[0002] 遺伝子導入法には人体に直接遺伝子を導入する方法 (in vivo法)と、一度体外に 取り出した細胞に遺伝子を導入して、人体に戻す方法 (ex vivo法)がある。
[0003] 遺伝子導入法は!ヽままで、物理的方法である微小注入法、電気穿孔法、遺伝子銃 法、化学的方法であるリボソーム法、膜融合蛋白質一リボソーム法、リポフエクシヨン 法、カチオン性ペプチド法、リン酸カルシウム共沈法、 DEAEデキストラン法、その他、 エンドサイト一シス法、ウィルスを使う方法など数多く開発され、応用されている。
[0004] これらの方法はそれぞれ長所と短所があり、 目的により方法を選択する必要がある 。物理的方法は、安全性が高ぐプラスミド DNAのサイズの制限がない。また、 in vivo の場合には、血液中の酵素の分解作用、および尿からの排泄により、 Naked核酸が 生体内に残る時間は短いため、 Naked核酸を細胞に導入する効率が非常に低い。
[0005] 化学的方法ではウィルスを使う方法より安全性が高ぐプラスミド DNAのサイズに制 限がなぐ物理的方法より導入遺伝子の発現効率が高いが、導入できる細胞は分裂 細胞に限られ、また、遺伝子発現は一過性である。
[0006] ウィルスを使う方法は上記の方法に比べ安全性が劣るが、導入遺伝子の発現効率 が最も高く非常に有用である。特に、レトロウイルス科のレンチウィルスベクターは分 裂細胞にも非分裂細胞にも導入ができ、遺伝子発現の持続性を持っているため、最 も遺伝子治療に応用されている。
[0007] 遺伝子を細胞内に導入し、効率よく発現させるためには細胞質内への導入、核内 移行、核内での遺伝子の維持および高効率の発現、これら 4つの段階を乗り越えな ければならないため、遺伝子の核内運送システムの開発はますます重要になってく る。
[0008] 例えば、核移行シグナル(nuclear localization signals)による核蛋白質の核内運送 システムを利用した人工ウィルスベクターが開発された。 Dzauおよびカネダらは HVJ( hemagglutinating virus of Japan, Sendai virus)リポソ ~~ムへクタ ~~を 発し 7こ。共辱人 する核蛋白質 HMG-1 (high mobility group 1 protein)を用いている(特許文献 1参照 ) o HMG-1は核内限定シグナルを持っておらず、 DNAと結合し凝縮させ核移行を補 助していると考えられる。しかし、 HMG-1の精製はコストが高ぐ労力が必要となる。

[0014] WPREはウッドチャック肝炎ウィルスゲノムに含まれる転写後調制因子で、 RNAを安 定にし、発現を高める作用があり、レトロウイルスベクターでは 293T細胞での遺伝子 発現効率がコントロールと比較し 5〜8倍となった (特許文献 3参照)。 WPREはレトロゥ ィルスベクターだけではなぐ AAV(adeno- associated virus)ベクター(非特許文献 1 参照)、アデノウイルスベクター (非特許文献 2参照)にも高 ヽ遺伝子発現効率が確認 された。
[0015] cPPT · CTS、 WPRE両者を搭載したレンチウィルスベクターはヒト幹細胞(非特許文 献 3参照)、ヒト造血幹細胞 (非特許文献 4参照)にも高 ヽ遺伝子導入、発現効率が報 告された。逆に、 Philippe- Emmanuel Mangeotが指摘したように、 WPRE力ヒト神経榭 状細胞 (Dendritic Cell)に対し、不利な作用を及ぼした (非特許文献 5参照)。 Priscill a Y. Yamも指摘したように、 WPREはヒトリンパ球 T細胞に対し、導入効率を少し高め、 GFPの発現効率を 2〜3倍上昇させた (非特許文献 6参照)。 cPPT'CTS、 WPRE両者 を搭載した場合は WPRE単独より遺伝子導入発現効率力 〜10倍上がる。ヒトリンパ 球 T細胞に対し、 cPPT'CTSは WPREより高い遺伝子導入発現効率を持つことが証明 された。

発明の開示
発明が解決しょうとする課題 [0016] 上記技術によりベクターにおける遺伝子導入効率および遺伝子発現レベルは向上 したが、遺伝子治療等を行う上で、まだ十分なものではなく更なる向上が望まれる。 そこで本発明の目的は、ベクター中の遺伝子発現レベルを高め、さらに、細胞への 遺伝子導入効率を高めるェンノヽンサ一を提供することにある。 課題を解決するための手段
[0017] 本発明者は上記課題に鑑み、鋭意検討した結果、特定の配列をェンハンサ一とし て挿入することにより、上記課題を達成し得ることを見出し、本発明を完成するに至つ た。
[0018] 即ち、本発明のェンハンサ一は、ベクターに挿入されたェンハンサ一であって、動 物ゲノムに存在する cis-acting elementのうち少なくとも一部を有することを特徴とする ものである。動物ゲノムとしては、マウス、ヒトまたはラット等のゲノムを挙げることができ る。例えば、 cis-acting elementとして、マウスのゲノムの GenBank no:AE008683の 214 722bp〜216731bp、 GenBank no:AF259074の 50041bp〜2050bpである配列番号 1、ヒ トのゲノムの GenBank no:HUAE000658の 209041bp〜211150bp、 GenBank no:NG001 332.1の 209041bp〜211150bpである配列番号 2またはラットのゲノムの GenBank no:N W_042967の 2162172bp〜2164672bpである配列番号 3で示される塩基配列を有する DNAが挙げられる。

