本発明の目的は、難溶性であるRebaudioside-Aを90重量%以上含有するステビア抽出物の溶解性を改善したステビア抽出物の混合物の提供、及び難溶性であるRebaudioside-Aを90重量%以上含有するステビア抽出物の溶解方法を提供することである。 すなわち本発明は、Rebaudioside-Aを90重量%以上含有するステビア抽出物及び該水溶性多糖を乾燥重量比がステビア抽出物:水溶性多糖=100:5~0.4:99.6となるように含むステビア組成物、及びRebaudioside-Aを90重量%以上含有するステビア抽出物に対して水溶性多糖を、上記乾燥重量比となるように混合し、得られる混合物を水性溶媒に溶解するステビア抽出物の溶解方法を提供する。
Description
本発明は、難溶解性であるRebaudioside-Aを90重量%以上含有するステビア抽出物を含む組成物、及び難溶解性であるRebaudioside-Aを90重量%以上含有するステビア抽出物の溶解方法に関する。
ステビア抽出物は、ステビア(キク科の植物。学名:ステビア レバウディアナ ベルトニー(Stevia rebaudiana BERTONI)の植物体の抽出物であり、通常はステビアの葉部の抽出物である。ステビア抽出物は甘味成分を含むので、ステビア甘味料とも呼ばれる。
ステビア抽出物の甘味成分としては、主成分であるSteviosideをはじめ、Rebaudioside-A、Rubusoside、Dulcoside-A、Steviobioside、Rebaudioside-C、Rebaudioside-B等のステビオール配糖体が知られている。
ステビア抽出物の甘味度は、ショ糖の200~300倍である。よって、ショ糖と同じ甘味を得るために使用されるステビア抽出物の量はショ糖と比較して大幅に少量でよいことから、実質的にノンカロリー甘味料として使用されている。また、ステビア抽出物は天然甘味料であることから、食品工業界でも多く使用されている(特許文献1)。
ステビア抽出物としては、Steviosideの含有比率の高いステビア抽出物(通常抽出品)、及びRebaudioside-Aの含有比率の高いステビア抽出物(調整品)が知られている。Steviosideの含有比率の高いステビア抽出物の甘味は比較的砂糖に似ているが、苦み等の異味が後味に残るという欠点がある。一方、Rebaudioside-Aの含有比率の高いステビア抽出物は、異味が少ない良質な甘味を呈する。
しかし、Rebaudioside-Aの含有比率の高いステビア抽出物においては、Rebaudioside-Aの含有比率が高いほど、甘味成分が高純度化されることになり、このステビア抽出物の水への溶解性が低下するという問題が生じる。
このため、Rebaudioside-Aの含有比率の高いステビア抽出物を溶液とする場合は、通常は、低濃度の溶液である。そのため、該溶液を乾燥又は固形化する場合には大量の水分を蒸発することが必要となり、生産効率が大幅に低下する。また、溶液からの異物除去のための濾過における作業が大幅に増加する。さらに、飲食品等の甘味料は、一定の濃度で保持した甘味料であることが一般的であるが、Rebaudioside-Aの含有比率の高いステビア抽出物は、再結晶化しやすいため、飲食品中で一定濃度で保持されることが困難であるといった問題がある。
そこで、本発明の目的は、難溶性であるRebaudioside-Aの含有比率の高いステビア抽出物の溶解性を改善し、該ステビア抽出物を水等の水性溶媒に溶解して溶液とした際に、該溶液の安定性を向上させるための手段を提供することを目的とする。
本発明は、以下の〔1〕~〔5〕を提供する。
〔1〕Rebaudioside-Aを90重量%以上含有するステビア抽出物及び水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように含む、ステビア組成物。
〔2〕水溶性多糖が、デンプン、アミロース、アミロペクチン及びデキストリンからなる群から選ばれる1種以上を含有する請求項1に記載のステビア組成物。
〔3〕水溶性多糖が、デキストリンである、請求項1又は2に記載のステビア組成物。
〔4〕上記〔1〕~〔3〕のいずれか一項に記載のステビア組成物を含有する飲食品。
〔5〕Rebaudioside-Aを90重量%以上含有するステビア抽出物に対して水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように混合し、得られる混合物を水性溶媒に溶解する、ステビア抽出物の溶解方法。
〔1〕Rebaudioside-Aを90重量%以上含有するステビア抽出物及び水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように含む、ステビア組成物。

