Abstract

Beschrieben wird ein HTS-geeignetes Testsystem zum Nachweis der Aktivität von cyclo-Nucleotid-abhängigen Proteinkinasen (cNPK) enthaltend a) mindestens eine Testverbindung, b) mindestens ein geeignetes cNPK-Substrat, c) mindestens eine zu inkubierende Zusammensetzung enthaltend cNPK und ATP, ggf. Phosphorylierungsreaktionsstopper, und d) ein geeignetes Detektionssytem zur Quantifizierung der Phosphorylierung des cNPK-Substrats. Ferner wird ein HTS-geeignetes Testsystem zum Nachweis der Aktivität von VASP-Phosphatasen beschrieben enthaltend e) mindestens eine Testverbindung, f) mindestens ein geeignetes VASP-Phosphatase-Substrat, g) mindestens eine zu inkubierende Zusammensetzung enthaltend VASP-Phosphatase, und h) ein geeignetes Detektionssystem zur Quantifizierung der Dephosphorylierung des VASP-Phosphatase-Substrats. Die beschriebenen Testsysteme lassen sich zum Auffinden von Verbindungen, die die Aktivität einer cNPK oder einer VASP-Phosphatase modulieren, aus chemischen oder natürlichen Substanzbibliotheken einsetzen.