発明の効果

[0022] 本発明の好適な実施形態について、詳細に説明する。本発明者は動物のゲノムに 存在する cis-acting elementをェンハンサ一として選択した。ェンハンサ一というのは 遺伝子の転写開始を促進し、転写開始点からかなり離れた上流または下流に位置し て、逆向きでも働く塩基配列と定義される。本発明のェンノ、ンサ一はベクター中の遺 伝子発現レベルを高めるための動物のゲノムに存在する cis-acting elementである。 このェンハンサ一は、例えば、マウスの嗅覚受容体遺伝子 MOR28(mouse olfactory r eceptor 28)の上流領域 75kb、ヒトの嗅覚受容体遺伝子 HOR28の上流領域 32kbに 存在している。マウスの嗅覚受容体 (OR)遺伝子は約 1000種類存在することが知ら れており、嗅神経細胞はこのうち 1種類のみを発現する。 YAC(yeast artificial chromo somes)に MOR28を含む数 lOOkb DNAを組み込み、マウスの受精卵前核へ微注射し 、 MOR28遺伝子を発現するトランスジヱニックマウスを作成し、このトランスジヱニック マウスを解析した結果、このェンハンサ一は MOR28遺伝子の発現を活性ィ匕すること が明らかになつている(Shou Serizawa et al. 2003 Science 2088-2094)。し力し、この ェンノヽンサ一を GFP遺伝子の転写を促進する CMVプロモーターの前に組み込んだ プラスミド DNAをリポフエクシヨン法により培養細胞 HEK293に導入した場合、コント口 ールと比較し GFP遺伝子発現の増加は見られな力つた。このェンハンサ一の遺伝子 発現を高めるメカニズムを解明するために、本発明者は HIV-1由来のレンチウィルス ベクターシステムを用いて研究を行った。
[0023] レンチウィルスベクターシステムの基本構造として、 4つの独立した発現プラスミドを 使用した。ベクター粒子形成に必須な蛋白質を供給するパッケージングベクター、ベ クタ一にパッケージングされる mRNAを供給し、標的細胞に対して目的の遺伝子を運 搬、導入するジーントランスファーベクター、シユードタイプベクター粒子を形成する ための外殻蛋白質供給ベクターの VSV-G供給ベクターである。ノ ッケージングベクタ 一は gag, polを含むプラスミド、 revを発現するプラスミド、この 2つの発現プラスミドから なる。
[0024] ジーントランスファーベクターは 3' LTRの配列を部分的に削除している。これは Self Inactivating Vector(SIN vector)と呼ばれ、標的細胞の染色体に侵入したレンチウイ ルスのプロウィルスは 3 ' LTRの僅かに残っている U3部分を 5 '端に結合した形となる。 それにより、標的細胞内では 5' LTRのプロモーター活性が無くなるため、全長の RNA や宿主遺伝子の転写が起こらない。そして、内部プロモーターからの目的遺伝子の 転写のみが行われることになり、安全性が向上する。
[0025] VSV- G供給ベクターは水庖性口内炎ウィルス (VSV: Vesicular stomatitis virus)の 表面糖蛋白質である VSV-Gを発現する。 VSV- Gのレセプターであるりん脂質、ホスフ ァチジルセリンがほとんどの動物細胞に存在するため、 VSV-Gによって、作られたレ ンチウィルスベクターは感染指向性が広くなり、ほとんどの動物細胞に感染できる。
[0026] パッケージングベクター、ジーントランスファーベクター、 VSV- G供給ベクターを細 胞にコトランスフエクシヨンすると、ジーントランスファーベクターの 5, LTRのプロモータ 一活性により mRNAが転写、ノ ッケージングベクターより供給されるウィルス蛋白によ りパッケージングシグナル(Packaging Signal)配列が認識され、ベクター粒子内に RN Aがパッケージングされる。次に、 VSV-Gによって、シユードタイプ化され、ベクター粒 子が完成する。ベクターが標的細胞に感染すると、ノ ッケージングされていた RNAは 逆転写酵素により DNAとなり、両端の LTRとインテグラーゼの働きで宿主のゲノムにィ ンテグレーシヨンされる。そして、内部プロモーターの活性により目的遺伝子が発現 する。
[0027] ジーントランスファーベクターは両端の LTRの間に、内在性プロモーター CMVと、そ れに次ぐ GFP遺伝子を含み、それをコントロールとする。 cPPT (cis- acting central pol ypunne tract およびし rS、cis— acting termination region)【こ り、 HIVの;!^ fe "過程 でトリプレックス DNA構造を形成し、 DNAの核内運送に関与している(米国特許第 66 82907号明細書)。本発明者はコントロールベクターの CMVプロモーター前に cPPT および CTS配列を^ aみ込んだジーントランスファーベクターを構築した。さらに、その cPPT及び CTS配列の前にマウス由来の 2kbェンハンサーを組み込んだジーントラン スファーベクター(以下、「ェンハンサージーントランスファーベクター」とも 、う)を構 築した。
