Abstract

The method for determination of catalytic efficiency of transglutaminase preparation provides that to the solution of dipeptide N-CZ-GLN-GLY in the mixture having pH in interval 6.00-6.10 containing NaOH, hydroxylamine and reduced glutathione in the ratio of 1:2:2 with total volume of 15 ml, 0.1 ml of transglutaminase sample is added. The mixture is thermostated at 37° C for 10 minutes, cooled and 15 ml of mixture of 3% solution is added into 0.1 N HCl, 12 % solution of trichloroacetic acid HCl and in the ratio in equal amount, is centrifuged. The value of optical density is determined in liquid part at 525 nm, using for comparison analogous solution without transglutaminase, activity values are calculated relative to calibrating diagram, built with using of chemically clean transglutaminase. The catalytic activity is determined according to the rate of penetration of gel of standardized 10% solution of food gelatin with pH 7.4. Evaluation in wide range of values mass fraction of enzymatic preparation in enzyme-substrate system is 0.5%, fermentation time at the temperature of 50 °C – 24 hours, and gel fermentation time at the temperature of 25 °C – 12 hours.
