Abstract

In this invention, we use DNA from hair roots as template, and a pair of oligonucleotide primers specifically designed based on the knowledge of the nucleotide sequences of ryanodine receptor gene. A small region of DNA arrounding the PSS mutation was amplified by using polymerase chain reaction (PCR). The DNA fragments were treated with restriction enzymes, and the genotype was determined by agarose gel electrophoresis. Adventages of using hair roots are collection without stress to pigs, it is easy to collection transportation and storage. This DNA-based test can clearly distinguish the genotypes of homozygous PSS, homozygous normal, and heterozygous pigs, and therefore could be used to create an improved lean, heavily-muscled but no PSS swine breed.
