Abstract
본 발명은 글루코사민 또는 그의 유도체의 신규한 용도에 관한 것으로서, 더욱 상세하게는 글루코사민, 글루코사민 유도체 또는 이들의 염을 포함하는 혈관 협착 및 재협착 예방 또는 치료용 조성물 및 상기 조성물을 이용하여 혈관협착 및 재협착을 예방 또는 치료하는 방법에 관한 것이다.
Description
본 발명은 글루코사민의 신규한 용도에 관한 것으로, 보다 상세하게는 글루코사민, 글루코사민 유도체 또는 이들의 염을 포함하는 혈관 협착 및 재협착 예방 또는 치료용 조성물 및 상기 조성물을 이용하여 혈관협착 및 재협착을 예방 또는 치료하는 방법에 관한 것이다.
혈관 질환은 혈관에 발생하는 질환으로 심장근육에 혈액 공급 장애를 일으킨다. 이러한 질환의 가장 흔한 원인은 동맥경화증이며, 동맥 내에 콜레스테롤과 다른 지방질, 혈전 생성(acute thrombosis), 혈액 내의 다른 여러 성분이 조합되어 발생한 플라크(augmented plaque)의 증가, 그리고 백혈구의 활성 및 흡착(leucocyte activation and adhesion)이 일어나면 동맥의 협착을 일으키게 되며 이에 따라 혈액 공급이 감소되어 영양분과 산소가 부족하게 된다(Libby P. et al., Circulation, 86(6). 47-52, 1992.; Lundgren CH et al., Circulation, 90(4), 1927-1934, 1994.). 이 결과로 협심증 또는 심근 경색증을 일으키게 되며 심한 경우 사망까지 초래하기도 한다.
현재 혈관 질환의 치료는 혈관 신생(Angiogenesis)을 위한 치료와 근육세포의 증식억제를 통한 혈관 협착 및 재협착방지(prevention for restenosis)로 나누어 생각할 수 있다.
경피적 관상동맥 성형술은 좁아진 관상동맥을 수술에 의하지 않고 확장시켜주는 방법으로서, 이의 시술 방법으로는 경피적 관상동맥 풍선 확장술, 경피적 관상동맥 스텐트 삽입술 등이 있다. 경피적 관상동맥 풍선 확장술은 대퇴부나 팔의 동맥을 통하여 가이드용 도관을 삽입한 후 도관을 대동맥을 통하여 병변이 있는 관상동맥의 입구에 위치시키고, 가이드용 도관의 위치를 확인 후 이 도관의 내부를 통하여 끝에 풍선이 부착되어 있는 도관을 관상동맥의 협착 부위에 위치시킨 후 풍선을 확장 시킨다. 확장된 풍선은 플라크 등을 압착하여 좁아진 관상동맥을 확장시켜 관상동맥의 혈류개선을 가져오는 방법이다. 또한, 스텐트 삽입술은 철망이 입혀진 풍선을 협착부위에 위치시킨 후 풍선을 확장하여 관상동맥의 내벽에 철망을 입히는 것이며, 이러한 스텐트는 풍선 확장술만 시술하였을 때보다는 재협착의 발생율이 낮으며, 스텐트가 혈관 내벽에 대한 지지대역할을 하는 특성이 있으므로 풍선 확장시 발생하는 합병증의 치료에 사용되고 있다.
이러한 관상동맥 성형술을 이용한 중재적 시술은 수술을 통한 방법보다, 간편하고 전신 마취에 의한 위험 부담을 줄일 수 있으며 성공률도 높아 세계적으로 널리 이용되고 있는 추세이다. 혈관의 경우 고령, 심장질환, 호흡기 질환 등의 원인으로 수술이나 마취 자체가 위험 부담이 큰 경우에 우선적으로 적용 할 수 있고 수술적 치료가 불가능한 위장관이나 담도의 말기 암 환자의 경우 잔여 생존 기간 동안 전신 상태를 개선 시켜 삶의 질을 향상시키는 역할을 할 수 있다.
