Description
本発明は、C型レクチンの一種であるデクチン-2遺伝子が欠損した非ヒト動物及びその用途に関する。
マクロファージ、樹状細胞は全身の末梢組織に広く存在しており、末梢から侵入する異物を殺傷、除去する自然免疫の担い手であるとともに、外来抗原を取り込んだ後に自身が移動してこれを二次リンパ器官へ運搬、Tリンパ球に抗原提示することにより獲得免疫を作動させる。近年、マクロファージ、樹状細胞に特異的に発現するC型レクチン(結合のためにカルシウムイオンが必要な動物レクチン)が多数同定され、マクロファージや樹状細胞の機能への関与について研究されている。
C型レクチンの中には、外来異物の捕捉のための受容体として機能するもの(DC-SIGNなど)、樹状細胞が血管外遊走する際のICAM-3との初期接着に関与するもの(DC-SIGN)等が知られている。
樹状細胞に特異的に発現するC型レクチンとしてデクチン-1及びデクチン-2が同定された(非特許文献1、2、特許文献1)。このうち、デクチン-1については、β-グルカンをリガンドとするレクチンであり、カンジダやザイモザンを認識する機能を有することが知られている(非特許文献3)。また、本発明者らはデクチン-1のノックアウトマウスを作成し、デクチン-1がβグルカンを認識し、サイトカイン産生や活性酸素種(ROS)を産生すること、カリニの感染防御に重要な役割を果たしていることを報告した(非特許文献4)。
しかしながら、デクチン-2については、遺伝子はクローニングされているものの、その機能については何ら明らかにされていない。
従って、本発明の課題は、C型レクチンの一種であるデクチン-2遺伝子欠損動物を提供し、さらにデクチン-2の機能を解明し、当該動物の用途を提供することにある。

そこで本発明者らは、遺伝子ターゲティング法を用いてデクチン-2遺伝子の機能を欠損させた動物を作製し、その機能を検討したところ、(1)デクチン-2遺伝子欠損非ヒト動物由来の樹状細胞がα-マンナン刺激によるサイトカイン産生能を消失していたことから、デクチン-2が、α-マンナン刺激による樹状細胞からのサイトカイン産生リセプターであること、(3)樹状細胞より産生されたサイトカインによるTh1又はTh17分化モデルになること、(4)さらにデクチン-2遺伝子欠損動物由来の樹状細胞が真菌刺激によるサイトカイン産生能を消失していたことから、真菌認識分子であること、(5)デクチン-2遺伝子欠損非ヒト動物では野生型に比べて真菌感染を防御できないこと、(6)デクチン-2遺伝子欠損非ヒト動物では野生型に比べて癌細胞の増殖を抑制できないこと、(7)デクチン-2遺伝子欠損非ヒト動物では野生型に比べて破骨細胞形成が抑制されること、(8)デクチン-2遺伝子欠損非ヒト動物では野生型に比べて関節炎が抑制されることを見出し、その結果デクチン-2遺伝子欠損非ヒト動物又は当該動物由来細胞は、樹状細胞によるサイトカイン産生、真菌感染症、癌、骨疾患及び関節炎のモデル動物又はモデル細胞として有用であり、またデクチン-2を発現している細胞を用いれば関節炎治療薬、骨疾患治療薬、Th1又はTh17分化調整剤のスクリーニングができることを見出し、本発明を完成するに至った。
すなわち、本発明は、ゲノム中のデクチン-2遺伝子の機能を欠損した非ヒト動物を提供するものである。
また本発明は、当該デクチン-2遺伝子欠損非ヒト動物又は当該動物由来細胞よりなる、樹状細胞によるサイトカイン産生、樹状細胞より産生されたサイトカインによるTh1又はTh17分化、真菌感染症、癌、骨疾患又は関節炎のモデル動物又はモデル細胞を提供するものである。
また本発明は、当該デクチン-2遺伝子欠損非ヒト動物又は当該動物由来細胞を用いることを特徴とする、樹状細胞によるサイトカイン産生関連疾患治療薬、真菌感染症治療薬、癌治療薬、骨疾患治療薬又は関節炎治療薬のスクリーニング方法を提供するものである。
さらに本発明は、被験物質をデクチン-2を発現している細胞に作用させる工程と、デクチン-2の活性を測定する工程とを含む、関節炎治療薬、骨疾患治療薬又はTh1もしくはTh17分化調節剤のスクリーニング方法を提供するものである。
また本発明は、当該デクチン-2遺伝子欠損非ヒト動物又は当該動物由来細胞よりなる、樹状細胞によるサイトカイン産生、樹状細胞より産生されたサイトカインによるTh1又はTh17分化、真菌感染症、癌、骨疾患又は関節炎のモデル動物又はモデル細胞を提供するものである。
また本発明は、当該デクチン-2遺伝子欠損非ヒト動物又は当該動物由来細胞を用いることを特徴とする、樹状細胞によるサイトカイン産生関連疾患治療薬、真菌感染症治療薬、癌治療薬、骨疾患治療薬又は関節炎治療薬のスクリーニング方法を提供するものである。
さらに本発明は、被験物質をデクチン-2を発現している細胞に作用させる工程と、デクチン-2の活性を測定する工程とを含む、関節炎治療薬、骨疾患治療薬又はTh1もしくはTh17分化調節剤のスクリーニング方法を提供するものである。
本発明のデクチン-2遺伝子欠損非ヒト動物又は当該動物由来細胞を用いれば、デクチン-2が関与する種々の疾患、例えば、樹状細胞によるサイトカイン産生関連疾患、真菌感染症、癌、骨疾患、関節炎等の治療薬のスクリーニングができる。また、さらに当該動物は、新たなデクチン-2の機能解明用の研究対象としても有用であり、例えばデクチン-2の内在性のリガンドの解明、デクチン-2が介在するシグナル経路の解明に利用できる。
本発明のデクチン-2遺伝子の機能を欠損した非ヒト動物としては、ゲノム中のデクチン-2遺伝子の一部又は全部を改変させた非ヒト動物が例示できる。
本発明において「ゲノム中のデクチン-2遺伝子の一部を改変する」とは、デクチン-2遺伝子の一部に変更、削除、挿入又は置換を生じさせることをいう。デクチン-2遺伝子を機能不能にするためには、デクチン-2遺伝子に対して、変更、削除、挿入又は置換のうち2又はそれ以上の方法を同時に使用してもよい。本発明において、デクチン-2遺伝子の変更、削除、挿入又は置換とは、デクチン-2遺伝子のゲノムDNA配列中の1又は2以上の塩基を、置換、削除、又は挿入し、当該変異させたデクチン-2遺伝子の発現産物がデクチン-2分子として機能しないようにすることをいう。
