Abstract

Die Erfindung betrifft ein Proteinkonstrukt zur Herstellung löslicher Polypeptide. Um eine Möglichkeit bereitzustellen, mit welcher sich auf einfache und damit kostengünstige Weise verschiedenste Polypeptide in löslicher Form herstellen lassen wird ein Proteinkonstrukt vorgeschlagen, umfassend ein Fusionsprotein mit einem ersten Aminosäuresequenzabschnitt und einem zweiten Aminosäuresequenzabschnitt, wobei der erste Aminosäure-sequenzabschnitt eine Sekundär-Amid Peptidbindungs spezifische cis/trans Isomerase (APIase) kodiert und der zweite Aminosäuresequenzabschnitt ein beliebiges erstes Polypeptid kodiert, oder umfassend ein Polypeptidassoziat, enthaltend ein mit einem ersten Tag versehenes erstes Polypeptidmolekül und ein mit einem zweiten Tag versehenes zweites Polypeptidmolekül, wobei das erste Polypeptidmolekül eine APIase ist und das zweite Polypeptidmolekül ein beliebiges zweites Polypeptid ist und wobei der erste Tag an dem zweiten Tag spezifisch bindet, vorzugsweise mit einer thermodynami sehen Affinitätskonstante von kleiner als 1 μM.
