Abstract

Description
本願発明は、特定遺伝子の発現抑制を作用機序とする膵臓がん治療用の組成物に関するものである。
遺伝子が転写するmRNAを標的として、遺伝子の発現を抑制する技術が知られている。RNA干渉法(RNA interference:以下「RNAi」と記載することがある)とは、短い相補的二重鎖RNA断片(siRNA)が相補的配列を有するmRNAの分解を促すことにより当該遺伝子の発現が特異的に抑制される現象を利用する方法である(非特許文献1)。また、アンチセンス核酸は、mRNAと2本鎖を形成することで、そのmRNAが担うべきタンパク質の合成を阻害するといった働きを持つ。これらの方法により、塩基配列が判明している任意の遺伝子の発現を人為的に特異的に抑制することが可能となり、アンチセンス核酸やsiRNAは核酸薬物としての有用性を示すことが明らかとなった。アンチセンス核酸やsiRNAの合成方法は既存の技術として確立しており、比較的安価に製造することが可能である。
一方、がんは遺伝子の産物である分子(核酸、タンパク)の機能異常によって正常細胞ががん細胞に変化することによりおこる。がん細胞の生存、増殖には多くの分子が関与しており、それらの中でどの分子が重要な役割を果たすかを明らかにし、それら重要な分子の発現を抑制することによってがん細胞の生存、増殖、造腫瘍性を阻止することが可能となる。よって、発現を抑制することによりがん細胞の生存、増殖、造腫瘍性を阻止する分子を同定することで、それを標的とした分子診療法の開発が可能となる。また、分子発現の抑制に特異的短二重鎖RNAを用いることにより、特異的配列を持つsiRNAを核酸薬物として使用することができる。
膵臓がんは世界的に難治性のがんとして知られており、日本における統計において臓器別がん死因の第5位であり、2000年の時点で罹患者は20045人、死亡者は19093人であり、罹患者のほとんどは死亡する極めて悪性の疾患である。また、罹患者は年々増加しており、1975-2000年の25年間に罹患者は3倍となっている(がんの統計、がん研究振興財団)。
また、世界的にみても、膵臓がんは西側世界において第5位のがん死因であり、悪性腫瘍の中で最も高い死亡率を示し、5年生存率は4%でしかない(例えば、非特許文献2)。これらの事実は現在の医療技術では膵臓がんを治癒させるのは困難であり、効果的な新しい診療法の開発が求められていることを示している。

膵臓がんの治療を目的として特定遺伝子の発現抑制を機序とする医薬に関する発明が知られている(特許文献1、2)。ただし、特許文献1は、アポトーシスの阻害に関連する遺伝子(Bcl-2ファミリー)の発現抑制を目的とするものであり、対象は膵臓がんに限定されない。また、がん細胞特異的にアポトーシスを生じさせるためには(すなわち、正常細胞を死滅させないためには)、RNA分子をがん細胞にのみ正確に導入する必要がある。一方、特許文献2は、膵臓がんに関連するCST6遺伝子およびGABRP遺伝子に関するものであり、これらの遺伝子が膵臓がん患者において発現亢進していることは確認されているものの、この遺伝子発現の亢進が膵臓がんの原因であることは未確認である。したがって、遺伝子発現を測定することによって膵臓がんの診断やその危険性の予見は可能であるが、遺伝子発現を抑制することによる膵臓がんの治療の可能性については全く不確定である。
なお、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子についてはそれぞれ以下が知られている。SON(SON DNA binding protein)はDNA結合分子として報告されているがその詳細な機能は明らかになっていない(非特許文献3-5)。MCM5(minichromosome maintenance complex component 5)はDNA 複製因子として機能する分子である(非特許文献6)。WDR5(WD repeat domain 5)はヒストンH3リジン4と相互作用し、発生に必須の機能を有している(非特許文献7)。PBK(PDZ binding kinase)はMAP2K群に属するリン酸化酵素であり、DNA障害性の反応に関与する(非特許文献8)。CENPA(centromere protein A)は染色体動原体を構成するヒストンH3に相当し、キネトコアに必須の働きをする(非特許文献9)。ただし、これらの遺伝子が膵臓がん細胞の生存、増殖、造腫瘍性に関係することは知られていない。