그러나, 관상동맥 성형술의 시술의 경우 손상에 의해 유도되는 혈관내막 증식증의 과정으로 혈관내피의 손상, 혈관막내 혈전증(mural thrombosis), 혈관 평활근세포와 섬유아세포의 이동과 단핵세포 및 임파구의 침윤, 신생내막(neointima)내의 세포의 증식, 세포외기질의 축적(reendothelialization), 세포사멸(apoptosis) 등을 통한 용해도 등에 의한 재협착(restenosis)이 발생한다. 약 6.8%의 환자에서 혈전증이나 혈관 경련 등에 위한 재협착이 발생할 수 있으며, 혈관 확장 수술 후 3 내지 6개월에 더 심한 혈관의 재협착이 일어날 수 있고, 40%의 환자에서 풍선확장술 부위에 재협착의 소견을 발견할 수 있다(Herrman J-PR et al., Drugs, vol. 46, 18-52, 1993). 동맥과 정맥의 우회로 수술한 부위의 혈관과 관상동맥과 대퇴혈관의 동맥내막절제술(endarterectomy) 부위의 20%에서 혈관의 이차적인 변화로 인하여 혈관이 막힐 수 있다(Volteas N et al., Int. 관상동맥 재협착 방지를 위해 죽종제거술(atherectomy), 레이저 혈관성형술, 고속회전 죽종제거술(rotablator), 절단풍선을 이용한 관상혈관 성형술(cutting balloon angioplast), 방사선 조사 등의 새로운 PTCA (Percutaneous Transluminal Coronary Angioplasty) 장비가 도입된 바 있으며, 항혈소판제, 항혈전제, 혈관확장제, 세포증식억제제, 지질대사 개선제, 항산화제 등의 여러 가지 전신적 및 국소약물 요법 및 유전자 요법과 같은 분자 생물학적 치료가 개발되어 시도되었다. 이 중 경구 투여나 정맥 투여와 같은 전신적인 약물요법은 가장 쉽게 적용할 수 있는 치료법이나, 동물실험에서의 재협착 방지 효과가 보고되었을 뿐, PTCA를 시행한 부위에서 목적하는 약물 농도에 도달되지 않고 약물의 부작용으로 인해 대부분 임상실험에서 재협착을 방지하지 못했다. 이론적으로 재협착은 PTCA가 시술된 국소부위의 관상동맥에만 일어나므로, 재협착의 방지를 위해서는 전신적인 약물요법보다는 부위특이적(site-specific)으로 고농도의 약물투여가 가능한 국소 약물 요법(local drug therapy)이 더욱 유용하다.
최근 PTCA를 시행한 부위에 직접 약물을 투여하기 위해, 이중풍선카테터, 디스패치(dispatch) 또는 미세구 풍선(microporous balloon) 등이 개발되어 임상에 이용되고 있으며, 장기간 PTCA시술부위에 약물을 전달하기 위해 서방성 미세입자(slow release microsphere)또는 스텐트에 약물을 입혀 치료하는 시도들이 늘고 있다.
한편, 글루코사민은 천연 아미노당의 하나로서 분자식 C6H13NO6이며 탄소가 6개 있으므로 갈락토사민과 함께 헥소사민으로 총칭된다. 무색의 침상결정으로, 110℃에서 분해하며, 물에 녹는 강염기성 물질이다. 천연으로는 키틴을 비롯하여 세균의 세포벽, 동물의 연골이나 피부를 구성하는 뮤코다당 등 다당류의 성분으로 널리 분포한다. 또한, 골관절염이나 상해로부터의 손상을 보수하기 위해 필요한 물질을 관절에 제공하며 특히 연골에서 발견되는 글리코사미노글리칸(glycosaminoglycan)이라 불리는 뮤코다당류를 만드는 물질이며, 그 외 소화기, 순환기 내의 기초점막, 관절부위의 활액, 인대 및 건을 포함한 신체조직의 대부분을 이루는 물질이다.
이에 본 발명자들은 글루코사민의 새로운 용도에 관하여 끊임없이 연구를 하는 과정에서 글루코사민이 새로운 혈관 협착 및 재협착 예방제 및 치료제로써 유용한 효과를 발견하고, 그 결과 본 발명을 완성하게 되었다.