非ヒト動物としてはげっ歯類が好ましく、特にマウスが好ましい。
当該遺伝子の一部又は全部の改変は、ターゲティングベクターの構築により行なわれる。得られたターゲティングベクターによるES細胞の相同組換えを行い、得られた相同組換えES細胞を胚内に移植し、ES細胞移植胚を偽妊娠仮親の子宮に移植して出産させることによりキメラ動物を作製することができる。以下、各工程毎に説明する。
デクチン-2遺伝子は、すでにクローニングされており、その塩基配列及びアミノ酸配列は配列番号1に示すとおりである。
ターゲティングベクターの構築
デクチン-2遺伝子のゲノムDNA配列を、デクチン-2遺伝子とは関連しないDNA配列、たとえば選択マーカー遺伝子DNA配列などと置換する方法により、ターゲティングベクターを作製する。これは、ターゲティングベクター中のDNAによって相同組換えされた相同組換え体を、選択マーカーなどを用いることで、より容易にスクリーニングすることができるようにするためである。より具体的には、単離されたデクチン-2遺伝子のゲノムDNAの一部を制限酵素処理により削除し、削除されたDNA領域の代わりにプロモーターを連結した選択マーカー遺伝子を挿入することにより、デクチン-2遺伝子のターゲティングベクターを作製する。
デクチン-2遺伝子のゲノムDNA配列を、デクチン-2遺伝子とは関連しないDNA配列、たとえば選択マーカー遺伝子DNA配列などと置換する方法により、ターゲティングベクターを作製する。これは、ターゲティングベクター中のDNAによって相同組換えされた相同組換え体を、選択マーカーなどを用いることで、より容易にスクリーニングすることができるようにするためである。より具体的には、単離されたデクチン-2遺伝子のゲノムDNAの一部を制限酵素処理により削除し、削除されたDNA領域の代わりにプロモーターを連結した選択マーカー遺伝子を挿入することにより、デクチン-2遺伝子のターゲティングベクターを作製する。
ES細胞中で強い発現活性を有するプロモーターとしてはホスホグリセリン酸キナーゼ-1(PGK-1)プロモーター、伸長因子2(EF-2)プロモーター、MC-1プロモーターなどを使用することができる。ターゲティングを行う遺伝子座、及びそのDNA領域により、プロモーター活性は大きく影響されるため、一般にはプロモーターは強力であるほどよい。従ってより望ましくは、PGK-1プロモータを使用する。
選択マーカー遺伝子としては、ネオマイシン耐性遺伝子、ハイグロマイシンBホスホトランスフェラーゼ遺伝子、ネガティブ選択用にはチミジンキナーゼ遺伝子、ジフテリア毒素A遺伝子断片などを目的に応じて使用することができる。
プロモーターと連結した選択マーカー遺伝子は、たとえばネオマイシン耐性遺伝子カセット(neoカセット、pKJ2として供与)、ハイグロマイシンBホスホトランスフェラーゼ遺伝子カセット(hphカセット)、チミジンキナーゼ遺伝子カセット(tkカセット、pMCtkとして供与)、ジフテリア毒素A遺伝子断片カセット(DT-Aカセット、pMCDT-A又はpMC1DT-Aとして供与)などの、プラスミドに挿入された状態で市販されている選択マーカー遺伝子を使用してもよい。ターゲティングベクターは、これらのプロモーターを連結した選択マーカーのうち、どの選択マーカーを使用して構築してもよい。
ターゲティングベクターによる相同組換え
次いで、上記の方法により作製したターゲティングベクターを使用して、ES細胞の相同組換えを行う。用いることができるマウス由来のES細胞は、TT2細胞、AB-1細胞、J1細胞、R1細胞、E14.1細胞などがある。キメラ動物を作出するために、これらのうちいずれのES細胞を用いるかは、採取したゲノムDNAの由来するマウス系統、キメラ動物の選別方法など、実験の目的や方法により任意に決定することができる。
次いで、上記の方法により作製したターゲティングベクターを使用して、ES細胞の相同組換えを行う。用いることができるマウス由来のES細胞は、TT2細胞、AB-1細胞、J1細胞、R1細胞、E14.1細胞などがある。キメラ動物を作出するために、これらのうちいずれのES細胞を用いるかは、採取したゲノムDNAの由来するマウス系統、キメラ動物の選別方法など、実験の目的や方法により任意に決定することができる。
デクチン-2遺伝子を機能不能な配列にしたターゲティングベクターをES細胞中に導入する。そしてES細胞中の目的とするデクチン-2遺伝子のゲノムDNA配列を、ターゲティングベクター中の機能不能にしたデクチン-2遺伝子DNA配列による相同組換えによって置換する。相同組換えは、デクチン-2遺伝子ゲノムDNA配列と、ターゲティングベクター中の非改変部分の配列との相同性を利用して、生じさせることができる。
ターゲティングベクターをES細胞に導入する方法としては、エレクトロポレーション法、リン酸カルシウム法、DEAE-デキストラン法などを使用することができる。効率、作業の容易性などを考慮して、エレクトロポレーション法を用いることが好ましい。
相同組換え体のスクリーニング
得られた組換えES細胞の中から、培養液中で、相同組換えによってES細胞に導入された選択マーカーを用いて、第一段階のスクリーニングである選択培養を行う。当該第一段階の選択工程においては、前述した選択マーカーのうち、複数の選択マーカー遺伝子を用いることもできる。

耐性クローンをピックアップし、増殖させる。その後、目的とするデクチン-2遺伝子がターゲティングされているかどうかを確実に調べるため、第二段階のスクリーニングを行うことができる。第二段階のスクリーニングは、DNAレベルで目的とするデクチン-2遺伝子がターゲティングされたかどうかを確認する工程である。このような方法としては、PCR法、サザンブロットハイブリダイゼーション法などが存在するが、そのうちのいずれを用いてもよく、またその中から2以上の方法を組み合わせて用いてもよい。
