自體免疫疾病係全球性問題。例如,乾癬係主要影響皮膚及常常影響關節的慢性自體免疫疾病;全世界流行率估計為2-3%,亦即,約1億2千5百萬人。此外,乾癬的發生率正在增加:在1970年至1974年間,在回溯性成年人同齡群中,乾癬的發生率為每10萬人約50.8例,且在1995年至1999年間,發生率為約每10萬人約100.5例。不管其流行率,自體侵襲性免疫細胞之鑑別及用於鑑別及/或治療乾癬的推定目標自身抗原有待發現。

[實例] [實例1:免疫細胞分離]
自標靶器官病灶分離未處理、未操縱的免疫細胞。替代將皮膚組織切碎並消化以分離免疫細胞,使用雷射單細胞擷取方法,雷射擷取顯微解剖(Laser Capture Microdissection;LCM)。在不受理論約束的情況下,咸信此類技術允許在不降解細胞外基質的情況下接近固體組織樣品中用於雷射擷取的未改變的免疫細胞。與皮膚生檢相比較而言,用於執行LCM的組織為約5-15微米厚,該皮膚生檢通常為6-8 mm厚。

[實例4:鑑別對抗原的周邊血液免疫細胞反應]
自受試者收集周邊血液樣品,同時收集組織樣品(例如,皮膚生檢;滑液或生檢;腦脊髓液或腦生檢;胰腺生檢),且自組織樣品的發現結果與患者的自身血液樣品相關聯。更特地而言,表徵血液樣品以測定在存在或不存在自標靶器官(例如,皮膚生檢)鑑別的T細胞及/或其相關抗原。若確認標靶器官中存在此類細胞及/或抗原,此類組分可在血液中得以鑑別,即使其以低頻率存在時亦如此。

[實例7:合成推定的(多種)抗原

[實例8:臨床前動物測試]

[實例9:人類試驗]
測試當前的免疫抑制藥物且將其用於所有具有潛在嚴重副作用的乾癬或其他自體免疫病狀治療。本文描述的基於抗原的方法係非免疫抑制性的,其具有最小至輕度的潛在副作用。

Claims (62)
- 一種自患有乾癬的受試者製備免疫細胞樣品的方法,前述方法包含以下步驟: 獲得乾癬組織樣品;及 使用雷射擷取顯微解剖自前述乾癬組織樣品原位分離單一免疫細胞。
- 一種自患有乾癬的受試者製備免疫細胞樣品的方法,前述方法包含以下步驟: 獲得乾癬組織樣品;及 使用實質上不改變免疫細胞之細胞生物化學概況的技術自前述乾癬組織樣品原位分離活的單一免疫細胞。

- 一種自患有自體免疫病症的受試者製備免疫細胞樣品的方法,前述方法包含以下步驟: 獲得組織樣品;及 使用實質上不改變免疫細胞之細胞生物化學概況的技術自前述組織樣品原位分離活的單一免疫細胞。
