Description

Composition for maintenance, prevention of decrease, suppression of decrease or improvement of living activity level

The present invention relates to a composition for maintaining, preventing, inhibiting, or improving the amount of living activity. In addition, the present invention relates to a method for maintaining, preventing decrease, suppressing decrease, or improving the amount of living activity, and the use of one or more sesamins for maintaining, preventing decrease, suppressing decrease, or improving the amount of living activity.

In recent years, how to improve living habits in countries around the world, including Japan, has become an important issue. In addition, for countries with a high proportion of elderly people in the population structure, such as Japan, extending healthy life expectancy has also become an important issue. Under such circumstances, Japan promotes continuous exercise to prevent diseases (civilization diseases) caused by lifestyle habits, and to increase muscle strength to prolong healthy life. Furthermore, in recent years, those who have low psychological barriers to start exercising and can easily take the first step have also recommended increasing the amount of physical activity in their daily lives. For example, some people recommend walking or housework during commuting or school as activities that most people can easily perform in their daily lives.

Here, according to the "Guidelines for Health Promotion Exercise 2006" issued by the Ministry of Health, Labour and Welfare of Japan, physical activity means "all behaviors that consume more energy than when staying still" and are defined as sports and life activities. By. Life activity is defined as "something other than exercise during physical activity." In addition, the intensity of physical activity is multiplied by the time spent on the activity, which is expressed as the amount of physical activity.

In addition, according to the Health and Labor White Paper (2014 edition) issued by the Ministry of Health, Labour and Welfare of Japan, healthy life span is defined as "a period during which one can live without restrictions on daily life due to health problems"; those who can live without care The period of life is synonymous with the period of unrestricted life activities. Therefore, with age, the possibility of obstacles in life activities will increase. For example, Non-Patent Document 1 discloses that with age, the amount of life activity decreases. Non-Patent Document 3 discloses that the higher the amount of physical activity, the longer the healthy life span. Therefore, in an aging society, it is required to maintain physical activity through exercise and maintenance of life activity, and to extend the healthy life span.

[先前技術文獻] [非專利文獻]In addition, the amount of physical activity of a person can be measured with the number of steps as a basic indicator. For example, in animal experiments, the mainstream is to measure the number of movement by infrared rays, or measure the movement distance by telemetry. However, the assessment of the amount of life activity that does not include exercise also requires low-intensity activity assessment. In recent years, some people have developed a measurement method that uses a built-in activity meter in a three-axis acceleration sensor as a measurement method of human life activity. By using this measurement method, low-intensity activities can also be accurately measured, so it can be used as an indicator to more accurately measure the amount of life activity. Using this measurement method, it has been confirmed that the amount of human life activity decreases with age (Non-Patent Document 1). [Prior Technical Literature] [Non-Patent Literature]

The problem to be solved by the invention]

It was confirmed in Non-Patent Document 2 that sesamin can inhibit the decrease in endurance when an exercise stress test is performed on rats fed a high-fat diet. However, no one has reported that sesamin can prevent the decrease in the amount of life activity during physical activity, rather than the decrease in the amount of exercise, and help maintain the amount of life activity. That is, it is not known that sesamins such as sesamin have the function of maintaining the amount of living activity. In particular, it has not been reported that sesamin, such as sesamin, contributes to the maintenance of the decrease in the amount of living activity associated with aging.

[解決課題之手段]The present invention aims to provide a composition for maintaining, preventing decrease, suppressing decrease, or improving the amount of living activity. In addition, the present invention aims at a method for maintaining, preventing, reducing, suppressing, or improving the amount of living activity, and the use of one or more sesamins for maintaining, preventing, preventing, suppressing, or improving the amount of living activity. [Means to solve the problem]

In order to solve the above-mentioned problems, the inventors of the present case have devoted themselves to research. By using the built-in activity meter of the three-axis acceleration sensor shown in Non-Patent Document 1, the activity level of rats was measured without exercise burden. Primitives have the function of maintaining the amount of physical activity other than exercise, that is, the amount of life activity. In addition, the inventors of the present application found that sesamin is useful for preventing or maintaining the decrease in the amount of living activity due to aging.

The increase in inactive time. The sesamin used in the present invention is an ingredient that has been taken in the form of food since ancient times, and it is also advantageous in terms of high safety and sustainable ingestion.

