本発明は、バイオマス原料の処理方法、並びに糖の製造方法、エタノールの製造方法、及び乳酸の製造方法に関する。
近年、地球温暖化対策の一環で、化石燃料の代替燃料として、植物資源から生産されるバイオエタノールが着目されてきている。
従来、バイオエタノールの原料としては、主にサトウキビなどの糖質や、穀物などのデンプン質が用いられていたが、これらの原料は、食物や飼料としても利用可能なものが多いため、各原料を用いたバイオエタノールの生産量を増加させると、食料や飼料の供給に影響を及ぼすことが懸念される。
一方、樹木や草、廃棄物などは、食料生産と競合しないため、エネルギー利用に有用なバイオマス原料である。
植物に由来するセルロース系バイオマスは、セルロース、ヘミセルロース、及びリグニンを主成分として含む。前記セルロースは、グルコースがβ-1,4-グルコシド結合して直鎖状に形成されている繊維状の多糖類であり、植物の細胞壁を構成する主要な成分である。前記ヘミセルロースは、前記セルロースと共に植物の細胞壁などを構成する多糖類である。前記リグニンは、ヒドロキシフェニルプロパンを基本単位として重合した高分子物質である。これらの中でも、前記セルロースは、地球上で最も存在量の多い天然高分子である。
そこで、近年、木質バイオマスや草本バイオマスなどのセルロースを含む原料を糖化し、各種燃料や化学原料として利用しようとする試みが広く行われている。

バイオマス原料からのエタノールの製造は、例えば、収集したバイオマス原料を糖化工程において糖に分解した後、発酵工程において酵母などの微生物を用いてエタノールに変換することにより行うことができる。
セルロース系バイオマスから糖を抽出するための糖化工程は、植物体内に高分子で存在しているセルロースと、ヘミセルロースとを可溶化すると同時に、グルコースなどの単糖類に分解する必要がある。糖化方法としては、従来、濃硫酸や濃塩酸などの強酸を用いて行われることが多かったが、環境負荷低減の観点から、これらの強酸の使用は避けることが望まれている。
セルロース系バイオマスから糖を抽出するための糖化工程は、植物体内に高分子で存在しているセルロースと、ヘミセルロースとを可溶化すると同時に、グルコースなどの単糖類に分解する必要がある。糖化方法としては、従来、濃硫酸や濃塩酸などの強酸を用いて行われることが多かったが、環境負荷低減の観点から、これらの強酸の使用は避けることが望まれている。
そこで、近年、濃硫酸や濃塩酸などの強酸による糖化に代わる手段として酵素を用いたバイオマス原料の糖化が広く研究されている。
酵素による糖化は、環境負荷低減、生成物の選択性、及び反応条件の穏和さの観点から望ましい手段であるが、セルロース系バイオマスの糖化に用いられる酵素は、既に実用化されているデンプンの酵素糖化に必要なアミラーゼ添加量と比較して、セルロースを糖化するために必要なセルラーゼの添加量が著しく多いことから、糖変換コストの増大が問題となっている。
酵素による糖化は、環境負荷低減、生成物の選択性、及び反応条件の穏和さの観点から望ましい手段であるが、セルロース系バイオマスの糖化に用いられる酵素は、既に実用化されているデンプンの酵素糖化に必要なアミラーゼ添加量と比較して、セルロースを糖化するために必要なセルラーゼの添加量が著しく多いことから、糖変換コストの増大が問題となっている。
また、この酵素糖化のためには、酵素を作用させ易くする目的から、予めバイオマス原料に対して前処理を行うことが必要となる。このバイオマス原料の前処理方法として様々な方法が知られているが、これらの中でも、希硫酸、加圧熱水などによる蒸煮処理などが一般的である(特許文献1~4参照)。しかしながら、前述したように硫酸の使用は好ましくない。また、バイオマス原料にこれらの前処理を行い、得られた処理物を酵素糖化に供する場合では、所望の程度の酵素糖化効率を得るためには該前処理を多段で行う必要があることや、200℃以上の高温にしなければならないことなどの点で問題である。
バイオマス原料を物理的手段により微細に粉砕することにより、化学的、生物化学的反応性が向上することも知られているが、粉砕のみにより充分な酵素糖化効率を得ようとすると、粉砕工程に多大なエネルギーを要し、経済合理性を失うおそれがある点で問題である。
更に、バイオマス原料をアンモニアあるいは有機アミンを用いて前処理することにより、その化学的、生物化学的反応性が向上することが知られている(特許文献5参照)が、前記前処理されたバイオマスであっても、その酵素糖化効率は未だ充分とはいえない点で問題である。
バイオマス原料を物理的手段により微細に粉砕することにより、化学的、生物化学的反応性が向上することも知られているが、粉砕のみにより充分な酵素糖化効率を得ようとすると、粉砕工程に多大なエネルギーを要し、経済合理性を失うおそれがある点で問題である。
更に、バイオマス原料をアンモニアあるいは有機アミンを用いて前処理することにより、その化学的、生物化学的反応性が向上することが知られている(特許文献5参照)が、前記前処理されたバイオマスであっても、その酵素糖化効率は未だ充分とはいえない点で問題である。
したがって、より酵素糖化効率を高めることのできる酵素糖化技術の開発、及び前記酵素糖化に適したバイオマス原料の前処理技術の開発が、未だ望まれているのが現状である。
本発明は、従来における前記諸問題を解決し、以下の目的を達成することを課題とする。即ち、本発明は、酵素糖化を効率的に行うことができ、そのため、糖の生産効率、エタノールの生産効率、及び乳酸の生産効率を向上させることが可能な、糖の製造方法、エタノールの製造方法、及び乳酸の製造方法、並びに前記糖の製造方法、エタノールの製造方法、及び乳酸の製造方法に用いられるバイオマス原料の処理方法を提供することを目的とする。
前記課題を解決するため、本発明者らは鋭意検討した結果、以下のような知見を得た。即ち、天然型セルロースであるセルロースI型を含むバイオマス原料を、アンモニアを含む処理剤で処理することにより得られる改質バイオマス原料を、セロビオヒドロラーゼII(Cel6A)を用いて酵素糖化することにより酵素糖化を効率的に行うことができ、したがって、糖の生産効率、エタノールの生産効率、及び乳酸の生産効率を格段に向上させることができるという知見である。
なお、本発明者らは以前に、天然型セルロースであるセルロースI型よりも低い結晶密度を有するセルロース(例えばセルロースIIII型)を酵素糖化の対象物として用いることにより、酵素糖化を効率的に行うことができること、及びセルロースI型を含むバイオマス原料を、アンモニア、特に超臨界アンモニアで処理することにより、セルロースIIII型を含む酵素糖化用セルロースを効率的に得ることができることを特許出願している(特開2008-161125号公報参照。)。
バイオマス原料を、アンモニアを含む処理剤で処理した後に、セロビオヒドロラーゼII(Cel6A)を用いて酵素糖化することにより、バイオマス原料の酵素糖化効率を格段に向上させることができることは、従来知られておらず、本発明者らによる新たな知見である。
本発明は、本発明者らによる前記知見に基づくものであり、前記課題を解決するための手段としては、以下の通りである。即ち、
<1> (a)バイオマス原料を、アンモニアを含む処理剤で処理することにより、改質バイオマス原料を得る工程、(b)該改質バイオマス原料を、セロビオヒドロラーゼII(Cel6A)で酵素糖化する工程、を含むことを特徴とするバイオマス原料の処理方法である。

本発明によれば、従来における前記諸問題を解決し、前記目的を達成することができ、酵素糖化を効率的に行うことができ、そのため、糖の生産効率、エタノールの生産効率、及び乳酸の生産効率を向上させることが可能な、糖の製造方法、エタノールの製造方法、及び乳酸の製造方法、並びに前記糖の製造方法、エタノールの製造方法、及び乳酸の製造方法に用いられるバイオマス原料の処理方法を提供することができる。
(バイオマス原料の処理方法)
本発明のバイオマス原料の処理方法は、(a)バイオマス原料を、アンモニアを含む処理剤で処理することにより、改質バイオマス原料を得る工程、(b)該改質バイオマス原料を、セロビオヒドロラーゼII(Cel6A)で酵素糖化する工程、を含み、必要に応じて、更にその他の処理を含む。
本発明のバイオマス原料の処理方法は、(a)バイオマス原料を、アンモニアを含む処理剤で処理することにより、改質バイオマス原料を得る工程、(b)該改質バイオマス原料を、セロビオヒドロラーゼII(Cel6A)で酵素糖化する工程、を含み、必要に応じて、更にその他の処理を含む。
<工程(a)>
前記工程(a)は、バイオマス原料を、アンモニアを含む処理剤で処理することにより、改質バイオマス原料を得る工程である。