さらに、本願発明者は膵臓がん細胞における信号伝達経路関連遺伝子群を同定しているが(非特許文献10)、これらの遺伝子と膵臓がん細胞の増殖、生存、造腫瘍性との関連性は全く未知である。

前記のとおり、膵臓がんに対する有効な治療方法が求められている。アンチセンス核酸やsiRNAを使用する方法はその有効性が期待されているが、標的となる遺伝子については十分に解明されていない。特に、膵臓がん細胞の増殖、生存、造腫瘍性に関する遺伝子については知られていない。
本願発明は、以上の通りに事情に鑑みてなされたものであって、特定遺伝子の発現抑制により、膵臓がん細胞の増殖抑制、生存阻害および造腫瘍性阻止を効果として現す膵臓がん治療用の組成物を提供することを課題としている。
本願発明者は膵臓がん細胞において先に同定した信号伝達経路関連遺伝子群(非特許文献10)を対象に、それらの発現抑制が膵臓がん細胞の生存、増殖、造腫瘍性に及ぼす効果を試験管内及び生体内で調べ、特定の遺伝子発現が膵臓がん治療に有効であることを確認して本願発明を完成させた。
本願発明は、膵臓がんを治療するための組成物であって、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子からなる群より選択される少なくとも1つの遺伝子の発現を抑制するアンチセンス核酸またはsiRNAの薬学的有効量を含む膵臓がん治療用の組成物である。
さらに詳しくは、SON遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号1または3のヌクレオチド配列を含み、MCM5遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号5のヌクレオチド配列を含み、WDR5遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号7のヌクレオチド配列を含み、PBK遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号9のヌクレオチド配列を含み、そしてCENPA遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号11のヌクレオチド配列をそれぞれ含む組成物である。
さらに本願発明は、膵臓がん治療薬の有効成分をスクリーニングする方法であって、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子からなる群より選択される少なくとも1つの遺伝子の発現レベルを低下させるか、または当該遺伝子がコードするタンパク質の生物活性を低下させる候補物質を目的物質として選択することを特徴とする方法である。
本願発明の組成物によって、膵臓がん細胞の増殖、生存、造腫瘍性を効果的に阻害することによって、膵臓がんの進行を抑制し、あるいは膵臓がん細胞を消滅させ、または外科手術や化学療法なとの予後を良好なものとすることができる。
また、本願発明のスクリーニング方法によって、膵臓がんの治療薬物の有効成分となりえる低分子化合物等を同定することが可能となる。
本願発明の膵臓癌治療用の組成物は、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子(以下、「標的遺伝子」と総称することがある)の少なくとも1つの遺伝子発現を抑制するアンチセンス核酸またはsiRNAの薬学的有効量を含むことを特徴とする。
これら標的遺伝子のmRNA配列は公知であり(SON: GenBank/NM_138927、MCM5: GenBank/NM_006739、WDR5:GenBank/NM_017588、PBK:GenBank/NM_018492、CENPA:GenBank/NM_001809)、これらの配列情報に基づいてアンチセンス核酸やsiRNAを調製することができる。
本願発明のアンチセンス核酸は、標的遺伝子の核酸またはそれに対応するmRNAに結合して、遺伝子の転写もしくは翻訳を阻害し、mRNAの分解を促進し、および/または標的遺伝子がコードするタンパク質の発現を抑制して、最終的にタンパク質の機能を抑制する。このアンチセンス核酸は、標的遺伝子またはそのmRNAの配列(以下、「標的配列」と記載することがある)に特異的にハイブリダイズすることができる限り、標的配列と完全に相補的であるヌクレオチド、または1以上のヌクレオチドのミスマッチを有するヌクレオチドであってもよい。例えば、本発明のアンチセンス核酸は、少なくとも15個連続したヌクレオチド配列において、標的配列に対して少なくとも70%以上、好ましくは80%以上、より好ましくは90%以上、さらにより好ましくは95%以上の相同性を有するポリヌクレオチドである。なお、相同性は、当技術分野で周知のアルゴリズムを用いて決定することができる。
