髓源性抑制細胞(MDSCs)是一群包含有早期髓系前驅細胞/粒細胞異質群體、巨噬細胞和樹突狀細胞等異細胞亞群的集合體。這些細胞通常由髓系標記Gr-1和CD11b表現其特徵。髓源性抑制細胞由於可藉由增加免疫抑制因子,例如精氨酸酶1(ARG-1)和一氧化氮合酶2(NOS2)之表現來抑制T細胞效應之功能,而在腫瘤免疫逃逸機制、自體免疫疾病、移植排斥反應,慢性炎症及感染等方面扮演重要角色。參見(例如),Gabrilovich與Nagaraj,Nat Rev Immunol 9:162-174(2009)。
由於髓源性抑制細胞所具有之免疫抑制特性,而使其能做為治療免疫性疾病的候選藥物。因此,亟需一種藉由得自活體培養方式產生之穩定且安全的髓源性抑制細胞。
本發明基於此發現,生長調節致癌基因(GRO)趨化激素,尤其是GRO-γ,對於人類周邊血單核細胞衍生的樹突狀細胞(MDDCs)之
分化及功能具有抑制作用。此外,發現GRO-γ可驅動MDDCs分化成為一種類似髓源性抑制細胞(MDSC)的表現型。
因此,本發明所描述係關於一種獲得免疫抑制細胞的方法。該方法包括:獲得一種能分化成樹突狀細胞之前驅細胞;及將該前驅細胞培養於含有一趨化激素之培養基中一段足夠使該前驅細胞分化為樹突狀細胞的時間,其中該樹突狀細胞係呈現一種免疫抑制表型,而藉此獲得該免疫抑制細胞。所述之趨化激素可為一種GRO趨化激素,例如,GRO-γ或GRO-α。
另一方面,本發明包含一種藉由上述方法得到之免疫抑制細胞,及含有該細胞的組成物。
本發明亦描述一種使用上述細胞或細胞組成物治療個體中各種病況之方法。例如,本發明之細胞或細胞組成物可用於有需要之個體中抑制免疫反應,調節與腫瘤發生相關之血管新生,治療自身免疫疾病,治療發炎性疾病,或治療移植物對抗宿主疾病(GVHD)。
圖1表示間葉系幹細胞(MSC)-條件培養基對於MDDC分化產生了抑制效果。將純化來自人類PBMCS的CD14+單核細胞培養在有或無MSC-條件培養基(MSC-CM)(1/2X體積)存在下培養於DC-分化培養基中。於第5天iDC再經無或以LPS(1mg/mL)處理(mDC)兩天。於第7天分析MDDCs的表型和功能。(A)針對與DC成熟相關之表面標記進行染色,並以流式細胞儀分析,測定10,000個細胞的平均螢光強度(MFI)。(B)未成熟之MDDCs以螢光標記葡聚糖試劑(1mg/mL)於37℃
下施予脈衝30分鐘,並藉由以流式細胞儀測量之FITC-葡聚醣攝取量分析其內吞能力。所顯示於FITC-葡聚醣攝入後之FACS圖譜為:無FTIC-葡聚醣存在下的MDDCs作為陰性對照組(NC,灰線),iMDDCs(iDC,虛線),及有MSC-CM存在下之iMDDCs具(iDC+ MSC-CM,黑色實線)。數據為於2位供者進行之三次獨立實驗的代表性結果。(C)成熟MDDCs與同種異體之T細胞共培養4天(DC:T=1:10),並於以1μCi/孔之[3H]-胸苷施予脈衝16小時後測量定與胸苷的併入量。藉由於三名供者各重複三次所測得之[3H]-胸苷併入量來測定T細胞增生。數據以相對於無MSC-CM補充組之倍數(MFI±SD),或以[3H]-胸苷攝取量之CPM(平均值±SD)表示。數據代表三次獨立實驗之結果。(n=3位供者,每次實驗)。*:p<0.05;**:p<0.01;***:p<0.0001。
圖2表示MSC-CM對MDDCs的抑制效果可藉由將抗-GRO-γ中和抗體加入MSC-CM而產生逆轉。(A)使用市售之人類細胞激素/趨化激素抗體陣列(RayBio)比較得自MSC條件培養基(MSC-CM)與含血清培養基對照組的細胞激素圖譜。培養基中所含細胞激素和趨化激素的量,係藉由將所述之陣列膜先與對抗特定細胞激素或趨化激素之生物素標記的抗體,之後再與HRP-共軛的卵白素進行培養,然後將其以X-射線底片曝光而測定得。實驗重複2次。(B)以流式細胞儀分析GRO趨化激素及IL-8對於未成熟MDDCs上之CD40之表現的影響。將人類CD14+單核細胞於IL-4(80ng/mL)和GM-CSF(80ng/mL)存在下,在含有10%人類AB+血清之α-MEM培養基中進行分化。於第7天,將CD14+單核細胞、單獨之未成熟MDDCs或補充以GRO-α、GRO-β、GRO-γ或IL-8之未成熟
MDDCs進行針對CD40表現之染色,並以流式細胞儀分析。灰線對應未染色的對照組。所示數據為對應所指定趨化激素之CD40陽性細胞的百分比。實驗結果來自三個獨立實驗。(C)以抗-GRO-γ(10μg/mL)抗體或同種型匹配的對照抗體於37℃下培養60分鐘,將MSC-CM中的GRO-γ活性中和,之後放置於4℃下隔夜。將MDDCs在有或不含MSC-CM(1/2X體積)之DC分化培養基中,於有或無中和性抗-GRO-γ抗體存在下進行分化。利用流式細胞儀進行MDDCs表型分析,結果以平均螢光強度相對於未經處理之iDC(MFI±SD)所獲得數值的倍數表示。結果來自兩個實驗,每組包括兩名供者。*:p<0.05;**:p<0.01。