-バイオマス原料-
前記バイオマス原料としては、特に制限はなく、目的に応じて適宜選択することができるが、セルロースI型を含むバイオマス原料が好ましい。
前記セルロースI型を含むバイオマス原料としては、特に制限はなく、目的に応じて適宜選択することができる。例えば、農業や林業などの生産活動に伴う残渣として得られる「廃棄物系バイオマス」や、エネルギーなどを得る目的で意図的に栽培して得られる「資源作物系バイオマス」などを使用することができる。
前記「廃棄物系バイオマス」としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、廃建材、間伐材、稲わら、麦わら、もみ殻、サトウキビバガスなどが挙げられる。
また、前記「資源作物系バイオマス」としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、サトウキビ、トウモロコシなどの食物としても栽培される糖質乃至デンプン系作物、及びセルロース類の利用を目的として栽培されるユーカリ、ポプラ、アカシア、ヤナギ、スギ、スイッチグラス、ネピアグラス、エリアンサス、ミスカンサス、ススキなどが挙げられる。
前記バイオマス原料としては、特に制限はなく、目的に応じて適宜選択することができるが、セルロースI型を含むバイオマス原料が好ましい。
前記セルロースI型を含むバイオマス原料としては、特に制限はなく、目的に応じて適宜選択することができる。例えば、農業や林業などの生産活動に伴う残渣として得られる「廃棄物系バイオマス」や、エネルギーなどを得る目的で意図的に栽培して得られる「資源作物系バイオマス」などを使用することができる。
前記「廃棄物系バイオマス」としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、廃建材、間伐材、稲わら、麦わら、もみ殻、サトウキビバガスなどが挙げられる。
また、前記「資源作物系バイオマス」としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、サトウキビ、トウモロコシなどの食物としても栽培される糖質乃至デンプン系作物、及びセルロース類の利用を目的として栽培されるユーカリ、ポプラ、アカシア、ヤナギ、スギ、スイッチグラス、ネピアグラス、エリアンサス、ミスカンサス、ススキなどが挙げられる。
前記セルロースI型を含むバイオマス原料は、木を原料とした「木質バイオマス」、草を原料とした「草本バイオマス」などにも分類される。本発明においては、木質バイオマス及び草本バイオマス共に使用することができるが、本発明の効果がより顕著に得られるとの観点から、セルロース含有量の多いバイオマスが好ましく使用される。
前記セルロースI型を含むバイオマス原料としては、前記したような各種バイオマスから精製などを行うことにより得られたセルロースI型そのものであってもよい。
また、前記セルロースI型を含むバイオマス原料は、1種単独で使用してもよいし、2種以上を併用してもよい。なお、天然型セルロースであるセルロースI型は、セルロースIα型とセルロースIβ型とに分類されるが、前記バイオマス原料に含まれるセルロースI型としては、これらのいずれであってもよく、またこれらの両者であってもよい。
また、前記セルロースI型を含むバイオマス原料は、1種単独で使用してもよいし、2種以上を併用してもよい。なお、天然型セルロースであるセルロースI型は、セルロースIα型とセルロースIβ型とに分類されるが、前記バイオマス原料に含まれるセルロースI型としては、これらのいずれであってもよく、またこれらの両者であってもよい。
前記バイオマス原料の状態としては、特に制限はなく、目的に応じて適宜選択することができ、収集されたものをそのまま使用してもよく、裁断、粉砕などにより適宜ある程度以下の大きさにしてから使用してもよい。
-アンモニアを含む処理剤-
前記アンモニアを含む処理剤としては、少なくともアンモニアを含有していれば、特に制限はなく、目的に応じて適宜選択することができ、必要に応じて、更にその他の化合物を含有する。

