Дію суміші пептидів, структурних аналогів фрагментів суперспіральної області молекули фібрин(іоген)уу Аа91-103 МЕП КСОЕ5БЗАММ, ВВ126-135 ОККОКОМКОМ, уб9-77 МРОЕБЗЗКРМ, в еквімолярному співвідношенні на полімеризацію фібрину визначають методом трансмісійної електронної мікроскопії негативно контрастованих зразків (13). Як негативний контрастер використовують 1 95 водний розчин уранілацетату. Сітки для електронного мікроскопу вкривають шаром 0,3 95 розчину формвару в дихлоретані та закріплюють розпиленням вуглецю за допомогою вакуумного випарювача. Для приготування зразків до стерильної скляної пробірки послідовно вносять 0,3 мг/мл фібриногену людини, 0,025 М Сасіг у 0,05 М амоній-форміатному буферному розчині (рН 7,9), загальний об'єм проби 220 мкл. Полімеризацію фібрину ініціюють внесенням тромбіну до кінцевої концентрації 0,25 МіІН/мл. Через 90 та 180 с із полімеризаційного середовища відбирають аліквоти, які розводять до кінцевої концентрації фібриногену - 7 мкг/мл, розчин якого по 10 мкл переносять на вкриті вуглецем гратки, які через 2 хв обробляють 1 95 розчином уранілацетату. Після інкубації протягом 5 хв сітку промивають спочатку 100 мМ амоній-ацетатним буферним розчином (рн 7,9), потім 10 мМ амоній-ацетатним буферним розчином (рН 8,5). Далі сітку накладають на краплину контрастера, попередньо нанесену на поверхню тефлонового блоку й через 1 хв контрастер видаляють за допомогою водноструменевого насосу. За допомогою електронного мікроскопу Н-600 при 75 КМ при збільшені 20000-50000 одержують електронно-мікроскопічні зображення середовищ, у яких відбувається полімеризація фібрину: через 60 с після внесення тромбіну в контрольній пробі спостерігають формування протофібрил фібрину та їхню латеральну асоціацію (фіг. 2), через 180 с спостерігають зрілу фібрилу (фіг. 3), за присутності трьох пептидів, структурних аналогів фрагментів суперспіральної області молекули фібрин(оген)уу Ас91-103 МЕІГВареЕББЗАММ,The effect of a mixture of peptides, structural analogs of fragments of the superspiral region of the fibrin(iogen) molecule Aa91-103 MEP KSOE5BZAMM, VV126-135 OKKOKOMKOM, уб9-77 MROEBZZKRM, in an equimolar ratio, on fibrin polymerization is determined by the method of transmission electron microscopy of negatively contrasted samples (13). Grids for the electron microscope are covered with a layer of 0.3 95 formvar solution in dichloroethane and fixed by spraying carbon using a vacuum evaporator. To prepare samples, 0.3 mg/ml human fibrinogen, 0.025 M Sasig in 0.05 M ammonium formate buffer solution (pH 7.9) are successively added to a sterile glass tube, the total sample volume is 220 μl. Polymerization of fibrin is initiated by introducing thrombin to a final concentration of 0.25 MiIN/ml. After 90 and 180 s, aliquots are taken from the polymerization medium, which are diluted to the final concentration of fibrinogen - 7 μg/ml, 10 μl of which solution is transferred to carbon-covered grids, which are treated with 1 95 uranyl acetate solution after 2 minutes. After incubation for 5 min, the grid is washed first with 100 mM ammonium-acetate buffer solution (pH 7.9), then with 10 mM ammonium-acetate buffer solution (pH 8.5). Next, the grid is placed on a drop of contrast agent previously applied to the surface of the Teflon block, and after 1 minute the contrast agent is removed using a water jet pump. With the help of an electron microscope H-600 at 75 KM at a magnification of 20,000-50,000, electron microscopic images of the environments in which fibrin polymerization takes place are obtained: 60 s after the introduction of thrombin in the control sample, the formation of fibrin protofibrils and their lateral association are observed (Fig. 2) , after 180 s a mature fibril is observed (Fig. 3), in the presence of three peptides, structural analogs of fragments of the superhelical region of the fibrin(ogen) molecule As91-103 MEIGVareEBBZAMM,
