Abstract

La presente invención se refiere a un procedimiento para aumentar el contenido endógeno de trans-resveratrol en vinos que consiste en la aplicación de tratamientos atóxicos cortos a la uva, preferentemente con nitrógeno seco, previamente al procedimiento de vinificación. El trans-resveratrol es una molécula producida de forma natural por la vid y otras plantas como medio de autodenfensa frente a las infecciones fungicas, diversos tipos de estrés, productos químicos, etc. La aplicación del tratamiento, caracterizado porque se somete la uva a cortos periodos de tiempo (inferiores a 24 horas) en atmósfera con muy bajo contenido en O2 , permite aumentar de forma importante su contenido endógeno de trans-resveratrol uva. Posteriormente, el trans-resveratrol pasa al vino durante el proceso de vinificación, produciendo de este modo un vino enriquecido en trans-resveratrol.

Description

TÍTULO
Método para aumentar el contenido endógeno de trans-resveratrol en el vino.
OBJETO DE LA INVENCIÓN. La presente invención se refiere a un procedimiento para aumentar el contenido endógeno de trans-resveratrol en vinos que consiste en la aplicación de tratamientos anóxicos cortos, preferiblemente con nitrógeno seco, a la uva previamente al procedimiento de vinificación. El trans-resveratrol es una molécula producida de forma natural por la vid y otras plantas como medio de autodenfensa frente a las infecciones fungicas, diversos tipos de estrés, productos químicos, etc. La aplicación del tratamiento, caracterizado porque se somete la uva a cortos periodos de tiempo (inferiores a 24 horas) en atmósfera con muy bajo contenido en O2, permite aumentar de forma importante su contenido endógeno de trans-resveratrol. Posteriormente, el trans-resveratrol pasa al vino durante el proceso de vinificación, produciendo de este modo un vino enriquecido en trans-resveratrol.
ANTECEDENTES.
En los últimos años, el vino ha sido ampliamente estudiado debido a los beneficios que presenta para la salud, principalmente en lo que respecta a la prevención de enfermedades cardiovasculares y la reducción del riesgo de carcinogénesis. Dichas propiedades se atribuyen principalmente a su elevado contenido en compuestos fenólicos y, muy especialmente, al trans-resveratrol (3,5,4'- trihidroxiestilbeno) que presenta múltiples propiedades beneficiosas para la salud (Burns, J., et al. J Agrie Food Chem. 50 (2002) 3337-3340; Mehta R.G., et al. Curr. Oncol Rep. 4 (2002) 478-86; Latruffe, N., et al. InL J. Mol Med. 10 (2002) 755-60).
El resveratrol es un derivado del estilbeno (3, 5, 4'-trihidroxiestilbeno) que presenta dos formas isómeras, cis y trans. Se ha descrito su actividad como fitoalexina producida por la vid como respuesta a las infecciones fungicas, especialmente a Ia Botrytis cinérea, (P. Jeandet et al. Am. J. Enol Vitic. 42 (1991) 41; M. Barlass et aL Am. J. Enol. Vitic. 38 (1987) 65). Se ha encontrado igualmente en uvas como consecuencia de infección o condiciones medioambientales extremas (LX. Creasy et al. J. Am. Soc. Hortic. Sel 113 (1988) 230; P. Langcake et al. Phytochemistry 16 (1977) 1193). El resveratrol se encuentra localizado en la piel de la uva principalmente (Orea, J.M. and González Ureña, A. Measuring and Improving the Natural Resistance in Fruits. En: W. Jongen (Ed.) Fruti and Vegetable Processing: Maximising Quality. Woodhead Publishing Ltd., Cambridge; págs. 233- 266 ) y su concentración depende de la climatología, así como de otros factores ambientales determinados por la variedad de la uva, las sucesivas infecciones sufridas por el viñedo, etc.
Durante el proceso de vinificación el trans-resveratrol es extraído de la piel de la uva y pasa a formar parte de la composición del vino. Se ha demostrado que las diferentes técnicas de vinificación influyen en el contenido final de trans-resveratrol en el vino, especialmente el tiempo de maceración con la piel dentro de la cuba (Mattivi et al. J. Agrie. Food Chem. 43 (1995) 1820-1823; Romero Pérez et al J. Ag. FoodChem. 47 (1999) 1533-1536).
En la presente invención se describe un procedimiento para aumentar el contenido endógeno de trans-resveratrol en el vino sometiendo a la uva a tratamientos anóxicos cortos, preferentemente con nitrógeno seco e inferiores a 24 horas, inmediatamente antes del prensado. En Ia solicitud de patente N0 200400427 (de los mismos inventores) se describe un método para aumentar el contenido de trans-resveratrol en la piel de la uva mediante tratamientos anóxicos cortos. En la presente invención se ha demostrado el aumento del contenido de trans-resveratrol en vinos fabricados a partir de uvas a las que se ha sometido a tratamiento anóxico previamente al proceso de vinificación.
DESCRIPCIÓN DE LA INVENCIÓN. Método para aumentar el contenido endógeno de trans-resveratrol en el vino.
Para llevar a cabo la presente invención se ha utilizado uva de vinificación de la variedad Tempranillo. Se han escogido diversos racimos de la vendimia que han sido seleccionados por presentar uvas con el hollejo más turgente con el fin de evitar que mosteen durante el pretratamiento con nitrógeno. Las uvas se cargaron en un depósito estanco (véase descripción de la Figura 1 a continuación) del que se evacúa el aire, introduciendo posteriormente un gas inerte a una presión ligeramente superior a la atmosférica. Las uvas deben permanecer en el citado depósito en atmósfera anóxica por un periodo no superior a 24 horas para no dañar la calidad de las uvas. En el modo de realización de la presente invención el tiempo del tratamiento anóxico ha sido de 24 horas.
Una vez terminado el tratamiento anóxico, se ventea el sistema y se extraen las uvas procediéndose al procesado habitual de la uva para su vinificación en tinto: despalillado, estrujado, encubado, fermentación alcohólica, remontados, descubado, prensado, fermentación maloláctica, trasiego y paso a barrica. El procesado de la uva debe realizarse lo más rápidamente posible después del tratamiento anóxico; preferiblemente, el procesado de la uva debe realizarse como máximo 18 horas después del tratamiento anóxico.