前記セルロースI型を含むバイオマス原料を、アンモニアを含む処理剤で処理する方法としては、特に制限はなく、目的に応じて適宜選択することができる。
前記アンモニアを含む処理剤で処理することにより、バイオマス原料中のセルロースI型の少なくとも一部が、より酵素糖化効率の高いセルロースIIII型へと変態する。
前記アンモニアとしては、特に制限はなく、目標とする糖化率、消費エネルギーなどを勘案し、それぞれに適した条件での処理を適宜選択することができ、例えば、液相、気相、亜臨界状態、超臨界状態などが挙げられるが、超臨界状態が、前記変態効率を向上する観点から好ましい。
前記アンモニアを含む処理剤で処理することにより、バイオマス原料中のセルロースI型の少なくとも一部が、より酵素糖化効率の高いセルロースIIII型へと変態する。
前記アンモニアとしては、特に制限はなく、目標とする糖化率、消費エネルギーなどを勘案し、それぞれに適した条件での処理を適宜選択することができ、例えば、液相、気相、亜臨界状態、超臨界状態などが挙げられるが、超臨界状態が、前記変態効率を向上する観点から好ましい。
--超臨界アンモニア流体による処理--
前記超臨界アンモニア流体を用いて処理する方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、前記セルロースI型を含むバイオマス原料と、アンモニアとをオートクレーブなどの反応器内に導入し、前記反応器内を加熱加圧して、アンモニアを超臨界状態にすることにより行うことができる。
前記超臨界アンモニア流体を用いて処理する方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、前記セルロースI型を含むバイオマス原料と、アンモニアとをオートクレーブなどの反応器内に導入し、前記反応器内を加熱加圧して、アンモニアを超臨界状態にすることにより行うことができる。
前記処理において、前記セルロースI型を含むバイオマス原料としては、収集されたものをそのまま使用してもよいが、ある程度小さくしてから使用することが、処理に必要なエネルギーを抑えることができる点で望ましい。
前記バイオマス原料のサイズとしては、特に制限はなく、目的に応じて適宜選択することができるが、例えば、5mm径以下が好ましく、1mm径以下がより好ましく、0.1mm径以下が更に好ましい。前記サイズが5mm径を超えると、処理が不十分となることがある。一方、前記サイズが前記更に好ましい範囲内であると、処理時間が短縮できる、使用するアンモニアの容量を少なくできるなどの点で有利である。
前記バイオマス原料のサイズとしては、特に制限はなく、目的に応じて適宜選択することができるが、例えば、5mm径以下が好ましく、1mm径以下がより好ましく、0.1mm径以下が更に好ましい。前記サイズが5mm径を超えると、処理が不十分となることがある。一方、前記サイズが前記更に好ましい範囲内であると、処理時間が短縮できる、使用するアンモニアの容量を少なくできるなどの点で有利である。
前記アンモニアを含む処理剤で処理を行う際の、前記アンモニアの使用量としては、特に制限はなく、目的に応じて適宜選択することができるが、前記セルロースI型を含むバイオマス原料1gに対して、10mg~300gが好ましく、100mg~150gがより好ましく、1g~50gが更に好ましい。
前記アンモニアの使用量が、前記セルロースI型を含むバイオマス原料1gに対して10mg未満であると処理が不十分となることがあり、300gを超えると処理の効率が悪くなることがある。一方、その使用量が前記更に好ましい範囲内であると、処理時間が短縮できる、使用する処理剤の量を少なくできるなどの点で有利である。
前記処理温度及び処理圧力としては、特に制限はなく、アンモニアが超臨界状態となる
範囲内で、目的に応じて適宜選択することができるが、0MPa~12.5MPaが好ましい。
前記アンモニアを含む処理剤で処理を行う処理温度としては、特に制限はなく、目的に応じて適宜選択することができるが、-35℃~140℃が好ましい。
前記アンモニアの使用量が、前記セルロースI型を含むバイオマス原料1gに対して10mg未満であると処理が不十分となることがあり、300gを超えると処理の効率が悪くなることがある。一方、その使用量が前記更に好ましい範囲内であると、処理時間が短縮できる、使用する処理剤の量を少なくできるなどの点で有利である。
前記処理温度及び処理圧力としては、特に制限はなく、アンモニアが超臨界状態となる
範囲内で、目的に応じて適宜選択することができるが、0MPa~12.5MPaが好ましい。
前記アンモニアを含む処理剤で処理を行う処理温度としては、特に制限はなく、目的に応じて適宜選択することができるが、-35℃~140℃が好ましい。
前記アンモニアを含む処理剤で処理を行う時間としては、特に制限はなく、用いる前記セルロースI型を含むバイオマス原料の量、前記処理圧力、前記処理温度などに応じ、所望の程度のセルロースI型からセルロースIIII型への変態が進行する範囲内で適宜選択することができるが、5分間~10時間が好ましく、10分間~5時間がより好ましく、30分間~2時間が更に好ましい。
前記処理時間が、5分間未満であると所望の程度のセルロースI型からセルロースIIII型への変態が進行しないことがあり、10時間を超えるとそれ以上セルロースI型からセルロースIIII型への変態は進行せず、全体として非効率となることがある。一方、前記処理時間が、前記より好ましい範囲内であると、効率よく、セルロースI型からセルロースIIII型への変態を進行させることができる点で有利である。
前記処理時間が、5分間未満であると所望の程度のセルロースI型からセルロースIIII型への変態が進行しないことがあり、10時間を超えるとそれ以上セルロースI型からセルロースIIII型への変態は進行せず、全体として非効率となることがある。一方、前記処理時間が、前記より好ましい範囲内であると、効率よく、セルロースI型からセルロースIIII型への変態を進行させることができる点で有利である。
--その他の化合物--
前記アンモニアを含む処理剤中のその他の化合物としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、二酸化炭素、窒素、エチレン、メタン、エタン、プロパン、ブタン、ペンタン、ヘキサン、トルエン、ベンゼン、フェノール、ジオキサン、キシレン、アセトン、クロロホルム、四塩化炭素、エタノール、メタノール、プロパノール、ブタノールなどが挙げられる。
なお、前記その他の化合物としては、水は使用しないことが好ましい。前記水を使用すると、得られたセルロースIIII型が、セルロースI型に戻ってしまう場合がある。
前記その他の化合物の含有量としても、特に制限はなく、目的に応じて適宜選択することができる。
前記アンモニアを含む処理剤中のその他の化合物としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、二酸化炭素、窒素、エチレン、メタン、エタン、プロパン、ブタン、ペンタン、ヘキサン、トルエン、ベンゼン、フェノール、ジオキサン、キシレン、アセトン、クロロホルム、四塩化炭素、エタノール、メタノール、プロパノール、ブタノールなどが挙げられる。
なお、前記その他の化合物としては、水は使用しないことが好ましい。前記水を使用すると、得られたセルロースIIII型が、セルロースI型に戻ってしまう場合がある。
前記その他の化合物の含有量としても、特に制限はなく、目的に応じて適宜選択することができる。
-改質バイオマス原料-
前記改質バイオマス原料は、前記アンモニアを含む処理剤で処理を行うことにより得ることができる。
本願における「改質バイオマス原料」とは、セルロースI型を含むバイオマス原料を、アンモニアを含む処理剤により処理したものを意味するが、バイオマス原料中に含まれるセルロースI型の少なくとも一部がセルロースIIII型へと変態したものであることが好ましい。前記セルロースIIII型は、前記セルロースI型より結晶密度の低いセルロースである。前記セルロースIIII型は、その結晶密度の低さから、酵素が作用し易い点で有利である。
また、前記改質バイオマス原料は、前記処理により前記バイオマス原料に含まれるヘミセルロースが改質され、より酵素が作用し易い状態へと変化しているため、酵素糖化効率を向上させることが可能となる。
前記改質バイオマス原料は、前記アンモニアを含む処理剤で処理を行うことにより得ることができる。
本願における「改質バイオマス原料」とは、セルロースI型を含むバイオマス原料を、アンモニアを含む処理剤により処理したものを意味するが、バイオマス原料中に含まれるセルロースI型の少なくとも一部がセルロースIIII型へと変態したものであることが好ましい。前記セルロースIIII型は、前記セルロースI型より結晶密度の低いセルロースである。前記セルロースIIII型は、その結晶密度の低さから、酵素が作用し易い点で有利である。
また、前記改質バイオマス原料は、前記処理により前記バイオマス原料に含まれるヘミセルロースが改質され、より酵素が作用し易い状態へと変化しているため、酵素糖化効率を向上させることが可能となる。
前記改質バイオマス原料中の前記セルロースIIII型の割合としては、特に制限はなく、目的に応じて適宜選択することができるが、前記セルロースIIII型が多い程、優れた酵素糖化効率が得られる点で好ましい。
また、前記改質バイオマス原料は、前記セルロースIIII型以外にも、例えば、セルロースI型(セルロースIα型、セルロースIβ型)や、その他の成分、例えば、ヘミセルロース、リグニンなどが含まれていてもよい。ただし、酵素糖化の効率向上の観点から、ヘミセルロース、リグニンは含まれない、あるいはその含有量が少ないことが好ましい。
また、前記改質バイオマス原料は、前記セルロースIIII型以外にも、例えば、セルロースI型(セルロースIα型、セルロースIβ型)や、その他の成分、例えば、ヘミセルロース、リグニンなどが含まれていてもよい。ただし、酵素糖化の効率向上の観点から、ヘミセルロース、リグニンは含まれない、あるいはその含有量が少ないことが好ましい。
前記改質バイオマス原料において、前記セルロースI型の少なくとも一部が前記セルロースIIII型へと変換されたことを確認する方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、X線回折、FT-IR、固体NMRなどにより確認することができる。
なお、前記改質バイオマス原料において、セルロースは、その分子構造の間に他の化合物を有していてもよい。例えば、改質バイオマス原料は、前記したように、天然型セルロースであるセルロースI型を含むバイオマス原料を、アンモニアを含む処理剤で処理することにより得ることができるが、その処理工程で生成するセルロースと、アンモニアとの複合体(以下、「セルロース・アンモニア複合体」と称することがある。)の状態であってもよい。しかしながら、前記セルロース・アンモニア複合体は、酵素糖化時におけるpHの調整が困難であり、また、水の作用を受けることによりセルロースI型に戻ってしまう性質を有することなどから、酵素糖化時には、前記セルロース・アンモニア複合体からアンモニアを除去した状態の改質バイオマス原料を使用することが好ましい。
したがって、前記アンモニアによる処理の後には、前記セルロース・アンモニア複合体から、アンモニアを除去する除去工程を設けることが好ましい。
前記アンモニアの除去方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、前記アンモニアによる処理後、得られた前記セルロース・アンモニア複合体を含む改質バイオマス原料を、メタノール、エタノール、アセトンなどで洗浄する方法、減圧乾燥する方法、処理剤の沸点以上の温度で乾燥させる方法などが挙げられる。これらの中でも、アンモニアの沸点以上の温度(例えば、常温~50℃)で、常圧又は減圧下にて乾燥させる方法が、有機溶媒を使用せず、安全性に優れる点で好ましい。

前記セロビオヒドロラーゼII(Cel6A)を入手する方法としては、特に制限はなく、目的に応じて適宜選択することができるが、前記担子菌を培養し、NCBIなどのデータベースより入手した前記セロビオヒドロラーゼII(Cel6A)の遺伝子配列を用いてクローニングすることにより、セロビオヒドロラーゼII(Cel6A)の組換体タンパク質を得る方法が好ましい。また、市販品を用いることもできる。
前記担子菌の培養の方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、寒天培地を用いた固体培養方法、液体培地を用いた液体培養方法などが挙げられる。これらの中でも、液体培養方法が、セロビオヒドロラーゼII(Cel6A)を多く製造できる点で好ましい。
前記液体培地としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、セルロースを含有するKremer and Wood培地(Kremer, S. M., and P. M. Wood., 1992, Evidence that cellobiose oxidase from Phanerochaete chrysosporium is primarily an Fe(III) reductase. Kinetic comparison with neutrophil NADPH oxidase and yeast flavocytochrome b2., Eur. J. Biochem., 205:133-138.)などが挙げられる。
前記液体培地としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、セルロースを含有するKremer and Wood培地(Kremer, S. M., and P. M. 前記培養の温度としては、特に制限はなく、目的に応じて適宜選択することができるが、20℃~45℃が好ましく、30℃~40℃がより好ましく、37℃が更に好ましい。前記培養の温度が20℃未満であると、担子菌の生育が遅くなることがあり、45℃を超えると担子菌が成育しないことがある。一方、前記培養の温度が前記更に好ましい範囲であると、セロビオヒドロラーゼII(Cel6A)を効率よく製造できる点で有利である。
前記培養の日数としては、特に制限はなく、目的に応じて適宜選択することができるが、1日~30日が好ましく、2日~14日がより好ましく、3日~7日が更に好ましい。前記培養の日数が1日未満であると、担子菌数が少なく、セロビオヒドロラーゼII(Cel6A)の製造量が少ないことがあり、30日を超えると、死菌となる担子菌数が多くなったり、製造されたセロビオヒドロラーゼII(Cel6A)が分解されてしまうことがある。一方、前記培養の日数が前記更に好ましい範囲であると、セロビオヒドロラーゼII(Cel6A)を効率よく製造できる点で有利である。
--酵素糖化--
前記酵素糖化の際の前記セロビオヒドロラーゼII(Cel6A)の使用量としては、特に制限はなく、目的に応じて適宜選択することができるが、例えば、改質バイオマス原料1gに対して、1mg~1,000mgが好ましく、2mg~500mgがより好ましく、5mg~200mgが更に好ましい。前記酵素の使用量が、前記酵素糖化用原料1gに対して1mg未満であると酵素糖化が不十分となることがあり、1,000mgを超えると酵素使用量が増大し効率が悪くなる。一方、前記酵素の使用量が、前記更に好ましい範囲内であると酵素添加量に対して得られる糖の量が多い点で有利である。