圖3顯示GRO-γ抑制MDDCs的分化。純化自人類PBMCs之CD14+單核細胞於有或無重組GRO-γ(250ng/mL)存在下,經過或未經CXCR2受體競爭型拮抗劑SB225002(250nm)預處理30分鐘的條件下進行分化。於第7天,分析MDDCs的表型和功能。(A)經過表面標記標定後,使用流式細胞儀進行MDDCs的表型分析。條形圖代表相對於未經處理之iDCS所獲得之MFI的MFI倍數(MFI±SD之倍數)。數據相當於三個獨立實驗之平均值,其中每個實驗包括兩至三名供者。(B)在37℃下,將未成熟之MDDCs以FITC-葡聚醣(1mg/mL)施予脈衝30分鐘,並以流式細胞儀測定其攝取情形。數據係來自五個不同實驗,且以相對於iDC組之MFI±SD的倍數表示。n=2-3。(C)將成熟MDDCs與同種異體T細胞(DC:T=1:10)共培養4天,並於以1μCi/孔之[3H]-胸苷施予16小時-脈衝後胸苷與測定胸苷併入量。以[3H]-胸苷併入量來測定T細胞增生。數據以相對於未經處理之MMDCs之CPM(平均值±SD)的倍數表示,且數據係來自

圖5顯示受GRO-γ處理之MDDCs啟動的T細胞具有耐受性性質。將人類CD3+ T細胞(3×105個)以被3×104個於有或無重組GRO-γ存在下,經過或未經SB225002預處理之條件下進行分化之MDDCs刺激。(A)以RT-PCR評估於純化自MDDC的CD3+ T細胞及T細胞共培養物中之選擇性基因IL-4、IL-10、IL-12和IFN-γ的mRNA表現。相對基因表現係對於GAPDH之表現正常化。數據以相對於經DC-啟動之T細胞組所得

圖6顯示GRO-γ處理之BMDC呈現MDSC細胞之細胞圖譜。(A)自C57BL/6小鼠採集骨髓細胞,並以單獨GM-CSF、GM-CSF/GRO-γ或GM-CSF/GRO-γ/SB225002處理六天使其分化為BMDCs。以FACS分析使用抗CD11b和GR-1抗體進行純化自BMDCs表面標定。然後使用FACS Aria細胞分選儀分離CD11b和GR-1雙陽性細胞,並以RT-PCR測定精氨酸酶1和誘導型一氧化氮合酶基因的轉錄程度。mRNA的表現係以BMDCs相對mRNA倍數表現(對GAPDH基因表現量正常化)。數據取自三次測量平均值±標準差(n=3)。結果來自三次獨立實驗。(B)以FACS Aria分選出OT-1/CD8+ T細胞,並於OVA257-264胜肽存在下與經GRO-γ處理或未經處理的BMDC共培養3天。使用3H-胸苷併入分析來測定抗原特定刺激之T細胞增生。(C)為於活體內分析GRO-處理細胞之免疫抑制活性的實驗流程圖。步驟1:BMDCs免疫前六天,準備不同組衍生自C57BL/6小鼠骨髓細胞的BMDCs。步驟2和步驟3:BMDCs免疫前一天將C57BL/6小鼠以1Gy照射,然後將小鼠以靜脈注射OT-1脾臟細胞(4×107細胞/鼠)。第4步:收集各種不同的BMDCs(5×104細胞/小鼠),然後於第0天以皮下注射到6-8週齡之OT-1脾臟細胞-重建的B6小鼠。步驟5:BMDCs刺激後第7天,犧牲各實驗組小鼠並從各小鼠收集脾臟細胞,然後置於96孔

生自各種不同次組骨髓之細胞的免疫抑制活性。將小鼠BMDCs於有或無GRO-γ存在下進行分化。從經過GRO-γ處理和未經處理之BMDCs分選出未經分離之細胞(總體細胞)、CD11b+細胞和CD11b-細胞。於各組分選細胞之存在下,估計同種異體反應性淋巴細胞的增生活性。所示數據為來自三個獨立實驗的平均值和標準差。使用Student’s t test確定顯著差異性,以p值表示。*:p<0.05;**:p<0.01;***:p<0.0001。ns:無統計學意義。
圖9顯示GRO-γ處理的BMDCs可改善GvHD小鼠的失重和存活率。於經過致死性照射之BALB/c小鼠(接受者,R)中,藉由以靜脈內注入分離自MHC不匹配的野生型B6小鼠(供者,D)之骨髓細胞和脾臟細胞來誘發GvHD。將不同的細胞組群分別注射入接受者:5×106個僅含分離自供者小鼠之骨髓細胞(BM,D)(-□-)(n=3),5×105個僅含分離自供者小鼠之脾臟細胞(SP,D)(-△-)(n=3),分離自供者小鼠之BM和SP的組合(BM(D)+SP(D))(-○-)(n=4),以及分離自供者小鼠之BM和SP的組合加上於移植後第0天(-■-)、第2天(-▲-)及第7天(-●-)注射1×107個經GRO-γ處理的BMDCs(GBMDC)。於移植後監測動物之體重變化(A)及存活率(B)達10天。以僅接受骨髓細胞的動物作為對照組。進行Log-rank(Mantel-Cox)測試,對於存活率之p=0.0255。表示三次實驗其中一次的結果。
本發明係基於發現為特定趨化激素,例如:生長調節腫瘤(GRO)化學趨素,可驅使人類周邊血液單核細胞衍生之樹突狀細胞分化,而成為具有類似髓源性抑制細胞(MDSC)的表現型。
據此,本發明係描述一種獲得可呈現類MDSC表現型之免疫抑制細胞的方法。該方法包括獲得可分化為樹突狀細胞(DC)之前驅細胞(例如,人類週邊血液單核細胞或老鼠含髓性前驅細胞之骨髓),並將該前驅細胞培養於一種含有趨化激素之培養基中一段足夠時間,以使該前驅細胞能分化為樹突狀細胞。以此培養得到之樹突狀細胞展現類似MDSC之表現型,例如具有免疫抑制之表現型。
適用於本發明方法之前驅細胞包括CD14+單核細胞、骨髓細胞及髓源性前驅細胞。較佳地,該前驅細胞為CD14+單核細胞或骨髓細胞。前驅細胞可以該項技術領域中習知的方法,或以如下所述之方法獲得。
將前驅細胞於含一或多種趨化激素之培養液中培養一段足以使該細胞分化成為可展現類似MDSC表現型之DC的時間(例如3-7天)。此可使用各種不同的培養基,例如:間質幹細胞(MSC)-條件培養基、α-MEM完全培養基及ROMI-1640培養基。所述之培養基可補充以其他因子,例如,粒細胞-巨噬細胞集落刺激因子(GM-CSF)或IL-4。該趨化激素可為GRO趨化激素。
