Abstract

Die Erfindung bezieht sich auf die vollständigen cDNA-Sequenzen der Cladosporium herbarum Allergene Clah8 und Clah12. Im Rahmen der molekularbiologischen Analyse der Allergene konnte Clah12 (wird von 22 % der Patienten erkannt) als ribosomales Protein Pl identifiziert werden. Dieses Protein ist insofern interessant, weil in Lupus Patienten Autoantikörper gegen ribosomale Proteine zu finden sind. Ob zwischen Schimmelpilzallergie und Autoimmunkrankheiten ein Zusammenhang besteht, muß erst gezeigt werden. Clah8 (wird von 9 % der Patienten erkannt) weist eine hohe Homologie zu einer Familie von Transkriptionsfaktoren auf, die durch eine sogenannte cold-shock Domäne gekennzeichnet sind. Mit Hilfe dieser rekombinanten Sequenz konnten mit Computeranalyse hoch potente B- und T-Zellepitope bestimmt werden. Die von den rekombinanten Proteinen abgeleiteten Peptide könnten für die Diagnose einer Cladosporium herbarum Schimmelpilzallergie verwendet werden. Im weiteren könnten die Peptide für eine in vitro aber auch in vivo allergenspezifische Stimulation von T-Zellen (Proliferation, Interleukinproduktion) eingesetzt werden, oder aber auch zu einer Blockierung von T-Zellen, die zu einer Toleranz der allergenspezifischen T-Lymphozyten führen.