本発明によれば、Rebaudioside-Aを90重量%以上含有するステビア抽出物の良質の甘味を保持しつつ、溶解性を改善し、該ステビア抽出物を水等の水性溶媒に溶解して溶液とした際に、該溶液の安定性を向上させることができる。
本発明は、Rebaudioside-Aを90重量%以上含有するステビア抽出物及び水溶性多糖を、乾燥重量比が、該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように含むステビア組成物、及びその製造方法に関する。また、本発明は、上記ステビア抽出物に対して上記水溶性多糖を、上記乾燥重量比となるように混合する、ステビア抽出物の溶解方法に関する。
本発明においては、Rebaudioside-Aを90重量%以上含有するステビア抽出物を用いる。Rebaudioside-Aを90重量%以上含有するステビア抽出物は、ショ糖と比較して異味が少なく良質な甘味を有するが、その半面、単独では水等の水性溶媒への溶解性が低い。ステビア抽出物とは、前述の通り、ステビアの植物体の抽出物を意味する。第69回JECFA(FAO/WHO合同食品添加物専門委員会)におけるステビア抽出物の規格は、Stevioside、Rubusoside、Dulcoside-A、Steviobioside、Rebaudioside-C、Rebaudioside-B、及びRebaudioside-Aを、これらの合計含量で95重量%以上含むものである。なお、この規格は、第69回JECFA(FAO/WHO合同食品添加物専門委員会)において、国際的に認知されたものである。本発明においても、上記JECFAの規格に適合したステビア抽出物が好ましく用いられるが、Rebaudioside-Aを90重量%以上含有するステビア抽出物であれば上記JECFAの規格に適合したステビア抽出物以外のものを用いてもよい。また、Rebaudioside-Aを90重量%以上含有するステビア抽出物に代えて、これと同様の組成を持つ化学合成品を用いてもよい。
Rebaudioside-Aを90重量%以上含有するステビア抽出物は、キク科植物であるステビアの植物体の全体又は一部、通常は葉部から、抽出されればよく、抽出条件に制限はない。製造方法の一例を挙げると以下の通りである。ステビアの葉部、例えば乾燥葉から、抽出溶媒(例えば、水、または含水有機溶媒(含水アルコール(メタノール、エタノール、多価アルコールなど)、含水アセトン等))にて抽出する。抽出液はそのまま、或いは濃縮して得られる濃縮液を、ステビア抽出物としてもよい。または必要に応じて、陽イオン交換樹脂、陰イオン交換樹脂、活性炭等の吸着剤を用いて上記抽出液或いはその濃縮液中のイオン性不純物を除去し、吸着剤に甘味成分を吸着させる。甘味成分を親水性溶媒で吸着剤から溶離して溶離液を濃縮して得られる濃縮液を得る。さらに該濃縮液を乾燥して乾燥物を得る。この乾燥物をステビア抽出物としてもよい。
このようにして得られるステビア抽出物が、Rebaudioside-Aを90重量%以上含んでいる場合には、本発明においてそのまま利用できる。一方、得られるステビア抽出物におけるRebaudioside-Aの含有量を90重量%以上に調整するために、さらに膜分離、アルコール抽出(例:メタノールを用いる抽出)、結晶化等の慣用手段を、適宜施してもよい。これらの操作は、必要に応じて、撹拌、加温(例えば30~70℃程度)、ろ過、乾燥粉砕等と組み合わせてもよい。また、以上の操作を繰り返し行ってもよい。結晶化にあたっては、結晶溶媒として、有機溶媒(エタノール、メタノール等)、上記有機溶媒に水を添加したもの、を適宣使用することができる。