GRO趨化激素,亦即CXCL1/GRO-α、CXCL2/GRO-β及CXCL3/GRO-γ含有Glu-Leu-Arg(ELR)基序,且係屬於IL-8血管新生係細胞激素家族。所述之趨化激素可得自化學來源,或使用該項技藝中習知的方法,例如:重組蛋白質技術而製備得。人類GRO-α、GRO-β(MIP2α)和GRO-γ(MIP2β)為三種不同、非等位人類基因之產物。GRO-β及GRO-γ分別與GRO-α共有90%及86%之胺基酸序列同源性。GRO趨化激素之胺基酸序列為該項技藝已知者。參見,例如:NCBI參考序列NP_001502.1(人

以上述方法獲得之免疫抑制細胞具有某些特定特徵(例如類MDSC表現型),以下做進一步詳述。舉例來說,這些細胞可刺激T細胞增生的能力減低,且藉由增加IL-10和IL-4分泌使T細胞直接分化呈現免疫耐受性表現型,增加Foxp3之表現,和減少IL-12和IFN-γ產生。免疫抑制細胞使用習知方法,及如下所述之方法,例如ELISA、流式細胞儀及定量RT-PCR進行特徵確定。
本發明亦包括含有上述免疫抑制細胞的組成物。該組成物可進一步包含醫藥上或生理上可接受之賦形劑。
本發明亦包含使用上述之細胞組成物,以於個體中方法對於抑制免疫反應、調控與腫瘤發生相關之血管新生及和治療各種其他免疫狀況病況,即移植物對抗宿主疾病(GVHD)、發炎性疾病、自體免疫疾病和移植排斥。所述之免疫抑制細胞可自異源或自體的前驅細胞產生。而以前例,可進行HLA-配對以避免或使宿主反應減至最低。
發炎性疾病包括(但不限定於)阿茲海默氏症、強直性脊柱炎、骨關節炎、類風濕關節炎、銀屑病關節炎、哮喘、動脈粥樣硬化,科
隆氏病、結腸炎、皮膚炎、憩室炎、纖維肌痛、肝炎,腸易激徵候群即全身性紅斑狼瘡。
自體免疫疾病括(但不限定於)阿狄森氏病、Celiac疾病、皮肌炎、葛瑞夫茲病、橋本氏甲狀腺炎、多發性硬化症、重症肌無力症、惡性貧血、類風濕性關節炎、乾燥徵候群,全身性紅斑狼瘡及第I型糖尿病。
個體係指分人類或非人類動物。非人類動物之實例包括所有具有免疫系統之脊椎動物,例如哺乳動物,如非人類靈長類(特別是高等靈長類)、狗、囓齒類(如小鼠或大鼠)、天竺鼠、貓、畜牧動物(如馬、牛、羊或豬),及非哺乳類如鳥類、兩棲類、爬蟲類等。於較佳具體態樣,所述之個體為人類。於另一具體態樣,該個體為實驗動物或為適合作為疾病模型之動物。
欲接受治療上述其中一病況之個體,可經由針對該特別疾病所定之標準診斷技術加以確定。“治療”意指將一種組成物(例如細胞組成物)投藥予已罹患,或具有發展成某一疾病之風險的個體,以治癒、減輕、緩解、補救、延緩發病、預防或改善該疾病、該疾病的症狀、該疾病之繼發狀態或導致該損傷/疾病之誘因。“有效量”係指能夠在受治療之個體中產生所希望的醫療結果之組成物的量。所述之治療方法可以單獨進行,或與其他藥物或療程結合。
上述之免疫抑制細胞可經由注入或注射(例如,靜脈內、鞘內、肌內、管腔內、氣管內、腹膜內或皮下注射)、口服、經皮或於該項技術領域中已知的其它方法進行投藥。
投藥可以兩個禮拜一次或一個禮拜一次,或其頻率更多或更
少。劑量和頻率有部份係取決於,預期以上述細胞進行治療之疾病或失調症的病理徵兆和臨床與亞臨床症狀的衰退狀況。如習於本技術領域者所能理解,可視各種例如病情的嚴重程度,個體之年齡、性別或身體狀況,副作用及醫師的判斷等因素,來調整劑量和投藥方案。在所有上述方法中,可將細胞以(例如)每次注射1×106至1×109個細胞之劑量施用予個體。
下列之特別實施例可被解釋為僅具有說明性,而不是以任何方式限制本發明所揭示的其餘部分。在無進一步闡述的情況下,相信熟悉本領域之技術人員可以根據本說明書的記載,將本發明利用至最充分的程度。所有於本說明書中引述的公開文獻,係以其全文通過引用的方式併入本文中作為參考。
實施例
材料與方法
(1)老鼠
C57BL/6(H2b)小鼠購買自台灣國家實驗動物中心並飼養於台灣國家衛生研究院之實驗動物中心。C57BL/6(H2b)/OT-1轉殖鼠(transgenic for the TCR-specific peptide OVA257-264,SIINFEKL)由台灣中央研究院Dr.John Kung贈與。本研究使用六到八週齡之小鼠。所有的動物實驗皆按照台灣國家衛生研究院的機構動物護理和使用委員會批准的協議(協定編號:NHRI-IACUC-100003和NHRI-IACUC-097077-A)進行。
(2)化學試劑
重組人類GRO-α、GRO-β、GRO-γ、IL-4、GM-CSF及重組小鼠GM-CSF購買自PeproTech Inc(Rocky Hill,NJ,USA)。重組小鼠GRO-α、GRO-β及GRO-γ購買自R&D系統(Minneapolis,MN,USA)。N-(2-羥基-4-硝基苯基)-N’-(2-溴苯基)脲(SB225002)購買自Calbiochem(San Diego,CA,USA)。脂多糖(LPS,分離自大腸桿菌055:B5)購買自Sigma-Aldrich(St.Louis,MO,USA)。R-Phycoerythrin(PE)標記之小鼠抗-人類CD11c、HLA-DR及CD83抗體購買自eBioscience(San Diego,CA,USA)。螢光異硫基氰酸酯(FITC)-共軛之小鼠抗-人類CD86、DC-SIGN、CD40和CD80抗體購買自BioLegend(San Diego,CA,USA)。
(3)MSC條件培養基(MSC-CM)之製備
