Abstract

Cette invention a trait à un procédé permettant d'introduire une séquence nucléotidique non virale susceptible d'expression dans une cellule possédant un récepteur commun de fixation non universelle, laquelle cellule est issue du groupe constitué par des lymphocytes T, B et des basophiles/mastocytes. Ce procédé consiste à mettre en contact cette cellule avec un vecteur viral comportant une séquence nucléotidique de recombinaison renfermant une séquence pour l'acide nucléique non viral susceptible d'expression et possédant une coque virale modifiée constituée de protéines de coque virale endogènes et d'une protéine de coque virale modifiée contenant une séquence aminoacide adénovirale provenant d'une protéine de sérotype adénoviral à 35 ou 51 fibres, cette séquence adénovirale de la protéine modifiée étant un ligand pour ledit récepteur de fixation. Dans une variante, le vecteur comporte une séquence codant une capside virale constituée de protéines de coque virale endogènes et d'une protéine de capside modifiée contenant une séquence aminoacide adénovirale provenant d'un sérotype adénoviral autre que le sérotype des protéines capsidiques endogènes, cette protéine modifiée étant un ligand pour ledit récepteur de fixation. Cette invention porte également sur un procédé de transduction cellulaire, cette cellule étant choisie dans le groupe constitué par des lymphocytes T, des lymphocytes B et des basophiles/mastocytes. Ce procédé consiste à mettre en contact ces cellules avec une particule adénovirale comportant une séquence nucléotidique non adénovirale et une protéine de capside chimère renfermant une séquence aminoacide dérivée d'au moins deux sérotypes adénoviraux, cette particule ayant un tropisme à l'égard de ces cellules supérieur à au moins l'un des sérotypes adénoviraux comportant ladite protéine de capside chimère. L'invention concerne, de plus, des cellules transduites, des ensembles de sous-populations de cellules, un procédé de transduction ex vivo d'une population cellulaire et une méthode d'administration à un être humain ou à un autre mammifère d'une population de cellules modifiées ex vivo par génie génétique. Elle a trait, en outre, à une technique d'identification de la fonction d'un premier acide nucléique dans des cellules hématopoïétiques.