前記酵素糖化の際のpHとしては、特に制限はなく、目的に応じて適宜選択することができるが、例えば、2~8が好ましく、3~7がより好ましく、4~6が更に好ましい。前記pHが、2未満又は8を超えると酵素が失活することがある。一方、前記pHが、前記更に好ましい範囲内であると酵素添加量に対して得られる糖の量が多い点で有利である。
なお、前記酵素の加水分解速度としては、特に制限はなく、目的に応じて適宜選択することができるが、前記改質バイオマス原料と、前記セロビオヒドロラーゼII(Cel6A)とが加水分解反応を行う表面密度が0.9以下のとき、該セロビオヒドロラーゼII(Cel6A)1分子あたりの加水分解速度が1分間に5回以上であることが好ましい。

<用途>
前記エタノールの製造方法により得られたエタノールは、例えば、燃料用エタノール、工業用エタノールなどとして好適に利用可能である。前記エタノールは前記バイオマス原料から得ることができるので、前記バイオマス原料となる植物を生産できる限りは再生産が可能であり、また、前記植物は栽培時に大気中の二酸化炭素を吸収するため、前記エタノールを燃焼させて二酸化炭素が発生したとしても、大気中の二酸化炭素濃度を増加させることにはならない。したがって、前記エタノールは、地球温暖化防止に望ましいエネルギー源ということができる。また、このようなエタノールは、近年特に、ガソリンに混合し、環境に優しい自動車燃料として使用することが期待されている。
前記エタノールの製造方法により得られたエタノールは、例えば、燃料用エタノール、工業用エタノールなどとして好適に利用可能である。前記エタノールは前記バイオマス原料から得ることができるので、前記バイオマス原料となる植物を生産できる限りは再生産が可能であり、また、前記植物は栽培時に大気中の二酸化炭素を吸収するため、前記エタノールを燃焼させて二酸化炭素が発生したとしても、大気中の二酸化炭素濃度を増加させることにはならない。したがって、前記エタノールは、地球温暖化防止に望ましいエネルギー源ということができる。また、このようなエタノールは、近年特に、ガソリンに混合し、環境に優しい自動車燃料として使用することが期待されている。
本発明の糖の製造方法により得られる糖を前記エタノールを産生する酵母などに代えて、それぞれ目的とするアルコール類を産生する微生物を使用して発酵せしめることにより、エタノール以外のアルコール類を製造することもできる。例えば、アセトン・ブタノール菌を使用した発酵を行うことにより、ブタノールを製造することができる。

前記乳酸菌としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、ラクトバチルス マニホティヴォランス(Lactobacillus manihotivorans)、ラクトバチルス プランタラム(Lactobacillus plantarum)、ストレプトコッカス サーモフィルス(Streptococcus thermophilus)、ラクトバチルス ブルガリカス(Lactobacillus bulgaricus)などが挙げられる。なお、前記乳酸菌は、天然の乳酸菌であってもよいし、遺伝子組換乳酸菌であってもよい。

<用途>
前記乳酸の製造方法により得られた乳酸は、例えば、化学的に重合させて、ポリ乳酸を製造することに好適に利用可能である。現在は、トウモロコシなどのデンプンから製造されることが多い乳酸を、食料には供し得ないセルロースを含むバイオマス原料から生産可能になることが望ましく、前記乳酸の製造方法によれば、このようなセルロースを含むバイオマス原料からの効率的なポリ乳酸の製造を可能とすることができる。
前記乳酸の製造方法により得られた乳酸は、例えば、化学的に重合させて、ポリ乳酸を製造することに好適に利用可能である。現在は、トウモロコシなどのデンプンから製造されることが多い乳酸を、食料には供し得ないセルロースを含むバイオマス原料から生産可能になることが望ましく、前記乳酸の製造方法によれば、このようなセルロースを含むバイオマス原料からの効率的なポリ乳酸の製造を可能とすることができる。
本発明の糖の製造方法により得られる糖を、前記乳酸菌に代えて、それぞれ目的とする有機酸を産生する微生物を使用して発酵せしめることにより、乳酸以外の有機酸、例えば、クエン酸、コハク酸、リンゴ酸、シュウ酸などを製造することもできる。
以下に本発明の実施例及び比較例を挙げて本発明を具体的に説明するが、本発明はこれらの実施例に何ら限定されるものではない。
(実施例1~6)
<Cel6Aによるバイオマス原料の処理>

後述する方法により、バイオマス原料を、アンモニアを含む処理剤で処理することにより、改質バイオマス原料を得た。

緑藻のシオグサ細胞壁(セルロースIα型/Iβ型=7:3)をバイオマス原料として用いた。
セルロースの精製は、5.0質量%水酸化カリウム水溶液中にて室温で一晩浸漬させる処理と、pH4.9の酢酸緩衝液で緩衝した0.3質量%NaClO2水溶液中にて80℃で3時間処理する操作とを3回繰り返して行った。精製後、水洗し、凍結乾燥して保存した(以下、「セルロースIα」と称することがある。)。
前記セルロースIαは、後述するセルロースIIIIを含む改質バイオマス原料の調製に用いた。

前記セルロースIαを、以下の操作により、超臨界状態のアンモニアによる処理に供した。
30mL容ポータブルリアクターに、前記シオグサ200mgを入れ、ドライアイス入りのメタノールバスで-13℃に冷却した。このリアクターにアンモニアガスを0.5MPaの定圧で30分間流入し、試料を完全に液体アンモニアに浸漬させた。リアクターの蓋を閉め、室温で15分間程放置した。次いで、140℃のオイルバス中にて1時間処理した。このとき、リアクター中の圧力はゆっくりと上昇して、20分間後に13MPaに達し、その後は一定であった(アンモニアの臨界温度Tc=405.6K、臨界圧力Pc=11.28MPa)。前記処理後、リアクターをオイルバスから取り出し、ドラフト中で直ちにアンモニアガスをリークした。前記処理後のセルロースをメタノールで洗浄し、乾燥させることにより、セルロースIIII型を含む改質バイオマス原料を得た(以下、「セルロースIIII」と称することがある。)。
前記セルロースIαを、以下の操作により、超臨界状態のアンモニアによる処理に供した。

Wood., 1992, Evidence that cellobiose oxidase from Phanerochaete chrysosporium is primarily an Fe(III) reductase. Kinetic comparison with neutrophil NADPH oxidase and yeast flavocytochrome b2., Eur. J. Biochem., 205:133-138.)で、Habu, N., K. Igarashi, M. Samejima, B. Pettersson, and K. E. Eriksson., 1997, Enhanced production of cellobiose dehydrogenase in cultures of Phanerochaete chrysosporium supplemented with bovine calf serum., Biotechnol. Appl. Biochem., 26:98の記載に基づいて、3日間培養した。

前記培養後の培養液をろ過し、ガラス繊維濾紙(ADVANTEC(登録商標) GA-100;東洋濾紙(株)製)を用いて菌糸体を分離した。次いで、分離した菌糸体を液体窒素で凍結し、ISOGEN((株)ニッポンジーン製)を用いて、製造者のマニュアルに基づいて、全RNAを約200mg抽出した。
前記抽出された全RNA 1μgから、Oligotex(TM)-dT30<Super>(タカラバイオ(株)製)を用いてmRNAを精製した。
次いで、前記mRNAから、逆転写酵素(ReverTraAce;東洋紡績(株)製)と、3’RACEアダプタープライマー(Invitrogen社製)とを用いて、製造者のマニュアルに基づいて、First-strand cDNAを合成した。
前記抽出された全RNA 1μgから、Oligotex(TM)-dT30<Super>(タカラバイオ(株)製)を用いてmRNAを精製した。
次いで、前記mRNAから、逆転写酵素(ReverTraAce;東洋紡績(株)製)と、3’RACEアダプタープライマー(Invitrogen社製)とを用いて、製造者のマニュアルに基づいて、First-strand cDNAを合成した。
NCBIデータベース上にあるファネロケーテ クリソスポリウム(Phanerochaete chrysosporium)由来Cel6A遺伝子(AAB32942)(配列番号:1)をもとに、以下のプライマーを設計した。