以先前已報告的方法(Lee et al.,2004)從臍帶血純化得到臍帶間質幹細胞(uMSCs)並加以特徵確定。將uMSCs保存於含有α-MEM與10% ES-FBS(HyClone,Logan,UT,USA)的培養基。將uMSCs(3x105個細胞/15mL置於15cm2-細胞培養盤中,第7代)於補充以混合的AB型血清(Invitrogen,Carlsbad,CA,USA)之培養基中在進行兩次繼代培養擴增。每次繼代培養時間為5天。將3×105uMSCs(p9)重新接種在含15ml完全培養基(α-MEM含有10%混合的AB型人類血清及1x青黴素-鏈黴素-穀胺酸(Invitrogen))的15cm2培養皿中,並於濕潤的二氧化碳培養箱中於37℃下
培養5天。將uMSCs的上清液在300×g及4℃下離心10分鐘,以除去細胞碎片,收集上清液並儲存於-80℃下,然後使用作為條件培養基(MSC-CM)。
(4)細胞激素/趨化激素列陣分析
使用人類細胞激素列陣C套組(傳訊人類細胞激素抗體列陣C系列1000.1,RayBio,Redwood City,CA,USA),按照製造商的指示操作,來建立於有或無uMSCs存在下之含有10%混合的AB型人類血清的α-MEM中,所含細胞激素和生長因子之分泌情形。使用ECL系統(Amersham Pharmacia Biotech,Aylesbury,UK),於Kodak BioMax光底片(Kodak,Rochester,NY,USA)上偵測化學發光信號。利用AlphaImager1220分析和文檔系統(Alpha Innotech,Braintree,UK)以點密度儀量化信號強度。將每一點信號對相鄰的背景值進行校正,並對該膜的陽性對照組正常化。
(5)產生人類MDDCs
從健康供者以白血球細胞除去法產生人類單核細胞-衍生的樹突狀細胞(MDDCs)。以Fycoll-Hypaque Plus(GE Health-Pharmacia,Uppsala,Sweden)密度離心純化得到周邊血液單核細胞(PBMC)。使用人類CD14+細胞分離套組(MACS,Miltenyi Biotec,Inc.,Auburn,CA,USA),依照製造商之操作指示,分離得到人類CD14+單核細胞。接著將純化的CD14+細胞(1×106/mL)在有或無GRO趨化激素存在下,
培養於2毫升含有10%混合的AB型人類血清,補充以rhGM-CSF(80ng/mL)和rhIL-4(80ng/mL)(Peprotech,NJ)之MSC-CM或α-MEM完全培養基中,補充加入誘導其分化成為DCs。於第3天,將額外1毫升含有相同濃度的rhGM-CSF和rhIL-4,含有或不含GRO趨化激素之培養基,添加到每組細胞。於第5天,將一半體積的細胞培養物,以同等體積含有相同濃度之rhGM-CSF和rhIL-4的新鮮培養基置換。於第5天,將1μg/mL的LPS加入iMDDCs的培養基中,再進行培養48小時,來並誘導MDDCs(成熟DCs,mMDDCs)的熟成。於第7天,利用FACS分析來監測未成熟DC(iMDDCs)和成熟DC(mMDDCs)間的表現型變化。該研究提案係經由中央研究院醫學研究倫理委員會(AS-IRB01-10113)和國家衛生研究院(EC1001101)研究倫理委員會機構審查委員會批准。
(6)FACS分析
將1×105個細胞以對抗CD11c、HLA-DR、CD80、CD86、CD83、CD40和DC-SIGN之螢光色團標記的抗體(Abs)進行染色,而獲得單核細胞、iMDDCs和mMDDCs的表現型圖譜。以流式細胞儀測定螢光強度。所使用相對應之同型-配對的對照為FITC-IgG1、FITC-IgG2a、PE-IgG2a及PE-IgG2b(BD Biosciences,San Jose,CA,USA)。使用FACS Calibur-流式細胞儀(BD Biosciences公司)分析其表面經標記的細
胞。關於細胞純化,係於FACS Aria細胞分選儀(BD Biosciences公司)上進行分選。各別經分選之細胞族群的純度係大於95%。
(7)細胞內吞測試
藉由分析FITC-葡聚醣(40KD,FD40S,Sigma-Aldrich公司)之細胞攝取量,來測量iMDDCs或mMDDCs的內吞活性,以流式細胞儀量化。在37℃下,將細胞(5×104細胞/樣本/96孔V底平盤)於1mg/mL之FITC-葡聚醣的RPMI-1640培養基中進行培養30分鐘。培育後,將細胞洗滌兩次以除去游離的試劑,然後用1%多聚甲醛固定。以FACS分析來自被細胞內吞之FITC-葡聚醣的信號。使用FlowJo5.7.2版軟體進行數據分析。取得自與不含FITC-葡聚醣之培養基進行培養之細胞的信號作為陰性對照。
(8)MLR測試
將處於不同分化階段之單核細胞衍生的細胞(3×104),在使用X-射線生物照射器(X-ray R-2000,Rad Source Technologies,Inc,Alpharetta,GA,USA)於30Gy下進行照射,然後與經純化的異體CD3+ T細胞(1:10),在完全培養基中,於96孔U底平盤中進行培養。將經X射線照射之髓細胞和同種異體T淋巴細胞於放置在37℃潮濕的CO2培養箱中。經過96小時後,將[3H]-胸苷(1μCi)加入每孔含有單核細胞、iMDDCs或mMDDCs與T細胞混合之槽中,並再培養
16小時。然後用Filtermate 96-孔收集器收取細胞,並於Packard微量平盤閃爍和發光計數器(Perkin-Elmer-Packard;Waltham,MA,USA)中測定放射活性(cpm)作為細胞增生指數。
(9)RNA製備及定量RT-PCR
