本発明は、 医療分野等において生体の高分解能を有する血管造影方法と、 その 方法に用いる高分解能血管造明影用 X線管に関する。 背景技術
血管造影用透過 X線装置は、低コスト、利便性の高い病気診断装置として、 臨 床医療分野でなくてはならない医療用分析装置である。 近年、 医療技術の進歩と 共に、 高分解能の血管造影用 X線装置が求められるようになってきている。 例え ば、 狭心症の診断には微細な心臓の環動脈を観察することが必要であり、 ま.た、 再生医療研究においては 1 0〃m以下といった高分解能の血管造影用 X線装置が 必要である。
再生医療の用語は、 従来は人工関節、輸血、皮膚移植、 臓器移植等を意味する ものであつたが、近年、幹細胞を所望の臓器に分化させ、 この臓器により難病を 治療するという新しい再生医療が注目されている。 この再生医療における臓器は 自分自身の細胞から再生される臓器であるため、 従来の他人の臓器の移植のよう に拒絶反応の心配がない。 このように幹細胞分化による再生医療は、遺伝子治療 と共に将来のもっとも注目される医療技術であり、世界中で熾烈な研究開発競争 が ί亍われている。
幹細胞分化による再生医療は、 まだ研究の途についたばかりであり、治療に生 かされるためには今後極めて膨大な研究開発を必要とする。 特に分化の過程が十 分に解明されておらず、 この過程を解明するためには分化過程中の微細な血管の 成長過程を観測することが必須である。
ところで、従来の透過 X線による血管造影方法は、 生体安全性の観点からヨウ 素を血管造影剤に使用するが、 使用する X線の波長を適切に選択できないために 血管部分と血管以外の部分との透過 X線の強度比、 すなわちコントラストが十分 でなく、 特に組織の深部にある血管像は組織を透過するにつれてコントラストが 低下し、人体においては、 せいぜい 1 0 0〃m程度の分解能しか得られない。 こ のため従来は、上記のような微細な血管像を観測する必要がある場合には、 シン クロトロン放射光を用いざるを得なかった。
しかしながら周知のように、 シンクロトロン放射光は電子を高エネルギーに加 速し、 この電子の制動に伴う輻射光を利用するものであるので、適切な波長の X 線を選択できるものの、 巨大な設備と膨大なエネルギーを必要とし、 コストが高 くまた利便性が悪い。 例えば、 シンクロトロン放射光設備はコストが高いため、 一つの設備を多目的に共同利用するが、 このため一人に割り当てられるマシン夕 ィムが短く、 また電力の不足する夏季には全く稼動できない。 このため再生医療 の研究開発が遅延する原因になっている。 発明の開示
上記課題に鑑み本発明は、 生体の血管の透過 X線による造影を高分解能で造影 できる、 低コスト且つ簡便な方法を提供することを一目的とする。 また、 この方 法に必要な、低コスト、 且つ利便性が高い高分解能血管造影用 X線管を提供する ことを他の目的とする。
上記目的を達成するため、本発明の高分解能血管造影方法は、 ヨウ素 ( I ) を 血管造影剤として使用し、 セリウム (C e ) の K a特性 X線を透過 X線源とする ことを特徴とするものである。 この方法によれば、 セリウムの 特性 X線がョ ゥ素に極めて良く吸収されるので、血管部分と血管以外の部分との透過 X線強度 のコントラストが高くなり、 従って、 高分解能で造影することができる。
また、本発明の高分解能血管造影用 X線管は、 X線管の対陰極物質がセリウム であることを特徴とする。 この X線管によれば、 セリウムの K a特性 X線が極め て低コスト、且つ簡便に発生できる。 セリウムの K a特性 X線のフォトンェネル ギ一は、 血管造影剤であるヨウ素の Κ吸収端エネルギーの近傍にあり、 且つわず かに大きいので極めて良くヨウ素に吸収される。 従って、本発明の X線管を、 ョ ゥ素を血管造影剤とする X線透過血管造影法に用いれば、 極めて微細な血管の造 影が可能になる。 本発明の高分解能血管造影用 X線管において、 対陰極物質がセリゥムである X 線管は、 熱陰極と、 熱陰極から発生する熱電子が加速されて衝突するセリウムか らなる板状の対陰極とを有する X線管であつても良い。

