Abstract

De nombreux antigènes de protéines et de glycoprotéines sécrétés par Mycobacterium tuberculosis (Mtb) ont été identifiés comme antigènes « précoces » de Mtb sur la base d'anticorps précoces présents chez les sujets infectés par la Mtb avant le développement d'une maladie clinique décelable. Des fragments de peptides porteurs d'épitopes desdits antigènes précoces de Mtb, en particulier d'une protéine secrétée de 88kDa, la GlcB (SEQ ID NO : 106) et d'un antigène de Mtb MPT51 (SEQ ID NO : 107) ont été identifiés. Lesdits peptides, des variants de ces derniers, des peptides multimères de ces derniers qui comprennent deux ou plusieurs répétitions d'un ou plusieurs peptides, et des peptides de fusion comprenant des protéines de Mtb antigéniques précoces, des peptides, ou les deux, sont utiles dans des méthodes d'immunoessais pour une détection précoce, rapide de la tuberculose (TB) chez un sujet. Des immunoessais préférés permettent de détecter les anticorps dans les urines d'un sujet. L'invention concerne en outre des compositions d'antigènes, des kits et des méthodes utiles dans la détection d'anticorps de Mtb précoces. Lesdites protéines et lesdits peptides antigéniques sont en outre utilisés dans des compositions de vaccins.
