Abstract

AGI strain of entomopathogenic fungus B . bassiana was isolated from cadaver of adult stage of red palm weevil Rhychophorus ferrugineus . The red palm weevil is destructive insect and it causes serious damage to date palms in Egypt . Recently , the weevil has spread widely and now is the most economically important pest of the date palm in Egypt . The biocontrol means by using entomopathogenic fungi is more needed to control such this insect .All stages of red palm weevil appeared highly susceptibility to AGI fungal strain . The fungal strain appeared highly pathogenicty , viability and sporulation compared with other 44 tested other strains of fungs B . bassiana . Genetic description of fungal strain was conducted . Field application of this fungal strain in large scale proved highly control to red palm weevil .

Description

Egyptian Strain Of Entomopathogenic Fungus Beauveria bassiana
Technical Field : Biological control of Agricultural insect pests by using the entomopathogenic fungi .
Background art : The date palm , is the most important fruit crop in Egypt as well as the Middle East. In the mid 1990 s1 a dreaded pest of palms , the red palm weevil Rhynchσphorus ferrugineus, was reported to cause serious damage to date palms in Egypt . Since , this time , the weevil has spread widely and now is the most economically important pest of the date palm in Egypt . Eggs of R . ferrugineus are laid into the trunk of the palm and the larval stage feed on within the trunk , which leads to the formation of tunnels inside the palm . Frequent and heavy attacks weaken and eventually kill the tree . The method currently employed to control red palm weevil are largely quantities of synthetic chemical insecticides , this lead to the pollution of soil and water course around areas with palm weevil infestation . However , other techniques of management such as sanitation , baits and traps of palm weevils have been used . Direct injection of insecticides into the trunk and intensive regular spraying of insecticides were employed to control this insed . The development of IPM for red palm weevil through the integration of biological control will reduce the using of insecticides . The prospects for the development of a biological control component for an integrated management strategy are good , given the evidence for the importance of natural enemies in controlling palm weevil population in natural environment . The fungus B . bassiana has been successfully used as biocontrol agent for the management of number of coleopteran insects . All red palm weevil stages showed highly susceptibility to fungal infection under laboratory tests . Disclosure of the invention :
The entomopathogenic fungus Beauveria bassiana strain ( AGl ) was isolated from insect cadaver of adult stage of red palm weevil Rhynchophorus ferrugineus in Ismailia Governorate during 2001 . Fungal strain proved highly pathogenicty to all stages of red palm weevil . The early instar larvae appeared high susceptibility to fungal infection follwed by adult stage . The bioassay procedures showed that the LC50 values ( the lethal mortality for 50 % of treaded insects ) of fungal spores concentration for 3rd larval instar , 9th larval instar and adult stage were 4.3 x 106 , 1.7 x 107 and 8.I x IO6 spores / ml respectvily . However , the LC50 ( lethal time mortality for 50 % of treated insects ) was 2.7 days for adult stage . Among 44 strains of fungus B . bassiana isolated from different regions in Egypt , the strains AGl proved highly pathogencity to red palm weevil than other strains . This strain appeared higher viability ( 99 % spore germination ) and highly sporulation ( 4.6 x 107 spores / ml ) compored with other tested fungal stains . Field application with fungus B . bassiana against red palm weevil were conducted through 2002 - 2005 . In the field using the fungal applied injection was an effective method for controlling red palm weevil in naturally infested palm trees . The success of the treatment was up 90 % . The periodical spray application of fungal spores on palm trees reduced red palm weevil population . Reduction of palm trees infestation following field application was noticed . Release of contaminated males of red palm weevil with fungal spores was carried out weekly in infested regions . This method proved significant reduction in the palm weevil population . These results suggests that fungus B . bassiana could be an effective and economically feasible control for the red palm weevil .
Genetic characterization :
Finger print of the fungus Beauveria basiana strain ( AGl ) was carried out as shown in fig ( 1 ) . The description of the fungal as following :

Claims 1- Egyptian strain of entomopathogenic fungus Beauveria basiana ( AGl ) was isolated from cadaver of red palm weevil . 2- Genetic characterization according to claim 1 , wherein said was carried out to describe the fungal strain . 3- The pathogenicity according to claim 1 , wherein said was highly against red palm weevil stages . 4- The spores viability according to claim 1 , wherein said was high reached to 99 % . 5- Fungal sporulation according to claim 1 , wherein said was high in insect cadavers . 6- Mass production of AGl according to claim 1 , wherein said can be produced in large scales . 7- The AGl strain of fungus B. hassiana used mainly to control the red palm weevil. 8- The AGl strain of fungus B . bassian according to claim 1 , wherein said can be used to control other pests .
