- a transient transactivation experiment was performed in which CMVrAR was cotransfected with the aforementioned enhancer/promoter/reporter construct (10:1 receptor to enhancer/promoter/reporter) in C2C12 cells using LipofectAMINE PlusTM reagent (GibcoBRL) according to the manufacturer's instructions was used to compare the activities of the enhancer/promoter/reporter constructs. Specifically, 10,000 cells/well were plated in growth media (Dulbecco's modified Eagle Medium (DMEM) high glucose supplemented with 10% FBS, IX sodium pyruvate and 0.5X antibiotic-antimycotic (all from GibcoBRL) ) .