本発明のステビア組成物において、Rebaudioside-Aを90重量%以上含むステビア抽出物は、1種類であってもよいし2種類以上の組み合わせであってもよい。2種類以上の組み合わせの例としては、抽出条件、Rebaudioside-Aの含有量、Rebaudioside-A以外の甘味成分の含有量等、が異なる2種類以上のステビア抽出物の組み合わせが挙げられる。
本発明において、水溶性多糖とは、水溶性の多糖(多数(例えば10個以上)の単糖が結合してなる糖)を意味する。水溶性多糖は、天然物および合成物のいずれであってもよい。水溶性多糖は1種類でもよいし、2種類以上の組み合わせであってもよい。
水溶性多糖としては、通常は食品または食品添加物として利用できる水溶性多糖が好ましい。水溶性多糖としては、例えば、ペクチン、グアーガム、アラビアガム、メチルセルロース、カルボキシメチルセルロース、プルラン、デキストラン、デンプン、アミロース、アミロペクチン、デキストリン、サイクロデキストリン等が挙げられる。このうち、単糖としてのグルコースが結合してなる水溶性多糖を含むことが好ましい。具体的には、デンプン、アミロース、アミロペクチン及びデキストリンからなる群から選ばれる1種以上が含まれることにより、Rebaudioside-Aを90重量%以上含有するステビア抽出物の溶解性をより顕著に向上させることができるので、好ましい。水溶性多糖としては、デキストリンが含まれることがより好ましく、デキストリンであることが更に好ましい。これにより、ステビア組成物は異味が少なくより良好な甘味を呈するものとなる。
本発明のステビア組成物においては、Rebaudioside-Aを90重量%以上含有するステビア抽出物と、水溶性多糖とを含む。両者の乾燥重量比は、ステビア抽出物:水溶性多糖=100:5~0.4:99.6であることが必要であり、好ましくはステビア抽出物:水溶性多糖=100:10~100:30である。これにより、Rebaudioside-Aを90重量%以上含有するステビア抽出物の溶解性を改善することができる。
本発明のステビア組成物は、さらに水性溶媒を含んでいてもよい。水性溶媒としては、水が例示される。ステビア組成物における水性溶媒の量は、水性溶媒に対する前記ステビア抽出物の濃度が0.5重量%以上となるような量であることが好ましく、4.0重量%以上となるような量であることがより好ましい。水性溶媒の上限は特に制限はないが、80重量%以下であることが好ましい。
本発明のステビア組成物は、本発明の目的に反しない範囲で、さらに他の成分を含んでいてもよい。係る成分としては、公知の添加剤が例示される。公知の添加剤としては例えば、本発明のステビア組成物以外の甘味料(例:糖アルコール、アセスルファムカリウム、スクラロース)が挙げられる。
本発明のステビア組成物の製造は、Rebaudioside-Aを90重量%以上含有するステビア抽出物及び水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように混合すればよく、それ以外の条件は特には限定されない。混合の際には、該ステビア抽出物と該水溶性多糖とをそのまま混合してもよいが、該ステビア抽出物の溶液(水性溶媒への溶液、通常は、水溶液)及び水溶性多糖の溶液(水性溶媒への溶液、通常は、水溶液)を混合してもよい。混合の際には、必要に応じて撹拌を加えてもよい。
本発明のステビア組成物は、優れた溶解性を有し、安定性に優れるほか、良質な甘味を呈するので、甘味料等の飲食品の一成分として利用することができ、また、飲食品添加物として利用することができる。飲食品の種類には特に制限はないが、水分を含む飲食品(例えば、ジュース、炭酸飲料、コーヒー飲料、茶飲料、乳飲料、アルコール飲料)であると、本発明の効果が発揮されるので、好ましい。飲食品添加物の種類には特に制限はないが、例えば、甘味料が挙げられる。
本発明の溶解方法においては、Rebaudioside-Aを90重量%以上含有するステビア抽出物に対して水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように混合し、得られる混合物を水に溶解する。
Rebaudioside-Aを90重量%以上含有するステビア抽出物は、単独では水への溶解性が非常に低いため、高濃度に溶解することが困難である。また、低濃度で溶解させる場合にも、水に溶解するまでに長時間がかかることが多い。さらに、一旦溶解しても、加熱、冷却、長時間放置等により容易に再結晶化する。しかし、本発明の溶解方法のように、水溶性多糖と共に特定の乾燥重量比となるように混合してから、得られる混合物を水に溶解することにより、水への溶解性は向上し、また、得られる溶液の安定性も向上する。
本発明の溶解方法においては、Rebaudioside-Aを90重量%以上含有するステビア抽出物及び水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように混合すればよく、それ以外の条件は特には限定されない。
Rebaudioside-Aを90重量%以上含有するステビア抽出物は、水性溶媒(例えば、水(常温))に対して、約0.5重量%未満の濃度で溶解させる場合であれば、溶解性及びその溶液の安定性をある程度保つことができる。しかし、上記濃度が、約0.5重量%以上である場合、溶解性、及びその溶液の安定性は著しく低下する。さらに上記濃度が約4.0重量%以上である場合、溶解性及びその溶液の安定性は劇的に低下する。つまり、本発明の溶解方法は、Rebaudioside-Aを90重量%以上含有するステビア抽出物を、水性溶媒に対し0.5重量%以上、中でも4.0重量%以上溶解させる手段として有用である。
本発明により、Rebaudioside-Aを90重量%以上含有するステビア抽出物の水性溶媒への溶解性の向上、再結晶化の防止が実現できる。また、Rebaudioside-Aを90重量%以上含有するステビア抽出物を、単独では溶解不可能な濃度において、水性溶媒に対し安定的に溶解させることができる。
以下に具体的な実施例を示すが、本発明はこれに限定されるものではない。
以下の実施例において、食品添加物公定書法に従って行われたステビアの定性試験とは、下記の試験を意味する。
試料0.06~0.12gを精密に量り、アセトニトリル/水混液(4:1)に溶かして正確に100mlとし、検液とする。別に定量用Steviosideを乾燥し、その約0.05gを精密に量り、アセトニトリル/水混液(4:1)に溶かして正確に100mlとし、標準液とする。検液及び標準液をそれぞれ10μlずつ量り、以下の操作条件で液体クロマトグラフィーを行う。検液のSteviosideのピーク面積Aa、Dulcoside-Aのピーク面積Ab、Rebaudioside-Cのピーク面積Ad、Rebaudioside-Aのピーク面積Ac、及び標準液のSteviosideのピーク面積Asをそれぞれ測定する。次式により各ステビオール配糖体の含量を求める。

