Abstract

Procédé d'obtention de cellules trophoblastiques d'un foetus d'un individu mammifère, à partir d'un échantillon du sang périphérique prélevé sur la mère portant ledit foetus, selon lequel, successivement: a) on sépare les érythrocytes dudit échantillon; b) on sépare au moins une partie de leucocytes, en marquant au moins une partie des leucocytes avec au moins un premier marqueur spécifique; c) on sépare des cellules trophoblastiques, en marquant ces dernières avec au moins un second anticorps substantiellement spécifique desdites cellules; d) on trie positivement par cytométrie de flux les cellules trophoblastiques; caractérisée en ce que le premier anticorps de l'étape de pré-enrichissement est un anticorps anti-leucocytaire commun, marquant spécifiquement au moins 80 % des leucocytes de la fraction leucocytaire, pour obtenir une fraction pré-enrichie en cellules trophoblastiques, permettant de distinguer ultérieurement lors de la cytométrie de flux les cellules trophoblastiques marquées uniquement par le deuxième anticorps.