上記プライマーと、ポリメラーゼとしてKOD-Plus(version2;東洋紡績(株)製)とを用い、前記のように調製したcDNAを鋳型としてPCRを行う(94℃、2分間を1サイクル、98℃、10秒間・68℃、1分30秒間を25サイクル、4℃で終了)ことで、PcCel6Aの成熟体タンパク質をコードする遺伝子(21番アミノ酸以降)を得た。
上記で得られたPCR産物は、Zero Blunt(登録商標) TOPO(登録商標) PCR cloning kit(Invitrogen社製)と、大腸菌E.coli JM109株(タカラバイオ(株)製)とを用いてクローン化した。

ミニプレップを行って調製したPcCel6A遺伝子を含むTOPOベクター、及び酵母発現用ベクターpPICZαA(Invitrogen社製)を、制限酵素EcoRI、及びNotI(タカラバイオ(株)製)によって切断し、得られたフラグメントをアガロース電気泳動で分離した後、ゲル抽出により前記フラグメントを得た。
制限酵素処理されたpPICZαAと、PcCel6A遺伝子とを、それぞれ20ng用い、DNA Ligation Kit<Mighty Mix>(タカラバイオ(株)製)によってライゲーションし、大腸菌E.coli JM109株(タカラバイオ(株)製)を用いてクローン化した。ミニプレップを行って調製したPcCel6A遺伝子を含む酵母発現用ベクターpPICZαA(以下、「pPICZαA/PcCel6A」と称することがある。)から、Ste13シグナル切断サイトを切除するために、以下のプライマーを設計し、pPICZαA/PcCel6Aを鋳型としてPCRを行った(94℃、2分間を1サイクル、98℃、10秒間・68℃、1分30秒間を15サイクル、4℃で終了)。

---形質転換体の作製---
前記ミニプレップにより得られた、Ste13シグナル切断サイトを切除したpPICZαA/PcCel6A-Ste(-)を、制限酵素BstXI(タカラバイオ(株)製)によって直鎖化し、エレクトロポレーション法によって酵母(Pichia pastoris KM-71H)株に導入した。形質転換体の選抜は、抗生物質(Zeocin)耐性を指標に行った。
前記ミニプレップにより得られた、Ste13シグナル切断サイトを切除したpPICZαA/PcCel6A-Ste(-)を、制限酵素BstXI(タカラバイオ(株)製)によって直鎖化し、エレクトロポレーション法によって酵母(Pichia pastoris KM-71H)株に導入した。形質転換体の選抜は、抗生物質(Zeocin)耐性を指標に行った。
---形質転換酵母の培養---
上述の形質転換体を、25μg/mLのZeocinを含むYPG培地(1質量%イースト エクストラクト、2質量%ポリペプトン、1質量%グリセロール)10mLで30℃、300rpmの条件により、往復振とう培養器で24時間培養した後、200mLのYPG培地の入った三角フラスコに接種し、回転振とう培養器(30℃、150rpm)で更に24時間培養した。遠心分離(3,000g、10分)によって菌体を回収した後、50mLのYPM培地(1質量%イースト エクストラクト、2質量%ポリペプトン、1質量%メタノール)に菌体を移し、24時間ごとに終濃度1質量%となるようにメタノールを加えながら、更に回転振とう培養器(26.5℃、150rpm)で96時間培養した。遠心分離(3,000g、10分)によって得られた培養上清を粗酵素液とした。
上述の形質転換体を、25μg/mLのZeocinを含むYPG培地(1質量%イースト エクストラクト、2質量%ポリペプトン、1質量%グリセロール)10mLで30℃、300rpmの条件により、往復振とう培養器で24時間培養した後、200mLのYPG培地の入った三角フラスコに接種し、回転振とう培養器(30℃、150rpm)で更に24時間培養した。遠心分離(3,000g、10分)によって菌体を回収した後、50mLのYPM培地(1質量%イースト エクストラクト、2質量%ポリペプトン、1質量%メタノール)に菌体を移し、24時間ごとに終濃度1質量%となるようにメタノールを加えながら、更に回転振とう培養器(26.5℃、150rpm)で96時間培養した。遠心分離(3,000g、10分)によって得られた培養上清を粗酵素液とした。
---PcCel6Aの製造及び精製---
前述のようにして得られた粗酵素液に、硫酸アンモニウムを70質量%飽和となるように加え、遠心分離(15,000g、30分)によって沈殿を回収した。前記沈殿を、1M硫酸アンモニウムを含む20mM酢酸ナトリウム緩衝液(pH5.0)に溶解させた。
1Mの硫酸アンモニウムを含有する20mMの酢酸ナトリウム緩衝液(pH5.0)で平衡化したPhenyl-Toyopearl 650Sカラム(26mm×120mm、東ソー(株)製)を用いて、前記溶液を分画した。得られたPcCel6Aは、300mLの逆勾配で、20mM酢酸ナトリウム緩衝液(pH5.0)へ溶出した。
その後、前記PcCel6Aを含む分画を集め、20mMトリス塩酸緩衝液(pH8.0)に対して平衡化した。次いで、20mMトリス塩酸緩衝液(pH8.0)で平衡化したSuperQ-Toyopearl 650S カラム(9mm×120mm、東ソー(株)製)に前記平衡化したPcCel6Aを含む溶液を加えた。PcCel6Aは、0M~0.5Mの塩化ナトリウム100mLの直線勾配でカラムから溶出した。
得られたPcCel6Aは、SDS-PAGE(12質量%ポリアクリルアミドゲル)で、単一のバンドを与えた。また、得られたPcCel6AのN末端のアミノ酸配列は、プロテインシーケンサー(Model 491 cLc;Applied Biosystems社製)により確認した。
その結果、形質転換酵母で得られたPcCel6AのN末端アミノ酸配列は、QASEWGQCGGIG(配列番号:6)であり、担子菌ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium)由来のCel6Aと同じであることが認められた。
精製したPcCel6Aは、50mM酢酸緩衝液(pH5.0)に緩衝液交換し、以下の糖取得工程に用いた。
前述のようにして得られた粗酵素液に、硫酸アンモニウムを70質量%飽和となるように加え、遠心分離(15,000g、30分)によって沈殿を回収した。前記沈殿を、1M硫酸アンモニウムを含む20mM酢酸ナトリウム緩衝液(pH5.0)に溶解させた。
1Mの硫酸アンモニウムを含有する20mMの酢酸ナトリウム緩衝液(pH5.0)で平衡化したPhenyl-Toyopearl 650Sカラム(26mm×120mm、東ソー(株)製)を用いて、前記溶液を分画した。得られたPcCel6Aは、300mLの逆勾配で、20mM酢酸ナトリウム緩衝液(pH5.0)へ溶出した。
その後、前記PcCel6Aを含む分画を集め、20mMトリス塩酸緩衝液(pH8.0)に対して平衡化した。次いで、20mMトリス塩酸緩衝液(pH8.0)で平衡化したSuperQ-Toyopearl 650S カラム(9mm×120mm、東ソー(株)製)に前記平衡化したPcCel6Aを含む溶液を加えた。PcCel6Aは、0M~0.5Mの塩化ナトリウム100mLの直線勾配でカラムから溶出した。
得られたPcCel6Aは、SDS-PAGE(12質量%ポリアクリルアミドゲル)で、単一のバンドを与えた。また、得られたPcCel6AのN末端のアミノ酸配列は、プロテインシーケンサー(Model 491 cLc;Applied Biosystems社製)により確認した。
その結果、形質転換酵母で得られたPcCel6AのN末端アミノ酸配列は、QASEWGQCGGIG(配列番号:6)であり、担子菌ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium)由来のCel6Aと同じであることが認められた。
精製したPcCel6Aは、50mM酢酸緩衝液(pH5.0)に緩衝液交換し、以下の糖取得工程に用いた。
--糖取得工程、及び糖の定量--
前記セルロースIIIIを、終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、前記PcCel6Aを、下記表1に従って添加し、30℃にて最大360分間反応させた。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、アセトニトリル/H2O(60/40~50/50、体積/体積)の直線勾配で、Shodex(登録商標) Asahipak NH2P-50(昭和電工(株)製)で分離した。前記分離された糖の量は、重合度(DP)=2~7のセロオリゴ糖(生化学工業(株)製)を標準として用いて定量した。なお、HPLCの装置は、Corona(登録商標) Chaged Aerosol Detector(登録商標)(ESA Biosciences社製)を用いたLC-2000(日本分光(株)製)を使用した。
前記セルロースIIIIを、終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、前記PcCel6Aを、下記表1に従って添加し、30℃にて最大360分間反応させた。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、アセトニトリル/H2O(60/40~50/50、体積/体積)の直線勾配で、Shodex(登録商標) Asahipak NH2P-50(昭和電工(株)製)で分離した。前記分離された糖の量は、重合度(DP)=2~7のセロオリゴ糖(生化学工業(株)製)を標準として用いて定量した。なお、HPLCの装置は、Corona(登録商標) Chaged Aerosol Detector(登録商標)(ESA Biosciences社製)を用いたLC-2000(日本分光(株)製)を使用した。