この X管によれば、 電子の加速方向と反対方向に X線を取り出せるので、制動 X線が少なくなり、 X線の単色性が高い。

また、 上記のプラズマ X線管は、 気体放電を生起するために一定圧力の気体を 必要とするが、 陰極と対陰極を一定気体圧力下に密封する密封管型であつても良 く、 あるいは、 陰極と対陰極とを内包する容器に一定圧力の気体を導入して使用 する組立型であってもよい。
上記のように、 本発明の高分解能血管造影方法、 並びに高分解能血管造影用 X 線管を用いれば、 1 0〃m以下といった微細な血管の造影が可能となり、 臨床医 療現場、 あるいは再生医療等の研究分野において有用である。 特に、再生医療の 研究分野においては、 装置が低コスト、且つ利便性が高いことから、装置のマシ ンタイム等の制約を受けずに多くの研究者が同時並列的に研究を進めることがで き、 もって再生医療研究の速度を速めることができる。 図面の簡単な説明

本発明は、 以下の詳細な発明及び本発明の幾つかの実施の形態を示す添付図面 に基づいて、 より良く理解されるものとなろう。 なお、添付図面に示す種々の実 施例は本発明を特定または限定することを意図するものではなく、単に本発明の 説明及び理解を容易とするためだけのものである。
図 1は、 本発明の高分解能血管造影方法を模式的に示す図である。 本発明の高 分解能血管造影方法は、従来の透過 X線による血管造影方法と比べて、 X線源に セリウム Κ α特性 X線を使用することだけが異なり、 他は同様である。 すなわち 図に示すように、本発明法は、 血液中にヨウ素を造影剤としてドープした被写体 1を挟んで、 X線源 2と X線写真フィルム等の X線撮像器 3を配置し、 透過 X線 強度のコントラストを利用して血管を造影するものである。
図 2は、 ョゥ素の Κ吸収端近傍の質量吸収係数の X線フォトンエネルギー依存 性を示す図である。 一般に、単一原子からなる物質の質量吸収係数は、 フオトン エネルギーの増加に対し、 原子に特有なフォトンエネルギーで急激に立ち上がり ( K殻電子の励起に対応する場合に K吸収端と呼ばれる) 、 その後指数関数的に 減少する。 従って、 造影剤の吸収端エネルギーよりも大きく、 且つ、 できるだけ 造影剤の吸収端に近いエネルギーを有する X線を使用すれば、 造影剤に吸収され る X線の割合が高まり、血管部分と血管部分以外との透過 X線のコントラストが 高くなるため、 高分解能の血管造影を得ることができる。
図に示すように、 セリウム Κ α特性 X線 (3 4 . 6 k e V) は、 ヨウ素の K吸 収端エネルギー (3 3 . 2 k e V) に極めて近いので、本発明の方法によれば、 透過 X線のコントラストが高くなる。
従来のタングステンを対陰極物質とした測定方法は図に示すように、 夕ングス テン 特性 X線(5 8 . 7 k e V ) が、 ヨウ素の K吸収端から離れているため にコントラストが小さい。 また従来、 モリブデンを対陰極物質とした X線源も使 用されているが、 モリブデンの 線はヨウ素の吸収端よりエネルギーが低いた めにほとんど吸収されず、 もっぱら制動 X線によるコントラスト像である。 制動 X線は波長が連続した X線源であるため十分なコントラストが得られない。 次に、本発明の方法に用いる高分解能血管造影用 X線管を説明する。
図 3は、本発明の高分解能血管造影用 X線管の主要部の構成を示す図である。 図 3 ( a ) は、 熱陰極と、熱陰極から発生する熱電子が加速されて衝突するセリ ゥムからなる板状の対陰極とを有する X線管を示している。 図 3 ( b ) は、環状 熱陰極と、環状熱陰極から発生する熱電子が加速されて衝突するセリゥムからな る棒状の対陰極とを有する X線管を示している。 また、 図 3 ( c ) は、 環状の炭 素冷陰極と、 セリウムからなる棒状の対陰極と、放電用気体とを有する X線管で あって、 セリゥムからなる棒状の対陰極物質を気体放電により蒸発させてプラズ マを形成し、 このプラズマから特性 X線を発生させるプラズマ X線管を示してい る。
