Abstract

L'invention porte sur un procédé pour obtenir le Lipide monophosphorylé A à partir de Bordetella pertussis (BpMPLA), comme sous-produit de la production du vaccin cellulaire anticoquelucheux, en vue d'une utilisation dans des applications immunostimulantes telles qu'un adjuvant de vaccin, et, plus particulièrement, sur un procédé de purification de BpMPLA à partir du traitement d'un extrait alcoolique, d'un surnageant ou d'un filtrat habituellement rejeté provenant de la masse finale du vaccin contre Bordetella pertussis (sous-produit de la fabrication du vaccin principal). Ce procédé de purification de BpMPLA comprend le traitement d'un extrait alcoolique habituellement rejeté et provenant de la masse finale du vaccin contre Bordetella pertussis, les opérations consistant à soumettre cette masse finale à une colonne d'affinité et à soumettre la fraction éluée par détergent à une hydrolyse acide, suivie d'une neutralisation, d'une lyophilisation et d'une remise en suspension avec de l'eau pour injection plus un détergent doux. Le nouveau BpMPLA est utilisé dans des applications immunostimulantes, plus particulièrement, en tant qu'adjuvant de vaccin, et, de façon encore plus particulière, en tant qu'adjuvant pour le vaccin contre la grippe, avec des résultats prometteurs.
