  • the method statistically significantly increases the fluorescent signal from the sample and thereby apparently improves the reaction yield and reduces the measurement scatter of parallel measurements.

Higher repeatability and shortened reaction time enable smaller consumption of chemicals, which reduces the price of the procedure and reduces the environment loading.

In RT-PCR the mineral oils are not added, since the detection of the reaction products is completely different from the classical procedures.

Abstract

L'invention a pour objet une amélioration de la collection de lumière fluorescente provenant de l'échantillon pendant une amplification par RT-PCR, par d'addition d'une couche translucide sur le trajet optique entre le mélange réactionnel RT-PCR et le détecteur de fluorescence, ainsi que le procédé de préparation de ladite couche translucide. L'invention concerne un procédé et des substances permettant de former la couche transparente qui améliore la sensibilité et la reproductibilité des mesures de la fluorescence dans l'amplification en chaîne par polymérase en temps réel, en RT-PCR en continu. L'invention améliore la collection de la lumière fluorescente provenant de l'échantillon pendant l'amplification RT-PCT. Elle repose sur le principe d'une substance qui est optiquement perméable et qui est placée sur le trajet optique entre le mélange réactionnel et le détecteur de fluorescence tout en ne se mélangeant pas à l'eau. Cette substance peut être par exemple de la paraffine ou une autre huile minérale. La substance est ajoutée au mélange réactionnel RT-PCR, dans lequel elle permet d'améliorer la collection de la lumière fluorescente dissipée et de la diriger dans le détecteur du cycleur RT-PRC. L'amélioration de la collection est réalisée par l'effet de formation d'une lentille au-dessus du mélange réactionnel, et en empêchant l'évaporation de l'eau hors du mélange réactionnel, et donc la condensation de l'eau évaporée sur la surface du trajet optique, comme par exemple un film qui couvre la plaque microtitre et empêche ainsi une dissipation additionnelle de la lumière à partir du mélange réactionnel.