본 발명의 하나의 목적은 글루코사민, 글루코사민 유도체 및 이들의 염을 포함하는 혈관 협착 및 재협착 예방 또는 치료용 조성물을 제공하는 것이다.
본 발명의 다른 하나의 목적은 상기 조성물을 이용하여 혈관협착 및 재협착을 예방 또는 치료하는 방법을 제공하는 것이다.
본 발명의 글루코사민, 글루코사민 유도체 또는 이들의 염은 혈관협착 및 재협착 예방 또는 치료에 우수한 효과를 나타내어, 경피적 관상동맥 성형술과 같은 수술적 방법을 사용하지 않고도 본 발명의 조성물을 섭취함으로써 심장 질환 등을 유효하게 예방 또는 치료할 수 있다.
도 1은 래트의 경동맥 내피조직을 풍선 카테터로 손상시킨 후, 1주, 2주 및 3주째에 혈관 중막(tunica media)의 증식 정도를 측정한 결과이다.
도 2는 래트의 경동맥 내피조직을 풍선 카테터로 손상시킨 후, 1주, 2주, 3주째에 혈관 조직을 채취하여 혈관내피 조직의 세포증식 정도를 PCNA 염색을 통해 확인한 결과이다.
도 3은 래트의 대동맥 중막 민무늬 근육세포에서 유래된 세포주를 대상으로 LPA를 처리하여 세포증식을 유도한 후 글루코사민을 처리하고 WST-1 assay를 이용하여 세포증식억제를 확인한 결과이다.
하나의 양태로서, 본 발명은 글루코사민, 글루코사민 유도체 또는 이들의 염을 포함하는 혈관 협착 및 재협착 예방 또는 치료용 조성물에 관한 것이다.
글루코사민은 바닷게 껍질이나 새우 껍질의 주성분이 키토산의 분해산물이다. 바닷게 또는 새우 껍질의 주성분은 키틴과 키토산으로, 키틴은 2-아세트아미도-2-데옥시-베타-D-글루코스(N-아세틸글루코사민)로 구성되어 있으며, 키토산은 키틴의 탈아세틸화가 일어난 다당류로서 폴리(베타-(1,4)-글루코사민)이다. 글루코사민은 다음과 같은 화학식1의 구조를 가진다.
본 발명에서 용어 "글루코사민 유도체"는 글루코사민의 하이드록시기의 수소가 아실 또는 알킬화합물로 치환된 것으로, 하기 화학식2의 구조를 가지는 것을 말한다.
상기 식에서 R은 탄소수가 2 내지 18인 아실기 또는 1 내지 5인 직쇄 또는 측쇄 알킬로서, 바람직하게는 아세틸, 프로피오닐, 부티릴(butyryl), 펜타노일(pentanoyl), 헥사노일(hexanoyl), 헵타노일(heptanoyl), 옥타노일(octanoyl), 노나노일(nonanoyl), 데카노일(decanoyl), 운데카노일(undecanoyl), 라우릴, 트리데카노일, 미리스틸(myristyl), 펜타데카노일, 팔미토일(palmitoyl), 마가릴(margaryl) 또는 스테아릴기 등의 아실기이거나, 또는 메틸, 에틸, 프로필, 부틸, 펜틸, 이소프로필, 이소부틸 또는 세크부틸(secbutyl)기 등의 알킬기일 수 있다.
상기와 같이 글루코사민의 1, 3, 4, 6번 알코올기에 소수성기를 도입시킬 경우, 글루코사민의 생리활성을 그대로 가지고 있다고 믿어지는 아민기는 그대로 보유하여, 글루코사민의 생리활성을 그대로 유지하면서 천연에서 분해가 잘 되고 무독성이며, 중금속에 대한 높은 흡착력을 가지며, 항균활성이 우수한 글루코사민 유도체를 얻을 수 있다.