用Trizol試劑萃取總體RNA,並使用ReverTra Ace set(Toyobo Life Science,Osaka,Japan),依據製造商之操作指示轉換成為cDNA。使用ABI棱鏡7900系統(Applied Biosystems,Foster City,CA,USA)進行RT-PCR分析。將樣品依照下列程序處理:5℃下2分鐘,94℃下10分鐘,以及40次包含95℃下15秒和60℃下1分鐘的循環。重複進行分析三次。對於每個樣品,皆測定循環閾值(Ct)值。將結果以同一平板上之GAPDH基因為基準歸一化。使用2△CT法計算不同細胞組的mRNA表現。設計對個別基因具特異性之跨越內含子的引子對。其序列如下所示:人類GAPDH,GAGTCAACGGATTTGGTCGT(前向引子,F,SEQ ID NO:4),TTGATTTTGGAGGGATCTCG(反向引子,R,SEQ ID NO:5);人類IL-10,ATGCCCCAAGCTGAGAACCAAGACCC(F,SEQ ID NO:6),AAGTCTCAAGGGGCTGGGTCAGCTATCCCA(R,SEQ ID NO:7);人類IDO,CGCCTTGCACGTCTAGTTCTG(F,SEQ ID NO:8),TGACCTTTGCCCCACACAT(R,SEQ ID NO:9);人類基質-金屬胜肽酶-9(MMP-9),GAAGATGCTGCTGTTCAGCG(F,SEQ ID NO:10),

(10)ELISA
從單獨的MDDCs(5×104)、單獨的CD3+純化T-細胞(人類CD3+細胞單離套組,MACS,Miltenyi Biotec,Inc.,5x105)或MDDCs(5×104)與CD3+純化T細胞(5×105)共同培養的培養物收取上清液,並使用市售之ELISA套組,根據製造商的操作指示(R&D systems)測定存在上清液中之IL-4、IL-10、IL-12、INF-γ、IL-6和TNF-α的濃度,重複進行三次。
(11)共聚焦顯微鏡
於有或無GRO-γ存在下產生MDDCs,然後分析細胞內IDO蛋白的表現。將細胞(5×104)平佈置於塗覆有聚-L-賴氨酸的玻片上15分鐘,之後迅速用0.5mL PBS洗滌兩次。加入0.5mL1%多聚甲醛(Sigma-Aldrich)固定細胞10分鐘,然後用PBS清洗3次,並在室溫下於0.5mL的1% BSA/PBS中靜置30分鐘。細胞以0.1%(v/v)NP-40/PBS洗滌三次,然後與0.5% BSA/PBS(含1:50稀釋後的小鼠抗人類IDO抗體(Chemicon Inc,Temecula,CA,USA))於室溫下反應1小時。細胞進一步用0.1%(體積/體積)NP-40/PBS清洗以除
去未結合的抗體,然後與1:150稀釋的與DyLight488共軛之抗小鼠IgG抗體(Sigma-Aldrich)在室溫下反應1小時。以0.1%(v/v)NP-40/PBS進行最終洗滌三次後,將玻片包封於10%甘油/PBS中,並用指甲油密封。使用Leica TCS SP5相機取得螢光圖像(Leica Camera AG,Solms,Germany)。
(12)老鼠BMDCs之生成
如先前所描述(Song et al,2011),收集小鼠骨髓衍生之樹突狀細胞(BMDCs)並將其分化成樹突狀細胞。簡單地說,將取自C57BL/6小鼠的骨髓細胞以2×105個細胞/mL之密度,在有或無重組小鼠GRO趨化激素存在下培養於含有10毫升添加有200U/mL(20ng/mL)重組小鼠GM-CSF之完全RPMI-1640培養基的培養盤中。於第3天,將一半體積的培養基更換以,由含有20ng/mL rhGM-CSF(有或無與GRO組合)之完全RPMI培養基。於第6天,從各培養盤收集來自不同處理組的BMDCs,將其清洗並進行特徵鑑定。
(13)於體外和體內測量經小鼠GRO-處理之細胞的免疫抑制活性
通過測量以OVA-啟動之DC刺激所產生對OVA特異性OT-1 CD8+純化T細胞的增生抑制作用,分析經GRO-γ處理細胞之體外免疫抑制活性。於第6天,收集在有或無GRO-γ存在下進行分化之小鼠(C57BL/6)BMDCs,然後再懸浮於LCM培養基(補充5%胎牛血清、50μg/mL慶大
黴素、20.25mM HEPES,50μM 2-ME和100U/mL青黴素、100μg/mL鏈黴素之RPMI1640培養基)。使用FACS Aria流式細胞儀進行細胞分選,來收集OT-1 CD8+ T細胞(2×105細胞/孔),然後單獨培養,或與BMDC(1×105細胞/孔)或BMDC/GRO-γ細胞(1×105細胞/孔)組合,在有OVA257-264CTL抗原表位(1μg/mL)或人類乳頭狀瘤病毒的RAH對照胜肽(1μg/mL)的存在下,培養於含200μL LCM培養基的96孔U底微量平盤中。將培養物在37℃,5% CO2培養箱中培養3天,將[3H]胸苷(1μCi)加入到各孔中,之後再額外培育16小時。然後使用半自動化的樣品收集器收取細胞,並在Packard微量平盤閃爍及發光計數器測定放射活性(cpm),作為細胞增生的指標。
對於活體內的免疫抑制分析,係於第(-6)天準備欲進行不同處理的小鼠BMDCs。於第(-1)天,製備OT-1臟細胞(4×107/小鼠)並將其以靜脈注射投藥予6-8週齡預先以X射線(1Gy)照射的B6小鼠。收集在有或無GRO-γ存在下進行分化之BMDCs,並於進行轉移當天,與或不與OVA257-264胜肽(1μg/mL)在37℃下進行培育1小時,然後加以洗滌以除去未結合的胜肽。然後在第0天,將各種BMDC製劑(5×104細胞/小鼠)以皮下注射入OT-1脾臟細胞重建的B6小鼠,來引發活體內的抗原特異性活化作用。於第7天,將各組以HBSS、BMDCs、BMDCs/GRO-γ、BMDCs/OVA或