[0028] 本発明者はこの 3つのジーントランスファーベクターを、それぞれレンチウィルスべ クタ一システムを利用し、リポフエクシヨン法により培養細胞 HEK293に導入し、シユー ドタイプ化されたレンチウィルスを作製し、 HIV- lp24 Antigen ELISA kit (ZepoMetrix Corporation製)により、それぞれのウィルスの gag蛋白質の量を測った。その結果、 2 kbェンハンサー、 cPPTおよび CTSを含むウィルスは、コントロールウィルスより 6倍以 上、 cPPTおよび CTSを含むウィルスよりも 3倍以上高い。おそらぐこの 2kbェンハン サ一は単独では、 cPPTおよび CTSを含むウィルスより遺伝子を細胞内に 2倍以上、コ ントロールより 3倍以上導入し発現させると考えられる。
[0029] 前述したように、遺伝子を細胞内に導入し効率よく発現させるためには細胞質内へ の導入、核内移行、核内での遺伝子の維持及び高効率の発現、これら 4つの段階を 乗り越えなければならない。この 2kbェンノ、ンサ一の高導入効率、高効率の発現を説 明するために、細胞核、核膜孔構造及びレンチウィルス DNA複合体の核内移行の複 雑なメカニズムを分析する必要がある。
[0030] 細胞核は最も大きい細胞小器官であり、細胞内の多くの情報が蛋白質や受容体結 合ホルモンとして核に入り、遺伝子の発現を制御している。核に必要なリボソーム蛋 白質、ヒストン、 DNAポリメラーゼ、 RNAポリメラーゼなども核膜孔カゝら入る。一方、リボ ソーム前駆体、 mRNA、 tRNAをはじめとしてその遺伝情報と翻訳の要素が細胞質内 に送り出される。そのために核には巨大分子を選択的に通す核膜孔がある。核膜孔 は内径力 ¾Onm、外径が 120nmのリングとそれに 8個のスポークがっき、中央に中心 顆粒があるのでこの全体を指して、核膜孔複合体という。核膜の裏にはラミナという構 造物が付着しており、ラミン A、 B、 Cという中間径フィラメントと相同性のある蛋白質を 成分としている。 1個の核当たり約 3000個の核膜孔がある。分子量 60kDaまでの分 子は拡散によって核膜孔を非選択的に通過する。しかし、これよりも大きい分子は特 定のシグナル配列を持ったものだけが通過することができる。
[0031] レンチウィルス DNA複合体の非分裂細胞核内移行システムの定説は以下のように なっている。逆転写された二本鎖 DNAは非分裂細胞質内に、 gag蛋白質に含まれるリ ン酸ィ匕された pl7(MA)蛋白、 Vpr蛋白及びインテグレース蛋白、逆転写酵素などとと もにプレインテグレーシヨン複合体(preintegration complex, PIC)を形成し核内に運 ばれる。その中に、リン酸ィ匕された pl7(MA)蛋白が強い核移行シグナルを持つことに よって、核輸送蛋白 Importinと結合し、 PIC複合体が核膜孔を通ることを促進する。 Vp r蛋白及びインテグレース蛋白も似たような作用を果たして 、る。
[0032] 目 ij し 7こよつに cPPT (cis— acting central polypunne tract)およびし rS (cis— acting te rmination region )はトリプレックス DNA構造を形成し、レンチウィルス DNA複合体の 細胞核内移行に機能している。一方、レンチウィルの Rev蛋白はその N端側に核移行 シグナルを有し、核内に移行し、ウィルス RNAに存在する RRE領域に結合し、ウイ ルス RNAのスプライシングを抑制する。その Rev蛋白 C端側に NES(nuclear export sig nal)と呼ばれる核外輸送シグナルを持ち、輸出蛋白 (exportin-l)、 Ran_GTPと結合し、 ウィルス RNAを核内力 細胞質内へ運ばれる。細胞質内に Ran- GTPase-activating p rotein(GAP)は Ran-GTPを分解することによって、 Rev蛋白は Rev-RRE複合体から離 れて、また繰り返し、核内に移行し、ウィルス RNAを核内から細胞質内へ運ぶ。
[0033] コントロールジーントランスファーベクターを、レンチウィルスベクターシステムを利 用し、リポフエクシヨン法により培養細胞 HEK293に導入し、ジーントランスファーべク ターの 5, LTRのプロモーター活性により GFPの mRNAが転写される。パッケージング ベクターは gag, polを含むプラスミド、 revを発現するプラスミド、 VSV- G供給ベクターも それぞれのプロモーター活性により mRNAが転写される。分裂細胞 HEK293の細胞 質内に導入し 2日目に、それらの蛋白質が合成される。合成された Rev蛋白が核内に 移行し、ある程度の量になると、ノ ッケージングベクターの RRE領域に結合し、スプラ イシングが抑制され、 gag, polの mRNAが転写される。 Rev蛋白がジーントランスファー ベクターの RRE領域に結合すると、スプライシングが抑制され、 RRE領域下流の GFP 遺伝子の転写を阻害する。同時に、 Rev蛋白それぞれ gag, polの mRNA後の RRE領域 に、すでにできた GFPmRNA前の RRE領域に結合し、核外輸送シグナルと輸出蛋白 の結合により、 RNAが核内力 細胞質内へ運ばれる。細胞質内に、パッケージングべ クタ一より供給されるウィルス蛋白により Packaging Signal配列が認識され、ベクター粒 子内に RNAがパッケージングされる。次に、 VSV-Gによって、シユードタイプ化され、 ベクター粒子が完成する。
