Abstract

Described is a method of displaying and selecting a protease molecule, the method comprising: (a) providing mRNA molecules, each mRNA molecule comprising a nucleotide sequence encoding a peptide; (b) incubating the mRNA molecules under conditions suitable for translation of the mRNA molecules to form complexes comprising ribosome, mRNA and encoded nascent peptide displayed on the ribosome; (c) contacting the complexes with a protease binding ligand under suitable binding conditions (i.e. for specific binding of a protease to a protease binding ligand), and selecting one or more complexes which are specifically bound to the protease binding ligand. Also described are methods of identifying proteases.
