The utility model relates to medicine, in particular to the composition of ingredients for an eye medicated film. 2 So-called ophthalmic drugs containing enzymes are used in the treatment of more than 30 eye pathologies, including severe forms of chemical burns.
Known composition of ingredients for ophthalmic drugs | see G.I. Bondarenko etc. Development of ophthalmic drugs based on immobilized proteolytic enzymes and biogenic stimulators. FARMAKOM 12. 93. State Research Center of Medicines. Kharkov. 1993 70 pp. 12-17). The composition contains an enzyme (alkaline protease or terrilithin), polyvinyl alcohol (PVA), peat and water.
But the eye preparation has the following significant drawbacks: 1. decrease in the proteolytic activity of terilitin and alkaline protease after simultaneous immobilization with peat by 94.49 and 31.895, respectively, possibly due to the inactivating effect of borax; 12 2. lack of data on preservation of proteolytic activity of the obtained immobilized preparations; 3. insufficient stabilization of immobilized proteolytic enzymes in the area of acidic pH values (shift of the pH-optimum by 1 unit), and in the area of alkaline pH values of 8.0-9.0 - b095 preservation of proteolytic activity; 4. low resistance to temperature conditions: the rate of thermal inactivation of immobilized proteinase compatible with peat decreased by half compared to free.
The closest to the claimed utility model is the composition of the ingredients for the medicated eye film, which is listed in Ukrainian utility model patent Mo13238).
«в)The composition contains the following components, wt. 9o: urea 1.3-4.76; -o papain 1.73-6.35; polyvinyl alcohol 17.8-19.4; the rest is water. "in)
This composition is chosen as the prototype of the claimed utility model. Fo
The prototype and the claimed useful model have the following common components: - protein component; - - polyvinyl alcohol; c - water.
But the specified composition has significant drawbacks: firstly, it does not prevent the formation of a burn scab, and it is used for its denaturation, and, secondly, when used in a high concentration, there is a pain syndrome and it can cause partial denaturation of healthy tissues of the cornea of the eye.
The basis of a useful model is the task of developing a composition of ingredients for an eye medicated film, "in which, by replacing the protein component and a different ratio of components, to ensure an increase in the effectiveness of the eye film in the treatment of burns." The task is solved by a composition of ingredients for an eye medicated film containing a protein component, polyvinyl alcohol and water, the fact that as a protein component the composition contains human serum albumin, according to the following ratio of the specified components, mass. for: human serum albumin 13.2-15.6; ke polyvinyl alcohol 79.4-81.8; the rest is water. - What is new in the proposed useful model is that human serum albumin is used as a protein component in the eye film for the first time in 5 years, as well as the mass ratio of the components of the composition. o Human albumin preparations (synonyms - human serum albumin, donor albumin - Zoiishio o AIritype) are made from blood plasma of donors or from placental blood serum. Sterile, viscous transparent solutions from yellow to light brown in color, containing 595, 10965 and 2095 protein.
Albumin is a white fraction of human blood plasma. With intravenous administration of albumin to the recipient
It maintains the osmotic pressure in the circulating blood, quickly increases blood pressure, promotes the flow of tissue fluid into the bloodstream and its preservation, participates in the exchange between tissues and blood, and is a source of protein nutrition for the body.
The mass ratio of the components was selected experimentally.
With a decrease in the concentration of albumin, the necessary therapeutic effect was not observed, and with an increase, allergic reactions occurred in the form of intense hyperemia of the conjunctiva with increased sensitivity.
The increase in the effectiveness of the eye film can be explained by the fact that it provides the following advantages, compared to the prototype: - prevents the formation of a burn scab; - there is no pain syndrome caused by the use of enzymes; 65 - there is no risk of denaturing effects of enzymes on healthy parts of the cornea.
95) водного -Д-The composition of the ingredients for the medicated eye film is prepared in the following way. 2 cm of 1096th (8.8 wt. 95) water -D-
Після цього одержують однорідні плівки, з яких висікають очні лікарські плівки у вигляді дисків, площею 0,б4см?, запаюють в поліетиленову плівку і зберігають при температурі 0-42С у холодильнику.solution of polyvinyl alcohol is mixed with 0.3-0.5 cm? (1.33-2.24 wt. 95) of a solution of human serum albumin in physiological saline. All operations are performed with constant stirring. The resulting mixture is placed on a substrate, dried at a temperature of 10-20 in the air. After that, homogeneous films are obtained, from which ophthalmic medicated films are cut out in the form of discs, with an area of 0.4 cm?, sealed in polyethylene film and stored at a temperature of 0-42C in a refrigerator.
5,0.Example 1. A composition of ingredients for an eye medicated film was prepared as described above. The components of the composition were taken in the following ratio, mass. 9b: 70 human serum albumin 12.94; polyvinyl alcohol 82.06; water 5.0.
Physico-chemical properties of the film obtained according to this example, 95 inclusions of serum albumin 75 people - are given in the table.
Examples 2-6 illustrate the physical and chemical properties of the film with different mass ratios of composition ingredients.
The data are given in the Table.
1-3.As can be seen from the data in the table, the highest percentage of inclusion of human serum albumin is observed in examples 1-3.
But in example 1, the use of a small amount of human serum albumin does not provide the necessary therapeutic effect.
Increasing the content of human serum albumin in the composition (examples 4-6) is impractical due to a violation of the physical and chemical parameters of the film (excess human serum albumin is not included in PVA and crystallizes on the surface of the film).
ЗIncreasing or decreasing the amount of PVA affects the inclusion of albumin. WITH
Experimental studies were carried out on both eyes of 15 rabbits of the Chinchilla breed weighing ЗО00ОГ, which "-- were in the same conditions.
During the experiments, the rabbits were divided into 3 groups. The first (control) group consisted of 5 rabbits (10 eyes), for the treatment of an experimental burn, Okubrax eye drops were used, two drops 3 times a day in both eyes.
In the second group of rabbits (5 rabbits, 10 eyes) for the treatment of burns of both eyes, in addition to instillations of eye drops "Okubrax", single applications of a solution of human serum albumin were performed during the day in an exposure of 5 minutes.
In the third group of rabbits (5 rabbits, 10 eyes), for the treatment of burns of both eyes, in addition to instillation of eye drops - with "Okubrax", medicated eye films with human serum albumin were used, which were placed in the conjunctival cavity of both eyes once, within 10 days. "» Treatment was carried out within 7 days from the moment of eye burn.
сThe use of selected treatment options was reflected in the course and consequences of the burn process in the eyes of experimental groups of animals. In the first (control) group of rabbits, within 3 days of observation from the moment of the burn, signs of inflammation were quite intense. Hyperemia of the eyelids, conjunctiva, as well as the level of the secreted corresponded to the highest level of inflammatory reaction. At the same time, when applying eye medicated films from human serum albumin in experimental groups of animals, the intensity of the manifestation of the inflammatory reaction began to gradually decrease after a day.