本発明においては、第一段階のスクリーニングにより選択されたES細胞クローンのプールのそれぞれからゲノムDNAを調製し、このゲノムDNAを鋳型としてPCR法を行うことで、ES細胞について第二段階のスクリーニングをするのが好ましい。このPCR法を用いたスクリーニングにより組換えが起こったクローンを単離する。
PCRプライマーは、ターゲティングにより目的とするゲノムDNA領域を外来性の遺伝子により置換することができたか否かをより効率的に確認できるように設計することが望ましい。そのためにはターゲティングベクターの配列のうち目的とする遺伝子の外側のゲノムDNA領域上と、選択マーカー遺伝子カセット領域上とに設定し、挿入部位を増幅することができるように設計するとよい。
第二段階のスクリーニングにおいて、ES細胞が相同組換え体であることを確認するため、ES細胞クローンに対してサザンブロットハイブリダイゼーション法による解析を行うことができる。サザンブロットハイブリダイゼーション解析は、ターゲティングベクターに含まれないゲノム領域を認識するプローブとターゲティングベクター中に存在する領域を認識するプローブを併用して行うことが望ましい。二種類のプローブを用いることにより相同組換えのほかに非相同組換えが存在するか否かを知ることができ、目的とするゲノム領域に相同組換えが起こったことをより正確に確認することができる。
デクチン-2遺伝子欠損(KOともいう)動物の作出 (Dectin-2 -/- )
次いで、相同組換えの結果得られた組換えES細胞を、8細胞期又は胚盤胞の胚内に移植する。このES細胞移植胚を偽妊娠仮親の子宮内に移植して出産させることによりキメラ動物を作製することができる。

ES細胞を胚内に移植する方法としては、たとえばマイクロマニピュレーション法、凝集法などが知られているがいずれを用いてもよい。マウスの場合、まず、ホルモン剤(たとえば、FSH様作用を有するPMSG及びLH作用を有するhCGを使用)により過排卵処理を施した雌マウスを、雄マウスと交配させる。その後、8細胞期胚を用いる場合には受精から2.5日目に、胚盤胞を用いる場合には受精から3.5日目に、それぞれ子宮から初期発生胚を回収する。このように回収した胚に対して、ターゲティングベクターを用いて相同組換えを行ったES細胞をin vitroにおいて注入し、キメラ胚を作製する。
一方、仮親にするための偽妊娠雌マウスは、正常性周期の雌マウスを、精管結紮などにより去勢した雄マウスと交配することにより得ることができる。作出した偽妊娠マウスに対して、上述の方法により作成したキメラ胚を子宮内移植し、妊娠・出産させることによりキメラ動物を作製することができる。
このようなキメラマウスの中から、ES細胞移植胚由来の雄マウスを選択する。このように、胚内に移植された組換えES細胞が生殖系列に導入された動物を選択し、そのキメラ動物を繁殖することにより目的とする遺伝子を欠損する個体を得ることができる。得られたデクチン-2遺伝子欠損ヘテロ接合体マウスどうしを交配させることにより、目的とする遺伝子欠損ホモ接合体マウスを得ることができる。
得られた本発明の非ヒト動物は、ゲノム中のデクチン-2遺伝子の機能が欠損している。従来、デクチン-2がどのような機能を有しているかを確認した報告はなく、デクチン-2遺伝子欠損動物がどのような特性を有しているかは不明であった。本発明者らは、本発明の非ヒト動物が次の機能を有することを確認した。
(1)デクチン-2遺伝子欠損動物由来の樹状細胞は、α-マンナン及びマンナン刺激によるサイトカイン(TNF、IL-12、IL-10、IL-6)産生能が消失していた。一方、野生型由来の樹状細胞は、α-マンナン刺激によりTNF、IL-12、IL-10、IL-6等を産生する。従って、デクチン-2は、樹状細胞におけるα-マンナン刺激によるサイトカイン産生のキーリセプターである。
(2)デクチン-2遺伝子欠損動物由来の樹状細胞は真菌刺激によるサイトカイン(TNF、IL-12、IL-10、IL-6、IL-1β、IL-23)産生能が消失する。一方、野生型由来の樹状細胞は真菌刺激によりTNF、IL-12、IL-10、IL-6、IL-1β、IL-23を産生する。
(3)真菌で刺激をしたデクチン-2遺伝子欠損動物由来の樹状細胞培養上清はT細胞をTh17に分化させなかった。一方、真菌で刺激をした野生型マウス由来の樹状細胞培養上清はT細胞をTh17に分化させた。従って、デクチン-2は、樹状細胞における真菌刺激によるサイトカイン産生のキーリセプターである。また、デクチン-2遺伝子欠損動物又は当該動物由来細胞は、樹状細胞により産生されたサイトカインによるTh1又はTh17分化モデルとして有用である。
(4)デクチン-2遺伝子欠損動物では、野生型に比べて真菌感染症に対する防御力が低下していた。従って、デクチン-2は、真菌感染症防御機構に関与していることがわかる。
(5)デクチン-2遺伝子欠損動物では、野生型に比べて移植された癌細胞の増殖を抑制できなかった。従って、デクチン-2は癌細胞の増殖抑制機構に関与していることがわかる。
(6)デクチン-2遺伝子欠損動物では、野生型に比べて破骨細胞形成が抑制されていた。従って、デクチン-2は破骨細胞形成に関与していることがわかる。
(7)デクチン-2遺伝子欠損動物では、野生型に比べて関節炎の発症を抑制されていた。従って、デクチン-2は関節炎の発症、進展に関与していることがわかる。
(3)真菌で刺激をしたデクチン-2遺伝子欠損動物由来の樹状細胞培養上清はT細胞をTh17に分化させなかった。一方、真菌で刺激をした野生型マウス由来の樹状細胞培養上清はT細胞をTh17に分化させた。従って、デクチン-2は、樹状細胞における真菌刺激によるサイトカイン産生のキーリセプターである。また、デクチン-2遺伝子欠損動物又は当該動物由来細胞は、樹状細胞により産生されたサイトカインによるTh1又はTh17分化モデルとして有用である。