BMDCs/GRO-γ/OVA處理之小鼠犧牲。從個別小鼠收取脾臟細胞(2×105個細胞/孔),並置於96孔U底平盤中以OVA(1μg/mL),或對照組RAH(1μg/mL)胜肽刺激。將培養物在37℃ 5% CO2下培育12、36及60小時。藉由3H-胸苷併入法測定得自HBSS組,或不同組BMDCs引發小鼠之脾臟細胞的增生。實驗重複兩次。數據為以每組三隻老鼠於三個獨立實驗中獲得之代表性結果。
(14)統計分析
使用GraphPad棱鏡,版本5.02(GraphPad Software,Inc)進行分析統計。數據係以得自至少三個獨立實驗的平均值±平均值標準偏差的(SEM)表示。使用one-tailed Student’s t-test分析各組間的統計學顯著差異。
我們認為p值<0.05是顯著的,且顯著程度如下所示:*,p<0.05;**,p<0.01;***,p<0.001。
結果
(1)MSC-條件培養基可對MDDC分化和功能產生抑制效果。
本發明建立一種用以誘導人類CD14+單核細胞分化為MDDCs之活體外標準操作程序,並建立用於在髓源性細胞之分化途徑的每一階段鑑定其表現型特徵之分析。
為研究MSC-條件培養基(MSC-CM)是否會影響MDDCs分化,遂在人類周邊血液CD14+單核細胞的分化
和成熟過程中,將培養基替換以MSC-CM。我們觀察到,相較於未經處理之未成熟MDDCs(iDCS),於經過MSC-CM處理之iDCS上的CD40、CD80、DC-SIGN、CD83、CD11C和HLA-DR表面表現顯著減少(圖1A,上列比較組)。在以經由於至少兩天培養期間施予LPS刺激而產生的成熟MDDCs(MDCS)所進行之實驗,也獲得類似的結果(圖1A,下列比較組)。
為了進一步檢查MSC-CM是否會影響iDCS之內吞活性,用流式細胞儀分析測定FITC-葡聚醣攝取。結果顯示,得自MSC-CM處理組之iDC的內吞能力顯著降低(圖1B)。在混合淋巴細胞反應中刺激同種異體T細胞增生的能力(MDDCs的另一項功能),也在從MSC-CM培養產生之mDC中受到減量調節(圖1C)。綜合上述,此等結果顯示MSC-CM對於MDDC分化和功能呈現抑制功效。
(2)GRO-γ在介導MSCs對於MDDCs分化和功能的抑制作用方面扮演關鍵性角色。
本發明研究在MSC-CM中是何種可溶性因子負責對於MDDC分化的抑制活性。於MSC-CM中檢測到GRO濃度顯著增加(圖2A)。我們進一步檢驗不同亞型的GRO對於人類iMDDCs之表現型的生物學效應。據發現,僅僅GRO-α和GRO-γ對於CD40表現具有顯著的抑制效果(圖2B)。由於GRO-α強度於細胞激素/趨化激素陣列中只有微小增加(圖
2A),故在後續的實驗中僅針對GRO-γ之特性進行研究。為了進一步確定存在於MSC-CM中之GRO-γ對於MDDC分化的作用,吾等顯示條件培養基對於iDC和mDC二者的抑制效果,部分可經由添加中和性抗-GRO-γ抗體而逆轉,但不受其同種型對照影響(圖2C)。在這種抗體的存在下,相較於以同種型對照組或MSC-CM處理的細胞,DC-SIGN、CD80和CD83於iDC的表面-表現量顯著增加(圖2C,上列比較組)。同樣,抗-GRO-γ抗體會實質上逆轉MSC-CM對於CD80、DC-SIGN、CD40和CD11C在成熟DC上之表面表現的抑制效果(圖2C,下列比較組)。
評估重組GRO-γ抑制MDDC分化及抑制其功能的能力。當單核細胞iDC分化時有GRO-γ存在下,相較於無GRO-γ存在下進行培養之細胞,CD40、CD83、CD80、CD11c、CD86和DC-SIGN在iDC上的表面表現量顯著降低(圖3A,上列比較組)。於mDC易觀察到類似的結果,惟,DC-SIGN表現並沒有受到添加GRO-γ影響(圖3(a),下列比較組)。另外,於培養基中加入CXCR2受體競爭型拮抗劑SB225002,會顯著逆轉GRO-γ對於此等所選擇之位於iDCS和mDC上的表面標記物表現的抑制效果(圖3A)。同樣地,DC-SIGN於iDC和mDC上的表現並沒有受到添加SB225002影響。關於MDDCs的功能,GRO-γ會分別顯著減量調節iDC的內吞活
性,及mDC於混合淋巴細胞反應中刺激T細胞增生的能力。此等功效亦會因添加SB225002而受阻斷(圖3B和圖3C)。
這些數據證明,由MSC分泌之GRO-γ在抑制MDDCs之分化和功能上扮演著關鍵的角色。
(3)GRO-γ驅動MDDC分化趨向類似髓源性抑制細胞的免疫表現型。
GRO-γ除了對MDDCs分化及功能具有抑制功效外,GRO-γ也影響MDDC分化前間的細胞激素表現。進行RT-PCR來檢測在得自7天培養物之MDDCs中所選定基因的相對mRNA濃度。結果顯示,當有GRO-γ存在時,發炎性細胞激素基因(即TNF-α、IFN-γ和IL-12)之mRNA,在MDDC分化期間被顯著下調(圖4A)。IL-10、IL-4、TGF-β、IL-1β和IL-6的基因及其他編碼COX-2、編程死亡配體(PD-L1和PD-L2)、基質-金屬胜肽酶9(MMP-9)及特別地IDO之基因的表現程度受到增量調節(圖4A)。人類MDSC特徵在於其具有能分泌IL-10、IL-4、IL-1β和IL-6,及表現COX-2、PD-L1、MMP-9和IDO的能力。定量PCR分析顯示,和未處理之MDDCs對照組相比,以GRO-γ處理之MDDCs會增加此等MDSC標記基因的表現(圖4A)。