また、本発明のアンチセンス核酸は、修飾オリゴヌクレオチドであってもよい。例えば、チオエート化オリゴヌクレオチドを用いることで、アンチセンス核酸にヌクレアーゼ抵抗性を付与することができる。
本発明のアンチセンス核酸は、標的タンパク質をコードするDNAまたはmRNAに結合し、転写または翻訳を阻害し、mRNAの分解を促進し、標的タンパク質の発現を抑制し、標的タンパク質の機能を抑制することによって、膵臓がん細胞の増殖、生存、造腫瘍性に作用する。
本願発明の組成物は、実質的にこのアンチセンス核酸それ自体であってもよいが、必要に応じて、賦形剤、等張剤、溶解剤、安定化剤、保存剤、鎮痛剤等を加えることによって、錠剤、粉剤、顆粒剤、カプセル剤、リポソームカプセル、注射剤、溶液などの剤形として調製することができる。
本願発明のアンチセンス核酸を含有する組成物は、患部に直接注入すること、または患部に達するように血管に注入することによって、患者に投与される。アンチセンス封入剤も、持続性および膜透過性を増加させるために使用される。例えば、リポソーム、ポリ-L-リジン、脂質、コレステロール、リポフェクチンまたはこれらの誘導体が含まれる。
本発明のアンチセンス核酸の用量は、患者の病態に応じて、例えば、0.1~100 mg/kg、好ましくは0.1~50 mg/kgの範囲とすることができる。
本願発明のsiRNAは、標的遺伝子mRNAの翻訳を防止する二本鎖RNA分子であり、標的mRNAに対するセンス鎖およびアンチセンス鎖からなる。siRNAは標準的な方法によって細胞に導入することができる。例えば、RNAの鋳型となるDNA分子を細胞内に導入する方法などである。その際に、DNA分子からの転写物が単一体となるように、センス鎖およびアンチセンス鎖がヘアピン型に一体化した構造であってもよい。
本願発明において、siRNAは好ましくは、センス鎖およびアンチセンス鎖が500、200、100、50、もしくは25ヌクレオチドまたはそれ未満の長さを有する。より好ましくは、siRNAは19~25ヌクレオチド長である。
siRNAの配列は、Ambionのウェブサイト(http://www.ambion.com/techlib/misc/siRNA_finder.html)から入手できるsiRNA設計コンピュータープログラムを用いて設計することができる。
また、siRNAに対しては、適宜に修飾をおこなってもよい。例えば、コレステロール結合siRNAは薬理学的特性を改善することが示されている(Song et al. Nature Med. 9:347-51, 2003)。
各標的遺伝子に対するsiRNAの具体例としては、センス鎖として、SONに対しては配列番号1または3のヌクレオチド配列、MCM5に対しては配列番号5のヌクレオチド配列、WDR5に対しては配列番号7のヌクレオチド配列、PBKに対しては配列番号9のヌクレオチド配列、そしてCENPAに対しては配列番号11のヌクレオチド配列を含む。アンチセンス鎖は、上記のセンス鎖と略同一長であり、かつセンス鎖とハイブリダイズするヌクレオチド配列を選択することができる。より具体的には、それぞれのsiRNAは以下のヌクレオチド配列からなる2本鎖RNAである。

また、本願発明のsiRNAは、アンチセンス鎖として前記の配列番号2、4、6、8、10、12のそれぞれのヌクレオチド配列を含み、センス鎖は、アンチセンス鎖と略同一長であり、かつアンチセンス鎖とハイブリダイズするヌクレオチド配列を選択することができる。さらに、後記の実施例に示したように、短ヘアピン型RNA(shRNA)を発現するベクター(例えば、pSUPERベクター、Oligoengine)を用いて、前記のセンス鎖とアンチセンス鎖の各々5'側19塩基を含むshRNAとして発現させるようにしてもよい。shRNAは細胞内でプロセスされてsiRNAに変換される事が知られている。

これらのsiRNAは、標的mRNAと結合し得る形態で、細胞内に直接導入することができる。または、siRNAをコードするDNAを組込んだ発現ベクターを細胞内に導入する方法を採用することもできる。ベクターを細胞に導入するために、FuGENE(Rochediagnostices)、Lipofectamine 2000(Invitrogen)、Oligofectamine(Invitrogen)、Nucleofector(和光純薬工業)等のトランスフェクション促進剤を使用することができる。