[0034] 2kbェンハンサー、 cPPTおよび CTSを含むベクター粒子はコントロールベクター粒 子より、 gag蛋白質の量が 6倍以上高いことの可能性としてはこのェンハンサ一は Rev 蛋白の RNA核外輸送を促進が挙げられる。 cPPTおよび CTSを含むベクター粒子は g ag蛋白質量がコントロールと比較して 2倍となった。この結果は米国特許第 668290 7号明細書の主張と一致している。つまり、 cPPT'CTSはトリプレックス DNA構造を形 成し、レンチウィルス DNA複合体の細胞核内移行に機能している。また、もう一つの 可能性として、このェンハンサ一は cPPT'CTSが機能しているレンチウィルス DNA複 合体(preintegration complex, PIC)の細胞核内移行に何らかの役割を果たしている ことが考えられる。遺伝子を細胞内に導入し、効率よく発現させるためには分裂細胞 HEK293に細胞周期の静止期でも、 DNA複合体 (PIC)の細胞核内移行システムが働 いていると考えられる。
[0035] 2kbェンハンサー、 cPPTおよび CTSを含まれているベクター粒子はコントロールべク ター粒子より、機能力価で約 9倍になった。単位ベクターあたりの遺伝子発現能を高 めたベクターは有効性の上昇だけでなぐ投与量の減少による副作用の軽減も期待 できることから、今後の応用が期待される。血液のリンパ球細胞に関わる遺伝子病は ADA欠損症、白血病、悪性リンパ腫、 HIV感染症などが知られている。リンパ球細胞 への治療用遺伝子の導入は現実的になっている。
[0036] cPPT'CTS/ジーントランスファーベクター、ェンハンサージーントランスファーべク ター由来の濃縮されたウィルスのリンパ球細胞への遺伝子導入効率を持つことを調 ベるために、ディプリーション(Depletion)法によるマウス Spleen mature B、 T細胞を 調製した。 cPPT'CTSZジーントランスファーベクター、ェンハンサージーントランスフ ァーベクター由来の濃縮されたウィルスを用いて、調製したマウス Spleen mature B細 胞を感染させ、フローサイトメーターによる解析を行った結果、ェンハンサージーント ランスファーベクター由来の濃縮されたウィルスは cPPT · CTS/ジーントランスファーべ クタ一ウィルスより、マウス Spleen mature B細胞への遺伝子導入効率を 2倍増加した
[0037] cPPT'CTS/ジーントランスファーベクター、ェンハンサージーントランスファーべク ター由来の濃縮されたウィルスを用いて、調製したマウス Spleen mature T細胞を感 染させ、フローサイトメーターによる解析を行った。その結果、ェンハンサージーントラ ンスファーベクター由来の濃縮されたウィルスは cPPT · CTS/ジーントランスファーべク ターウィルスより、マウス Spleen mature T細胞への遺伝子導入効率を若干増加した。 これよりェンハンサ一が cPPT'CTSより Spleen mature B、 T細胞への高い遺伝子導入 効率を持つことが確認された。
[0038] また、上記実験の感染条件は感染 3時間後、ウィルス液を排除し、 24時間培養で あった。 Spleen mature B、 T細胞がウィルスの感染により、傷害を受けて、感染時間 の経過に伴い、死んでいくことを配慮し、感染時間を短くした。適当な感染時間を検 討するために、本発明者は感染時間を 3時間、 24時間と設定し、ェンハンサージ一 ントランスファーベクター由来の濃縮されたウィルスを用いて、調製したマウス Spleen mature B細胞を感染させ、フローサイトメーターによる解析を行った。その結果、 MOI (multiplicity of infection) 1、 24時間感染の場合は、遺伝子導入効率が 42%に達成し 、 3時間感染の場合より 4倍高くなつた。ェンノヽンサ一と cPPT'CTSと組み合わせて、 MOKmultiplicity of infection) 1のウィルス量で、高い遺伝子導入効率が得られており 、高い MOIの感染はより高い遺伝子導入効率が得られることが予想され、本発明は 遺伝子治療の応用に関し、高く評価されると考えられる。
[0039] 本発明はェンハンサーである動物のゲノムに存在する cis-acting elementを含む組 換えウィルスベクターを用いて、動物細胞を感染し、遺伝子発現レベル及び導入効 率を高めるものである。ウィルスベクターに含まれるこのェンハンサ一はそのウィルス ベクターに組み込まれる単一の遺伝子の発現だけではなぐ 2つの遺伝子の同時的 な発現にも作用し、遺伝子の発現レベルを高めることが考えられる。なお、ウィルスべ クタ一に含まれるこのェンノヽンサ一を遺伝子の転写を開始するプロモーターの前に 置いても、遺伝子の 3 '側に置いても、遺伝子の発現レベルを高めることができる。

