Description
The useful model belongs to agricultural biotechnology and can be used in selection and genetic research for the purpose of studying biological features, microclonal reproduction of valuable genotypes and creation of new breeding material of monoecious non-narcotic hemp (Sappabrikh zayn 1.) of the Central Russian ecological and geographical type.
Quite often in selection work, there is a need to increase the reproduction ratio of valuable genotypes of monoecious non-narcotic hemp (Sapparbiv zayima I.) of the Central Russian ecological and geographical type. For this purpose, microclonal propagation of plants is used under conditions of miigo. In addition, the cultivation of somatic tissue structures and the regeneration of plants from them allows obtaining genetically modified regenerative plants, which can differ genotypically and phenotypically significantly from their original forms and become valuable breeding material. The passage of cells through the stage of callusogenesis leads to the appearance of various types of changes in the genotype, and the level of somaclonal variability depends on various factors, in particular, the starting material, the conditions of cultivation of ip myo, the composition of nutrient media, the presence of exogenous growth regulators, etc. At the same time, unlike artificially induced mutagenesis, it is possible to select useful and eliminate harmful or unwanted mutant forms. Obtaining haploids with subsequent doubling of the chromosomal set is of particular importance for selection.
The method of doubled haploids allows you to create homozygous material and increase its productivity. Thus, in the breeding and genetic work with monoecious non-narcotic hemp (Sapparib zayima I.) of the Central Russian ecological-geographical type, the use of biotechnological methods to obtain fundamentally new selection material is increasingly in demand, however plants of monoecious non-narcotic hemp of the Central Russian ecological-geographical type are not sufficiently sensitive to the cultivation of human cells in general and the culture of isolated cells and tissues in particular. They are weakly affected by the photoperiod and are able to bloom with a significant range of variation in the length of the light day (phenological phases are only slightly lengthened or shortened), and therefore they quickly finish their development and die, which makes their further use as an ip mio impossible.
Taking into account the above arguments, the development of a nutrient medium for the cultivation of monoecious non-narcotic hemp (Sappabiv Zayima I.) of Central Russia is relevant
Of the ecological-geographical type in the conditions of ip miigo, which would improve this process.
Дане відоме середовище потребує модифікації свого складу для покращення культивування конопель (найближчий аналог).The known nutrient medium is universal and can be used for the cultivation of monoecious non-narcotic hemp (Sapparie occupies 1.) of the Central Russian ecological and geographical type. The disadvantage of the known nutrient medium is that, provided it is used for the cultivation of monoecious non-narcotic cannabis (Sappabiv Zaima I. of the Central Russian ecological and geographical type, explants grow rather slowly, necrotic spots appear on the leaves under intense light, the shoots quickly enter the flowering phase, end their development and die, which makes their further use impossible, the induction of callusogenesis and organogenesis is complicated, the accumulation of phenolic compounds is observed.
The basis of a useful model is the task of developing, on the basis of an analogue, a new basic nutrient medium, which would be more than the closest analogue, suitable for the cultivation of monoecious non-narcotic hemp (Sapparie occupies 1.) of the Central Russian ecological and geographical type in the conditions of ip mio, that is, such a nutrient medium , which would improve the process and result of cultivation due to the intensification of the growth of shoots and improvement of their viability, contributed to the extension of the duration of the vegetative stage of development and ontogenesis in general, inhibited the onset of the generative stage, and would act as an inhibitor of the accumulation of phenolic compounds.
середньоросійського еколого- географічного типу в умовах іп міїго, яке включає 440 мг/л (З мМ) Сасі».2НегоО, 370 мг/л (1,5 ММ)The task is solved by the fact that the nutrient medium for the cultivation of monoecious non-narcotic hemp (Sappariz zayima!.) of the Central Russian ecological and geographical type in conditions of ip miigo, which includes 440 mg/l (Z mM) of Sasi".2NegoO, 370 mg/l (1 .5 MM)

Nitrogen is one of the extremely important biogenic macroelements for plants, as it is part of amino acid molecules and, accordingly, proteins, phytohormones, DNA, RNA, chlorophyll, etc. Monoecious non-narcotic hemp (Sappabriv zayima I.) of the Central Russian ecological-geographical type is quite demanding in terms of availability of compounds of this element available for nutrition. The presence of Nitrogen in the composition of the medium is mainly ensured by the presence of MNi" and MOH ions. It is the increased concentration of MNAMO" (2400 mg/l) in comparison with the nutrient medium of Murashige and Skug that intensifies the growth of explants and calli of monoecious non-narcotic cannabis of the Central Russian ecological and geographical type, promotes better development of vegetative organs, at the same time inhibits the development of generative organs, prolonging the duration of their ontogenesis and miigo as a whole.
The demand for monoecious non-narcotic hemp (Sappabibh zayima I.) of the Central Russian ecological and geographical type in Kaliya is somewhat less, compared to other cultures. The reduced concentration of K ions in the nutrient medium, the source of which are KMOz salts (1768 mg/l) and KNePO»x (136 mg/l), harmonizes the intensive growth of plants, which is observed due to increased nitrogen concentration, prevents excessive elongation of the stem and internodes, causes the formation of additional nodes, which is important for increasing the reproduction ratio of microclones. At the same time, due to the sufficient amount of the carbohydrate glucose (which gives better results, compared to sucrose), the water balance (osmotic pressure) is not significantly disturbed.
Decreased, in comparison with the nutrient medium of Murashige and Skuga, to 136 mg/l concentration
Phosphorus in the form of orthophosphoric acid ions PO43- (136 mg/l КНегРОх) to a certain extent inhibits the development of plants and their transition to a generative state, and also prevents the appearance of necrotic spots on leaves under intense lighting.

It was experimentally established that the cultivation of monoecious non-narcotic hemp of the Central Russian ecological-geographical type in the conditions of ip mio gives better results with an increased content in the environment of such microelements as Boron, Cuprum and
Cobalt, therefore, compared to the nutrient medium of Murashige and Skuga, the compounds containing them are doubled, and HzVOz is complexed (chelated) with CzH5(OH)z, which contributes to better assimilation of the element.
Inclusion in the medium of 5 mg/l of ascorbic acid, which is an antioxidant, and 1 g/l of activated carbon prevent the formation of phenolic compounds that cause inhibition of growth and development or lead to the death of explants.
Nicotinic acid was excluded from the composition of the medium, as it had a negative effect on the growth and development of shoots, somatic and generative callusogenesis and organogenesis in hemp of this type.