発現ベクターを使用する場合は、例えば、標的mRNAに対するアンチセンス鎖を第1のプロモーター(例えば、クローニングされたDNAの3'側にあるプロモーター配列)によって転写させ、センス鎖を第2のプロモーター(例えば、クローニングされたDNAの5'側にあるプロモーター配列)によって転写させる。この場合、センス鎖とアンチセンス鎖は、細胞内においてハイブリダイズして2本鎖を構成する。または、2つのベクターを利用して、siRNAのセンス鎖およびアンチセンス鎖をそれぞれ別個に転写させることもできる。鋳型となるDNAは、例えばヘアピンのような二次構造を有するsiRNAをコードしてもよく、この場合は単一の転写産物がセンス鎖およびアンチセンス鎖の両方を有する。なお、ヘアピンループ構造を形成させるためには、任意のループ配列をセンス鎖とアンチセンス鎖との間に配置する。ループ配列は、前記Abionのウェブサイトのリストから選択することができる。
次に、本願発明のスクリーニング方法について説明する。
本願発明のスクリーニング方法は、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子の少なくとも1つの遺伝子の発現レベルを低下されるか、または当該遺伝子がコードするタンパク質の生物活性を低下させる候補物質を膵臓がん治療薬の有効成分として特定する。
すなわち、本願発明のスクリーニング方法は、標的遺伝子、その遺伝子がコードするタンパク質、もしくはその遺伝子の転写調節領域を利用して、遺伝子の発現もしくは遺伝子がコードするタンパク質の生物学的活性を抑制する化合物等をスクリーニングする。
例えば、標的タンパク質を対象とするスクリーニング方法は、以下の工程を含む。
(1)候補物質を、標的タンパク質と接触させる工程;
(2)標的タンパク質と候補物質との結合活性を検出する工程;および
(3)標的タンパク質へ結合する物質を選択する工程。
(1)候補物質を、標的タンパク質と接触させる工程;
(2)標的タンパク質と候補物質との結合活性を検出する工程;および
(3)標的タンパク質へ結合する物質を選択する工程。
工程(2)における標的タンパク質と候補物質との結合活性の検出は、免疫沈降法(例えば、Harlow and Lane, Antibodies, 511-52, Cold Spring Harbor Laboratory publications, New York, 1988)、ウェスト-ウェスタンブロット解析(Skolnik et al., Cell 65: 83-90, 1991)、ツーハイブリッドシステム(Dalton and Treisman, Cell 68: 597-612, 1992、Fields and Sternglanz, Trends Genet 10: 286-92, 1994)などによって行うことができる。また、アフィニティークロマトグラフィーや、表面プラズモン共鳴現象を用いたバイオセンサーを用いてスクリーニングすることもできる。
さらに、固定化された標的タンパク質を、低分子化合物ライブラリー、天然物質バンク、またはランダムファージペプチドディスプレイライブラリーと接触させ、結合する物質をスクリーニングする方法、標的タンパク質に結合する物質を単離するためのコンビナトリアルケミストリー技術に基づくハイスループットスクリーニング法(Wrighton et al., Science 273: 458-64, 1996; Verdine, Nature 384: 11-13, 1996; Hogan, Nature 384: 17-9, 1996)等を採用することもできる。
さらに、標的タンパク質の生物活性を低下させる物質をスクリーニングする場合には、例えば、以下の工程:
(1)候補物質を標的タンパク質と接触させる工程;
(2)標的タンパク質の生物学的活性を検出する工程;および
(3) 候補物質の非存在下で検出される標的タンパク質の生物学的活性と比較して、標的タンパク質の生物学的活性を抑制する物質を選択する工程、
を含む。
(1)候補物質を標的タンパク質と接触させる工程;
(2)標的タンパク質の生物学的活性を検出する工程;および
(3) 候補物質の非存在下で検出される標的タンパク質の生物学的活性と比較して、標的タンパク質の生物学的活性を抑制する物質を選択する工程、
