. Un material nanoestructurado según la reivindicación 16, caracterizado porque la molécula terapéutica es CPT o un derivado de esta que además incorpora un ligando orgánico.
18. Un material nanoestructurado según la reivindicación 17, caracterizado porque el ligando orgánico es glicina.
19. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque la concentración del agente bioactivo oscila entre el 0,1 y el 40% en peso.
20. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque comprende además una molécula de un PEG unida por enlace covalente.
21 . Un material nanoestructurado según la reivindicación 20, caracterizado porque el peso molecular de la molécula de PEG oscila entre 200 y 20.000 Da y su concentración oscila entre el 1 y el 30% en peso.
22. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque comprende además una molécula de un agente fluorescente, unida por enlace covalente, seleccionado entre 5-amino fluoresceina, fluoresceina isotiocianato, NHS-fluoresceina, rodamina B isotiocianato, tetrametilrodamina B isotiocianato, NHS-Cy5 y/o NHS-Cy5.5 o un derivado de estos que incorpore un ligando orgánico.
23. Un material nanoestructurado según la reivindicación 22, caracterizado porque la concentración del agente fluorescente oscila entre el 0,01 y el 10% en peso.
24. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque comprende además un radionúcleo de 18F unido por enlace covalente a la matriz inorgánica.
25. Un material nanoestructurado según la reivindicación 24, caracterizado porque la concentración del radionúcleo de 18F oscila entre el 0,0001 y el 1 % en peso.
26. Un material nanoestructurado según cualquiera de las reivindicaciones anteriores, caracterizado porque comprende además una molécula directora a receptores específicos, células o tejidos, unida por enlace covalente, seleccionada entre proteínas, carbohidratos, lípidos, ácidos nucleicos y pequeñas moléculas seleccionadas entre hormonas y análogos hormonales, vitaminas, co-factores enzimáticos y metabolitos de la síntesis de proteínas, o un derivado de estos que incorpore un ligando orgánico.
27. Uso de un material nanoestructurado descrito según cualquiera de las reivindicaciones anteriores en una aplicación biotecnológica, caracterizado porque dicho uso comprende poner en contacto una o más células con las nanopartículas del material.
28. Uso de un material nanoestructurado según la reivindicación 27, caracterizado porque dicha aplicación biotecnológica es un tratamiento o profilaxis de una patología in vivo que comprende poner en contacto los tejidos afectados o supuestamente afectados por la patología con nanopartículas del material.
29. Uso de un material nanoestructurado según la reivindicación 28, caracterizado porque dicha patología está seleccionada entre una infección viral, infección fúngica, infección bacteriana o tumor.
30. Uso de un material nanoestructurado según la reivindicación 28, caracterizado porque las nanopartículas son administradas por vía oral, parenteral, respiratoria, transdérmica o por inyección directa en la zona lesionada.
