Abstract

Die Erfindung betrifft ein Verfahren zur Erzeugung einer Zufallsanordnung klonaler Inseln auf einer Oberfläche, umfassend die Schritte a1) Bereitstellung einer Oberfläche aufweisend mindestens einen Primer-Typ, dessen Primer-Moleküle mindestens zum Teil irreversibel an der Oberfläche immobilisiert sind; b1) Hybridisierung von zu amplifizierenden Nukleinsäuren mit den Primern aus Schritt a1); c1) Amplifikation der zu amplifizierenden Nukleinsäuren aus Schritt b1), wobei mindestens ein Gegenprimer-Typ zur Amplifikation eingesetzt wird, dessen Primer-Moleküle mindestens zum Teil nicht immobilisiert sind und dessen Primer-Moleküle mit den Primern aus Schritt a1) in bezug auf die zu amplifizierenden Nukleinsäuren mindestens ein PCR-Primerpaar bilden, und wobei der Reaktionsraum der Amplifikation durch einen Raum gebildet wird, der durch die Oberfläche und durch eine an die Oberfläche angrenzende mikrokompartimentierende Matrix begrenzt wird.