前記トリコデルマ リーセイ(Trichoderma reesei) Cel7A(以下、「TrCel7A」と称することがある。)は、セルラーゼ製剤(セルクラスト、ノボザイム社製)より、カラムクロマトグラフィーにより精製した。
即ち、セルラーゼ製剤を、20mMトリス塩酸緩衝液(pH8.0)に対して平衡化した。次いで、20mMトリス塩酸緩衝液(pH8.0)で平衡化したSuperQ-Toyopearl 650S カラム(9mm×120mm、東ソー(株)製)に前記平衡化したTrCel7Aを含む溶液を加えた。TrCel7Aは、0M~0.5Mの塩化ナトリウム100mLの直線勾配でカラムから溶出した。
その後、前記TrCel7Aを含む分画を集め、1M硫酸アンモニウムを含む20mM酢酸ナトリウム緩衝液(pH5.0)に対して平衡化した。次いで、1Mの硫酸アンモニウムを含有する20mMの酢酸ナトリウム緩衝液(pH5.0)で平衡化したPhenyl-Toyopearl 650Sカラム(26mm×120mm、東ソー(株)製)を用いて、前記溶液を分画した。得られたTrCel7Aは、300mLの逆勾配で、20mM酢酸ナトリウム緩衝液(pH5.0)へ溶出した。
精製したTrCel7Aは、50mM酢酸緩衝液(pH5.0)に懸濁し、以下の糖取得工程に用いた。
前記トリコデルマ リーセイ(Trichoderma reesei) Cel7A(以下、「TrCel7A」と称することがある。)は、セルラーゼ製剤(セルクラスト、ノボザイム社製)より、カラムクロマトグラフィーにより精製した。
即ち、セルラーゼ製剤を、20mMトリス塩酸緩衝液(pH8.0)に対して平衡化した。次いで、20mMトリス塩酸緩衝液(pH8.0)で平衡化したSuperQ-Toyopearl 650S カラム(9mm×120mm、東ソー(株)製)に前記平衡化したTrCel7Aを含む溶液を加えた。TrCel7Aは、0M~0.5Mの塩化ナトリウム100mLの直線勾配でカラムから溶出した。
その後、前記TrCel7Aを含む分画を集め、1M硫酸アンモニウムを含む20mM酢酸ナトリウム緩衝液(pH5.0)に対して平衡化した。次いで、1Mの硫酸アンモニウムを含有する20mMの酢酸ナトリウム緩衝液(pH5.0)で平衡化したPhenyl-Toyopearl 650Sカラム(26mm×120mm、東ソー(株)製)を用いて、前記溶液を分画した。得られたTrCel7Aは、300mLの逆勾配で、20mM酢酸ナトリウム緩衝液(pH5.0)へ溶出した。
精製したTrCel7Aは、50mM酢酸緩衝液(pH5.0)に懸濁し、以下の糖取得工程に用いた。
-糖の製造、及び糖の定量-
前記セルロースIIIIを、終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、前記TrCel7Aを、下記表2に従って添加し、30℃にて最大360分間反応させた。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、実施例1~6と同様の方法で定量した。
前記セルロースIIIIを、終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、前記TrCel7Aを、下記表2に従って添加し、30℃にて最大360分間反応させた。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、実施例1~6と同様の方法で定量した。
図2に、TrCel7Aと、超臨界アンモニア処理をしたシオグサ由来セルロース(セルロースIIII)との反応液中のセロビオース濃度(μM)の経時変化を示す。

図1(実施例1~6)及び図2(比較例1~6)より、セルロースIIIIを、PcCel6Aを用いて加水分解した場合(実施例1~6)も、TrCel7Aを用いて加水分解した場合(比較例1~6)も、酵素濃度依存的に加水分解産物(セロビオース)の量が増加した。しかしながら、セロビオース生成量は、TrCel7Aを用いて加水分解した場合と比較して、PcCel6Aを用いて加水分解した場合の方が、顕著に多かった。また、PcCel6Aを用いて加水分解した場合は、TrCel7Aを用いて加水分解した場合と比較して反応速度も速かった。
(実施例3、及び比較例3、7~8)
<PcCel6A及びTrCel7AによるセルロースIα及びセルロースIIIIの糖化>
-セロビオース生成速度及び吸着酵素1分子あたりの加水分解速度の検討-
PcCel6Aと、TrCel7Aとのセルロースの糖化効率を比較するため、実施例3及び比較例3と同様の方法でセルロースIIIIの糖化を行なった。
また、PcCel6A及びTrCel7AによるセルロースIαの糖化効率を検討するため、セルロースIIIIをセルロースIαに変えた以外は、実施例3及び比較例3と同様の方法でセルロースIαの糖化を行なった。
即ち、セルロースIα又はセルロースIIIIを、下記表3に従って終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、実施例1~6で調製したPcCel6A 1.0μM又は比較例1~6で調製したTrCel7A 1.0μMを、下記表3に従って添加し、30℃にて最大360分間反応させた。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、実施例1~6と同様の方法で定量し、セロビオース生成量(図3)、及び基質(セルロースIα又はセルロースIIII)に吸着した酵素1分子あたりの加水分解速度(図4)を求めた。
<PcCel6A及びTrCel7AによるセルロースIα及びセルロースIIIIの糖化>
-セロビオース生成速度及び吸着酵素1分子あたりの加水分解速度の検討-
PcCel6Aと、TrCel7Aとのセルロースの糖化効率を比較するため、実施例3及び比較例3と同様の方法でセルロースIIIIの糖化を行なった。
また、PcCel6A及びTrCel7AによるセルロースIαの糖化効率を検討するため、セルロースIIIIをセルロースIαに変えた以外は、実施例3及び比較例3と同様の方法でセルロースIαの糖化を行なった。
即ち、セルロースIα又はセルロースIIIIを、下記表3に従って終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、実施例1~6で調製したPcCel6A 1.0μM又は比較例1~6で調製したTrCel7A 1.0μMを、下記表3に従って添加し、30℃にて最大360分間反応させた。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、実施例1~6と同様の方法で定量し、セロビオース生成量(図3)、及び基質(セルロースIα又はセルロースIIII)に吸着した酵素1分子あたりの加水分解速度(図4)を求めた。
図3は、PcCel6AとセルロースIIII(以下「PcCel6A-セルロースIIII」と称することがある)との加水分解反応(実施例3)、TrCel7AとセルロースIIII(以下「TrCel7A-セルロースIIII」と称することがある)との加水分解反応(比較例3)、PcCel6AとセルロースIα(以下「PcCel6A-セルロースIα)」と称することがある)との加水分解反応(比較例7)、及びTrCel7AとセルロースIα(以下「TrCel7A-セルロースIα」と称することがある)(比較例8)との加水分解反応における、それぞれの反応液中のセロビオースの生成量(μM/分間)と、反応表面密度(ρ)との関係を示す。