(4)デクチン-2遺伝子欠損動物では、野生型に比べて真菌感染症に対する防御力が低下していた。従って、デクチン-2は、真菌感染症防御機構に関与していることがわかる。
(5)デクチン-2遺伝子欠損動物では、野生型に比べて移植された癌細胞の増殖を抑制できなかった。従って、デクチン-2は癌細胞の増殖抑制機構に関与していることがわかる。
(6)デクチン-2遺伝子欠損動物では、野生型に比べて破骨細胞形成が抑制されていた。従って、デクチン-2は破骨細胞形成に関与していることがわかる。
(7)デクチン-2遺伝子欠損動物では、野生型に比べて関節炎の発症を抑制されていた。従って、デクチン-2は関節炎の発症、進展に関与していることがわかる。
本発明のデクチン-2遺伝子欠損動物又はその由来細胞は、上記のような性質を有することから、樹状細胞によるサイトカイン産生、樹状細胞より産生されたサイトカインによるTh1又はTh17分化真菌感染症、癌、骨疾患及び関節炎のモデル動物又はモデル細胞として有用である。ここでサイトカインとしてはTNF、IL-12、IL-10、IL-6、IL-23、IL-1β、IL-17F等が挙げられるが、ここに挙げたサイトカインに限定されるものではなく、デクチン-2が起因する他のサイトカインを含めることができる。真菌としては、マイクロスポラム、トリコフィトン、アコリオン、エピダーモフィトン、ケラチノマイセス、カンジダ、カリニ、クリプトコッカス、アスペルギルス等が挙げられ、特にカンジダ、カリニ、クリプトコッカス、アスペルギルス等が好ましい。癌としては、腺癌、肉腫、白血病、上皮性腫瘍等が挙げられ、より具体的には、咽頭癌を代表とする口唇、口腔及び咽頭癌;食道癌、胃癌、結腸・直腸癌などを代表とする消化器癌;肺癌を代表とする呼吸器及び胸腔内臓器癌;骨及び関節軟骨癌、皮膚の悪性黒色腫、有棘細胞癌及びその他の皮膚の癌;中皮腫を代表とする中皮及び軟部組織癌、乳癌、子宮癌、卵巣癌を代表とする女性性器癌;前立腺癌を代表とする男性性器癌;膀胱癌を代表とする尿路癌:脳腫瘍を代表とする眼、脳及び中枢神経系癌、甲状腺及びその他の内分泌腺癌、非ホジキンリンパ腫やリンパ性白血病を代表とするリンパ組織、造血組織及び関連組織癌、及びこれらを原発巣とする転移組織の癌等が挙げられる。骨疾患としては、骨代謝性疾患、特に骨粗しょう症等が挙げられる。また、関節炎としては、関節リウマチ等が挙げられる。
また、本発明のデクチン-2遺伝子欠損動物又はその由来細胞を用いれば、サイトカイン産生関連疾患、真菌感染症、癌、骨疾患及び関節炎の治療薬をスクリーニングすることができる。これらの医薬のスクリーニングは、本発明のデクチン-2遺伝子欠損動物又はその由来細胞に、被験物質を投与又は添加し、各疾患の発症又は進展を観察すればよい。ここで、各疾患の発症及び進展の観察は、各疾患のマーカーとなるサイトカインを測定することにより行うこともできる。同時にコントロール物質を投与した群をコントロール群とするのが好ましい。さらに野生型も同時に試験するのが好ましい。
本発明の関節炎治療薬又は骨疾患治療薬のスクリーニングは、被験物質をデクチン-2を発現している細胞に作用させる工程と、デクチン-2の活性を測定する工程によっても行うことができる。ここで、デクチン-2を発現している細胞としては、デクチン-2KO樹状細胞でなければ特に制限されず、広く動物細胞の樹状細胞が挙げられる。また、このスクリーニング方法には、インビトロだけでなくインビボも含まれるため、被験物質をデクチン-2を発現している細胞に作用させる工程には、動物から採取した細胞や動物由来の株化細胞に直接被験物質を作用させる工程だけでなく、そのような細胞を保有している動物に被験物質を投与する場合も含まれる。次の工程であるデクチン-2活性の測定手段としては、例えばデクチン-2遺伝子の発現量の測定、デクチン-2の作用により変動するサイトカイン量の測定等が挙げられる。ここでデクチン-2の作用による変動するサイトカインとしては、前記TNF、IL-12、IL-10、IL-6、IL-23、IL-1β、IL-17F等が挙げられる。かかるデクチン-2活性を測定した結果、デクチン-2活性を抑制した被験物質を選択すれば、関節炎治療薬、骨疾患治療薬がスクリーニングできる。また、デクチン-2からのシグナルによりTh1、Th17の分化を誘導することができる。そのため、本スクリーニングにより、デクチン-2の活性を測定した結果、デクチン-2の活性を抑制する物質を選別することにより、Th1やTh17の分化を抑制し得る物質を得ることができる。逆に、デクチン-2の活性を促進する物質を選別することにより、Th1やTh17の分化を促進し得る物質を得ることができる。Th1やTh17の分化を抑制する物質は、上記関節炎治療薬以外にも、Th1やTh17分化促進が起因する疾患の治療薬になり得る。Th1やTh17の分化促進する物質は、Th1やTh17分化促進により治療効果がもたらされる疾患、例えば、感染症の治療薬になり得る。
本発明のデクチン-2遺伝子欠損動物又はその由来細胞は、デクチン-2に対する抗体を作製するための動物又は細胞として利用できる。すなわち、デクチン-2遺伝子欠損動物は、デクチン-2を発現しないので、良好なヒト型抗デクチン-2抗体産生材料として有用である。例えば、ヒト型抗デクチン-2抗体を産生するマウスと、デクチン-2遺伝子欠損マウスとを交配して、抗デクチン-2抗体を産生し、かつデクチン-2遺伝子欠損マウスを作製すれば、当該マウスはヒト型抗デクチン-2抗体として良好な抗体を産生する。また、ヒト型抗デクチン-2抗体を産生するマウス由来のES細胞を用いて本発明と同様の手段によりデクチン-2遺伝子欠損マウスを作製すれば、そのマウスはヒト型抗デクチン-2抗体として良好な抗体を産生する。このようにして得られる抗デクチン-2抗体は、前記各種疾患の治療に有用な医薬となり得る。例えば、骨疾患、関節炎についてはデクチン-2の活性を抑制し得る抗体を、癌や真菌感染症に対してはデクチン-2の活性を促進し得る抗体を選択すれば、これらの疾患又は治療薬となり得る。