El efecto del tratamiento anóxico ha sido evaluado en primer lugar mediante el análisis del contenido polifenólico total (índice de Folin-Ciocalteau) durante la vinificación. Los resultados obtenidos permiten concluir que la extracción de los polifenoles durante la fermentación alcohólica es mucho más rápida en el mosto obtenido a partir de la uva tratada, alcanzándose valores próximos a 30 puntos en los inicios de la fermentación en comparación con los 10 puntos del mosto obtenido a partir de las uvas sin tratar.
El efecto del tratamiento anóxico en la calidad del vino ha sido estudiado mediante diversos análisis comparativos del vino vinificado a partir de uvas tratadas y del vino de control que ha sido elaborado con uvas de la misma vendimia y ha seguido el mismo proceso de vinificación, pero sin ningún tratamiento anóxico. El análisis de los espectros de absorción UV-visible de ambos vinos, permite comprobar la existencia de una mayor absorbancía en Ia región entre 200 y 400 nm, que es la zona característica de absorción de los diversos polifenoles presentes en el vino. Por otra parte se ha realizado el análisis del contenido de trans-resveratrol en el vino obtenido a partir de las uvas tratadas en comparación con los valores obtenidos en el vino de referencia. La técnica empleada para dicho análisis es la desarrollada para el análisis de trans-resveratrol mediante la combinación de la desorción láser con la ionización multifotónica resonante (generalmente denominada REMPI por sus iniciales en inglés: Resonant Multiphoton Ionization) y posterior detección por espectrometría de masas de tiempo de vuelo (generalmente denominada TOFMS por sus iniciales en inglés: Time-of-Flight Mass Spectrometry). (ES2137896;WO9957748; C. Montero et al. Appl. Phys. B (2000) 601-605; y J.M. Orea et al. Anal Chem. (2001) 5921-5929). El análisis del contenido de trans- resveratrol en ambos vinos, permite concluir (véase figura 2 más adelante) que un tratamiento anóxico de 24 horas permite aumentar dicho contenido de forma importante.
Por último, es de destacar, que no se han apreciado diferencias significativas en cuanto a las principales propiedades físico-químicas y organolépticas entre ambos vinos, teniendo en cuenta el análisis de los parámetros enológicos estudiados, principalmente: densidad, pH, acidez, graduación alcohólica, color, SO2, antocianos, etc.
BREVE DESCRIPCIÓN DE LAS FIGURAS
La Figura 1 muestra un esquema del sistema desarrollado para la realización de los tratamientos anóxicos objeto de la presente invención. La uva se deposita entera en un depósito siempre-lleno (a) cuya tapa hermética (b) dispone de una junta de goma que se puede hinchar y permite el cierre estanco del depósito con el exterior. La válvula de teflón convencional que presentan las tapas de este tipo es reemplazada por un racor, al cual se acopla una válvula de inflado (c). Este dispositivo, completamente estanco, permite provocar una sobrepresión en el interior del depósito que se mantiene con el tiempo. Tras cerrar el depósito con la tapa, comienza su llenado con nitrógeno por la válvula acoplada a la bombona de nitrógeno seco (e). La presión de nitrógeno introducido en el depósito se controla con el manómetro (d). Para extraer el aire acumulado entre los racimos, se realiza un purgado con nitrógeno introduciendo nitrógeno por Ia válvula superior y sacando el aire por el grifo de la parte inferior del depósito (f). Tras varios minutos purgando con nitrógeno, se cierra el grifo de la parte inferior y se comienza a producir una sobrepresión de nitrógeno en el interior del depósito, ajustándola a 1 bar. Esta sobrepresión se mantiene durante 24 horas. La Figura 2 muestra los resultados obtenidos en los análisis del contenido de trans-resveratrol en el vino de referencia obtenido a partir de la uva sin tratar (arriba) y en el vino elaborado según la presente invención (abajo). Los análisis han sido realizados mediante la combinación de la desorción por láser con REMPI y posterior detección por TOFMS. Como se puede observar, en el vino obtenido a partir de las uvas sometidas a anoxia durante 24 horas se produce un aumento en el contenido de trans-resveratrol; el incremento neto en el contenido de trans-resveratrol en este vino es más de 3 veces el contenido en trans-resveratrol en el vino de referencia, sin tratar.
MODO DE REALIZACIÓN DE LA INVENCIÓN
La presente invención se ilustra mediante el siguiente ejemplo operativo que no pretende ser en ningún caso limitante de su alcance, el cual viene definido exclusivamente por las reivindicaciones adjuntas. Las uva se introduce en un recipiente estanco, de 700 litros de capacidad, similar al mostrado esquemáticamente en la Figura 1, o cualquier otro recipiente de volumen adecuado a la cantidad de uva que se quiera tratar y provisto de las conexiones adecuadas para extracción del aire e introducción de nitrógeno.
Una vez introducidas las uvas en el recipiente, se extrae el aire de forma suave, mediante purgado con nitrógeno. A continuación, sin que entre aire en el recipiente, se rellena dicho recipiente con un gas inerte, por ejemplo: nitrógeno seco, hasta que la presión en el interior del recipiente sea ligeramente superior a la presión atmosférica, por ejemplo 1 bar aproximadamente. Las uvas se mantienen en estas condiciones durante un período de aproximadamente 24 horas, controlando que durante dicho tiempo se mantenga la sobrepresión de nitrógeno en el interior la cámara.
Transcurrido dicho periodo de anoxia, se ventea el sistema, y se extraen las uvas. El procesado de las uvas debe realizarse de la forma más rápidamente posible después del tratamiento anóxico; así de forma rápida se procede al despabilado y estrujado de las uvas, bombeándose la pasta resultante al depósito de fermentación. Tras esperar 48 horas se realiza una siembra de levaduras, lo que origina el arranque de la fermentación alcohólica. Se establece un programa diario de remontados con el fin de extraer el mayor contenido de compuestos polifenólicos de los hollejos de las uvas, consistiendo en un bazuqueo a primera hora de la mañana (rotura y hundimiento del sombrero) junto con un remontado de 15 minutos de duración. Concluida la fermentación alcohólica se procede al descubado y prensado de los orujos, juntando el vino de sangrado con el vino prensa y dejando el depósito convenientemente cerrado para que realice la fermentación maloláctica de manera espontánea.