為了進一步分析所釋出之細胞激素在各組已分化細胞中之分布情形,遂於培養7天後收集MDDCs,洗淨後再培養3天。進行ELISA,測量於從不同的實驗組取得之上
清液中的細胞激素濃度。與RT-PCR之數據相符,於得自在有GRO-γ存在下進行分化之MDDCs的上清液中,偵測到IL-4和IL-10濃度升高,而IFN-γ和IL-12濃度降低(圖4B)。以DyLight 488-共軛之抗-IDO抗體進行之胞內染色亦顯示,於GRO-γ存在下進行分化之MDDCs中,IDO的細胞內表現量增加(圖4C)。
這些結果指出,GRO-γ不僅抑制MDDC分化,也會驅動朝向類似MDSC的免疫表現型進行分化。
(4)GRO-γ引發之MDDCs驅動T細胞趨向耐受性免疫表現型進行分化
為進一步探討經GRO-γ處理之MDDCs對於T細胞反應的影響,遂將使用人類CD3+細胞分離套組純化得之CD3+ T細胞暴露於有或無以GRO-γ處理之已分化自體MDDCs細胞。進一步以相同的方法,自T/MDDCs混合物分離出CD3+ T細胞。萃取出RNA,並以RT-PCR檢測所選細胞激素基因之表現。實驗發現,相較於與未經處理的MDDCs共同培養之T細胞,與經GRO-γ-引發之MDDCs共培養的CD3+T細胞會表現較高量之IL-4和IL-10 mRNA,但表現較低量的IFN-γ和IL-12 mRNA(圖5A)。亦使用ELISA分析MDDCs/T細胞共培養培養基中的細胞激素分布。與RT-PCR數據一致,吾等發現,相較於取自未經引發之MDDCs的T對照組上清液,與GRO-γ引發之MDDCs共培養的T細胞之
上清液中IL-4和IL-10的分泌量顯著升高(圖5B)。在GRO-γ引發之MDDCs的存在下,IL-12和IFN-γ於共培養培養基中的濃度顯著降低(圖5B)。
這些數據表示,GRO-γ引發之MDDCs可驅動T細胞朝向更具耐受性的表現型。
(5)GRO-γ處理的BMDCs顯示於活體內具有類MDSC特徵。
為了進一步於活體內探討GRO-γ-引發之DCs的功能,我們於老鼠系統中研究DCs在有或無GRO-γ存在下的分化。收集分化的BMDCs,並使用CD11b和GR-1螢光抗體針對小鼠MDSC標記進行染色。分選出雙陽性CD11b+ Gr-1+細胞做進一步分析。我們發現,BMDCs中雙陽性CD11b+ Gr-1+細胞的百分比不因GRO-γ的存在下而變化(圖6A,上列比較組)。不過,我們分析ARG-1基因和誘導型一氧化氮合成酶(iNOS)基因的表現,彼等已知於小鼠會在MDSCs中受到增量調節,而抑制T細胞增殖和細胞凋亡(Condamine與Gabrilovich,2011)。正如預期,於經GRO-γ處理的BMDC中ARG-1和iNOS的mRNA濃度升高培養上清中(圖6A,下列比較組),且CXCR2受體競爭型拮抗劑SB225002可逆轉於經GRO-γ處理之BMDC中對於這兩個基因的誘導作用。我們的數據顯示,GRO-γ之免疫抑制活性不因CD11b+ Gr-1+雙陽性
細胞數量增加而影響,卻是屬於BMDC特性中功能改變的結果。
接著,我們使用卵清蛋白(OVA)特異-挑戰系統,來分析取自活體受GRO-γ處理而誘導之MDSCs是否於體外和活體內具有耐受性。收集OT-1小鼠脾臟細胞,並且使用FACS Aria細胞分選儀分離出OT-1/CD8+ T細胞。將分選出之OT-1/CD8+ T細胞置於96-孔平盤中,進一步於OVA257-264胜肽存在下以經GRO-γ處理或未經處理之BMDCs進行刺激。以[3H]胸苷併入法測定T細胞增生。結果顯示,於第3天以OVA257-264胜肽脈衝下,經由活體外分化之BMDCs刺激OT-1/CD8+ T細胞增生的作用,在呈現受到GRO-γ-引發之BMDCs減量調節(圖6B)。
接著在活體內試驗GRO-γ產生的MDSCs對T細胞的抑制效果。圖6C列示所設計用於分析GRO-γ產生的MDSCs之功能的實驗流程圖。將骨髓細胞於有或無GRO-γ存在下,以GM-CSF進行分化六天。將小鼠以各種經過OVA257-264胜肽施予脈衝之BMDC製劑進行免疫。於BMDCs注射前一天,將受體小鼠施予照射(1Gy),並以取自相同單倍型(haplotype)之OT-1脾臟細胞重建。然後收集各種不同的BMDCs製劑,並以皮下注射入適應OT-1脾臟細胞之B6小鼠。然後在以經過OVA257-264胜肽施予脈衝之BMDCs免疫後第7天將動物犧牲,從免疫之小鼠收集脾臟細胞,並於活
體物以OVA257-264胜肽刺激12、36和60小時。結果指出,取自以GRO-γ引發之BMDCs注射的老鼠之脾臟細胞,其受OVA257-264胜肽刺激的增生作用,相較於取自接受BMDCs對照組之細胞接受在OVA257-264胜肽刺激後,於36和60小時的增生顯著降低(圖6D)。亦使用ELISA分析取自不同實驗組以OVA257-264胜肽刺激的脾臟細胞上清液中細胞激素的分泌情形。我們觀察到,於經GRO-γ引發之BMDCs組中之IFN-γ和TNF-α濃度,相較於未經處理之BMDCs組有降低的現象(圖6E)。
這些數據顯示,由GRO-γ引發之iDC在活體內表現出類MDSC-表現型和功能。
(6)經GRO-γ處理的BMDC之CD11b+和CD11b-次組呈現免疫抑制效果。
我們分析經GRO-γ處理和未處理的BMDCs中CD11b+Gr1+細胞的數量和特性。研究結果表明,於BMDC分化期間有GRO-γ存在時,不僅Arg-1基因、iNOS基因及MDSC相關之基因的表現受到增量調節,經過GRO-γ處理之BMDCs中Arg-1和iNOS之蛋白表現和酵素活性也增加。另一方面,在CD11b+Gr1+細胞的出現頻率方面,控制組和經GRO-γ處理之BMDCs相似。