[0044] 人の組織の細胞、例えば、鼻腔または呼吸系統の気管支の粘膜上皮細胞、特に 嚢胞性繊維症 (cystic fibrosis, CF)を治療することが好ましい。嚢胞性繊維症とは、 白人種に極めて高頻度にみられる常染色体劣性遺伝子病で、欧米では出生児 250 0人当たり 1人の発症頻度を示し、気道上皮では気道管腔内に粘稠な痰が多く溜まり 、呼吸困難を起こす。このェンハンサー配列を含むパラミクソウィルスエンベロープシ ユードタイプレチウィルスベクターを作製し、鼻腔または呼吸系統の気管支の粘膜上 皮細胞での高い遺伝子発現レベルを得られることが考えられる。

ェンハンサー、 cPPT'CTSによるシユードタイプレンチウィルスベクターの調製
プラスミド DNA(p EYFP- Nl、 Clontech社製)を制限酵素 Hindlll, BamHIで切断し、 ブランティング(Bluntting)させ、セルフライゲーシヨン(self-ligation)により環状の Has mid DNAとした。その環状の Plasmid DNAの Asel siteに制限酵素 Hindlll, BamHIの 認識配列を含む LinkerAselを組み込んで、コントロールベクターを得た。
[0046] 2kbェンハンサー断片はマウスのゲノム DNAである GenBank no:AE008683の 21472 2bpから 216731bpまでの配列を元にして、 primer EcoRV F (5, -GATATCcagaatagtac ttcattatg)、 primer EcoRV R (5,— GATATCTTGATCATCATTTCTATCTT)を用いた PCR反応により 2kbェンハンサー断片を増幅させた。増幅させた 2kbェンハンサー断 片をコントロールベクターに組み込み、発現ベクターを得た。

[0053] レンチウィルス産生に用いたヒト胎児腎細胞由来細胞株 293FT細胞(Invitrogen社 製)を、 10%非動化ゥシ胎児血清、 500 μ g/ml neomycine dnvitrogen社製)を含む D- M EM (Invitrogen社)で培養した。ベクターのトランスフエクシヨンは LIPOFECTAMINE P LUS (Invitrogen社製)を用い、添付説明書に従!、行った。 [0054] 293FT細胞を直径 15cm dish (コラーゲンタイプ I コート製品、 IWAKI社製)へ 3 X 10 6個の細胞密度でまき、炭酸ガスインキュベータ一中(37°C、 5%炭酸ガス存在下)で 48 時間培養した。トランスフエクシヨンの 30分前に培養液を 10mlの OptiMEM (Invitrogen 社製)に培地交換し培養を続けた。トランスフエクシヨンに使用する DNA量はそれぞれ 20 μ gの 3つのジーントランスファーベクターと、 6 μ gのパッケージングベクター pLP 1 、 pLP2 (Invitrogen社製)とを使用し、それらに加えて 2 μ gの VSV- G供給ベクター pLP VSVGを使用した。 DNAを 3mlの OptiMEM (Invitrogen社製)に溶解させた後に 400 μ 1 の PLUS reagent (Invitrogen社製)をカ卩え、攪拌後 15分室温で静置した。 DNAと PLUS reagent (Invitrogen社製)との混合液に、 3mlの OptiMEMで希釈した 100 μ 1の LIPOF ECTAMINEを添加し、攪拌後さらに 15分室温で静置した。以上の方法により調製し た DNAと LIPOFECTAMINEとの複合体を含む溶液を直径 15cm dishで培養して!/、る 2 93FT細胞へ滴下して緩やかに攪拌した後に炭酸ガスインキュベータ一中(37°C、 5% 炭酸ガス存在下)で 3時間培養した。培養後 30mlの 10%非動化ゥシ胎児血清を含む D -MEM (Invitrogen社製)を添カ卩し、炭酸ガスインキュベータ一中(37°C、 5%炭酸ガス存 在下)で 48時間培養した後に培養上清を 0.45 μ mのポアサイズのフィルター(DISMIC -25CSフィルター、 ADVANTEC)で濾過してベクター溶液調製した。

ρ24蛋白質の発現量の測定は HIV- lp24 Antigen ELISA kit (ZepoMetrix Corporati on製)を用いて添付説明書に従い行った。まずは、 HIV-1 P24 Antigen Standardを作 り、次は 225 1のコントロールジーントランスファーベクター、 cPPT'CTS/ジーントラ ンスファーベクター、ェンハンサージーントランスファーベクター由来のウィルス液を 5 倍希釈し、順番に 5回希釈系列を作って、添付説明書に従って反応を行った。最後 に 450nmのフィルターを持つマイクロプレートリーダー(BIO- RAD社製)を用いて測定 し、解析ソフト ΜΡΜΠΙにより解析を行った。その結果を図 2に示す。これにより CPPT配 列の後、レンチウイノレスベクターの CMVプロモーターの前に、マウスの cis-acting ele ment配列を挿入することによって、遺伝子発現レベルが CPPT配列なしのベクターと 比較して 6倍、 CPPT配列ありのベクターと比較して 3倍以上上昇することが確認され た。