図3より、PcCel6A-セルロースIαの加水分解反応(比較例3)及びTrCel7A-セルロースIαの加水分解反応(比較例8)では、いずれもほとんどセロビオースの生成は認められなかった。TrCel7A-セルロースIIIIの加水分解反応(比較例3)はセロビオースの生成は認められたものの、その生成速度は遅かった。一方、PcCel6A-セルロースIIIIの加水分解反応(実施例3)によるセロビオースの生成速度は、比較例3、7~8と比較して顕著に速かった。
図4は、セロビオースの生成量の向上が、単に加水分解反応を行うセルロースIIIIの反応表面密度が広がったことによるものではないことを確認するため、基質(セルロースIα又はセルロースIIII)に吸着した酵素1分子あたりの加水分解速度を求めたグラフである。図4より、セルロースIIIIの反応表面密度が0.4のとき、TrCel7A 1分子あたりの加水分解速度が1分間に2.5回であった(比較例3)のに対し、PcCel6A 1分子あたりの加水分解速度は1分間あたり11.5回であった(実施例3)。これらの結果より、PcCel6Aでは、酵素1分子あたりの加水分解速度が顕著に速いことが認められた。
-酵素量の定量-
前記PcCel6A-セルロースIIIIの加水分解反応(実施例3)、TrCel7A-セルロースIIIIの加水分解反応(比較例3)、PcCel6A-セルロースIαの加水分解反応(比較例7)、及びTrCel7A-セルロースIαの加水分解反応(比較例8)において、それぞれ反応中にセルロースIIII又はセルロースIαに吸着したPcCel6A又はTrCel7Aの量を、DEAE-5PW(東ソー製)で分離し、定量した。緩衝液は、A:20mM トリス塩酸緩衝液(pH8.0)、B:20mM トリス塩酸緩衝液(pH8.0)+0.5M塩化ナトリウムを用い、A/B=100/0(体積/体積)を5分間、A/B=100/0~40/60(体積/体積)の直線勾配で10分間、A/B=0/100(体積/体積)を5分間で分離した。なお、タンパク質定量用のHPLC装置は、可視紫外検出器(UV-2075)を用いたLC-2000(日本分光(株)製)を使用した。
図5は、反応液中の遊離タンパク質量濃度(μM)と、吸着酵素量(nmol/mg-セルロース)との関係を示す。

前記PcCel6A-セルロースIIIIの加水分解反応(実施例3)、TrCel7A-セルロースIIIIの加水分解反応(比較例3)、PcCel6A-セルロースIαの加水分解反応(比較例7)、及びTrCel7A-セルロースIαの加水分解反応(比較例8)において、それぞれ反応中にセルロースIIII又はセルロースIαに吸着したPcCel6A又はTrCel7Aの量を、DEAE-5PW(東ソー製)で分離し、定量した。緩衝液は、A:20mM トリス塩酸緩衝液(pH8.0)、B:20mM トリス塩酸緩衝液(pH8.0)+0.5M塩化ナトリウムを用い、A/B=100/0(体積/体積)を5分間、A/B=100/0~40/60(体積/体積)の直線勾配で10分間、A/B=0/100(体積/体積)を5分間で分離した。なお、タンパク質定量用のHPLC装置は、可視紫外検出器(UV-2075)を用いたLC-2000(日本分光(株)製)を使用した。
図5は、反応液中の遊離タンパク質量濃度(μM)と、吸着酵素量(nmol/mg-セルロース)との関係を示す。

図5より、PcCel6A-セルロースIαの加水分解反応(比較例7)で吸着したPcCel6A量及びTrCel7A-セルロースIαの加水分解反応(比較例8)で吸着したTrCel7A量は同程度であり、その酵素量は少なかった。
一方、図3より、PcCel6A-セルロースIIIIの加水分解反応(実施例3)は、セロビオース生成速度が速かったにも関わらず、PcCel6A-セルロースIIIIの加水分解反応(実施例3)で吸着したPcCel6A量は、TrCel7A-セルロースIIIIの加水分解反応(比較例3)で吸着したTrCel7A量と比較して顕著に少なかった。
これらの結果より、PcCel6AをセルロースIIIIと反応させた場合は、吸着したPcCel6Aの酵素1分子あたりの活性が上昇していると考えられる。
一方、図3より、PcCel6A-セルロースIIIIの加水分解反応(実施例3)は、セロビオース生成速度が速かったにも関わらず、PcCel6A-セルロースIIIIの加水分解反応(実施例3)で吸着したPcCel6A量は、TrCel7A-セルロースIIIIの加水分解反応(比較例3)で吸着したTrCel7A量と比較して顕著に少なかった。
これらの結果より、PcCel6AをセルロースIIIIと反応させた場合は、吸着したPcCel6Aの酵素1分子あたりの活性が上昇していると考えられる。
(実施例3、7、及び比較例7~11)
<PcCel6AとPcCel45Aとの相乗効果>
酵素としては、PcCel6Aと、メタノール資化性酵母(Pichia pastoris)で発現させた組換ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium) Cel45Aとを用い、基質としては、実施例1~6のセルロースIIII及びセルロースIαのいずれかを用いた場合の加水分解生成物について調べ、PcCel6AとPcCel45Aとの相乗効果を以下のようにして試験した。
<PcCel6AとPcCel45Aとの相乗効果>
酵素としては、PcCel6Aと、メタノール資化性酵母(Pichia pastoris)で発現させた組換ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium) Cel45Aとを用い、基質としては、実施例1~6のセルロースIIII及びセルロースIαのいずれかを用いた場合の加水分解生成物について調べ、PcCel6AとPcCel45Aとの相乗効果を以下のようにして試験した。
-組換PcCel45Aの製造-
ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium) Cel45Aの組換体タンパク質(以下、「PcCel45A」と称することがある。)を、以下の方法で製造した。

--PcCel45A遺伝子のクローニング--
NCBIデータベース上にあるファネロケーテ クリソスポリウム(Phanerochaete chrysosporium)由来Cel45A遺伝子(AB378504)をもとに、以下のプライマー(Pccel45A-F1、Pccel45A-F2)を設計した。また、リバースプライマー(Invitrogen社製)は、以下の配列を用いた。

上記プライマーを用い、PcCel6Aと同様に調製したcDNAを鋳型にしてPCRを行い(94℃、2分間を1サイクル、98℃、10秒間・68℃、1分30秒間を25サイクル、4℃で終了)、前記PcCel45Aをコードする領域と3’非翻訳領域をPCRによって増幅した。
また、5’非翻訳領域の塩基配列は、GeneRacer(トレードマーク) Kit(Invitrogen社製)と、SuperScript(登録商標)III RT(Invitrogen社製)と、以下の遺伝子特異的プライマーとを用いて増幅した(94℃、2分間を1サイクル、98℃、10秒間・70℃、30秒間を5サイクル、98℃、10秒間・68℃、30秒間を5サイクル、98℃、10秒間・66℃、30秒間・68℃、30秒間を20サイクル、4℃で終了)。
得られたPCR産物は、前記PcCel6Aと同様の方法でクローン化した。

PcCel45A遺伝子を含むTOPOベクターを鋳型とし、上記オリゴヌクレオチドプライマーと、ポリメラーゼとしてKOD-Plusとを用いてPCRを行い(94℃、2分間を1サイクル、98℃、10秒間・68℃、30秒間を20サイクル、4℃で終了)、発現ベクターに挿入するDNA断片を増幅した。
前記増幅したDNA断片を酵母の発現ベクターpPICZαのXhoIとNotI部位に挿入し、組換ベクター(発現ベクター)を得た。

--形質転換体の作製--
上記で得られた発現ベクター約5μgを、Bpu1102I(タカラバイオ(株)製)を用いて、直鎖化した。次いで、エレクトロポレーションにより、酵母(Pichia pastorisKM71H株)へ、前記直鎖化した組換ベクターを導入した後、形質転換体の選別を行った。前記エレクトロポレーション、及び形質転換体の選別は、EasySelect(トレードマーク) Pichia expression kit(version G;Invitrogen社製)のマニュアルに基づいて行った。これにより、前記発現ベクターにより形質転換された形質転換体を得た。
上記で得られた発現ベクター約5μgを、Bpu1102I(タカラバイオ(株)製)を用いて、直鎖化した。次いで、エレクトロポレーションにより、酵母(Pichia pastorisKM71H株)へ、前記直鎖化した組換ベクターを導入した後、形質転換体の選別を行った。前記エレクトロポレーション、及び形質転換体の選別は、EasySelect(トレードマーク) Pichia expression kit(version G;Invitrogen社製)のマニュアルに基づいて行った。これにより、前記発現ベクターにより形質転換された形質転換体を得た。
--形質転換酵母の培養--
上述の形質転換体を、実施例1~6と同様の方法で培養し、遠心分離(3,000g、10分)によって得られた培養上清を粗酵素液とした。