さらに本発明のデクチン-2遺伝子欠損動物は、デクチン-2の内在性リガンドの解明、デクチン-2が外在するシグナル経路の解明のためのツールとしても有用である。
次に実施例を挙げて本発明をさらに詳細に説明する。
実施例1
Dectin-2-/-マウスの作製
デクチン-2をコードする遺伝子Clec4nのエキソン1と2(デクチン-2の細胞内及び膜貫通領域をコード)をネオマイシン耐性遺伝子(neor)で置換するためのターゲティングベクターを構築した(図1)。ターゲティングベクターは、neorとネガティブ選抜遺伝子であるジフテリア毒素(DT)遺伝子を持つカセットプラスミドpBl-lx-neo-DTを用い、そのBamHI部位に及びHindIII部位にそれぞれES細胞(E14.1)から抽出したゲノムDNAを鋳型としてPCR法にて増幅した5’側相同領域(プライマーセット:TTG GAT CCA AGA AGG TCA GTC TGG TCC ATG AC(配列番号2)及びAGG GAT CCT GGC TTC TGT CAA AGA GG(配列番号3))と3’側相同領域(プライマーセット:TTT AAG CTT CCC ATT ATC TGG TCT GAC TTC ACA(配列番号4)及びCCT CTA TGA ATA TAG TTG CTC CAG ACG(配列番号5))を対応する制限酵素で処理後、挿入した。NotI 消化により直鎖化したターゲティングベクター25μgを1×107細胞のE14.1にGENE PULSER II(BioRad)を用いて電気穿孔法(500μF/0.25V)により導入した。培養24時間後より、300μg/mlのG418(ナカライ)を含むES培地(15%ウシ胎児血清(FCS)(Biowest)、1,000U/ml LIF、10mMピルビン酸ナトリウム(Gibco)、非必須アミノ酸(Gibco)、55μMメルカプトエタノール(Gibco)及び50μg/mlペニシリン(Gibco)、50U/mlストレプトマイシン(Gibco)を含有するDMEM(Gibco))にて1週間薬剤選抜を行ない、耐性クローンの単離を行なった。相同組換えクローンはPCR法(プライマーセット:AAA CAG CTG GTA ATG CCT TGG TGG(配列番号6)及びCAT TAT ACG AAG TTA TCT CGA GTC GC(配列番号7))及びサザンハイブリダイゼーション法により同定した。なお、サザンハイブリダイゼーション法はBamHI 消化したゲノムDNAに対して、プローブとしては、PCR法によって増幅したゲノム領域のDNA断片を用いて、5’側(TTG GCA TAC CAA CCC ATG AGA AGT T(配列番号8)AAC TAG AAG CCT GGG ATG TAC GTG(配列番号9)及びAAA CAG CTG GTA ATG CCT TGG TGG(配列番号10)とAAT GAG GTA ACT CTC ATC AGT CCC T(配列番号11))3’側(ACT GGT GTA CAA AGC TGT AGG CTT C(配列番号12)TTG GCA GAG CCA AGA AAT GCT CAG(配列番号13))についてそれぞれ行なった。相同組換えESクローンは、凝集胚キメラ法を用いてBDF1オスマウスとC57BL/6Jメスマウス(日本SLC)との交配により得られた8細胞期胚とのキメラ胚を作製した後、偽妊娠処理をした仮親のICRメスマウス(日本SLC)の子宮に移植することにより、キメラマウスを誕生させた。キメラマウスはC57BL/6Jマウスと交配し、ヘテロ接合体のF1マウスを得た。Dectin-2-/-マウスを得るため、ヘテロ接合体のF1子孫を兄妹交配した。また、C57BL6/J背景のDectin-2-/-を得るためにヘテロ接合体のF1子孫をさらに5世代又は7世代C57BL/6Jマウスに戻し交配を行い、最終的に6世代目あるいは8世代目のヘテロ接合体マウスを兄妹交配した。BLAB/cA背景のDectin-2-/-マウスを得るためにはBALB/cAマウスとキメラマウスを交配し、得られたF1へテロ接合体マウスを最終的に8世代BALB/cAマウスに戻し交配した。得られたヘテロ接合体マウスを兄妹交配し、BALB/cA背景のDectin-2-/-マウスを得た。
Dectin-2-/-マウスの作製
デクチン-2をコードする遺伝子Clec4nのエキソン1と2(デクチン-2の細胞内及び膜貫通領域をコード)をネオマイシン耐性遺伝子(neor)で置換するためのターゲティングベクターを構築した(図1)。ターゲティングベクターは、neorとネガティブ選抜遺伝子であるジフテリア毒素(DT)遺伝子を持つカセットプラスミドpBl-lx-neo-DTを用い、そのBamHI部位に及びHindIII部位にそれぞれES細胞(E14.1)から抽出したゲノムDNAを鋳型としてPCR法にて増幅した5’側相同領域(プライマーセット:TTG GAT CCA AGA AGG TCA GTC TGG TCC ATG AC(配列番号2)及びAGG GAT CCT GGC TTC TGT CAA AGA GG(配列番号3))と3’側相同領域(プライマーセット:TTT AAG CTT CCC ATT ATC TGG TCT GAC TTC ACA(配列番号4)及びCCT CTA TGA ATA TAG TTG CTC CAG ACG(配列番号5))を対応する制限酵素で処理後、挿入した。NotI 消化により直鎖化したターゲティングベクター25μgを1×107細胞のE14.1にGENE PULSER II(BioRad)を用いて電気穿孔法(500μF/0.25V)により導入した。培養24時間後より、300μg/mlのG418(ナカライ)を含むES培地(15%ウシ胎児血清(FCS)(Biowest)、1,000U/ml LIF、10mMピルビン酸ナトリウム(Gibco)、非必須アミノ酸(Gibco)、55μMメルカプトエタノール(Gibco)及び50μg/mlペニシリン(Gibco)、50U/mlストレプトマイシン(Gibco)を含有するDMEM(Gibco))にて1週間薬剤選抜を行ない、耐性クローンの単離を行なった。