Abstract
Винахід належить до способу детекції нуклеїновокислотної послідовності-мішені в аналізі з РТОСЕ (розщепленням та подовженням РТО). В даному винаході здійснюють детекцію нуклеїновокислотної послідовності-мішені, при цьому РТО (олігонуклеотид, що зондує та мітить), гібридизований з нуклеїновокислотною послідовністю-мішенню, розщеплюється з вивільненням фрагмента, і цей фрагмент гібридизується з СТО (захоплюючим та матричним олігонуклеотидом) з утворенням подовженого дуплекса, після чого відбувається детекція присутності подовженого дуплекса. Подовжений дуплекс забезпечує одержання сигналів (генерацію, підсилення, гасіння або ослаблення сигналів) від міток, що вказують на присутність подовженого дуплекса, і має регульовану величину Т, яка є добре придатною для детекції присутності нуклеїновокислотної послідовності-мішені.The invention relates to a method for detecting the target nucleic acid sequence in an assay with RTOS (cleavage and elongation of RT).
