On ajoute de la glace (200g) puis on laisse remonter à 10°C. La solution est clarifiée sur Clarcel. La phase organique est soutirée. La phase aqueuse est extraite deux fois par du dichlorométhane (200ml). Les phases organiques sont combinées et lavées 3 fois par une solution de chlorure de sodium 5% (200ml). La solution est concentrée et sous vide (8mb) à 20°C pour donner 85,4g de 3-Chloro-l- ( 2-thienyl ) -propanone . Rendement 91,4%. Pureté CGV : 98%. A partir du composé obtenu à cette première étape, on prépare le racémique l-(2-thienyl)-3-Chloro-l- propanol. On charge de l'éthanol (350ml), du borohydrure de sodium (17,1g) et on rince par de l'éthanol (50ml). On agite pendant 15 minutes à température ambiante puis
on refroidit à -6°C. On coule en 40 minutes le produit 3-Chloro-l-(2-thienyl) -propanone en maintenant la température en dessous de —2°C. On rince par du dichlorométhane (10ml). On laisse sous agitation à —3°C pendant une heure. On hydrolyse la réaction par une solution de NH4C1 (107g) sous agitation vigoureuse en 40 minutes en maintenant la température entre —3°C et 0°C. On laisse remonter à température ambiante en 2 heures. L'éthanol est chassé à l'évaporateur rotatif sous 60mb. On ajoute de l'eau (250ml) et on extrait par du DCM 4 fois (200ml, 3x70ml). Les phases organiques sont jointes et lavées par de l'eau trois fois (100ml, 2x70ml). On sèche sur sulfate de magnésium et on concentre à sec pour obtenir 75g de l-(2-thienyl)-3-Chloro-l-propanol . La phase aqueuse est extraite trois fois par du n-butanol (100ml + 70ml + 50ml). On sèche sur sulfate de magnésium et on concentre sous vide (4mb, 60°C). On obtient 46,1g. Teneur : 69,5%.