Claims

REIVINDICACIONES
1. Método para aumentar el contenido endógeno de trans-resveratrol en el vino caracterizado porque se realiza mediante la aplicación de un tratamiento anóxico a las uvas, previo al procedimiento de vinificación.
2. Método para aumentar el contenido endógeno de trans-resveratrol en el vino, según la reivindicación 1, caracterizado porque consta de las siguientes etapas. i.- Introducir las uvas en un recipiente estanco provisto de las conexiones adecuadas para extracción de aire e introducción de un gas inerte adecuado. iL~ Extraer el aire de dicho recipiente mediante purgado con nitrógeno o empleando una bomba rotatoria de poca potencia, hasta una presión inferior a 1 mbar. iii.- Insuflar un gas inerte en el interior de dicho recipiente estanco hasta que la presión en su interior sea ligeramente superior a la atmosférica, iv.- Mantener las uvas en el interior de dicho recipiente durante un tiempo comprendido entre 6 y 24 horas, controlando que durante dicho tiempo se mantiene la sobrepresión de gas inerte en el interior de dicho recipiente. v.- Ventear el recipiente y extraer las uvas. vi.- Procesar la uva lo más rápidamente posible después del tratamiento anóxico. viL- Realizar le procedimiento de vinificación en tinto habitual.
3. Método para aumentar el contenido endógeno de trans-resveratrol en el vino, según la reivindicación 2, caracterizado porque el gas inerte utilizado para obtener una atmósfera con un contenido muy bajo en oxígeno es N2, un gas noble, o cualquier gas no reactivo que no afecte a la calidad y propiedades organolépticas de las uvas.
4. Método para aumentar el contenido endógeno de trans-resveratrol en el vino, según reivindicación 3, caracterizado porque dicho gas inerte es nitrógeno seco.
5. Método para aumentar el contenido endógeno de trans-resveratrol en el vino, según reivindicaciones anteriores, caracterizado porque la presión del gas inerte en el interior de Ia cámara durante el período de anoxia es de 1,1 bares.
6. Método para aumentar el contenido endógeno de trans-resveratrol en el vino, según reivindicación 2, caracterizado porque el período de anoxia es de 24 horas.
7. Método para aumentar el contenido endógeno de trans-resveratrol en el vino, según reivindicaciones anteriores, caracterizado porque le procesado de la uva ser realiza como máximo en el plazo de 18 horas después del tratamiento anóxico.