ベクターの濃縮
実験例 1にて回収された 3種類のウィルス液をそれぞれ 20000rpm (45 Ti rotor, Bee kman)、 4°Cにて、 2時間遠心し、ペレットを PBS (5% FCS, 2 μ g/ml polybrene)に溶解 し、使用まで- 80°Cに保存した。

ベクターのカ価沏 I定
実験例 2の HIV-lp24 Antigen ELISAの結果〖こより、 lOOOpgの p24蛋白質の量を持 つ、コントロールジーントランスファーベクター由来のウィルス液 140 1、 cPPT-CTS/ ジーントランスファーベクター由来のウィルス液 70 μ 1、ェンハンサージーントランスフ ァーベクター由来のウィルス液 23 1を 293FT細胞に感染させ、遺伝子導入される細 胞の数によって、力価を計算した。 293FT細胞を 1 X 106/wellで 6wellカルチャープ レートにまき、 48時間培養した。光学顕微鏡で 200倍の倍率で検鏡、視野内の遺伝 子導入細胞数を測定し、 3視野の平均を求め、視野の面積とプレートの面積よりもと めた係数 854.865をかけることにより力価の算出を行った。その結果は図 3に示す。図 3 (a)は lOOOpgあたりの力価(T. U. : Transducing Unit)を表し、コントロール、 cPPT' CTS、ェンハンサージーントランスファーベクターの力価は夫々、 6 X 104T.U./1000pg 、 1.4 X 105T.U./1000pg、 1.2 X 105T.U./1000pgであった。また、図 3 (b)は、 1mlあた りの力価(T. U./ml)に換算したものであり、ェンハンサージーントランスファーベクタ 一の力価は 5.14 X 106T.U./mlとなり、コントロール(4.3 X 105T.U./ml)の約 11倍、 cP PT'CTS (1.9 X 106T.U./ml)の約 2. 5倍となった。

ディプリーション(Depletion)法によるマウス Sdeen mature B細胞の調製
3週齢の BALB/c系のマウスより脾臓を採取し、フロスト付きスライドガラスのフロスト 部分で脾臓を挟み、 5%FCSを含む RPMI1640中にスライドガラスを摩擦させることによ り、脾臓細胞浮遊液を調製した。得られた細胞浮遊液をスピヅッ遠心管 (Falcon)に 回収し、 1500 rpm, 5分間遠心して、細胞のぺレートを得た。次に、赤血球溶血用緩 衝液 (0.83%塩ィ匕アンモ-ゥムを含む)を細胞のぺレートに添カ卩してよく攪拌し、氷に 1分間置き、 10mlの 2%FCSを含む HBSS溶液(Hanks, balanced salt solution, Sigma社 製)をカ卩えて 2回遠心洗浄してから、 5mlの最終濃度 20 μ g/mlの anti- Fc y RII/II (2.4 G2;PharMingen,San Diego,CA)をカ卩えて氷上に 15分間置いた。この操作により低親 和性 Fc受容体への抗体の非特異的結合を阻害する。細胞浮遊液を 2 X 107個/ mlに 調整した。
[0059] ピオチン標識抗体混合物(Miltenyi Biotec社製)に含まれる antト CD90、 antト CD43 、 anti- CD5、 anti- CD3、 anti- CDllb(Mac- 1)、 anti- Gr- 1、 anti- Ter 119を最適濃度に 希釈し、調整された細胞浮遊液に添加し、氷上で 30分間反応させた。 HBSS溶液で 細胞浮遊液を遠心洗浄して余分な抗体を除き、メッシュを通す。 100 1の標識ストレ プトアビジン microbeads (Miltenyi Biotec社製、 Gladbach、 Germany)を細胞に添カロし 、氷上で 15分間反応させた。また、 HBSS溶液で細胞浮遊液を遠心洗浄して 500 1 浮遊液とし、 MACS clumn (Miltenyi Biotec社製)により分離し、 15mlの細胞とした。
[0060] 得られた 15 mlの細胞にマウス Spleen mature B細胞がどの割合で存在して!/、るかを 調べるために、フローサイタメーター FACS Calibur (Becton Dickinson, Mountain Vie w, CA)を用い、以下の 6つの細胞検体を使用した。 1は細胞のみであり、抗体を添加 しないネガティブコントロールである。 2、 3、 4の細胞は 2.4G2抗体を添カ卩したコント口 ールである。 5の細胞はピオチン標識抗体混合物(Miltenyi Biotec社製)に含まれる a nti— CD90、 anti— CD43、 anti— CD5、 anti— CD3、 anti— CDllb(Mac— 1)、 anti— Gr— 1、 anti— Terll9を添カ卩し、 MACS clumnを掛ける前のコントロールである。 6の細胞は MACS cl umnを掛けた後のサンプルである。
[0061] 50 μ 1ずつ各 6つの細胞(1 X 106個/ ml)を 96 wellプレートに載せて、 HBSS溶液で細 胞を遠心洗浄した。 2、 3、 4の細胞にはそれぞれ 100 /z lの anti- CD45R/B220- FITC 標識(BD PharMingen) , anti- TCR jS -PE標識(BD PharMingen)、 anti- CD45R/B220 - APC標識(BD PharMingen)を添カ卩した。 5、 6の細胞は anti- CD45R/B220- FITC標 識(BD PharMingen) , anti- TCR jS -PE標識(BD PharMingen)、 anti- CD45R/B220- A PC- streptavidin標識(BD PharMingen)を含む抗体の混合物をそれぞれ 100 μ 1添カロ した。そのプレートを氷上で遮光し、 30分間反応させた。その後、 150 1の HBSS溶液 で細胞を 2回遠心洗浄し、 1mlの PI staining buffer (1 X HBSS溶液、 2% FCS, 1 ug/ml Propidiumiodide)に浮遊させ、メッシュを通し、 FACSチューブに載せてフローサイタメ 一ター FACS Calibur(Becton Dickinson)にて蛍光強度を測定した。 6の細胞はマウス Spleen mature B細胞の割合力 ^7.4%を示して、良い結果になった。