Rebaudioside-A含量が91重量%であり、ステビア4成分(Stevioside、Rebaudioside-A、Rebaudioside-C及びDulcoside-A)の総含量が95重量%であるステビア抽出物(以下、ステビア甘味料1という。)の溶解性を調査した。
ステビア甘味料1の調製は以下のようにして行った。Rebaudioside-Aを豊富に含有する市販のステビア抽出物(守田化学工業、レバウディオP9)1部に対してメタノール9部を添加して攪拌懸濁させた。攪拌を続けながら室温から55℃に加温した後、攪拌放冷にて室温まで冷却した。その後12時間以上攪拌を続け、懸濁液を得た。この懸濁液をろ過し、得られた結晶を乾燥粉砕して粉末を得た。
Rebaudioside-A含量が91重量%であり、ステビア4成分(Stevioside、Rebaudioside-A、Rebaudioside-C及びDulcoside-A)の総含量が95重量%であるステビア抽出物(以下、ステビア甘味料1という。)の溶解性を調査した。

ステビア甘味料2の調製は以下のようにして行った。ステビア甘味料1(Rebaudioside-A含量が91重量%)を1部に対してメタノール9部を添加して攪拌懸濁させた。攪拌を続けながら室温から55℃に加温した後、攪拌放冷にて室温まで冷却した。その後12時間以上攪拌を続け、懸濁液を得た。この懸濁液をろ過し、得られた結晶を乾燥粉砕して粉末を得た。

ii)溶液4-iを12時間攪拌し続けても、未溶解物は発生しなかった。
iii)溶液4-iを95℃に加温、攪拌した後、65℃で攪拌を12時間継続した結果、未溶解物は発生しなかった。得られた溶液を、以下、溶液4-iiiという。
iv)溶液4-iiiを4℃で12時間放置した結果、未溶解物は発生しなかった。
[実施例5]
<ステビア甘味料2とアミロースとを含むステビア組成物の高濃度溶解性、並びに得られるステビア組成物高濃度溶液の安定性の調査>

以上のことから、水溶性多糖を混合することで、Rebaudioside-Aを90重量%以上含有するステビア抽出物の溶解性が改善されることが確認された。
[実験5]
ステビア甘味料と水溶性多糖とを含むステビア組成物の水溶液の甘味質試験を行った。なお、甘味質試験は5人のパネラーが下記の基準で評価した。

1)甘味立ちの速さ:ショ糖を基準にした官能評価

上記ステビア甘味料1(Rebaudioside-A含量91重量%)56重量%とアミロペクチン(和光純薬工業製)44重量%からなるステビア組成物を甘味強度がBrix7(ショ糖換算7%甘味)になるように超純水を添加して、ステビア組成物の溶解品を調整し、評価した。
表1から明らかなように、実施例7~9では、甘味立ちの早さ、甘味の切れの良さの点で、高い評価を得た。また、後味の点では実施例7のものが特に良好な評価であった。この結果は、本発明のステビア組成物が良質の甘味を呈すること、特にデキストリンと組み合わせた場合には異味が少なくより良質の甘味を呈することを示すものである。
Claims (5)
- Rebaudioside-Aを90重量%以上含有するステビア抽出物及び水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように含む、ステビア組成物。
- 水溶性多糖が、デンプン、アミロース、アミロペクチン及びデキストリンからなる群から選ばれる1種以上を含有する請求項1に記載のステビア組成物。
- 水溶性多糖が、デキストリンである、請求項1又は2に記載のステビア組成物。
- 請求項1~3のいずれか一項に記載のステビア組成物を含有する飲食品。
- Rebaudioside-Aを90重量%以上含有するステビア抽出物に対して水溶性多糖を、乾燥重量比が該ステビア抽出物:該水溶性多糖=100:5~0.4:99.6となるように混合し、得られる混合物を水性溶媒に溶解する、ステビア抽出物の溶解方法。
