Abstract
Cette invention concerne un moyen et un procédé permettant d'augmenter l'efficacité de l'expression de l'hormone somatotrope dans une cellule transformée d'Escherichia coli et aussi d'augmenter la vitesse de collecte d'une protéine active à partir d'un corps d'inclusion produit. L'invention concerne spécifiquement : un vecteur d'expression de l'hormone somatotrope comportant, intégré dedans, de l'ADN comprenant l'une quelconque des séquences nucléotidiques choisies dans le groupe constitué par la séquence nucléotidique présentée en SEQ ID NO:3 et une séquence nucléotidique ayant une substitution d'un nucléotide dans la séquence nucléotidique présentée en SEQ ID NO:3 telle que le pourcentage de GC dans la séquence nucléotidique contenant la substitution est compris dans l'intervalle de 48 à 52 % et qu'aucune modification n'est causée par une séquence d'acides aminés codée par la séquence nucléotidique comportant la substitution et qui, par ailleurs a un promoteur PL et un promoteur PR et un gène répresseur sensible à la température ; un procédé de production de l'hormone somatotrope en utilisant une cellule d'Escherichia coli qui a été transformée avec le vecteur d'expression ; et un procédé de production d'une forme active de l'hormone somatotrope à partir de l'hormone somatotrope produite sous forme d'un corps d'inclusion.