The form of implementing the invention]

The composition system for maintaining, preventing decrease, suppressing decrease, or improving the amount of living activity of the present invention contains one or more kinds of sesamin. Sesamin is one of the main lignan compounds in sesame, and contains about 0.5 to 1.0% by weight in sesame. Long-term intake of synthetic compounds is not good based on unpredictable side effects. On the other hand, sesamin, which can ensure safety, is suitable for long-term intake.

In the present invention, physical activity, exercise, and life activity have the same meaning as the definitions of physical activity, exercise, and life activity in the "Health Promotion Guide 2006" issued by the Ministry of Health, Labour and Welfare of Japan. In other words, physical activity refers to all behaviors that consume more energy than when remaining still, and life activity refers to all activities other than exercise. In addition, exercise refers to those who are planned or deliberately performed for the purpose of maintaining or improving physical strength, for example. The amount of living activity can be measured with a built-in activity meter (Actimarker manufactured by Panasonic) for humans, for example. For animals, it can be measured using a three-axis acceleration sensor built-in activity meter (nanotag manufactured by KISSEI COMTEC Co., Ltd.) without applying exercise such as treadmills.

The life activities in the present invention are not particularly limited. For example, humans may include housework, work, and/or childcare activities. As far as animals are concerned, the amount of activity measured under the condition of no exercise is equivalent to the so-called living activity amount of human beings.

Furthermore, in the present invention, the amount of life activity can be measured by the amount of life activity and/or the time of life activity. That is, the maintenance, prevention of reduction, inhibition of reduction or improvement of the amount of living activity refers to the maintenance, prevention of reduction, inhibition of reduction or improvement of the amount of living activity and/or life activity time. In addition, since the decrease (decrease) of life activity time will lead to the increase of inactive time, the maintenance, prevention of decrease, suppression of decrease or improvement of life activity amount in the present invention also includes prevention, suppression or improvement of increase of inactivity time. In addition, the inactive time mentioned in this manual refers to the time during which no physical activity is performed, that is, the time during which sleep or static behavior remains static.

The maintenance, prevention of decrease, suppression of decrease or improvement of the above-mentioned living activity level is preferably to prevent or restrain the decrease of living activity level, improve the reduced living activity level, prevent or inhibit the increase of inactive time, or improve the increased inactive time.

Furthermore, the maintenance, prevention, reduction, suppression, or improvement of the above-mentioned living activity level is preferably to prevent or inhibit the reduction in living activity level accompanying aging, to improve the reduction in living activity level accompanying aging, and to prevent or inhibit aging. Increase in inactive time due to growth, improve the inactive time that increases with age. In addition, the term “age increase” in the present invention refers to the changes that occur with the increase of age after the age of about 40 years old and above.

1:0.5~1:2。(Sesamin) In the present invention, sesamins refer to the general term for compounds including sesamin and its analogs. Sesamin is one of the main lignan compounds contained in sesame. As the sesamin analogs, in addition to episesamin, for example, dioxabicyclo[3.3.0]octane derivatives described in JP 4-9331 A can be cited. Regarding one or more types of sesamin, one type of compound may be used alone, or two or more types may be used. Specific examples of sesamin include sesamin, episesamin, sesame lignan, episesame lignan, sesamol, sesalin, etc., and these stereoisomers or racemates can be used alone or in combination. . Moreover, sesamin metabolites (such as those described in JP 2001-139579 A), as long as they can show the effects of the present invention, are also sesamin analogs included in the sesamin class of the present invention, and can Used in the present invention. In the present invention, one or more types of sesamin may preferably use sesamin and/or episesamin, more preferably use sesamin and episesamin, or episesamin, and even more preferably use episesamin . When using sesamin and episesamin, these ratios are not particularly limited. For example, sesamin: episesamin (weight ratio) is preferably 1:0.1 to 1:9, more preferably 1:0.3 to 1:3, More preferably, it is 1:0.5 to 1:2.

