Abstract

D-leucine repeats in the C-terminal transactivation domain (C-TAD) of hypoxia-inducible factor-1 alpha (HIF-1 alpha) are critical for transactivation, coactivator interaction and intranuclear trafficking. Further, CBP regulates the intranuclear distribution of HIF-1 alpha and SRC-1. These findings provide new ways to increase HIF-1 alpha transactivation capacity, inhibit HIF-1 alpha activity, and screen substances that may be capable of altering the HIF-1 alpha transactivation response. Further, they provide novel methods and products that can be used to cause substances to be hypoxia-inducible when in their native state they are not.
