Abstract

La présente invention concerne un procédé d'obtention in vitro de cellules insulino-sécrétrices de mammifère, caractérisé en ce qu'il comporte les étapes suivantes: a) la préparation des tissus pancréatiques de mammifère par prélèvement de pancréas, b) la dissociation des tissus pancréatiques obtenus à l'étape (a) en cellules pancréatiques isolées, c) éventuellement l'élimination des cellules endocrines des cellules pancréatiques isolées à l'étape (b), d) l'induction de la dédifférenciation des cellules isolées à l'étape (b) en cellules précurseur canalaires, e) l'induction de la rédifférenciation des cellules précurseurs canalaires obtenues à l'étape (d) en cellules insulino-sécrétrices. Elle concerne également l'utilisation des cellules insulino-sécrétrices ainsi obtenues pour la préparation d'une composition pharmaceutique utile pour le traitement des pathologies pancréatiques et notamment du diabète.