を含む。
すなわち、本願発明における標的タンパク質は、膵臓がん細胞の増殖、生存、造腫瘍性を担っているため、これらの活性を阻害する物質を特定することができる。
このスクリーニングによって特定される物質は、例えば、標的タンパク質に対するアンタゴニストである。
本願発明のスクリーニング方法は、標的遺伝子の発現レベルを低下させる物質をスクリーニングする。この方法は、例えば、以下の工程:
(1)標的遺伝子を発現する細胞に候補物質を接触させる工程;
(2)細胞の標的遺伝子発現レベルを測定する工程;および
(3)標的遺伝子の発現レベルを、対照と比較して低下させる物質を選択する工程、
を含む。
(1)標的遺伝子を発現する細胞に候補物質を接触させる工程;
(2)細胞の標的遺伝子発現レベルを測定する工程;および
(3)標的遺伝子の発現レベルを、対照と比較して低下させる物質を選択する工程、
を含む。
標的遺伝子を発現する細胞には、例えば、膵臓がん細胞から樹立された細胞株が含まれる(例えば、KLM1、PK1、PK59、MIA PACa-2など)。また、正常細胞に標的遺伝子DNAをトランスフェクションしてもよい。発現レベルは、当該技術分野における公知の方法によって検出することができる。
本願発明のスクリーニング方法は、標的遺伝子の転写制御領域に対する作用物質を対象とすることもできる。この方法は、例えば、以下の工程:
(1)標的遺伝子の転写制御領域の下流にレポーター遺伝子を連結した融合DNAを含むベクターを導入した細胞に候補物質を接触させる工程;
(2)レポーター遺伝子の発現を測定する工程;および
(3)レポーター遺伝子の発現を低下させる物質を選択する工程、
を含む。
(1)標的遺伝子の転写制御領域の下流にレポーター遺伝子を連結した融合DNAを含むベクターを導入した細胞に候補物質を接触させる工程;
(2)レポーター遺伝子の発現を測定する工程;および
(3)レポーター遺伝子の発現を低下させる物質を選択する工程、
を含む。
このようなレポーターアッセイは、当該技術分野において公知の方法であり、レポーター遺伝子や宿主細胞は適宜に選択することができる。
なお、前記のスクリーニング方法における候補物質は、例えば、細胞抽出物、細胞培養上清、発酵微生物の産物、海洋生物からの抽出物、植物抽出物、精製タンパク質または粗タンパク質、ペプチド、非ペプチド化合物、合成低分子化合物、天然化合物などである。
以下、実施例を示して本願発明をさらに詳細かつ具体的に説明するが、本願発明は以下の例に限定されるものではない。
膵臓がん細胞の増殖に対するsiRNAの効果を検証した。
ヒト膵臓がん細胞株MIA PaCa-2を5x103/wellの濃度で96-well plateに播き、10% fetal bovine serum 入りDulbecco's Modified Eagle Medium (DMEM+10% FBS) を用いて37°C, 5% CO2下で一晩培養した。翌日、以下のsiRNAをoligofectamine (Invitrogen)を用いて10、 50、100nMの濃度で導入した。導入方法の詳細はInvitrogen社の使用説明書に依った。
SONに対するsiRNA:センス鎖-1(配列番号1)とアンチセンス鎖-1(配列番号2)
MCM5に対するsiRNA:センス鎖(配列番号5)とアンチセンス鎖(配列番号6)
WDR5に対するsiRNA:センス鎖(配列番号7)とアンチセンス鎖(配列番号8)
PBKに対するsiRNA:センス鎖(配列番号9)とアンチセンス鎖(配列番号10)
CENPAに対するsiRNA:センス鎖(配列番号11)とアンチセンス鎖(配列番号12)
導入した細胞についてDMEM+10% FBSで5日間培養し、細胞増殖の程度を3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromideを用いた比色測定法(MTT法)により毎日計測した。
結果として図1に導入4日目の増殖程度を示す。いかなるヒト遺伝子に対しても相補的でない配列を持つsiRNA (Negative control)導入細胞と比較して、各siRNAを導入した細胞の増殖は有意に抑制された。

導入した細胞についてDMEM+10% FBSで5日間培養し、細胞増殖の程度を3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromideを用いた比色測定法(MTT法)により毎日計測した。