図 3 ( a ) において、 X線管 3 1は、 フィラメントでなる熱陰極 3 2と、熱陰 極 3 2から発生する熱電子が加速されて衝突するセリウムからなる板状の対陰極 3 3とを有している。 セリウムからなる対陰極 3 3は、 導電性が良く、且つ熱伝 導性の良い金属ブロック 3 4、 あるいは水冷パイプを有する金属ブロック 3 4に 埋め込まれて固定されている。 熱陰極 3 2と金属ブロック 3 4との間に高電圧を 印加し、熱電子をセリゥムの K吸収端以上のエネルギーに加速してセリゥム Κ α 特性 X線を発生する。 X線は熱電子の加速方向に対して横方向 (3 5 ) に取り出 す。 この X線管 3 1は、 若干の制動 X線も生じるが、 必要とされる分解能が 1 0 u m程度であれば、 血管造影に支障はない。
図 3 ( b ) において、 X線管 3 6は、環状のフィラメントである環状熱陰極 3 7と、環状熱陰極 3 7かち発生する熱電子が加速されて衝突するセリウムからな る棒状の対陰極 3 8とを有している。 この X線管 3 6は、 熱電子の加速方向と反 対方向 (3 9 ) から X線を取り出すので制動 X線が少なくなり、単色性が高い。 また、 対陰極 3 8を気化させながら使用することもできる。
図 3 ( c ) において、 プラズマ X線管 4 0は、環状の炭素冷陰極 4 1と、 セリ ゥムからなる棒状の対陰極 4 2と、 炭素冷陰極 4 1と対陰極 4 2を含む空間に充 填した気体放電用の気体とを有している。 炭素冷陰極 4 1と対陰極 4 2との間に コンデンサを接続し、 このコンデンサに電荷を蓄え、 炭素冷陰極 4 1の近傍で気 体放電を起こすことにより、 コンデンサに蓄えた電荷を瞬時に放電し、 セリウム からなる棒状の対陰極 4 2を蒸発させ、 セリウムからなる弱電離プラズマを生起 する。 このプラズマに電子が衝突、 及び制動されて X線が発生する。 プラズマの 長軸方向 (符号 4 3で示す) に沿って伝搬するセリウムの Κ α:特性 X線は誘導放 出によって強度が強くなり、 プラズマの長軸方向 (4 3 ) に沿って伝搬する制動 X線はプラズマに吸収されて強度が減衰する。 このため、強度が極めて強く、且 つ単色性に優れたセリゥム K a特性 X線パルスが発生する。 プラズマを X線発生 媒質とするこのタイプの X線管はブラズマ X線管と呼ばれている (本発明者によ る特開 2 0 0 2 - 3 1 3 2 6 7号公報参照) 。
以下、本発明を幾つかの実施例に基づいて説明する。 実施例 1
実施例 1は、本発明の方法及び本発明の X線管によって、 高分解能な血管造影 ができることを実証するものである。
ヨウ素のプラスチック被覆微少球またはセリゥムのプラスチック被覆微少球を 血管造影剤としたゥサギの心臓の血管造影を比較した。 X線源として図 3 ( a ) に示した X線管を用いた。
図 4は、 血管造影剤の異なるゥサギの心臓の血管造影を示す透過 X線写真であ る。 (a) はヨウ素のプラスチック被覆微少球を用いた場合、 (b) はセリウム のプラスチック被覆微少球を用いた場合を示している。
図 4 (a) から明らかなように、 ヨウ素を造影剤とした場合には、 セリウム α特性 X線がヨウ素に良く吸収されるために、 細い冠状動脈を明瞭に造影するこ とができる。 なお、 図の左下の白い線は分解能を確認するために、 心臓に挿入し た 1 00 m径のタングステン線による造影である。
一方、 図 4 (b) から明らかなように、 セリウムを造影剤とした場合には、 セ リウム Κ α特性 X線のフォトンエネルギーがセリゥムの吸収端エネルギーより低 く、 セリウム Κ«特性 X線がセリウムにほとんど吸収されないために、 環状動脈 はほとんど造影できない。 '
実施例 1から、 ヨウ素を造影剤とした場合には、 セリウム K a特性 X線を使用 することによって極めてコントラストの高い血管造影ができることがわかる。 実施例 2
実施例 2は、 本発明の方法及び X線管によって、 生体深部の血管であっても鮮 明に造影できることを実証するものである。
ョゥ素のプラスチック被覆微少球を血管造影剤として犬の心臓の血管造影を行 い、 生体の X線の吸収、 散乱を模擬する媒質としてアクリル板 (5 Omm厚) を 使用した。 他の実施条件は実施例 1と同様である。