相同組換えクローンはPCR法(プライマーセット:AAA CAG CTG GTA ATG CCT TGG TGG(配列番号6)及びCAT TAT ACG AAG TTA TCT CGA GTC GC(配列番号7))及びサザンハイブリダイゼーション法により同定した。なお、サザンハイブリダイゼーション法はBamHI 消化したゲノムDNAに対して、プローブとしては、PCR法によって増幅したゲノム領域のDNA断片を用いて、5’側(TTG GCA TAC CAA CCC ATG AGA AGT T(配列番号8)AAC TAG AAG CCT GGG ATG TAC GTG(配列番号9)及びAAA CAG CTG GTA ATG CCT TGG TGG(配列番号10)とAAT GAG GTA ACT CTC ATC AGT CCC T(配列番号11))3’側(ACT GGT GTA CAA AGC TGT AGG CTT C(配列番号12)TTG GCA GAG CCA AGA AAT GCT CAG(配列番号13))についてそれぞれ行なった。相同組換えESクローンは、凝集胚キメラ法を用いてBDF1オスマウスとC57BL/6Jメスマウス(日本SLC)との交配により得られた8細胞期胚とのキメラ胚を作製した後、偽妊娠処理をした仮親のICRメスマウス(日本SLC)の子宮に移植することにより、キメラマウスを誕生させた。キメラマウスはC57BL/6Jマウスと交配し、ヘテロ接合体のF1マウスを得た。Dectin-2-/-マウスを得るため、ヘテロ接合体のF1子孫を兄妹交配した。また、C57BL6/J背景のDectin-2-/-を得るためにヘテロ接合体のF1子孫をさらに5世代又は7世代C57BL/6Jマウスに戻し交配を行い、最終的に6世代目あるいは8世代目のヘテロ接合体マウスを兄妹交配した。BLAB/cA背景のDectin-2-/-マウスを得るためにはBALB/cAマウスとキメラマウスを交配し、得られたF1へテロ接合体マウスを最終的に8世代BALB/cAマウスに戻し交配した。得られたヘテロ接合体マウスを兄妹交配し、BALB/cA背景のDectin-2-/-マウスを得た。
デクチン-2ターゲティングベクターを129/Ola由来のES細胞であるE14.1に導入し、257クローンについてスクリーニングを行ない、4つの相同組換えクローンの単離・同定に成功した。これらは凝集胚キメラ法によりキメラマウスの作製を行ない、交配によりDectin-2KOマウスの樹立・系統化を行なった。デクチン-2欠損遺伝子座については、尻尾より抽出したゲノムDNAをBamHI消化した後、サザンハイブリダイゼーション法により確認した。さらに、脾臓より抽出したRNAを用いたノザンハイブリダイゼーション法により、Dectin-2KOマウスではデクチン-2mRNAの発現が消失していることを確認し(図2)、デクチン-2遺伝子が完全に破壊されていることを確認した。
実施例2
樹状細胞によるサイトカインの産生
(1)骨髄由来樹状細胞(BMDC)及び試薬
Dectin-2-/-マウスあるいは野生型C57BL/6Jマウスの大腿骨、脛骨、腰骨より骨髄細胞を採取し、MEM培地(大日本製薬)中に拡散させた。0.017M Tris-HCl(pH7.2),、0.14M NH4Clを含む緩衝液で、赤血球を除去後、2×106細胞を20ng/mlのGM-CSF(ペプロテック)、10%ウシ胎児血清(BioWest)を含むRPMI-1640培地(シグマ)10mlに懸濁し、直径10cmのペトリ皿で培養した。3日後に同じ組成の培地を10ml加え、培養液を20mlとした。6日後に半量である10mlの培地をとり、新しく調整した10ng/mlのGM-CSF(ペプロテック)、10%FCS(BioWest)100μg/mlペニシリン、100U/mlストレプトマイシンを含むRPMI-1640培地(シグマ)に交換した。8日後に再度20mlの培養液のうち10mlを新しい培地と交換した。10日後の細胞をBMDCとして利用した。2×106細胞を10%ウシ胎児血清を含むRPMI-1640培地に懸濁し、Candia albicans(C.albicans)由来のαマンナン及びマンナンを最終濃度0μg/ml、1μg/ml、10μg/mlになる様に加え24時間培養し、培養上清を回収した。
樹状細胞によるサイトカインの産生
(1)骨髄由来樹状細胞(BMDC)及び試薬
Dectin-2-/-マウスあるいは野生型C57BL/6Jマウスの大腿骨、脛骨、腰骨より骨髄細胞を採取し、MEM培地(大日本製薬)中に拡散させた。0.017M Tris-HCl(pH7.2),、0.14M NH4Clを含む緩衝液で、赤血球を除去後、2×106細胞を20ng/mlのGM-CSF(ペプロテック)、10%ウシ胎児血清(BioWest)を含むRPMI-1640培地(シグマ)10mlに懸濁し、直径10cmのペトリ皿で培養した。3日後に同じ組成の培地を10ml加え、培養液を20mlとした。6日後に半量である10mlの培地をとり、新しく調整した10ng/mlのGM-CSF(ペプロテック)、10%FCS(BioWest)100μg/mlペニシリン、100U/mlストレプトマイシンを含むRPMI-1640培地(シグマ)に交換した。8日後に再度20mlの培養液のうち10mlを新しい培地と交換した。10日後の細胞をBMDCとして利用した。2×106細胞を10%ウシ胎児血清を含むRPMI-1640培地に懸濁し、Candia albicans(C.albicans)由来のαマンナン及びマンナンを最終濃度0μg/ml、1μg/ml、10μg/mlになる様に加え24時間培養し、培養上清を回収した。