[0063] ビォチン標識抗体混合物(PharMingen)に含まれる anti- IgM、 anti- IgD、 anti- CD45 R/B220、 anti-CD llb(Mac-l), anti- Gr- 1、 anti- Ter 119を最適濃度に希釈し、調整さ れた細胞浮遊液に添加し、氷上で 30分間反応させた。 HBSS溶液で細胞浮遊液を遠 心洗浄して余分な抗体を除き、メッシュを通した。 100 1の標識ストレプトアビジン micr obeads (Miltenyi Biotec社製、 Gladbach、 Germany)を細胞に添カ卩し、氷上で 15分間 反応させた。また、 HBSS溶液で細胞浮遊液を遠心洗浄して 500 1浮遊液とし、 MAC S clumn (Miltenyi Biotec)により分離し、 10 mlの細胞とした。

マウス Sdeen mature B細胞への遣伝子導人及びフローサイトメーターによる解析 実験例 5で調製されたマウス Spleen mature B細胞を 10%FCS, 10"5M 2 メルカプト エタノール(2- ME: 2- mercaptoethanol)、 25mM HEPESおよびペニシリン一ストレプト マイシン(penicillin- streptomycin)を含む RPMI1640培地で交換し 2.5 X 105/wellで 6w ellカルチャープレートに撒いた。実験例 3にて濃縮された cPPT'CTS/ジーントランス ファーベクター由来のウィルス液、ェンハンサージーントランスファーベクター由来の ウィルス液をそれぞれ MOI(multiplicity of infection) 1, 0で調製した。つまり、 MOIが 1 の場合は、 2.5 X 105/wellの細胞に対して、 2.5 X 105TU/mlにて調製した。調製された ウィルス液をそれぞれマウス Spleen mature B細胞に 3時間感染させてから、 10%FCS, 10— 5M 2- ME、 25mM HEPESおよびペニシリン ストレプトマイシンを含む RPMI1640 培地で交換し、 24時間培養し、 PI staining buffer (1 X HBSS溶液、 2% FCS, 1 ug/ml Propidiumiodide)に浮遊させ、メッシュを通し、 FACSチューブに載せてフローサイタメ 一ター FACS Calibur (Becton Dickinson)にて YFP及び PEの 2蛍光により蛍光強度を 測定し、解析した。その結果を図 4に示す。ェンノヽンサ一ジーントランスファーベクタ 一(ΜΟΙ = 1)の場合は Spleen mature B細胞での EYFP陽性発現細胞割合は 12%で 、 cPPT'CTS (6%)の 2倍になっている。これにより本発明のェンハンサ一が cPPT'C TSより Spleen mature B細胞への高い遺伝子導入効率を持つことが証明された。

マウス Sdeen mature T細胞への遣伝子導入及びフローサイトメーターによる解析
実験例 6にて調製されたマウス Spleen mature T細胞を 10%FCS、 10"5M 2—メルカプ トエタノール(2- ME: 2- mercaptoethanol) , 25mM HEPESおよびペニシリン一ストレプ トマイシン(penicillin- streptomycin)を含む RPMI1640培地で交換し 2.5 X 105/wellで 6 wellカルチャープレートに撒いた。実験例 3で濃縮された cPPT'CTS/ジーントランス ファーベクター由来のウィルス液、ェンハンサージーントランスファーベクター由来の ウィルス液をそれぞれ MOI(multiplicity of infection) 1, 0で調製した。つまり、 MOI(mul tiplicity of infection) 1の場合は、 2.5 X 105/wellの細胞に対して、 2.5 X 105TU/mlにて 調製した。調製されたウィルス液をそれぞれマウス Spleen mature T細胞を 3時間感染 させてから、 10%FCS、 10— 5M 2- ME, 25mM HEPESおよびペニシリン—ストレプトマイ シンを含む RPMI 1640培地で交換し、 24時間培養し、 PI staining buffer ( 1 X HBSS溶 液、 2% FCS, 1 μ g/ml Propidiumiodide)に浮遊させ、メッシュを通し、 FACSチューブ に載せてフローサイタメ一ター FACS Calibur(Becton Dickinson)にて YFP及び PEの 2 蛍光により蛍光強度を測定し、解析した。結果を図 5に示す。ェンハンサージーントラ ンスファーベクター(ΜΟΙ = 1)の場合は Spleen mature T細胞での EYFP陽性発現細 胞割合は 12%で、 cPPT'CTSの場合は 9.7%であった。これにより本発明のェンハン サ一が cPPT'CTSより Spleen mature T細胞への高い遺伝子導入効率を持つことが証 明された。

