Abstract

The present invention relates to methods for producing a recom­binant eukaryotic organisms, preferably a recombinant plant, comprising the step of decreasing the chromatin assembly capacity of said organism. The method may be used for introducing sequences into the chromosomal DNA of a eukaryotic cell either by illegitimate insertion or, preferably, by homologous recombina­tion. The method results in increased transformation efficacy and/or increased frequency of homologous recombination. The invention furthermore relates to expression constructs suitable for expressing at least part of a nucleic acid sequence encoding for a chromatin assembly factor protein (e.g., in form of antisense or double-stranded RNA) suitable for decreasing expression or functionality of at least one chromatin assembly factor protein in plant or plant cells. Object of the invention are furthermore recombinant plant cells or plants comprising a decreased chromatin assembly capacity, preferably a decreased expression or functionality at least one chromatin assembly factor and the use of said recombinant plants for the production of food, feeds, seed, pharmaceuticals or fine chemicals.
