Definitions

  • THIS INVENTION is related to the decontamination and/or sterilisation of equipment. More particularly, the present invention is of use in the decontamination and/or sterilisation of equipment which comes into contact with biological tissue, especially plant tissue.
  • Microbial contamination is critical for success in the micro-propagation of plants by tissue culture.
  • Microbial contaminants can originate from the environment wherein the tissue culture is performed, from operators undertaking the tissue culture, or from the plants themselves.
  • the equipment used to cut and manipulate plants is a key vector for the spread of this microbial contamination between plants.
  • tweezers and scalpels used to cut and manipulate plants are sterilised between each tray of plants processed by the operator. Sterilisation is usually achieved by heat and/or chemicals.
  • Sterilisation is usually achieved by heat and/or chemicals.
  • tweezers and scalpels are dipped in a solution of ethanol and then heated using (a) small ovens known as "bacti-cinerators", (b) heat beads, (c) d e flame of a gas burner, or (d) high pressure steam in an autoclave.

  • This control of contamination is more important for automated than manual tissue culture because of the larger number of plants processed through the automated procedures and the consequent increased potential to spread micro-organisms from one plant to another.