-PcCel45Aの製造及び精製-
前述のようにして得られた粗酵素液を、実施例1~6と同様の方法で精製した。
得られたPcCel45AのN末端のアミノ酸配列は、プロテインシーケンサーにより確認した。
その結果、形質転換酵母で得られたPcCel45AのN末端アミノ酸配列は、ATGGYVQQAT(配列番号:13)であり、担子菌ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium)で得られた天然のものと同じであることが認められた。
PcCel45Aは、50mM酢酸緩衝液(pH5.0)に緩衝液交換し、以下の相乗効果試験に用いた。
前述のようにして得られた粗酵素液を、実施例1~6と同様の方法で精製した。
得られたPcCel45AのN末端のアミノ酸配列は、プロテインシーケンサーにより確認した。
その結果、形質転換酵母で得られたPcCel45AのN末端アミノ酸配列は、ATGGYVQQAT(配列番号:13)であり、担子菌ファネロケーテ クリソスポリウム(Phanerochaete chrysosporium)で得られた天然のものと同じであることが認められた。
PcCel45Aは、50mM酢酸緩衝液(pH5.0)に緩衝液交換し、以下の相乗効果試験に用いた。

前記セルロースIα又は前記セルロースIIIIを、下記表4に従って終濃度が0.1質量%となるように50mM酢酸ナトリウム溶液(pH5.0)250μL中に添加した。次いで、実施例1~6で調製したPcCel6A、比較例1~6で調製したTrCel7A、及びPcCel6A+PcCel45Aのいずれかを添加し、30℃にて最大360分間反応させた。なお、前記PcCel6A及びTrCel7Aの終濃度は、それぞれ1.0μM、PcCel6A+Cel45Aの全酵素濃度は2.0μM(PcCel6A 1.0μMに、Cel45A1.0μMを添加して調製した)で行った。
反応終了後、反応液を5分間遠心分離(×15,000g)して反応を停止した後に得られた上澄みを、実施例1~6と同様の方法で分析した。

図6は、TrCel7A-セルロースIαの加水分解反応(比較例8)、PcCel6A-セルロースIIIIの加水分解反応(実施例3)、TrCel7A-セルロースIIIIの加水分解反応(比較例3)、及びPcCel6A+PcCel45AとセルロースIIII(以下「PcCel6A+PcCel45A-セルロースIIII」と称することがある)の加水分解反応(実施例7)における、反応液中のセロビオース濃度(μM)の経時変化を示す。
図6中、「■」はTrCel7A-セルロースIαの加水分解反応(比較例8)を示し、
「▲」はPcCel6A-セルロースIIIIの加水分解反応(実施例3)を示し、
「●」はTrCel7A-セルロースIIIIの加水分解反応(比較例3)を示し、
「◆」はPcCel6A+PcCel45A-セルロースIIIIの加水分解反応(実施例7)を示す。
なお、PcCel6A-セルロースIαの加水分解反応(比較例7)、PcCel45A-セルロースIαの加水分解反応(比較例9)、PcCel6A+PcCel45A-セルロースIαの加水分解反応(比較例10)、及びPcCel45A-セルロースIIIIの加水分解反応(比較例11)は、反応液中のセロビオース濃度(μM)が低い、若しくはセロビオースが生成していないため、図6には示していない。
図6中、「■」はTrCel7A-セルロースIαの加水分解反応(比較例8)を示し、
「▲」はPcCel6A-セルロースIIIIの加水分解反応(実施例3)を示し、
「●」はTrCel7A-セルロースIIIIの加水分解反応(比較例3)を示し、
「◆」はPcCel6A+PcCel45A-セルロースIIIIの加水分解反応(実施例7)を示す。
なお、PcCel6A-セルロースIαの加水分解反応(比較例7)、PcCel45A-セルロースIαの加水分解反応(比較例9)、PcCel6A+PcCel45A-セルロースIαの加水分解反応(比較例10)、及びPcCel45A-セルロースIIIIの加水分解反応(比較例11)は、反応液中のセロビオース濃度(μM)が低い、若しくはセロビオースが生成していないため、図6には示していない。
図7は、TrCel7A-セルロースIαの加水分解反応(比較例8)、PcCel6A-セルロースIαの加水分解反応(比較例7)、PcCel45A-セルロースIαの加水分解反応(比較例9)、PcCel6A+PcCel45A-セルロースIαの加水分解反応(比較例10)、TrCel7A-セルロースIIIIの加水分解反応(比較例3)、PcCel6A-セルロースIIIIの加水分解反応(実施例3)、PcCel45A-セルロースIIIIの加水分解反応(比較例11)、及びPcCel6A+PcCel45A-セルロースIIIIの加水分解反応(実施例7)の、それぞれの加水分解反応におけるセロビオース生成速度(μM/分間)を示す。
図6より、PcCel6A+PcCel45A-セルロースIIIIの加水分解反応(実施例7)では、PcCel6A-セルロースIIIIの加水分解反応(実施例3)と比較して、セロビオースが多く得られた。
また、図7より、PcCel45A-セルロースIIIIの加水分解反応(比較例11)ではセロビオースの生成は全く認められなかったが、PcCel6A+PcCel45A-セルロースIIIIの加水分解反応(実施例7)におけるセロビオース生成速度は、PcCel6A-セルロースIIIIの加水分解反応(実施例3)におけるセロビオース生成速度と比較して顕著に早かった。一方、PcCel6A+PcCel45A-セルロースIαの加水分解反応(比較例10)におけるセロビオースの生成速度は遅かった。
これらの結果から、PcCel6Aは、他のセルラーゼと併用するとセルロースIIIIの分解に関し、相乗効果を有することが認められた。PcCel45Aは、PcCel6Aの活性を増強すると考えられる。
また、図7より、PcCel45A-セルロースIIIIの加水分解反応(比較例11)ではセロビオースの生成は全く認められなかったが、PcCel6A+PcCel45A-セルロースIIIIの加水分解反応(実施例7)におけるセロビオース生成速度は、PcCel6A-セルロースIIIIの加水分解反応(実施例3)におけるセロビオース生成速度と比較して顕著に早かった。一方、PcCel6A+PcCel45A-セルロースIαの加水分解反応(比較例10)におけるセロビオースの生成速度は遅かった。
これらの結果から、PcCel6Aは、他のセルラーゼと併用するとセルロースIIIIの分解に関し、相乗効果を有することが認められた。PcCel45Aは、PcCel6Aの活性を増強すると考えられる。
本発明の、バイオマス原料の処理方法、糖の製造方法、エタノールの製造方法、及び乳酸の製造方法によれば、糖の生産効率、エタノールの生産効率、及び乳酸の生産効率を格段に向上させることができる。
したがって、本発明の、バイオマス原料の処理方法、糖の製造方法、エタノールの製造方法、及び乳酸の製造方法は、近年注目されている、環境に優しい燃料を産出することを目的としたバイオマス原料からのエタノール製造に、また環境に優しい生分解性プラスチックの製造などに好適に利用可能である。
したがって、本発明の、バイオマス原料の処理方法、糖の製造方法、エタノールの製造方法、及び乳酸の製造方法は、近年注目されている、環境に優しい燃料を産出することを目的としたバイオマス原料からのエタノール製造に、また環境に優しい生分解性プラスチックの製造などに好適に利用可能である。
Claims (7)
- (a)バイオマス原料を、アンモニアを含む処理剤で処理することにより、改質バイオマス原料を得る工程、
(b)該改質バイオマス原料を、セロビオヒドロラーゼII(Cel6A)で酵素糖化する工程、
を含むことを特徴とするバイオマス原料の処理方法。 - バイオマス原料が、セルロースI型を含む請求項1に記載のバイオマス原料の処理方法。
- 工程(a)が、超臨界アンモニア流体で処理することを含む請求項1から2のいずれかに記載のバイオマス原料の処理方法。
- 改質バイオマス原料とセロビオヒドロラーゼII(Cel6A)とが加水分解反応を行う表面密度が0.9以下のとき、該セロビオヒドロラーゼII(Cel6A)1分子あたりの加水分解速度が1分間に5回以上である請求項1から3のいずれかに記載のバイオマス原料の処理方法。
- 請求項1から4のいずれかに記載のバイオマス原料の処理方法により糖を得ることを特徴とする糖の製造方法。
- 請求項5に記載の糖の製造方法により得られた糖を発酵させてエタノールを得ることを特徴とするエタノールの製造方法。
- 請求項5の記載の糖の製造方法により得られた糖を発酵させて乳酸を得ることを特徴とする乳酸の製造方法。