Chem. Pharm. Bull. (Tokyo), 56 (11) 1611-2 (2008))來合成。The sesamin used in the present invention is not subject to any restriction on its form or production method. For example, in the case of sesamin, sesamin extracted from sesame oil (referred to as a sesamin extract or purified product) by a known method (for example, the method described in Japanese Patent Application Laid-Open No. 4-9331) can be used. Moreover, you may use a commercially available sesame oil (liquid form) as it is. However, when sesame oil is used, it is possible to be evaluated as the characteristic flavor of sesame oil as poor sensory; therefore, it is advisable to use a tasteless and odorless sesamin extract (or purified sesamin) extracted from sesame oil. In addition, when sesame oil is used, since the content of sesamin is low, if a suitable amount of sesamin is to be mixed, the volume per unit of the formulated composition will become too large, which may cause ingestion problems. In particular, when the formulation is formed into oral administration, the formulation (tablet, capsule, etc.) is too large and ingestion problems occur. Therefore, it is also preferable to use a sesamin extract (or a purified sesamin) derived from sesame oil from the viewpoint that the intake can be reduced. In the present invention, one or more kinds of sesamins that have been purified or separated can also be used. Sesamins can also be obtained by synthesis. 生活活動量的測定係使用加速度計測器nanotag(KISSEI COMTEC股份有限公司製)。 <Example 1: The effect of the sesamin/episesamin mixture (1:1) on inhibiting the decrease in the amount of living activity (in vivo test)> In this experiment, it was carried out under the condition of no exercise load, so all the activities measured by the study are equivalent to the so-called living activities of human beings. The general feed group and the feed group containing sesamin and episesamin are divided into groups in a way that the amount of living activity is equal. The measurement of the amount of living activity uses an accelerometer nanotag (manufactured by KISSEI COMTEC Co., Ltd.). For a 14-month-old Slc: SD male rat, under isoflurane anesthesia, an accelerometer was implanted under the skin of the neck, and after confirming recovery, the amount of activity during the active period (light-off time) was measured for 3 days. For the measurement of the amount of living activity, the number of vibrations above a certain intensity is continuously measured, and the total number of vibrations is used as the amount of living activity. This is divided into general feed (AIN-93M, manufactured by CLEA Japan) or feed containing 0.1% by weight of sesamin and episesamin mixture (sesamin: episesamin (weight ratio) = 1:1) The group of (test food) (each 8 animals/group) was allowed to take each feed (test food) for 8 months. The group that consumes general feed is called the general feed group for the elderly (general feed group), and the group that consumes the feed containing 0.1% by weight of sesamin and episesamin is called the feed group containing sesamin and episesamin for the elderly (contains sesamin and episesamin). Table sesamin feed group). In addition, the feed containing sesamin and episesamin, in which the total content of sesamin and episesamin is 0.1% by weight, is a mixture of sesamin and episesamin (sesamin: episesamin (weight ratio) )=1: 1). The life activity level during the active period was measured at the time points of 17 months and 22 months old for 1 week. In order to grasp the animal's health status, the food intake and body weight were measured regularly; rats that were judged to be difficult to continue the test were given appropriate euthanasia and excluded from the analysis. The final analysis targets are 4 old-age general feed groups and 6 old-age feed groups containing sesamin and episesamin.

Fig. 1 shows the change in the relative amount of living activity when the amount of living activity before the start of the test food is set to 1. In the graph of Figure 1, ● is the general feed group for the elderly, and ▲ is the feed group containing sesamin and episesamin for the elderly (*: p<0.05, compared to the general feed group for the elderly, repeated measures two-way ANOVA) . The lower the displayed value, the lower the activity. As shown in Figure 1, for the general feed group, the amount of life activity decreases with age. On the other hand, the feed group containing sesamin and episesamin can inhibit the decrease in the amount of living activity associated with aging. As a result of repeated analysis, an interaction can be seen between the test food and time ( p=0.014). At 22 months of age, compared with the general feed group, the living activity of the feed group containing sesamin and episesamin was significantly higher (Bonferoni test, p=0.023).

<Example 2: The effect of inhibiting the decrease in living activity level by administering sesamin or episesamin alone (in vivo test)> Example 2 is a test for exploring which of sesamin or episesamin is more helpful in inhibiting the decrease in life activity. The basic operation is implemented in accordance with Example 1. The 14-month-old Slc: SD male rats are divided into general feed (CE-2, manufactured by CLEA Japan Co., Ltd.) or feed containing 0.1% sesamin or episesamin in a way that equalizes their activity levels. Groups (3 animals/group) were allowed to take each feed (test food) for 1 month. The life activity level during the active period was measured at the 16-month-old time point after 1 month of ingestion of the test food. The final analysis targets are 2 general feed groups, 3 sesamin-containing feed groups, and 3 episesamin-containing feed groups. Table 1 shows the amount of living activity of each group after taking the test food for 1 month when the amount of living activity before the start of the test food is set to 1 hour. The lower the value, the lower the amount of life activity. As shown in Table 1, for the general feed group, the amount of living activity decreased. On the other hand, the feed group containing sesamin and the feed group containing episesamin, compared with the general feed group, can suppress the decrease in the amount of life activity caused by aging. Regarding the feed group containing episesamin, no reduction in the amount of life activity due to age increase is seen.