また、センス鎖-2(配列番号3)とアンチセンス鎖-2(配列番号4)からなるSONに対するsiRNAも、センス鎖-1およびアンチセンス鎖-1からなるsiRNAと同様の細胞増殖抑制効果を示した。
以上の結果から、本願発明のsiRNAは膵臓がん細胞の増殖を効果的に抑制することが確認された。
比較的長期にわたる膵臓がん細胞の生存、増殖に対するsiRNAの効果を検証するため、短ヘアピン型RNA(shRNA)を発現するベクターであるpSUPERベクター(Oligoengine)を用いて配列番号1と2、5と6、7と8、9と10、11と12の各siRNAの5'側19塩基と同様の配列を含むshRNAを発現するベクターをOligoengine社使用説明書に沿って構築した。各々のshRNA発現ベクターを膵臓がん細胞株MIA PACa-2, PCI-35にLipofectamine (Invitrogen)を用いて導入し、G418による選択培養を4週間行って生存コロニー数を測定した。導入方法の詳細はInvitrogen社の使用説明書に依った。
結果として図2にMIA PaCa-2細胞の生存コロニー数を示す。Negative controlのsiRNAに相当するshRNA発現ベクターを導入した細胞と比較して、配列番号1-2、5-6、7-8、9-10、11-12のsiRNAに相当するshRNA発現ベクターを導入した細胞の生存コロニー数は有意に抑制された。
以上の結果から、本願発明のsiRNAは膵臓がん細胞の生存を効果的に阻害することが確認された。
膵臓がん細胞の造腫瘍性に対するsiRNAの効果を検証するため、配列番号1と2、7と8、9と10、11と12の各siRNAの5'側19塩基と同様の配列を含むshRNA発現ベクターを導入したMIA PACa-2細胞をG418入り選択培地で選択してshRNA恒常発現クローンを樹立し、それをヌードマウス皮下に移植して6週間観察し、形成された腫瘍径を測定した。
結果は図3に示したとおりである。Negative controlのsiRNAに相当するshRNA発現クローンと比較して、各shRNA発現ベクターを導入した細胞の造腫瘍性は有意に抑制された。
以上の結果から、本願発明のsiRNAは膵臓がん細胞の造腫瘍性を効果的に阻止することが確認された。なお、配列番号5と6のMCM5を標的とするsiRNA配列を含むshRNAを恒常的に発現するクローンは得られなかったため配列番号5と6のsiRNAの膵臓がん細胞の造腫瘍性に対する効果は不明である。
正常膵管上皮細胞と膵臓がん細胞のそれぞれの増殖に対するsiRNAの効果を検証した。
正常膵管上皮細胞(HPDE)と膵臓がん細胞株(MIA PACa-2)のそれぞれに、実施例1と同様の方法で各siRNAを導入し、実施例1と同様の方法で細胞増殖の程度を測定した。
結果は図4に示したとおりであり、各siRNAは正常細胞の増殖には有意な抑制効果を示さなかった。
Claims (7)
- 膵臓がんを治療するための組成物であって、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子からなる群より選択される少なくとも1つの遺伝子の発現を抑制するアンチセンス核酸またはsiRNAの薬学的有効量を含む膵臓がん治療用の組成物。
- SON遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号1または3のヌクレオチド配列を含む請求項1の組成物。
- MCM5遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号5のヌクレオチド配列を含む請求項1の組成物。
- WDR5遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号7のヌクレオチド配列を含む請求項1の組成物。
- PBK遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号9のヌクレオチド配列を含む請求項1の組成物。
- CENPA遺伝子の発現を抑制するsiRNAのセンス鎖が、配列番号11のヌクレオチド配列を含む請求項1の組成物。
- 膵臓がん治療薬の有効成分をスクリーニングする方法であって、SON遺伝子、MCM5遺伝子、WDR5遺伝子、PBK遺伝子およびCENPA遺伝子からなる群より選択される少なくとも1つの遺伝子の発現レベルを低下させるか、または当該遺伝子がコードするタンパク質の生物活性を低下させる候補物質を目的物質として選択することを特徴とする方法。