由於目前對於MDSC細胞的體內發育調節機轉並不清楚,大部分研究者僅能從帶有腫瘤的老鼠體內獲得
MDSC。為了瞭解利用GRO處理的免疫抑制細胞其抑制免疫的能力與體內生成之MDSC的異同,如先前之報告所述,吾等建立能夠在活體內發展MDSCs之帶有腫瘤的小鼠。參見Youn et al.,J Immunol 181:5791-5802(2008)。將EL4細胞以皮下注射投入8週齡的C57BL/6小鼠,以產生帶有腫瘤的小鼠。接受PBS緩衝液的小鼠當做無腫瘤對照組。於第28天犧牲動物,並評估脾臟中CD11b+ Gr1+細胞之生成比例(圖7A)。在正常小鼠中,CD11b+Gr1+MDSCs約占脾臟細胞的2%左右,但是在帶有腫瘤的小鼠中增加到19%(圖7A)。評估各種細胞(包括總體脾細胞及CD11b+和CD11b-分選次組(純度>95%))在帶有腫瘤和無腫瘤小鼠中的抑制活性。數據顯示,只有CD11b+次組(而非總體脾臟細胞或CD11b-次組)在無腫瘤小鼠中能實質地抑制同種異體混合型淋巴細胞反應(同種異體的MLR)(圖7B)。另一方面,從帶有腫瘤的小鼠收取之總體脾臟細胞不論CD11b+細胞及CD11b-次組都表現出驚人的抑制MLR能力(圖7B)。
此外,本發明亦證明,收取自沒有GRO-γ之BMDCs的總體脾臟細胞、CD11b+和CD11b-細胞,對同種異體之MLR沒有顯著影響(圖8)。然而,所有三組取自經GRO-γ處理的BMDCs之細胞,皆表現出顯著抑制淋巴細胞增生(圖8)。我們的數據顯示,經GRO-γ處理之BMDCs其CD11b+和
CD11b-次組皆具有免疫抑制功效,此組細胞呈現的免疫抑制能力與從帶有腫瘤小鼠體內分離的細胞效果相近。
(7)經GRO-γ處理之BMDCs可改善GvHD mice小鼠的失重和生存率。
藉由以靜脈內灌流將分離自MHC不匹配的野生型B6小鼠(供者,D)之5×106骨髓細胞和5×105脾臟細胞,注入經致死照射之BALB/c小鼠(接受者)來誘發GvHD。將得自供者小鼠之不同細胞組以靜脈內注射到接受者,即單獨5×106個小鼠骨髓細胞、單獨5×105個脾臟細胞、骨髓細胞與脾臟細胞之組合,及與脾臟細胞與1×107經GRO-γ處理之與接受者同源之BMDCs,於移植後第0、2及7天注射到小鼠中。對照組小鼠只接受骨髓細胞。監測接受者之生存率、食物攝取量、失重及臨床的GvHD。
對照組小鼠在移植後顯示體重量有小量變化(圖9A)。而接受脾臟細胞的小鼠顯示其在移植後體重顯著減少(圖9A)。於移植後第2天和第7天,接受經與接受者同源之GRO-γ處理之BMDCs的老鼠,體重恢復(圖9A)。對照組小鼠相較於同時接受骨髓細胞和脾臟細胞的接受者,有顯著較高的存活率(100%)(圖9B)。接受經與接受者同源之GRO-γ處理之BMDCs的小鼠,其存活率高於只接受骨髓和脾臟細胞的小鼠(圖9B)。
數據表明,經GRO-γ處理之BMDCs可改善GvHD小鼠的失重情形和生存率。
其它實施例

Claims (16)
- 一種免疫抑制細胞的製造方法,該方法包括取得一能夠分化成樹突狀細胞之前驅細胞,其中該前驅細胞為血球髓性單核球細胞;及將該前驅細胞培養於含有生長調節致癌基因(GRO)趨化激素GRO-γ或GRO-α之培養基中一段足夠使該前驅細胞分化為樹突狀細胞的時間,其中該樹突狀細胞係呈現一種類似髓源性抑制細胞(MDSC)具免疫抑制的表現型,而藉此獲得該免疫抑制細胞。
- 如請求項1所述的方法,其中該培養基為間葉系幹細胞(MSC)-條件限制培養基或α-MEM完全培養基,含有5%~20%混合的人類血清,人類GM-CSF(20~250ng/mL)及人類IL-4(20~250ng/mL)。
- 如請求項1所述的方法,其中該GRO趨化激素為人類GRO-γ或人類GRO-α。
- 如請求項1所述的方法,其中該前驅細胞是人類外周血CD14+單核細胞,且該培養基含有重組人類GRO-γ。
- 如請求項1所述的方法,其中該前驅細胞是小鼠骨髓細胞,且該培養基為是含有小鼠GM-CSF(2~100ng/mL)之完全RPMI-1640培養基。
- 如請求項1或5所述的方法,其中該GRO趨化激素為小鼠GRO-γ或小鼠GRO-α。
- 如請求項1所述的方法,其中該免疫抑制的表現型包括(a)增加以下的一個或多個基因的表現:IL-10、IL-4、TGF-β、IL-1β、IL-6、COX-2、PD-L1、PD-L2、MMP-9、IDO、ARG-1及iNOS;(b)減少以下的一個或多個基因表現:TNF-α,IFN-γ,IL-12;及(c)能夠抑制T細胞反應。
- 一種藉由如請求項1所述的方法獲得之免疫抑制細胞,其中該細胞係選自CD11b+、HLA-DR-/low及TLRlow細胞。
- 如請求項8所述的免疫抑制細胞,其中於該細胞之CD40、CD83、CD80、CD11c、CD86、DC-SIGN及TLR表現降低。
- 一種組合物,其包含如請求項8所述的免疫抑制細胞。
- 如請求項10所述的組合物,其係用於抑制個體之免疫反應。
- 如請求項10所述的組合物,其係用於製備用於治療個體之病況的醫藥品。
- 如請求項12所述的組合物,其中該病況為自體免疫性疾病。
- 如請求項12所述的組合物,其中該病況為炎症性疾病。
- 如請求項12所述的組合物,其中該病況為移植物抗宿主病(GVHD)。
- 如請求項12所述的組合物,其中該病況為移植排斥反應。