본 발명의 글루코사민 및 글루코사민 유도체의 염은 약제학적 및 의학적으로 허용가능한 염으로서, 그 글루코사민 또는 글루코사민 유도체의 염은 글루코사민의 생리학적 활성을 유지하는 한 어떠한 염이라도 가능하다. 그러한 예로서, 염산염, 브롬산염, 황산염, 인산염, 아세트산염, 시트르산염, 푸마르산염, 락트산염, 말레산염, 숙신산염, 타르타르산염 등의 유기 또는 무기산과의 염, 나트륨염, 칼륨염 등의 알칼리금속과의 염 또는 암모늄염, 트리메틸아민, 트리에틸아민, 피리딘, 피콜린, 2,6-루티딘, 에탄올아민, 디에탄올아민, 트리에타올아민, 사이클로헥실아민, 디사이클로헥실아민 등의 유기 염기 등의 모든 약제학적으로 허용가능한 염을 들 수 있다.
본 발명에 유용한 글루코사민은 글루코사민의 어느 기원으로부터라도 얻을 수 있으며, 천연물질에서 분리 및 정제될 수 있고, 시판되는 것을 이용할 수도 있으며, 또는 약제학적으로 허용 가능한 글루코사민의 합성에 적합하다면 어느 방법에 의하여도 합성될 수 있다. 또한, 글루코사민은 글루코사민 또는 글루코사민의 유도체를 포함하는 키틴, 히아루론산, 헤파린 및 케라토설페이트의 가수분해 산물 및 다른 유도체로부터 글루코사민을 분리 및 정제함으로써 제공될 수 있다.
내피상해 후의 신생내막의 형성에 대해서, 본 발명의 글루코사민을 투여한 1주, 2주 및 3주째의 래트의 혈관조직을 헤마톡시린 에오신으로 염색해 본 바, 내막이 형성되지 않고, 외막 다음에 중막이 그대로 증대되어 있었다. 이에 반하여, 아무런 처리를 하지 않은 래트의 경우 혈관이 깨끗하게 외막, 중막 및 내막으로 형성되어 있었다(도 1 및 2 참조).
또한, 본 발명의 글루코사민이 혈관협착의 주요 병리학적 기전인 중막(tunica media)의 비후를 억제하는 것을 확인하기 위해, 래트의 대동맥 중막 민무늬 근육세포에서 유래된 세포주(rat vascular smooth muscle cells: A-10)의 증식을 유도한 후 글루코사민을 처리하여 WST-1 assay를 이용하여 조사한 결과 24시간 후 혈관민무늬근육 세포의 증식을 억제하는 효과가 있음을 확인할 수 있었다(도 3 참조).
이들의 결과로부터, 본 발명의 글루코사민은 혈관 내막의 형성을 억제하는 효과가 있음이 명백하다. 따라서 글루코사민을 개체에 투여하여 혈관협착을 예방 또는 치료할 수 있을 것이며, PCTA시술 후의 재협착의 예방을 실현할 수 있을 것이다.
본 발명의 글루코사민 또는 글루코사민 유도체를 포함하는 약제학적 조성물은 단일제로도 사용할 수 있으며, 공인된 약학 조성물을 포함하여 복합제로 제조하여 사용할 수도 있다.
글루코사민 또는 글루코사민의 유도체를 포함하는 약제학적 조성물은 글루코사민 또는 글루코사민의 유도체를 포함하는 캅셀에 부형제 없이 충전하거나 미립된 고체 담체, 액체 담체 또는 그 양자와 균일하게 충분히 접촉시켜 제조될 수 있다. 그 후에 필요한 경우 생성물을 바람직한 제제로 성형하여 사용할 수 있다. 적합한 담체 부형제의 예로서, 전분, 물, 식염수, 에탄올, 글리세롤, 링거액 및 덱스트로스 용액 등을 들 수 있으며, 문헌(Remington's pharmaceutical science, 19th Ed., 1995, Mack Publishing Company, Easton PA) 등에 개시되어 있는 바에 따라 당해 기술 분야에 알려진 적합한 제제로 제제화할 수 있다.
