Abstract

In der Durchflußzytometrie ergibt sich das Problem, daß man gleichzeitig zwei Fluorochrome mit überlappenden Emissionspeaks messen möchte. Dies gelingt mit Hilfe eines Ein-Laser-Durchflußzytometers mit zwei Fluoreszenzkanälen unterschiedlicher Empfangswellenbereiche, wobei das Verfahren die folgenden Schritte aufweist: a) Anregen des ersten Fluorochroms von mit diesem Fluorochrom markierten Referenzzellen mit einem für beide Fluorochrome geeigneten Laserstrahl; B) Einstellen der Photomultiplier-Röhrenspannung des zweiten Fluoreszenzkanals, welcher an die Emission des zweiten Fluorochroms angepasst ist, auf einen Wert, bei dem kein vom ersten Fluorochrom im zweiten Fluoreszenzkanal erzeugten Fluoreszenz-Emissions-Signal detektiert wird; C) Anregen der Fluorochrome von mit erstem und zweitem Fluorochrom markierten, zu messenden Zellen mit dem Laserstrahl; D) Messen des Fluoreszenz-Emissions-Signals des ersten Fluorochroms in den zu messenden Zellen im ersten Fluoreszenzkanal, welcher an die Emission des ersten Fluorochroms angepasst ist; und E) Messen des Fluoreszenz-Emissions-Signals des zweiten Fluorochroms in den zu messenden Zellen im zweiten Fluoreszenzkanal.