C.albicans(NBRC1385株)はBMDCの刺激に用いる2日前にポテトデキストロース寒天培地(栄研化学)に播種し、30℃の恒温槽で培養した。生えて来たC.albicansのうち、独立したコロニーを40個程度エーゼでかき取り、20mlの生理的食塩水に懸濁した。C.albicansを生理的食塩水で2回洗った後、1×106細胞/ml及び5×105になる様に濃度を調整し、65℃の恒温槽で2時間培養した。これをC.albicans死菌としてBMDCの刺激に用いた。BMDCにC.albicans死菌を加え、48時間後に培養上清を回収し、サイトカイン濃度の測定、あるいはT細胞の培養に用いた。
T細胞は、C57BL/6Jマウスのリンパ節細胞を採取し細胞を単離後、抗マウスCD4抗体、抗マウスCD62L抗体で染色し、CD4陽性、CD62L陽性細胞をセルソーターで分離した。あらかじめ抗マウスCD3抗体を個層化した48穴プレートにT細胞を4×105細胞になる様に播種し、無血清培地とC.albicans死菌で刺激を行ったBMDCの培養上清を1対1になるように混合し、5日間培養した。5日目にT細胞を回収し、細胞内のIL-17とIFN-γの存在量をフローサイトメーターで測定した。
T細胞は、C57BL/6Jマウスのリンパ節細胞を採取し細胞を単離後、抗マウスCD4抗体、抗マウスCD62L抗体で染色し、CD4陽性、CD62L陽性細胞をセルソーターで分離した。あらかじめ抗マウスCD3抗体を個層化した48穴プレートにT細胞を4×105細胞になる様に播種し、無血清培地とC.albicans死菌で刺激を行ったBMDCの培養上清を1対1になるように混合し、5日間培養した。5日目にT細胞を回収し、細胞内のIL-17とIFN-γの存在量をフローサイトメーターで測定した。
(2)TNF、IL-12p40、IL-6、IL-10、IL-23、IL-1β産生量の測定
酵素免疫吸着測定(ELISA)法で回収した培養上清中のTNF、IL-12p40、IL-6、IL-10、IL-23、IL-1βの産生量の測定を行った。キットはIL-23はeBioscience社のものを使用し、IL-23以外は全てBDBiosciences社のものを使用し、添付の測定方法に従い、測定した。
酵素免疫吸着測定(ELISA)法で回収した培養上清中のTNF、IL-12p40、IL-6、IL-10、IL-23、IL-1βの産生量の測定を行った。キットはIL-23はeBioscience社のものを使用し、IL-23以外は全てBDBiosciences社のものを使用し、添付の測定方法に従い、測定した。
(3)C.albicans 由来マンナンに対する反応性の低下
Dectin-2-/-マウス及び野生型C57BL/6Jマウス由来BMDCを用いて、C.albicans由来αマンナン及びマンナンで刺激し24時間後の培養上清中のサイトカイン産生量を測定した。その結果、TNF、IL-6、IL-12p40、IL-10全てにおいて、野生型マウス由来BMDCからはαマンナン、マンナンの用量依存的にサイトカイン産生が見られたが、Dectin-2-/-マウス由来BMDCは反応性を失い、ほぼ完全にサイトカイン産生が消失した。また、マンナンで刺激をした場合には野生型マウス由来BMDCは用量依存的にTNF、IL-12p40を産生したが、Dectin-2-/-マウス由来BMDCは反応性を失い、ほぼ完全にサイトカイン産生が消失した(図3)。この結果からデクチン-2はC.albicans由来αマンナン及びマンナンを認識し、サイトカイン産生するために必須であることが示された。

Dectin-2-/-マウス及び野生型C57BL/6Jマウス由来BMDCを用いて、C.albicans死菌で刺激し48時間後の培養上清中を採取し、抗CD3抗体と抗CD28抗体存在下にナイーブT細胞に添加すると野生型マウス由来BMDC培養上清はT細胞をTh17細胞とTh1細胞に分化させたが、デクチン-2-/-由来BMDCの培養上清はT細胞をTh17細胞に分化させなかった(図5)。この結果からデクチン-2はC.albicansを認識し、T細胞をTh17細胞に分化させるために必須であることが示された。
実施例3
感染実験
C57BL/6背景のDectin-2-/-と野生型C57BL/6Jマウスを用い、C.albicansの感染実験を行った。C.albicans(THK519株)はマウスに感染させる2日前にポテトデキストロース寒天培地(栄研化学)に播種し、30℃の恒温槽で培養した。生えて来たC.albicansのうち、独立したコロニーを40個程度エーゼでかき取り、20mlの生理的食塩水に懸濁した。C.albicansを生理的食塩水で2回洗った後、1.25×106細胞/mlになる様に濃度を調整し、そのうち1匹当たり200μlを尾静脈より投与し、その後毎日生死の確認を行った。
感染実験
C57BL/6背景のDectin-2-/-と野生型C57BL/6Jマウスを用い、C.albicansの感染実験を行った。C.albicans(THK519株)はマウスに感染させる2日前にポテトデキストロース寒天培地(栄研化学)に播種し、30℃の恒温槽で培養した。生えて来たC.albicansのうち、独立したコロニーを40個程度エーゼでかき取り、20mlの生理的食塩水に懸濁した。C.albicansを生理的食塩水で2回洗った後、1.25×106細胞/mlになる様に濃度を調整し、そのうち1匹当たり200μlを尾静脈より投与し、その後毎日生死の確認を行った。
野生型マウスと比較し、Dectin-2-/-マウスでは有意に生存率が低下していた。この結果からデクチン-2はC.albicansに対する感染防御に重要な役割を果たしていることが示された(図6)。
実施例4
コラーゲン誘導関節炎(CIA;collagen-inducedarthritis)
CIAの誘導には129×C57BL/6J F1のDectin-2-/-及びDectin-2+/+マウスを使用した。CFA(Difco)と等量の4mg/mlチキンII型コラーゲン(IIC)(シグマ)をエマルジョン化し、1匹当たり0.1mlを後背部の皮下に投与した。CFAには1mlに対して結核死菌(Difco)5mgを予め添加した。初回免疫から21日後に同様に調整したIICを用いて追加免疫を行った。