본 발명의 글루코사민 또는 글루코사민 유도체를 포함하는 약제학적 조성물은 글루코사민 또는 글루코사민 유도체를 유효성분으로 포함하는 어떠한 제형으로도 적용가능하며, 경구용 또는 비경구용으로도 제조할 수 있다. 본 발명의 글루코사민 또는 글루코사민 유도체를 포함하는 약제학적 조성물은 바람직한 순도 하에 약제학적으로 허용가능한 담체, 즉 사용되는 농도와 투여량에서 수용체에게 비독성이고 다른 제제 성분과 화합할 수 있는 담체와 혼합하여 단위 투여량의 제형으로 조제하는 것이 바람직하다. 특히 제제는 산화제 및 인체에 유해한 것으로 알려진 기타 다른 화합물을 포함하지 않는 것이 바람직하다. 또한 본 발명의 글루코사민 또는 글루코사민 유도체는 1종 이상의 약제학적으로 허용가능한 붕해제, 결합제, 활택제 등에서 1종 또는 2종 이상을 선택적으로 함께 약제학적 조성물로 투여될 수 있다. 예를 들어, 본 발명의 조성물을 정제 또는 경질 캅셀제 등의 고형 제형으로 제조할 경우, 부형제로서 미결정 셀룰로오즈, 유당, 저치환도 히드록시셀룰로오즈 등이 사용될 수 있고, 붕해제로서 전분글리콜산 나트륨, 무수인산일수소 칼슘 등이 사용될 수 있다. 또한 결합제로는 폴리비닐피롤리돈, 저치환도 히드록시프로필셀룰로오즈, 히드록시프로필셀룰로오즈 등이 사용될 수 있고, 활택제로서는 스테아린산 마그네슘, 이산화규소, 탈크 등에서 선택하여 사용할 수 있다. 또한, 무수 이염기성 인산칼슘과 같은 정제에 광택을 제공하는 첨가제를 사용할 수 있으며, 수불용성 물질로 피복시킴으로서 공기 중의 수분이 정제 내로 침투하는 것을 방지할 수 있다. 이때 피막기제는 분자구조가 치밀해야 하고, 수용액에 쉽게 용해되지 않는 것이 바람직하다. 이러한 피막제로서 메타크릴산 코폴리머, 히드록시프로필메틸셀룰로오즈프탈레이트, 셀룰로오즈아세테이트프탈레이트, 히드록시프로필메칠셀룰로오즈아세테이트석시네이트, 폴리비닐알코올 등의 고분자물질을 사용할 수 있다. 이들 고분자 물질은 단독 또는 혼합하여 피복에 사용할 수 있다. 또한, 상기 피막은 당업계에서 통상적으로 피막기제에 첨가할 수 있는 첨가제, 예를 들면 가소제, 방부제, 착색제, 차광제 등을 포함할 수 있다.
본 발명의 조성물은 멸균 수용액 등의 액제 및 주사제의 형태일 수 있으며, 필요시 10 내지 40%의 프로필렌글리콜 및 용혈현상을 방지하는데 충분한 양(예로, 약 1%)의 염화나트륨을 함유할 수 있다.
본 발명의 또 다른 양태는, 글루코사민, 글루코사민 유도체 또는 이들의 염을 사용하여 혈관협착 및 재협착을 예방 또는 치료하는 방법에 관한 것이다.
구체적인 일 양태로서, 본 발명의 치료방법은 상기 글루코사민, 글루코사민 유도체 또는 이들의 염을 포함하는 약제학적 조성물을 혈관협착 또는 재협착을 예방 또는 치료하고자 하는 개체에 투여하여 혈관협착 및 재협착을 예방 또는 치료하는 단계를 포함한다. 상기 개체는 인간, 영장류 및 가축, 사육, 애완용 또는 스포츠 동물, 예컨대 개, 말, 고양이, 양, 돼지, 소 등을 비롯한 혈관흡착 및 재협착의 예방 또는 치료가 필요한 임의의 포유동물을 말한다. 특히, 본 발명의 예방 또는 치료는 인간이 바람직하다.