3時間及び 24時間感染の条件でマウス Sdeen mature B細胞への遣伝子導入および フローサイトメーターによる解析
実験例 5で調製されたマウス Spleen mature B細胞を 10%FCS、 10"5M 2—メルカプト エタノール(2- ME: 2- mercaptoethanol)、 25mM HEPESおよびペニシリン一ストレプト マイシン(penicillin- streptomycin)を含む RPMI1640培地で交換し 2.5 X 105/wellで 6w ellカルチャープレートに撒 、た。実験例 3で濃縮されたェンハンサージーントランスフ ァーベクター由来のウィルス液をそれぞれ MOI(multiplicity of infection) 1, 0.1, 0で 調製した。つまり、 MOKmultiplicity of infection) 1の場合は、 2.5 X 105/wellの細胞に 対して、 2.5 X 105TU/mlにて調製した。調製されたウィルス液をそれぞれマウス Spleen mature B細胞を 3時間、 24時間感染させてから、 10% FCS, 10"5M 2- ME、 25 mM H EPES及びペニシリン—ストレプトマイシンを含む RPMI1640培地で交換し、 PI staining buffer (1 X HBSS溶液、 2% FCS, 1 μ g/ml Propidiumiodide)に浮遊させ、メッシュを通 し、 FACSチューブに載せてフローサイタメ一ター FACS Calibur(Becton Dickinson) にて YFP及び PEの 2蛍光により蛍光強度を測定し、解析した。その結果を図 6に示す 。ェンハンサージーントランスファーベクター(感染 3時間)の場合は Spleen mature B 細胞での EYFP陽性発現細胞割合は 10.4% (ΜΟΙ = 1)、 1.1% (MOI = 0.1)となった。 ェンハンサージーントランスファーベクター(感染 24時間)の場合は Spleen mature B 細胞での EYFP陽性発現細胞割合は 42.2% (ΜΟΙ = 1)、 11.7% (MOI = 0.1)になった。 感染時間が 3時間から 24時間変わると、ェンハンサージーントランスファーベクター は Spleen mature B細胞での EYFP陽性発現細胞割合は 4倍(ΜΟΙ = 1)、 10倍(MOI = 0.1)増加した。これにより、ェンハンサ一による Spleen mature B細胞での高発現レ ベルが感染時間延長により促進されていることが分かる。
本発明者はこのェンハンサ一と WPREとの比較的な実験を行って!/ヽな 、が、厚生労 働科学研究費補助金 (エイズ対策研究事業)総合研究報告書にて、 SIVベクターの 開発と血友病 A遺伝子治療への応用に関し、株式会社ディナベック研究所により以 下の報告がなされて ヽる。ベクター自身の'性能を高めるためにレンチウイノレスべクタ 一の骨格配列に細胞への導入効率を上昇させる目的で SIVゲノムに含まれる cPPT配 列、および導入遺伝子の発現効率を高める WPRE配列を挿入し、緑色蛍光蛋白質( GFP)遺伝子をマーカーに用い、 293T細胞に遺伝子導入実験を行った結果、 cPPT 配列の搭載により遺伝子導入効率が、 WPRE配列の搭載により遺伝子発現効率が上 昇したことが明らかになった。さらに両者を同時搭載することにより従来の SIVベクター に比べて機能力価で約 10倍ベクターの生産性を上昇させた。 本発明の実験例 4によると、ェンハンサー、 cPPT'CTSを含むベクター粒子はコント ロールベクター粒子より、機能力価で約 9倍になっている。即ち、 293T細胞の場合は このェンハンサ一と WPREとは相当なレベルになっている。ヒトリンパ球 T細胞の場合 はこのェンハンサ一は cPPT · CTS、 WPREより高 ヽ遺伝子導入効率を持つ進歩性で、 遺伝子治療における応用が力なり期待される。

Claims

請求の範囲
[I] ベクターに挿入されたェンハンサ一であって、動物ゲノムに存在する cis-acting ele mentのうち少なくとも一部を有することを特徴とするェンノヽンサ一。
[2] 前記動物ゲノムがマウス、ヒトまたはラットのゲノムである請求項 1記載のェンノヽンサ
[3] 前記 cis-acting elementが配列番号 1、配列番号 2または配列番号 3で示される塩基 配列を有する DNAである請求項 1または 2記載のェンノヽンサ一。
[4] 遺伝子の転写を開始するプロモーターの前、または遺伝子の 3 '側に挿入した請求 項 1〜3のうちいずれか一項記載のェンハンサー。
[5] 前記ベクターが組換えウィルスベクターである請求項 1〜4のうち 、ずれか一項記 載のェンハンサー。
[6] 前記組換えウィルスベクターがレトロウイルスベクターである請求項 5記載のェンノヽ ンサ一。
[7] 前記レトロウイルスベクターがレンチウィルスベクターである請求項 6記載のェンハ ンサ一。
[8] 前記レンチウィルスベクターがヒト免疫不全ウィルス (HIV)ベクターである請求項 7 記載のェンハンサー。
