- K 2 CO 3 (0.135 g, 0.97 mmol) was added to a solution of 3-(4-sec- butylphenyl)-2-(4-hydroxy-3-methylphenyl)quinazolin-4(3H)-one (0.250 g, 0.65 mmol) in DMF (15 ml_). The mixture was stirred at room temperature for 30 min, then (2-bromoethoxy)(tert-butyl)dimethylsilane (0.16 ml_, 0.78 mmol) was added and the reaction was heated at reflux temperature for 3 hours, before concentrating in vacuo and dissolving the residue in EtOAc.
- reaction mixture was cooled to room temperature, filtered through Celite®, with THF washings, concentrated, and purified by flash chromatography on silica gel, eluting with 50% ethyl acetate/heptane to 100% ethyl acetate, to afford the title compound as a white solid (0.195 g, 59%).
- reaction mixture was cooled to room temperature, filtered through Celite®, with THF washings, concentrated, and purified by flash chromatography on silica gel, eluting with 40% ethyl acetate/heptane to 100% ethyl acetate. Further purification by reverse-phase HPLC, eluting with 10% to 90% CH 3 CN in water with 0.1% TFA, afforded the title compound as a white solid (0.025 g, 13%).
- Phenylboronic acid (0.041 g, 0.34 mmol) and Pd(PPh 3 ) 4 (0.003 g) were added to a mixture of 2-(6-bromopyridin-3-yl)-3-(4-sec-butylphenyl)quinazolin- 4(3H)-one (0.150 g, 0.34 mmol) in 2 M Na 2 CO 3 (0.3 ml_) and DME (5 ml_). After refluxing overnight, the reaction mixture was concentrated in vacuo. The residue was dissolved in CHCI 3 and washed with H 2 O, then brine, dried (Na 2 SO 4 ), filtered, and concentrated in vacuo.

- Pd(OAc) 2 (0.026g , 0.12 mmol), DPPF (0.055 g, 0.10 mmol) and NaOfBu (0.049 g, 0.52 mmol) were added to a mixture of 2-(5-bromopyridin-3-yl)-3- (4-sec-butylphenyl)quinazolin-4(3H)-one (0.175 g, 0.40 mmol) and Et 2 NH (0.06 ml_, 0.60 mmol) in toluene (2 mi_). The mixture was microwaved at 90 0 C and 300W (max. power) with cooling for 3 hours, before being concentrated in vacuo.
- Pd(PPh 3 ) 4 (0.150 g, 0.12 mmol), K 2 CO 3 (0.270 g, 1.90 mmol), and trimethylboroxine (0.18 ml_, 1.20 mmol) were added to a solution of 2-(6-chloropyridin-3-yl)-3-(4- cyclopentylphenyl)quinazolin-4(3/-/)-one (0.260 g, 0.65 mmol) in dioxane (15 mL) and heated to reflux temperature for 20 minutes. The reaction mixture was filtered through Celite®, washed with EtOAc.
- Methanesulfonyl chloride (0.026 ml_, 0.33 mmol) was added to a mixture of 3-(4-aminophenyl)-2-(6-methylpyridin-3-yl)quinazolin-4(3/-/)-one (0.11 g, 0.33 mmol) and TEA (0.093 ml_, 0.67 mmol) in CH 2 CI 2 (5 mL). The mixture was stirred for 2 hours and concentrated in vacuo. The residue was redissolved in 1 :1 dioxane and H 2 O (5 mL) and LiOH*H 2 O (0.050 g, 1.2 mmol) was added.
- BoC 2 O (0.142 g, 0.65 mmol) and DMAP (0.6 mg, 0.005 mmol) were added to a solution of 3-(4-bromophenyl)-2-(1H-indol-5-yl)quinazolin-4(3/-/)-one (0.225 g, 0.54 mmol) in THF (15 mL). After stirring at room temperature for 4 hours, the mixture was concentrated in vacuo.
- Trifluoroacetic anhydride (0.19 mL, 1.30 mmol) and TEA (0.37 ml_, 2.60 mmol) were added to a solution of 4-amino-3-iodobenzaldehyde (0.330 g, 1.30 mmol) in CH 2 CI 2 (15 mL). After stirring at room temperature for 1 hour, the reaction mixture was concentrated in vacuo. Purification by flash chromatography on silica gel, eluting with 5% to 50% EtOAc in heptane, afforded 2-amino- ⁇ /-(4-sec- butylphenyl)benzamide (0.350 g, 79%).
