Abstract

La présente invention concerne la production de substances bioactives et peut être utilisée dans des processus de production biotechnologiques, en médecine et dans l'industrie pharmaceutique, en particulier pour produire des préparations médicinales et diagnostiques à partir de venin de serpent. Le procédé selon l'invention est caractérisé en ce qu'il consiste : à réaliser une chromatographie d'affinité sur une colonne contenant un support Blue Sépharose FF, à diluer une fraction de matériau non conjugué au support Blue Sépharose FF et à l'appliquer sur une colonne remplie d'un échangeur d'anions de DEAE Sépharose FF, puis à laver le sorbant pour éliminer le matériau non conjugué, à faire passer un tampon de Tris HCl contenant du NaCl présentant un pH de 8,2 à travers la colonne, à éluer la phospholipase A2 du sorbant au moyen d'un gradient discontinu de force ionique de NaCl, et enfin à recueillir le pic protéique présentant une activité phospholipase, à le concentrer et à le faire sécher par lyophilisation.