図 5は生体中の血管の深さを模擬した犬の心臓の血管造影を示す透過 X線写真 である。 図 5 (a) は摘出した犬の心臓を直接血管造影した場合、 (b) は摘出 した犬の心臓と X線撮像器との間に、 厚さ 5 Ommのアクリル板を挿入した場合 を示している。
図 5 (a) から明らかなように、 微細な冠状動脈が鮮明に造影できる。 また、 図 5 (b) から明らかなように、 アクリル板を揷入しても微細な冠状動脈が鮮明 に造影されることができる。 なお、 図 5の (a)及び (b) の左下の白い線は分 解能を確認するために、 心臓に揷入した 1 00 m径のタングステン線による造 影である。 また右上の棒状の造影は、 撮影に際し犬の心臓を固定するために用い た支持棒の造影である。
アタリル板を揷入しても、 すなわち造影する血管が生体深部の血管であっても コントラストの減衰無しに造影できることがわかる。 これは X線の波長が単色で あるためである。 実施例 3
実施例 3は、 本発明の方法及び X線管によれば、 径が数十/ m以下の血管造影 が可能であることを実証するものである。 ゥサギの下肢の微細血管を造影した。 他の実施条件は実施例 1と同様である。
図 6はゥサギの耳の微細血管の造影を示す透過 X線写真である。 図において、 右上の白い線は 5 0 a m径のタングステン線の造影である。 このタングステン線 の造影と比較することにより、 数十 m径以下の血管の造影が可能であることが わかる。
図 7はゥサギの下肢の微細血管の造影を示す透過 X線写真である。 図 7 ( a ) はゥサギの下肢の微細血管の造影を示す図であり、 (b ) は (a ) を拡大した図 である。 中央やや下の白い線は 1 0 0 m径のタングステン線の造影である。 こ のタングステン線の造影と比較することにより、 数十/ m径以下の血管の造影が 可能であることがわかる。
なお、 上記実施例で用いた X線管は、 図 3 ( a ) に示した構成の X線管である が、 図 3 ( b ) 及び図 3 ( c ) に示した構成の X線管はさらに X線の単色性が高 いので、 これらの X線管を用いた場合にはさらに 1 0 m以下の高解像度の造影 が得られることは明かである。 産業上の利用可能性
上記説明から理解されるように、本発明によれば、 高分解能の血管造影ができ る。 従って、本発明は、 微細な血管の観察を必要とする、 臨床医療分野、 あるい は再生医療等の医療研究分野に用いれば極めて有用である。

Claims

請 求 の 範 囲
1 . ヨウ素を血管造影剤として使用し、 セリウムの Κ α特性 X線を透過 X 線源とすることを特徴とする、高分解能血管造影方法。
2 . X線管の対陰極物質がセリウムであることを特徴とする、 高分解能血 管造影用 X線管。
3 . 前記 X線管は、熱陰極と、 この熱陰極から発生する熱電子が加速され て衝突するセリウムからなる板状の対陰極と、 を有することを特徴とする、請求 項 2に記載の高分解能血管造影用 X線管。
4 . 前記 X線管は、環状熱陰極と、 この環状熱陰極から発生する熱電子が 加速されて衝突するセリウムからなる棒状の対陰極とを有することを特徴とする 、請求項 2に記載の高分解能血管造影用 X線管。 ' ·
5 . 前記 X線管は、 陰極と対陰極を真空に密封した真空管型、 または上記 陰極と対陰極を内包する真空容器を真空引きして使用する組立型であることを特 徴とする、請求項 3または 4に記載の高分解能血管造影用 X線管。
6 . 前記 X線管は、環状の炭素冷陰極と、 セリウムからなる棒状の対陰極 と、 上記環状の炭素冷陰極と対陰極とを含む空間を満たす気体放電用の気体と、 を有し、 上記気体の気体放電により上記対陰極物質を蒸発させてプラズマを形成 し、 このプラズマから特性 X線を発生させることを特徴とする、 請求項 2に記載 の高分解能血管造影用 X線管。
7 . 前記 X線管は、前記炭素冷陰極と対陰極とを含む空間に前記気体を密 封する密封管型または上記空間に上記気体を導入して使用する組立型であること を特徴とする、請求項 6に記載の高分解能血管造影用 X線管。