본 발명의 치료방법은 글루코사민 또는 글루코사민 유도체를 약학적 유효량으로 투여하는 것을 포함한다. 적합한 총 1일 사용량은 올바른 의학적 판단범위 내에서 처치의에 의해 결정될 수 있다는 것은 당업자에게 자명한 일이다. 특정 환자에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성물, 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료 기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다.
또한, 본 발명의 치료방법은 글루코사민 또는 글루코사민 유도체를 경구 또는 비경구의 다양한 경로를 통해 통상적인 방식으로 투여할 수 있다. 바람직하게는 혈관에 국소적으로 투여하는 것이다.
이하, 본 발명을 하기 실시예를 참조하여 구체적으로 설명한다. 다만, 하기 실시예는 본 발명의 이해를 돕기 위한 것으로, 이에 의해 본 발명의 범위가 한정되는 것은 아니다.
실시예 1 : 래트 경동맥 풍선 확장술(balloon injury) 모델의 제작
이하의 방법은 Ciro Indolfi, et al.의 방법을 참조하였다(Ciro Indolfi, et al., Effect of Hydroxymethylglutaryl Coenzyme A Reductase Inhibitor Simvastatin on Smooth Muscle Cell Proliferation In Vitro and Neointimal Formation In Vivo After Vascular Injury, Journal of the American College of Cardiology, Vol. 35, No. 1, 2000).
오리엔트 바이오(대한민국)으로부터 구입한 300 내지 320 g (12주령)의 스플라그-돌리 웅성 래트 25마리에 케타민 100mg/kg를 복강 내 주사하여 마취시켰다. 경동맥(carotid artery)의 혈관형성술을 풍선 색전제거술용 카테터(balloon embolectomy cathter)를 사용하여 수행하였다. 구체적으로, 상기 풍선 카테터 (2F Fogarty, Edwards Laboratories, Santa Ana, California)를 우측 외경동맥을 통하여 도입하고, 상기 풍선을 1ml 주사기를 사용하여 부풀렸다. 혈관은 내강(lumen)을 통하여 세 번 부풀려진 풍선을 통과시킴에 의해 파괴되었다.
실시예 2 : 약물 용량 및 투여
조직의 트랜스글루타미나제 저해제인 글루코사민 (2.5Mm)를 플루로닉(pluronic) 겔 (n=15)을 사용하여 풍선 확장술시 손상된 동맥으로 국소 투여하였다. 한편, 대조군으로서 10마리에서는 약물의 투여 없이 풍선 확장술을 수행하였다.
글루코사민의 서방형 제제는 플루로닉 (Pluronic®)을 사용하여 제조되었다. 약물(글루코사민/Sigma)을 물에 용해한 후, 폴록사머를 적절한 중량으로 부가시켜 혼합하였다. 상기 혼합물은 사용하기 전에 4℃에서 보관하였다.
실시예 3 : 글루코사민에 의한 혈관협착에 대한 효과
상기 실시예 2의 글루코사민을 투여하고 1, 2 및 3주 후에 래트의 경동맥에 대한 혈관협착을 관찰하였다. 구체적으로, 래트에 케타민 자일라진(ketamin xylazine) 100mg/kg을 복강 내 주사하여 마취시킨 다음, 경동맥을 제거하고 횡단면이 동맥의 대략 중간 부분에서 절단된 후, 그 3개의 횡단면은 세포 유형을 구별하기 위해서 헤마톡실린-에오신(hematoxylin-eosin)을 사용하여 염색하였다. 상기 횡단면은 저출력하에서 촬영되었고, 맹검적으로 비디오디지털화하고, 이미지 분석 시스템 내에 저장하였다. 상기 중막(media) 및 신생혈관내막 및 혈관벽을 조심스럽게 규명하였고, 신생혈관내막 및 중막의 비율을 산출하였다. 그 결과를 도 1 및 2에 나타내었다.