野生型マウスと比較し、Dectin-2-/-マウスではCIAの発症率が低下した(図7)。この結果から、デクチン-2は関節炎の発症に重要な役割を果していることが示された。

実施例5
破骨細胞分化
野生型C57BL/6Jマウス及びC57BL/6J背景のDectin-2-/-マウスの大腿骨、脛骨より骨髄細胞を採取し、5×105細胞を500μlの培地(M-CSF(ペプロテック)10ng/ml、10%FCS、50μg/mlペニシリン、50U/mlストレプトマイシンを含むαMEM(Gibco))に懸濁し、24ウェルプレートで2日間培養した。その後、10ng/mlのM-CSF、100ng/mlのRANKL(ペプロテック)、10%FCS、50μg/mlペニシリン、50U/mlストレプトマイシンを含むαMEMで3日間培養した。培地をのぞき、10%中性緩衝ホルマリン(ナカライ)を加え、2分間放置後、除去した。その後、50%アセトン(ナカライ)、50%エタノール(ナカライ)を加え1分間放置後除去した。TRAP染色液を加え、20分間放置した。その後水洗、乾燥し、顕微鏡で観察、計数した。TRAP染色液は45mM酢酸(ナカライ)、50mM酒石酸(ナカライ)、0.01%ナフトール AS-MX フォスファターゼ(ナカライ)、0.06%Fast Red Violet LB Salt LB(ナカライ)。
破骨細胞分化
野生型C57BL/6Jマウス及びC57BL/6J背景のDectin-2-/-マウスの大腿骨、脛骨より骨髄細胞を採取し、5×105細胞を500μlの培地(M-CSF(ペプロテック)10ng/ml、10%FCS、50μg/mlペニシリン、50U/mlストレプトマイシンを含むαMEM(Gibco))に懸濁し、24ウェルプレートで2日間培養した。その後、10ng/mlのM-CSF、100ng/mlのRANKL(ペプロテック)、10%FCS、50μg/mlペニシリン、50U/mlストレプトマイシンを含むαMEMで3日間培養した。培地をのぞき、10%中性緩衝ホルマリン(ナカライ)を加え、2分間放置後、除去した。その後、50%アセトン(ナカライ)、50%エタノール(ナカライ)を加え1分間放置後除去した。TRAP染色液を加え、20分間放置した。その後水洗、乾燥し、顕微鏡で観察、計数した。TRAP染色液は45mM酢酸(ナカライ)、50mM酒石酸(ナカライ)、0.01%ナフトール AS-MX フォスファターゼ(ナカライ)、0.06%Fast Red Violet LB Salt LB(ナカライ)。
野生型マウスと比較し、Dectin-2-/-マウスでは破骨細胞形成が抑制されていた。このことから、デクチン-2は破骨細胞の分化・成熟に重要な役割を果たしていることが示された(図8、図9)。
実施例6
可移植性腫瘍細胞株としてBALB/c由来繊維肉腫細胞であるMethA(理研BRCより入手)を用いた。MethAは、10%ウシ胎児血清及び100μg/mlペニシリン、100U/mlストレプトマイシンを含むRPMI1640培地(SIGMA)を用い、5%CO2、37℃の炭酸ガス恒温槽内で培養を行なった。
実験用マウスとしては、MethAと同系であるBALB/cA背景Dectin-2-/-あるいは野生型BALB/cAマウス(日本クレアより導入)の10週齢のオスを用いた。
それぞれのマウスの右後肢足蹠皮下にPBSに懸濁したMethA1.5×106細胞を接種し、対照として左後肢足蹠皮下にはPBSのみを注射した。MethA接種右足蹠の測定値から対照左足蹠の測定値を引いたものをMethAによる腫瘤の大きさとした。
可移植性腫瘍細胞株としてBALB/c由来繊維肉腫細胞であるMethA(理研BRCより入手)を用いた。MethAは、10%ウシ胎児血清及び100μg/mlペニシリン、100U/mlストレプトマイシンを含むRPMI1640培地(SIGMA)を用い、5%CO2、37℃の炭酸ガス恒温槽内で培養を行なった。
実験用マウスとしては、MethAと同系であるBALB/cA背景Dectin-2-/-あるいは野生型BALB/cAマウス(日本クレアより導入)の10週齢のオスを用いた。
それぞれのマウスの右後肢足蹠皮下にPBSに懸濁したMethA1.5×106細胞を接種し、対照として左後肢足蹠皮下にはPBSのみを注射した。MethA接種右足蹠の測定値から対照左足蹠の測定値を引いたものをMethAによる腫瘤の大きさとした。
MethAを接種した4日後より2日毎に足蹠の厚さを測定した。Dectin-2-/-マウスでは野生型BALB/cAマウスに比較してMethAの腫瘤形成が有意に促進されて果たしていることが示された(図10、図11)。
Claims (11)
- ゲノム中のデクチン-2遺伝子の機能を欠損した非ヒト動物。
- 非ヒト動物がマウスである請求項1記載の非ヒト動物。
- 請求項1又は2記載の非ヒト動物又は当該動物由来細胞よりなる、樹状細胞によるサイトカイン産生モデル動物又はモデル細胞。
- サイトカインが、TNF、IL-12、IL-10、IL-6、IL-23、IL-1β又はIL-17Fである請求項3記載のモデル動物又はモデル細胞。
- 請求項1又は2記載の非ヒト動物又は当該動物由来細胞よりなる、樹状細胞より産生されたサイトカインのTh1又はTh17分化モデル。
- 請求項1又は2記載の非ヒト動物又は当該動物由来細胞よりなる、真菌感染症モデル動物又はモデル細胞。
- 請求項1又は2記載の非ヒト動物又は当該動物由来細胞よりなる、癌モデル動物又はモデル細胞。
- 請求項1又は2記載の非ヒト動物又は当該動物由来細胞よりなる、骨疾患モデル動物又はモデル細胞。
- 請求項1又は2記載の非ヒト動物又は当該動物由来細胞よりなる、関節炎モデル動物又はモデル細胞。
- 被験物質をデクチン-2を発現している細胞に作用させる工程と、デクチン-2の活性を測定する工程とを含む、関節炎治療薬又は骨疾患治療薬のスクリーニング法。
- 被験物質をデクチン-2を発現している細胞に作用させる工程と、デクチン-2の活性を測定する工程とを含む、Th1又はTh17分化調節剤のスクリーニング法。