도 1에서 볼 수 있는 바와 같이, 래트의 경동맥 내피조직을 풍선 카테터로 손상시킨 후 1주, 2주 및 3주 후에 혈관 중막(tunica media)의 증식 정도를 측정한 결과. 글루코사민을 처리한 경우는 아무런 처리를 하지 않은 대조군과 비교하여 혈관 중막의 증식이 통계적으로 유의하게 감소하였음을 알 수 있었다. 또한, 손상된 경동맥 내피조직의 세포증식 정도를 PCNA 염색을 통해 확인한 결과, 글루코사민을 처리한 경우에 아무런 처리를 하지 않은 대조군과 비교하여 혈관의 증식 정도가 현저하게 억제되었음을 알 수 있었다(도 2 참조). 이러한 결과들로 볼 때, 본 발명의 글루코사민은 혈관 흡착 및 재흡착의 억제에 유용함을 알 수 있다. 또한, 글루코사민과 동일한 생리활성을 가지고 있는 글루코사민 유도체도 혈관 흡착 및 재흡착의 억제에 유용할 것이다.
실시예 4 : 대동맥 중막 민무늬 근육세포 유래 세포주(A-10)에서의 글루코사민에 의한 세포증식억제 효과
글루코사민의 세포증식 억제 효과를 알아보기 위해, WST-1 assay를 이용하여 래트의 대동맥 중막 민무늬 근육세포에서 유래된 세포주(rat vascular smooth muscle cells: A-10)를 대상으로 세포증식을 측정하였다. 구체적으로, 먼저 래트의 세포주 A-10에 lysophosphatidic acid(LPA)를 처리하여 세포증식을 유도하였다. 그런 다음, 실시예 2의 글루코사민을 각각 별도로 처리하고, MTS(3-(4,5-디메틸티아졸-2-일)-5(3-카르복시메토시페닐)-2-(4-설포페닐)-2H-테트라졸리움 이너 솔트)를 400 ㎍/㎖의 농도가 되도록 세포 배양액에 첨가하고, 37℃의 온도에서, 12시간, 24시간 동안 동일한 배양조건에서 각각 배양한 후, 시료를 100㎕씩 취하여 96-웰 멀티플레이트에 옮긴 후 마이크로 플레이트 리더(Microplate reader)를 이용하여 440nm에서 흡광도를 측정하였다. 또한, 상기의 과정을 글루코사민 대신 세포사멸억제제로서의 시스타민을 처리하여 반복하였다. 그 결과를 도 3에 나타내었다.
도 3에서 볼 수 있는 바와 같이, 글루코사민을 처리한 경우, 대조군과 비교하여 혈관 민무늬근육 세포의 증식을 억제하는 것을 확인할 수 있었다. 이를 통해, 혈관협착의 주요병리학적 기전인 중막(tunica media) 유래 민무늬근육 세포의 증식을 억제함으로써 혈관의 협착을 방지할 수 있음을 알 수 있다.
본 발명의 글루코사민, 글루코사민 유도체 또는 이들의 염을 포함하는 조성물은 혈관흡착 및 재협착의 예방 또는 치료를 위하여 유용하게 사용할 수 있다.

- 글루코사민, 글루코사민 유도체 또는 이들의 염을 포함하는 혈관 협착 및 재협착 예방 또는 치료용 조성물.
- 제1항에 있어서, 상기 조성물은 혈관 내막의 형성 억제능을 갖는 것을 특징으로 하는 혈관 협착 및 재협착 예방 또는 치료용 조성물.
- 제1항 또는 제2항의 어느 한 항의 조성물을 혈관협착 및 재협착을 예방 또는 치료하고자 하는 개체에 투여하여 혈관협착 및 재협착을 예방 또는 치료하는 방법.
- 제1항 또는 제2항의 어느 한 항의 조성물을 이용하여 혈관 내피조직의 손상 후 신생내막 형성을 억제하는 방법.
- 제5항에 있어서, 상기 혈관은 경동맥 또는 관상동맥인 것을 특징으로 하는 방법.
- 제1항 또는 제2항의 어느 한 항의 조성물을 이용하여 관상동맥 성형술의 시술 후 내피조직의 손상에 의해 유도되는 혈관내막증식증을 예방 또는 치료하는 방법.
- 혈관협착 및 재협착을 예방 또는 치료하기 위한 제1항 또는 제2항의 어느 한 항의 조성물의 용도.
